The Whitening Effect by Water Extracts of Bombyx Mori Linne (백강잠(Bombyx Mori Linne) 물추출물의 미백 효능)

  • Numerous novel ingredients have been introduced for the hight functionality of whitening cosmetics. Tough the preliminary research, we have found water extracts Bombyx mori have high whitening efficacy. The results of the research for the whitening effect of Bombyx Mori L. are as follow 1. Bombyx Mori L. inhibited concentration dependently the generation of melanin increased by the stimulation of $\alpha$-MSH and protoporphyrin IX, and $IC_{50}$ value was 8.3, 9.2 ${\mu}M$ respectively. 2. Melanin increased by the stimulation of $\alpha$-MSH and protoporphyrin IX was five to seven times superior in the inhibiting effect, compared with kojic acid used as positive control group. 3. Bombyx Mori L. did not have a decolorizing effect on melanin already generated. 3. Bombyx Mori L. was observed to have toxicity of over 100 ${\mu}M$ for the mouse melanoma B16 cells. Therefore, These results suggest that water extracts of Bombyx mori have inhibitory activity against mushroom tyrosinase and inhibitory activity of melanin synthesis in B16 melanoma cells in vitro.

Morphological and Biochemical Characterization of the Chorion in Interspecific Hybrid Between Bombyx mori and Bombyx mandarina (집누에(Bombyx mori)와 멧누에(Bombyx mandayina)의 종간교잡에 있어서 란각구조 및 Chorion 단백질)

  • choChorion(egg-shell) morphology of the F1 hybrid between Bombyx mori and Bombyx mandarina has been observed by scanning electrom microscope and chorion protein was analyzed by electrophoresis. The chorion surface structure of F1 hybrids in the lateral(flat) region was similar to that of maternal line. The F1 hybrids chorion was found to have basically a three layer structure. The middle and inner layer were very much like those of the Bombyx mandarina and Bombyx mori. There were many conic pillar structures in the outer layer of the F1 hybrid, which was similar to Bombyx mandarina. This conic pillar structure had a thin cover layer was more clear in the dorsal and ventral side of the F1 hybrid chorion. The conic pillar structure of Bombyx mandarina was found to be dominant in F1 hybrid chorion irrespective of their maternal line. Major components of chorion protein were analyzed by two-dimensional electrophoresis and found to have isoelectric points in the range of pH 4.0-6.5 and molecular weight 10 to 50 kd. F1 hybrid chorion protein components related directly to those of the maternal line. The conic pillar structure was dominat characteristic and it was present in all F1 hybrid.

Molecular Phylogenetics of Silkworm (Bombyx mori) Based on Mariner-Like Elements (MLEs) (Mariner-Like Elements (MLEs)를 이용한 누에의 분자적 계통 분석)

  • In order to understand molecular phylogenetics of silkworm (Bombyx mori), we analyzed the sequences of BmoMAR isolated from Bomhyx mori that is partial coding gene of transposase of mariner-like element(MLE). By pairwise comparing nucleotide sequences of BmoMAR with ten previously reported insect MLEs accessed in GeneBank, the average genetic distance was estimated to be 0.4840. The phylogenetics tree constructed from nine insect species except for human MLE(Hsmarl) by UPGMA method indicated that MLEs are divided into three clusters, and Drosophila mariutiana was independently subgrouped. Bombyx mori(BmoMAR) was subgrouped with microcaddishfly (Orthotrichia cristata), webworm(Atteva punctella), almond moth(Ephestia cautella), Hyalopora cecropia which we lepidoptera. Phylogenetics tree according to UPGMA principle, on the basis of informative nucleotide sequences of nine insect MLEs, indicated that Bombyx mori was more closely related to microcaddishfly(Orthotrichia cristata) and webworm (Atteva punctella) of lepidoptera. We suggest that insect MLEs are a useful key for studying molecular phylogenetics among intra species of insects.

  • The baculovirus egt gene encodes an ecdysteroid UDP-glucosyltransferase(EGT) which catalyzes the transfer of glucose from UDP-glucose to the insect moltion hormone ecdysteroid resulting in a functionally inactive ecdysteroid. In baculovirus-infected insect larvae, EGT has been shown block molting and pupation. In this study, we compared the development of 4th and 5th instar silkworm, Bombyx mori, larvae injected with either wild-type bombyx mori nucleopolyhedrovirus (BmNPV) or a mutant BmNPV(BmEGTZ) in which the egt gene was disrupted by the insertion of a lacZ gene cassette. Larvae injected with BmEGTZ died roughly 12 h more rapidly compared to indentical larvae infected with BmNPV. In addition, BmEGTZ- infected larvae prematurely stopped feeding and gain less weight compared to BmNPV-infected larvae. In order to investigate why BmEGTZ-infected larvae died more rapidly than BmNPV-infected larvae, the array of hemolymph proteins in BmEGTZ-or BmNPV-infected larvae were analyzed by SDS-PAGE. The hemolymph of BmEGTZ-infected larvae showed virus-specific proteins, including polyhedrin, about 12 h earlier than the hemolymph of BmNPV-infected larvae

Bombyx mori β-tubulin Promoter for High-level Expression of Heterologous Genes

  • We previously isolated 9 clones that show stronger signal compared to Bombyx mori cytoplasmic actin gene (BmA3) by using a dot blot hybridization. In this study, we focused on one clone among these clones which has high amino acid similarity with ${\beta}$-tubulin gene of B. mori. This clone was ubiquitously expressed in all tissues and developmental stage of B. mori. As result of promoter assay using dual luciferase assay system, we found the highest transcription activity region (-750/-1) in the 5'-flanking region of ${\beta}$-tubulin gene, which has about 47 fold more intensive promoter activity than BmA3 promoter. Moreover, the ${\beta}$-tubulin promoter was normally regulated in Bm5, Sf9, and S2 cells. Therefore, we suggest that ${\beta}$-tubulin promoter may be used more powerful and effectively for transgene expression in various insects containing B. mori as a universal promoter.

  • A mutagenicity testing system using a specific locus mutation of Bombyx mori was introduced from National Institute of Genetics in Japan. In the system, mutagenicity could be detected by the egg color manifested by the pe and/or re genes, which is a kind of recessive visible mutation of the insect. The efficiency to detect mutagenicity of the system was examined and improved. To promote the sensitivity of the system to mutagens, eight varieties of Bombyx mori were tested for their sensitivity to two mutagens, EMS and MMC. Two varieties of the silkworm, N12 and C5, were finally selected as the most sensitive ones. The most sensitive development stage of the silkworm to mutagens was mid and late pupal stages for female and male, respectively. The system will be applied to test unknown mutagens after some more detail examination about its sensitivity.

  • The present investigation reports a phenomenon hitherto unknown in tropical sericulture, wherein dia-pause nature of bivoltine eggs is overcome through a cross with a non-diapausing race of silkworm, Bombyx mori, L. Eggs of bivoltine silkworm Bombyx mori, L. generally do not hatch under tropical conditions. To prevent diapause, they are subjected to acid treatment or low temperature hibernation scheduled. A race developed at KSSRDI is found to prevent the diapause nature of bivoltine eggs when crossed as male parent, without any acid treatment or hibernation schedule. This phenomenon is reported for the first time, being unique, opens up interesting area of research in silkworm genetics of commercial implications in the industry.

Cloning and Characterization of hydroxypyruvate isomerase (EC 5.3.1.22) gene in silkworm Bombyx mori

  • The sequence of hydroxypyruvate isomerase gene was obtained in NCBI. In this study, the hydroxypyruvate isomerase gene of Bombyx.mori was identified and annotated with bioinformatics tools. The result was confirmed by RT-PCR, prokaryotic expression, mass spectrographic analysis and sub-cellular localization. The hydroxypyruvate isomerase cDNA comtains a 783bp ORF, and has 4 exons. The deduced protein has 260 amino acid residues with the predicted molecular weight of 29169.30 Da, isoelectric point of 6.10, and contains conserved PRK09997 and Hfi domains. The hydroxypyruvate isomerases of Nasonia vitripennis and Bombyx mori have a high homology. Through RTPCR analysis, we found that this transcript was present in testis, ovary, blood-lymph, fat body, midgut, silk gland and tuba Malpighii. This protein was located in cytoplasm through immunohistochemistry. We submitted the cloned gene under the accession number EU344910. The enzyme has been classified under accession number EC 5.3.1.22.

Biochemical Properties of the Lectin Isolated from Bombyx mori (누에(Bombyx mori)로부터 분리한 렉틴의 생화학적 특성)

  • A new lectin was purified from Bombyx mori (BML) by physiological saline extraction, ammonium sulfate precipitants, anion exchange column chromatography on DEAE Sephadex A-50 and gel filtration column chromatography on Sephadex G-200. BML agglutinated trypsinized and glutaraldehyde-fixed erythrocytes, and was observed the most high activity with rabbit, chicken erythrocytes and rat splenic lymphocytes. Agglutinability was markedly affected at highly acidic pH, but was relatively stable with high temperature. The effect of metal ions was observed and BML was affected by bivalaent cations, especially depending on $Ca^{2+}$, $Fe^{2+}$, $Mn^{2+}$, whereas, inhibited by $Mg^{2+}$. Agglutination was strongly inhibited by heparin and glucuronic acid. BML was proved to be a glycoprotein which contains 17.16% of sugars. By mass spectrometry analysis, we found 2 bands that were considered as lectin subunits.
