Solid-state NMR Studies of Miscibility and Morphology in Blends of Bisphenol-A type Polycarbonate and Poly (ester-ether) Elastomer

  • Miscibility and morphology in blends of bisphenol-A type polycarbonate and poly (ester-ether) elastomer with different compositions are studied by solid-state NMR spectroscopy. $^{13}$ C Solid-state NMR of CP/MAS/TOSS/DD, CP/MAS/DD, inversion recovery CP/MAS/DD, and 2D rotor driven spin diffusion techniques are used to identify the miscibility, morphology, and transesterification in blends. The blends of PC /BT elastomer with 15% to 42% of soft segment seem to be single phase miscible mixing and those of PC/PBT and PC/PBT elastomer with 62% of soft segment are cocontinuous two phase immiscible mixing. No significant transesterification reactions are observed in blends with different compositions.

  • Melt blending of Bisphenol A polycarbonate (PC) and poly(trimethylene terephthalate) (PTT) was carried out over the entire composition range. The mixing time was varied up to 90 min. The resulting samples were analyzed by FT-IR, DSC, XRD, DMTA, $^{1}H NMR$, and SEM. The process of transesterification between the two polymers and their resulting compatibilization were observed. The behaviors of the PTT-rich and PC-rich blends were different and an equilibrium was found to exist. Peculiar behavior, which was different from that of the PTT-rich and PC-rich blends, was exhibited by the 50/50 (PTT/PC) blend.

  • We investigated the education process of an environmental experiment in middle and high school. The major objective was that students understand about biological toxicity of endocrine disruptor. We used bisphenol A in environmental experiment to educate them about the effects of endocrine disruptor. The test animal was zebrafish (Danio rerio, wild type) which is very easy to use in the school experiment class. We educated the high school students (n=25) in Suwon through this teaching program. Finally we analyzed the effect of research education between two groups, education students (test) and non-education students (control). This experiment showed there were some positive effects on environmental reorganization of students. They understood the affect of endocrine disruptors in fish and man. Specifically, understanding the importance of preservation of ecosystem increased in the test-education group.

Review Newly Adopted OECD Acute Oral Toxicity Test Guideline 420 (OECD test guideline 420 고정용량 급성경구독성시험법에 대한 고찰)

  • The OECD acute toxicity guideline has been revised recently to protect animal welfare. The GLP authority of the Ministry of Environment, the National Institute for Environmental Research, recommended GLP laboratories in Korea to ufo the revised acute toxicity guideline. This study was carried out to optimize newly adopted OECD test guideline 420 (TG 420). Bisphenol A was selected for test chemical. Following TG420, Bisphenol A was classified as class 5/unclassified group. The revised TG 420 was very effective test in minimizing animal number and classifying chemicals. The method, however had short-coming in evaluation of test results statistically because the test had no control group, and the test should be stopped when animals were dead at the lowest dose or alive at the highest dose. TG 420 required at Least 20 animals to complete the test, but it could result in producing unused animals that need to sacrifice.

  • Poly(4-morphonylphosphonate of bisphenol A) (PMPB) was synthesized by the interfacial polymerization of phosphonic dichloride, 4-morphonyl (PDCM) and bisphenol A in an aqueous-organic biphasic system. Synthesized polymer was found to be PMPB by using EA, FT-IR and $^{1}$H-NMR. Prepared PMPB had different values of molecular weight depending on the solubility in the organic phase: the higher the solubility, the higher the molecular weight. PMPB is amorphous and its thermal decomposition temperature is about 35$0^{\circ}C$. PMPB is congenial to application to polypropylene by blending because of no reaction with polypropylene.

  • Bisphenol A (BPA) mimics estrogen and its activity is one third to one quarter that of estradiol. BPA, an ubiquitous environmental contaminent has been shown to cause development reproductive toxicity and carcinogenic effect. BPA may do physiological action through ER$\alpha$ and ER$\beta$ which are expressed in central nerve system. We previously found that expose of BPA to immature mice resulted in behavial alternation, suggesting that overexposure of BPA could be neurotoxic. (omitted)

  • This study investigated the relative degradation of commonly known endocrine-disrupting compounds such as bisphenol A (BPA) and 17${\beta}$-estradiol (E2) using ultrasound (US) and pulsed ultraviolet (PUV) in water. The removal efficiency of BPA and E2 was determined as a function of generating power and $H_{2}O_{2}$ production. The ultrasound and PUV irradiation of the aqueous solution was performed in 3 L and 90 L stainless reactor at a constant temperature of $20^{\circ}C$ with an applied power of 200 W and 2000 W, respectively. The removal of BPA and E2 by US and PUV varied with catalysts. The experiments were conducted under the following conditions: total operating time, 30 min; initial concentration, 1 uM. In the case of E2 (10 min), % removal was 92.5/95.8/87.6/82.4, while % removal of BPA (10 min) was 62.3/82.3/91.1/67.0/64.3 in various conditions (PUV, $PUV+H_2O_2$, PUV+wire mesh, $PUV+TiO_2$ coated wire mesh), respectively.

Interactive Effect of Bisphenol A (BPA) Exposure with -22G/C Polymorphism in LOX Gene on the Risk of Osteosarcoma

  • Background: Osteosarcomas have many established risk factors, both genetic and environmental, but by themselves these explain only part of the total cancer incidence. Bisphenol A (BPA) is an environmental estrogen associated with risk of several kinds of tumour. The lysyl oxidase gene (LOX) may also contribute to risk of tumours including osteosarcomas. Here, we investigated possible interactions of BPA and a LOX polymorphism on the risk of osteosarcoma. Method: The present hospital-based case-control study included 106 cancer patients and 112 controls from a Chinese population. Internal burden of BPA exposure was assessed using high-performance liquid chromatography-mass spectrometry (HPLC-MS) method. Genotypes were determined using PCR-RFLP methods. Results: Compared with those in low BPA exposure group, subjects with BPA more than or equal to median value had significant increased risk of osteosarcoma among subjects who carried GC or CC genotypes. A significant interaction with BPA level and the -22G/C polymorphism was observed for osteosarcoma overall, osteosarcoma affecting knee and osteosarcoma affecting hip, as $P_{forinteraction}$ = 0.036 for osteosarcoma overall; $P_{forinteraction}$ = 0.024 for osteosarcoma affecting knee; and $P_{forinteraction}$ = 0.017 for osteosarcoma affecting hip. Conclusions: The results suggest that BPA exposure interacts with the -22G/C polymorphism of the LOX gene to increase the risk of osteosarcoma.

Decomposition Characteristics of Bisphenol A by a Catalytic Ozonation Process (오존촉매산화공정에 의한 비스페놀 A의 분해특성)

  • Bisphenol A (BPA) in aqueous solution was measured using HPLC technique, which was established by acetonitrile analysis and KDP solution analysis methods. In these experiments the decomposition characteristics of BPA were compared using the ozone alone, ozone/pH 10, and ozone/hydrogen peroxide processes. About 70% of 10 mg/L of BPA was removed during 60 min by the ozone alone process, while 10 mg/L of BPA was completely removed by the ozone/pH 10 and ozone/hydrogen peroxide processes in 40 min and 60 min, respectively. The final decomposition efficiency drawn from results of TOC and HPLC analyses showed that the ozone/hydrogen peroxide process was the best among them, whereas the concentrations of TOC and reaction intermediates when using the ozone/pH 10 process were higher than those of the ozone alone process after 60 min of reaction. The ozone/hydrogen peroxide process was the most efficient among them when oxidizing organic carbons in water to $CO_2$ and $H_2O$.

Bisphenol A Bis(2,3-dihydroxypropyl) ether (BADGE.2H2O) Induces Orphan Nuclear Receptor Nur77 Gene Expression and Increases Steroidogenesis in Mouse Testicular Leydig Cells

  • Bisphenol A bis (2,3-dihydroxypropyl) ether ($BADGE.2H_2O$) is a component of commercial liquid epoxy resins commonly used in the food-packing industry and in dental sealants. There is evidence that it has significant estrogenic activity. Nur77 plays a crucial role in the regulation of certain genes involved in LH-mediated steroidogenesis in testicular Leydig cells. It was previously demonstrated that Bisphenol A (BPA) stimulates Nur77 gene induction and steroidogenesis. In this study, we investigated the effects of $BADGE.2H_2O$ on Nur77 gene expression and steroidogenesis. Northern blot analysis showed that it increased the expression of Nur77 mRNA and protein, and transient transfection assays demonstrated that it increased the promoter activity and transactivation of Nur77. It also increased the expression of certain steroidogenic genes, such as StAR and $3{\beta}$-HSD. Finally, over-expression of a dominant negative Nur77 cDNA via adenoviral infection reduced $BADGE.2H_2O$-mediated progesterone biosynthesis. These results indicate that $BADGE.2H_2O$ disrupts testicular steroidogenesis by increasing Nur77 gene expression.
