Effect of PLGA/Silk Fibroin Hybrid Film on Attachment and Proliferation of Schwann Cells (실크피브로인을 함유한 PLGA 하이브리드 필름이 슈반세포의 부착과 증식에 미치는 영향)

  • Poly(lactic-co-glycolic acid) (PLGA) is a biodegradable synthetic polymer with acceptable mechanical strength and well-controlled degradation rate. Also, it can be easily fabricated into many shapes. Silk fibroin contains powerful bioactive molecules. We fabricated natural/synthetic hybrid films using 0, 10, 20, 40 and 80 wt% of silk fibroin. Schwann cells (SCs) were seeded on PLGA/silk fibroin hybrid films and confirmed the effects of adhesion and proliferation on SCs according to the content of silk fibroin. In this study, we confirmed PLGA/silk fibroin hybrid film containing 40% and 80% of silk fibroin interrupted adhesion and proliferation of SCs. Films containing 10% and 20% of silk, however, provided suitable environment for growth and proliferation of SCs. These results suggest that silk fibroin provides suitables surface to neural cells and its proper content provides proper culture conditions to improve cell adhesion and proliferation.

  • Iron oxides ($Fe_3O_4$) are metabolically secreted after endocytosed by cells, indicating no cytotoxicity. Therefore, they are widely used as a contrast agent before photographing of magnetic resonance imaging. In this study, iron oxide nanoparticles are synthesized by the co-precipitation method and subsequently immobilized with a homing peptide (AP), which specifically interacts with interleukin-4 receptor located on the membrane of endothelial and bladder cancer cells. The size of AP-immobilized iron oxide particle is about 39 nm. Intracellular uptake of the AP-immobilized iron oxide nanoparticles was investigated using bladder cancer cells and fibroblasts as the control. As the result, the nanoparticles are specificially uptaken by bladder cancer cells. However, the nanoparticles are not specificially uptaken by fibroblast. It could be said that the AP-immobilized iron oxide nanoparticles have a potential to be used as a contrast agent for early diagnosis of cancer.

A Study on Awareness and Degree of Practice about Infection Control by Dental Hygienics Student's in Some Ares (일부지역 치위생과 학생의 감염관리 인식 및 실천도에 관한 조사)

  • This research surveyed 324 college students majored dental hygiene in Gwangju and Chonnam province to analyze their awareness and degree of practice about infection control. Through the study for recognition about infectious disease related with the grade, the sophomore students are the group who is the most aware. In addition, the study shows students who experienced teeth cleaning have high degree of awareness (p<0.05). The research of perception about infectious disease based on experiences for vaccinations and education about infection prevention says hepatitis B has the highest rate when it comes to level of occurrence risk and tuberculosis and acquired immune deficiency syndrome are the lowest (p<0.05). According to the research, the group who had vaccination, knowledge about infection prevention and experience for scaling shows high practice rate for hand washing according to whether students receive protective inoculation or not. Depending on what the result were, since student learned about infection control has high degree of recognition and practice about infection management if we could emphasize the importance to students through regulative education about infection control and then increase the degree of practice, it would make big contributions to the effective infection control.

Anti-inflammatory and antifatigue effect of Korean Red Ginseng in patients with nonalcoholic fatty liver disease

  • Background: Korean Red Ginseng (KRG) is a well-known natural product with anticarcinogenic and antioxidant effects. We evaluated the antifatigue effect of KRG in patients with nonalcoholic fatty liver disease (NAFLD). Methods: Eighty patients with NAFLD were prospectively randomized to receive 3 wk of KRG or placebo in addition to counseling on healthy eating and regular exercise. Liver function test, proinflammatory cytokines, adiponectin, antioxidant activity, and fatigue score were measured and compared according to the body mass index between the KRG and placebo groups. Results: The liver function tests were significantly improved after 3 wk of treatment in both groups. The mean levels (at baseline and after treatment) of tumor necrosis factor-${\alpha}$ were $108.0pg/mL{\pm}54.8pg/mL$ and $92.7pg/mL{\pm}39.0pg/mL$ (p = 0.018) in the KRG group and $123.1pg/mL{\pm}42.1pg/mL$ and $127.5pg/mL{\pm}62.2pg/mL$ (p = 0.694) in the placebo group, respectively. There was a significant difference in change of adiponectin levels between the KRG ($7,751.2pg/mL{\pm}3,108.1pg/mL$ and $8,197.3pg/mL{\pm}2,714.5pg/mL$) and placebo groups ($7,711.6pg/mL{\pm}3,041.3pg/mL$ and $7,286.1pg/mL{\pm}5,188.7pg/mL$, p = 0.027). In patients with overweight, the fatigue score was significantly decreased in the KRG group ($35.0{\pm}13.2$ and $24.5{\pm}8.9$, p = 0.019). Conclusion: Our results show that KRG might be effective in reducing proinflammatory cytokine and fatigue in overweight patients with NAFLD, in addition to improvements in adiponectin levels.

  • Bartonella henselae is the causative agent of cat scratch disease. Although cats are the main zoonotic reservoirs of Bartonella spp., unusual cases of cat scratch disease caused by a domestic dog scratch have been recently reported. For the in vivo B. henselae infection, eight dogs were inoculated intradermally with $2{\times}10^8CFU$ of B. henselae Houston-1 suspended in 1 ml of phosphate buffered saline on day 0 and subsequent injections of the same amount given intradermally on days 21, 28, 36, 58 and 64. After in vivo canine B. henselae infection was confirmed by nested PCR, the IFN-$\gamma$ levels of the culture supernatant of PBMC stimulated with B. henselae was significantly higher in the B. henselae-PCR positive group than the B. henselae-PCR negative group. Our results showed that the canine immune responses against B. henselae were different from those of cats. Th1 activation by B. henselae stimulation was characterized in dog peripheral blood mononuclear cells, whereas Th2 activation was reported in B. henselae-infected cats.

A Study on Emotional Labor, Emotional Burnout, Turnover Intention of Dental Hygienist (치과위생사의 감정노동, 감정소진, 이직의도에 관한 연구)

  • The purpose of this study was to examine dental hygienist's emotional labor, burnout and turnover intention. Data were collected from 215 dental hygienists working at hospitals and dental clinics through self-administered questionnaire, and analyzed by statistical analysis using t-test, ANOVA, Pearson's correlation coefficient and multiple regression analysis. The main findings of the study are as follows: the higher the educational level of dental hygienists, and the bigger the size of dental institutions they work, and the lower the monthly income of dental hygienists, the higher the level of emotional labor of dental hygienists. Burnout was influenced on emotional labor and turnover intention. Emotion labor and turnover intention were influenced on burnout. Tenure under 5 years, five-day workweek (non), emotional burnout were found to have significant effects on the turnover intention. Dental hygienists were doing relatively high emotional labor, and it influences his burnout experience and turnover intention. It is needed to make the workplace better environment which can secure appropriate work load and rewards, and education programs etc.

Antifungal Activities of Pseudomonas spp. Strains Against Plant Pathogens and Optimization of Culture Conditions (식물병원성 진균에 항균 효과를 지닌 슈도모나스 균주의 항진균 활성 증진을 위한 배양조건의 최적화)

  • To define the optimum conditions for the mass production of four antifungal Pseudomonas spp. isolated from soil, we have investigated culture conditions and effects of various nutrient sources on the bacterial growth and evaluated antagonistic activity against Rhizoctonia solani and Sclerotinia homoeocarpa, plant pathogens. The optimum temperature and pH for the growth of these isolates were determined as pH 7.0 and $20^{\circ}$ or $25^{\circ}C$, respectively. Sucrose, tryptone, and $K_2HPO_4$ generally were more adequate for better growth as carbon, nitrogen and mineral source, respectively. The nutrient sources were also found to be very effective for high antifungal activities against R. solani and S. homoeocarpa. It was elucidated that YUD-F group (P. mandelii and P. fluorescens), which inhabit regions at relatively low temperature, had more broad spectrum and higher antifungal activity than YUD-O group (P. trivialis and P. jessenii) generally against R. solani and S. homoeocarpa. It is thought that the differences of the average temperature in the various habitats of Pseudomonas spp. influence the optimal growth temperature and antifungal activity. Especially, Pseudomonas spp. of YUD-O group showed the better antifungal activity against dollar spot caused by S. homoeocarpa, but showed relatively weaker antifungal activity against brown patch caused by R. solani.

TNF$\beta$ Induces Cytotoxicity of Antibody-Activated CD$4^+$T-lymphocytes Against Herpes Virus-Infected Target Cells

  • We have extended our previous work that cross-linking CD4 molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$We have extended our previous work that cross-linking CD$4^+$ molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$ T cells. The killing activity of antibody activated CD$4^+$T cells was completely blocked by herbimycin A, a protein tyrosine kinase (PTK) inhibitor, but not by bisindolylamaleimide, a protein kinase C (PKC) inhibitor. Herbimycin A treated human or bovine peripheral blood CD$4^+$T cells lacked PTK activity and failed to kill virally infected target cells even after cross-linking of CD4 molecules. The CD$4^+$cross-linking failed to induce effector cell proliferation or the transcription of TNF${\beta}$ Upregulation of TNF${\beta}$ was induced by incubating the antibody activated effector cells with BHV-1 infected D17 target cells for 10 h. Anti-TNF${\beta}$ antibody partially abolished (13-44%) the direct effector cell-mediated antiviral cytotoxicity. However, this antibody neutralized 70 to 100% of antiviral activity of effector and target cell culture supernatants against BHV-1 infected D17 cells. The inhibition level of the antiviral activity by the antibody was dependent on the effector and target cell ratio. These results support the hypothesis that increased p$56^ICK enzyme activity in effector cells transduces a signal critical for effector cell recognition of viral glycoproteins expressed on the target cells. Following target cell recognition, lytic cytokines known to participate in target cell killing were produced. A better understanding of the killing activity displayed by CD$4^+$T lymphocytes following surface receptor cross-linking will provide insight into the mechanisms of cytotoxic activity directed toward virally-infected cells.T cells. Preparation of Chitosan-coated Magnetite Nanoparticles (키토산이 피복된 나노 크기의 자성체 분말 제조)

  • Magnetic nanoparticles can be used for a variety of biomedical applications. They can be used in the targeted delivery of therapeutic agents in vivo, in the hyperthermic treatment of cancers. in magnetic resonance (MR) imaging as contrast agents and in the biomagnetic separations of biomolecules. We have synthesized magnetite $(Fe_3O_4)$ nanoparticles using chemical coprecipitation technique with sodium oleate as surfactant. Nanoparticle size can be varied from 2 to 8nm by controlling the sodium oleate concentration. Magnetite phase nanoparticles could be observed from X-ray diffraction. Magnetic colloid suspensions containing particles with sodium oleate and chitosan have been prepared. Nanoparticles, both oleate-coated and chitosan-coated, have been characterized by several techniques. Atomic farce microscope (AFM) was used to image the coated nanoparticles. Magnetic hysteresis measurement were performed using a superconducting quantum interference device (SQUID) magnetometer at room temperature to investigate the magnetic properties of the magnetite nanoparticles. The SQUID measurements revealed superparamagnetism of nanoparticles.

The Roles of Amino and Carboxyl Domains in the Mouse Wee1 Kinases (생쥐 Wee1 인산화효소들의 각 도메인의 역할에 관한 연구)

  • The molecular machinery controlling cell cycle is centered around the regulation of the activity of maturation-promoting factor (MPF), a complex composed of a catalytic Cdc2 and the cyclinB regulatory subunit. Cdc2 kinase is inactivated by phosphorylation of inhibitory kinase, Wee1. It has been known that there are three different Wee1 kinases in the mammalian cell, Wee1A, Wee1B and Myt1. To investigate the regulatory mechanism of Wee1 kinases, the phosphorylation and degradation of Wee1A and Wee1B were checked in the Xenopus oocyte cell cycle. When Wee1 kinases were injected into frog oocyte, Wee1B was more stable than Wee1A. Wee1A and Wee1B kinase were phosphorylated by many kinases such as PKA and Akt. The roles of amino or carboxyl terminal in mouse Wee1A or Wee1B kinase were investigated using chimeric constructs. The degree of protein phosphorylation, degradation and cell cycle progression were different between chimeric constructs. The amino domain of Wee1A was implicated in the protein phosphorylation and degradation while amino domain of Wee1B and carboxyl domain of Wee1A were involved in the activity regulation. These results suggested that the domains of Wee1 kinase have different and significant roles in regulating the Wee1 kinases in the cell cycle progression.
