Dipterans are excellent insects to serve as forensic indicators to determine the postmortem interval of a human corpse. Therefore, we undertook a study to identify the major dipterans that are attracted to and develop in vertebrate carcasses. To determine what dipterans occur in Korea, chicken carcasses were placed in various habitats including the forest, open field, stream bank, greenhouse, rooftop, rice paddy, Chinese cabbage field, vacant hut, and pond at different times of the year. The dipterans were collected at various time intervals after placement in each habitat until the end of decomposition and identified. Our results showed that 49 identified and unidentified dipteran species from 36 genera in 19 families were collected from the carcasses. Habitat placement influenced the number of species isolated from the carcasses. For example, 39 species were collected from the forest habitat, 28 species from the open field, stream bank, or greenhouse each, 16 species from a vacant home, and 12 species from the pond. The dipteran species that were collected from all habitats were Phaenicia sericata, Lucilia illustris, and Lucilia sp. in the family Calliphoridae and Boettcherisca peregrine in the family Sarcophagidae. Other dipteran species that were isolated depended on carcass placement that is, fewer dipteran species were observed from carcasses in the backpack and much more species were recorded from unburied carcasses than buried carcasses. P. sericata, L. illustris, Lucilia sp., B peregrine and an unidentified sarcophagid species were observed year round, but some dipterans showed seasonal differences.

Chemokines is a small protein that plays a major role in inflammatory reactions and viral infections as a chemotactic factor of cytokines involved in innate immunity. Most of the chemokines belong to the chemokine groups CC and CXC. To investigate the immune system of the olive flounder (Paralichthys olivaceus), an expression pattern specifically induced in the early developmental stages of analysis is examined using qRT-PCR. We also examined tissue-specific expression of both CC and CXC chemokine in healthy olive flounder samples. CC and CXC chemokine shows increased expression after immune-related organs are formed compared to expression during early development. CC chemokine was more highly expressed in the fin, but CXC chemokine showed higher expression in the gills, spleen, intestines, and stomach. Spatial and temporal expression analysis of CC and CXC chemokine were performed following viral hemorrhagic septicemia virus (VHSV) infection. CC chemokine showed high expression in the gills, which are respiratory organs, whereas CXC chemokine was more highly expressed in the kidneys, an immune-related organ. These results suggest that CC and CXC chemokine play an important role in the immune response of the olive flounder, and may be used as basic data for the immunological activity and gene analysis of it as well as other fish.
In order to investigate the changes of free amino acids during the differentiation and development of young spike in naked barley, a typical spring grain, Wanju, and two winter grains, Sedohadaka and Nonsankwa No. 1-6 differing in their spring habits, were analyzed at different growth stages by thin layer chromatography. In all the varieties 22 ninhydrin positive components were detected at the sowing time of March 5 and 20 components in the sowing plots of March 30. In case of the latter plot, β-alanine was identified only in both Wanju and Sedohadaka, whereas pipecolic acid was detected only in Nonsankwa No. 1-6. Particularly, it is interesting that β-alanine was observed only in the case showing the normal heading independent of the varieties and sowing times. Whether these components are directly related to the physiology of spring habits in barley or not is also a question to be answered. Of the major amino acids, alanine and γ-aminobutyric acid were always detected in appreciably large spots, and serine, leucine, aspartic acid, valine and asparagine were somewhat larger. In the plot of march 30, glutamic acid was also detected in very large spot in both Wanju and Sedohadaka at the stage of spikelet differentiation and in Nonsankwa No. 1-6 at the stage of bract differentiation. Histidine, which showed the only qualitative difference among the varieties during seed germination, cannot be observed at all. Proline observed considerably large spot during seed germination was always detected but very small except that it was observed in large spot at the stage of floret differentiation in Nonsankwa No. 1-6 in the plot of March 5.
In this study, thirty-two stands in Kwangnung forest located in the central part of Korea were preferentially selected. In each stand, all stems for trees and shrubs were recorded by species and their girths were measured down to 5cm. In addition, several enviromental factors such as field soil pH, field soil moisture, soil compressibility, depth of soil, thickness of litter layer, elevation and basal area were measured. Three soil cores were sampled and various physical and chemcial properties was determined. The vegetational data were subjected to three kinds of multivariate ordination(PO, PCA, RA). The results suggested that Kwangnung forest was consisted of three forest types: coniferous, mixed and broad leaved forest communities. The relation between the stand scores of ordination and several environmental factors were investigated in terms of correlation analysis in order to examine the relationships between the vegetation and certain environmental factors. As a result of this analysis, the amount of sand content in A1 horizon decreased frm the coniferous to broad leaved forest, while maximum field capacity, pore space, exchangeable cations, loss on ignition, soil pH nad the amount of total nitrogen had a tendancy to increase significantly. However, easily soluble phosphorus appeared to have little to do with the forest types. The result of species ordination of centered-standardized PCA suggested that the major successional pathway in Kwangnung forest was; Pinus densifloralongrightarrowQuercus mongolica, Q. serrata, Q. alienalongrightarrowCarpinus laxifloralongrightarrowC. erosa in sequence. This trend is in good agreement with the past studies. In three kinds of ordination (centered PCA, centered-standardized PCA and RA) based on nineteen species and twenty-five stands, the total variances accounted for the first three axes were 77%, 46% and 63% respectively. The estimated beta diversity in Kwangnung forest assumed as a coenocline, was 1.5~1.8 HC. Increasing the effect of the sampling errors on ordination perfermance, this low heterogeneity seems to cause the poor concentration of the total variance. The results from the four kinds of ordination were in good agreement with each other, especially between PO, centered-standardized PCA and RA appeared robust. It seems to be worthy of applying multivariate method for analyzing other forest communities in Korea.

Baculoviruses are characterized by large double-stranded circular DNA genomes and rod-shaped enveloped virions. Bombyx mori nucleopolyhedrovirus(BmNPV) is a major pathogen, which causes severe damage in sericulture. Currently, BmNPV is recogtnized as an improtant tool in molecular biology, especially for expression of useful genes in B.mori cells and silkworm larvae. Our laboratories have focused on the studies of the molecular mechanisms of BmNPV replication and the application of BmNPV to agriculture and medicine. The entire nucleotide sequence of the BmNPV genome has recently determined. The BmNPV genome possessed 135 putative genes and 7 homologous repeated sequence (hrs) regions. Relatively little space, a few to a few hundred base-pairs, was observed between the open reading frames and hrs. Termination codons often overlapped. These results showed a compactly packde BmNPV genome. Based on comparative sequence analyses, we speculated that the ancestor of BmNPV was a baculovirus similar to Autographa californica NPV(AcNPV). The function of the BmNPV genes were characterized by gene deletion analysis; p35 was found to be involved in blocking apoptosis and cysteine proteinase was found to be involved in horizontal virus transmission by degrading viral-infected larval host. By AcNPV and BmNPV coinfection experiments, we identified a BmNPV gene involved in expanding host specificity of AcNPV. The identified gene was likely encoded a DNA helicase based on the amino acid sequence analysis; a few amino acid substitutions in the putative DNA helicase gene resulted in the expansion of host range of AcNPV. These findings indicate that BmNPV evolved within a short period from an AcNPV-like ancestral virus due to rapid evolution including specific amino acid substitutions and gene deletions/insertions.
A recent genetic study with Brucella abortus revealed the secretion activator gene A (SagA) as an autolysin component creating pores in the peptidoglycan (PGN) layer for the type IV secretion system (T4SS) and peptidoglycan hydrolase inhibitor A (PhiA) as an inhibitor of SagA. In this study, we determined the crystal structures of both SagA and PhiA. Notably, the SagA structure contained a PGN fragment in a space between the N- and C-terminal domains, showing the substrate-dependent hinge motion of the domains. The purified SagA fully hydrolyzed the meso-diaminopimelic acid (DAP)-type PGN, showing a higher activity than hen egg-white lysozyme. The PhiA protein exhibiting tetrameric assembly failed to inhibit SagA activity in our experiments. Our findings provide implications for the molecular basis of the SagA-PhiA system of B. abortus. The development of inhibitors of SagA would further contribute to controlling brucellosis by attenuating the function of T4SS, the major virulence factor of Brucella.
Riptortus clavatus, one of the many insects in major crops, damages pods and seeds, which reduces seed vigor and viability in soybeans. This study was conducted to examine the effect of diversely damaged seeds by R. clavatus on seed germination and seedling emergence and to determine the association of damaged seed with quality and yield of soybean sprouts. All seeds damaged by R. clavatus significantly (P<0.05) reduced seed vigor as measured by the rates of seed germination, germination speed, and seedling emergence. Mean seed germination rate of non-damaged seeds in sprout-soybean varieties was 97.8%, whereas the rates of seeds damaged at different levels, 31-50% and 51-80%, were 23.0 and 5.4%, respectively. The rates of seedling rot and abnormal, incomplete germination significantly (P<0.05) increased as the amount of seeds damaged by R. clavatus increased to 5, 10 and 15% against the total seeds for sprout production. Yield of soybean sprouts from seeds damaged at different levels decreased up to 13% as compared to that in normal seeds. In customer preferences on soybean sprout produce, 84% of customers participated in survey preferred to purchase sprouts from seeds with 5% of damaged seeds, but sprouts produced from seeds with 15% of damaged seeds were intended to purchase only by 22% of the customers. Areas of the seed damaged by R. clavatus were readily infected by pathogens as the seed germinated, resulted in deteriorated quality and reduced yield of sprout produce.

Osteoarthritis (OA) is a naturally occurring, irreversible disorder and a major health burden. The disease is multifactorial, involving both physiological and mechanical processes, but calcium crystals have been associated intimately with its pathogenesis. This study tested the hypothesis that these crystals have a detrimental effect on the differentiation of osteoclasts and bone homeostasis. This study employed an osteoblastosteoclast coculture system that resembles in vivo osteoblastdependent osteoclast differentiation along with $Ca^{2+}$-phosphate-coated culture dishes. The calcium-containing crystals upregulated the expression of RANKL and increased the differentiation of osteoclasts significantly as a result. On the other hand, osteoblast differentiation was unaffected. MicroRNA profiling showed that dual-specificity phosphatases 1 (DUSP1) was associated with the increased RANKL expression. DUSP1 belongs to a family of MAPK phosphatases and is known to inactivate all three groups of MAPKs, p38, JNK, and ERK. Furthermore, knockdown of DUSP1 gene expression suggested that RANKL expression increases significantly in the absence of DUSP1 regulation. Microarray analysis of the DUSP1 mRNA levels in patients with pathological bone diseases also showed that the downregulated DUSP1 expression leads to increased expression of RANKL and consequently to the destruction of the bone observed in these patients. These findings suggest that calcium-containing crystals may play a crucial role in promoting RANKL-induced osteoclastogenesis via DUSP1.
Distribution of Salmonella enterica serovars and their associated virulence determinants is wide-spread among food animals, which are continuously implicated in periodic salmonellosis outbreaks globally. The aim of this study was to determine and evaluate the diversity of five Salmonella serovar virulence genes (invA, pefA, cdtB, spvC and iroN) isolated from food animals and humans. Using standard microbiological techniques, Salmonella spp. were isolated from the feces of humans and three major food animals. Virulence determinants of the isolates were assayed using PCR. Clonal relatedness of the dominant serovar was determined via pulsed-field gel electrophoresis (PFGE) using the restriction enzyme, Xbal. Seventy one Salmonella spp. were isolated and serotyped into 44 serovars. Non-typhoidal Salmonella (NTS; 68) accounted for majority (95.8%) of the Salmonella serovars. Isolates from chicken (34) accounted for 47.9% of all isolates, out of which S. Budapest (14) was predominant (34.8%). However, the dominant S. Budapest serovars showed no genetic relatedness. The invA gene located on SPI-1 was detected in all isolates. Furthermore, 94% of the isolates from sheep harbored the spvC genes. The iroN gene was present in 50%, 100%, 88%, and 91% of isolates from human, chicken, sheep, and cattle, respectively. The pefA gene was detected in 18 isolates from chicken and a single isolate from sheep. Notably, having diverse Salmonella serovars containing plasmid encoded virulence genes circulating the food chain is of public health significance; hence, surveillance is required.
The white button mushroom (Agaricus bisporus) is one of the most widely cultivated species of edible mushroom. Despite its economic importance, relatively little is known about the genetic diversity of this species. Illumina paired-end sequencing produced 43,871,558 clean reads and 69,174 contigs were generated from five offspring. These contigs were subsequently assembled into 57,594 unigenes. The unigenes were annotated with reference genome in which 6,559 unigenes were associated with clusters, indicating orthologous genes. Gene ontology classification assigned many unigenes. Based on genome data of the five offspring, 44 polymorphic simple sequence repeat (SSR) markers were developed. The major allele frequency ranged from 0.42 to 0.92. The number of genotypes and the number of alleles ranged from 1 to 4, and from 2 to 4, respectively. The observed heterozygosity and the expected heterozygosity ranged from 0.00 to 1.00, and from 0.15 to 0.64, respectively. The polymorphic information content value ranged from 0.14 to 0.57. The genetic distances and UPGMA clustering discriminated offspring strains. The SSR markers developed in this study can be applied in polymorphism analyses of button mushroom and for cultivar discrimination.
