Zinc has been considered as a vital constituent of proteins, including enzymes. Mobile reactive zinc ($Zn^{2+}$) is the key form of zinc involved in signal transductions, which are mainly driven by its binding to proteins or the release of zinc from proteins, possibly via a redox switch. There has been growing evidence of zinc's critical role in cell signaling, due to its flexible coordination geometry and rapid shifts in protein conformation to perform biological reactions. The importance and complexity of $Zn^{2+}$ activity has been presumed to parallel the degree of calcium's participation in cellular processes. Whole body and cellular $Zn^{2+}$ levels are largely regulated by metallothioneins (MTs), $Zn^{2+}$ importers (ZIPs), and $Zn^{2+}$ transporters (ZnTs). Numerous proteins involved in signaling pathways, mitochondrial metabolism, and ion channels that play a pivotal role in controlling cardiac contractility are common targets of $Zn^{2+}$. However, these regulatory actions of $Zn^{2+}$ are not limited to the function of the heart, but also extend to numerous other organ systems, such as the central nervous system, immune system, cardiovascular tissue, and secretory glands, such as the pancreas, prostate, and mammary glands. In this review, the regulation of cellular $Zn^{2+}$ levels, $Zn^{2+}$-mediated signal transduction, impacts of $Zn^{2+}$ on ion channels and mitochondrial metabolism, and finally, the implications of $Zn^{2+}$ in health and disease development were outlined to help widen the current understanding of the versatile and complex roles of $Zn^{2+}$.
The calcium-activated $K^+$ ($BK_{Ca}$) channel is one of the potassium-selective ion channels that are present in the nervous and vascular systems. $Ca^{2+}$ is the main regulator of $BK_{Ca}$ channel activation. The $BK_{Ca}$ channel contains two high affinity $Ca^{2+}$ binding sites, namely, regulators of $K^+$ conductance, RCK1 and the $Ca^{2+}$ bowl. Lysophosphatidic acid (LPA, 1-radyl-2-hydroxy-sn-glycero-3-phosphate) is one of the neurolipids. LPA affects diverse cellular functions on many cell types through G protein-coupled LPA receptor subtypes. The activation of LPA receptors induces transient elevation of intracellular $Ca^{2+}$ levels through diverse G proteins such as $G{\alpha}_{q/11}$, $G{\alpha}_i$, $G{\alpha}_{12/13}$, and $G{\alpha}s$ and the related signal transduction pathway. In the present study, we examined LPA effects on $BK_{Ca}$ channel activity expressed in Xenopus oocytes, which are known to endogenously express the LPA receptor. Treatment with LPA induced a large outward current in a reversible and concentration-dependent manner. However, repeated treatment with LPA induced a rapid desensitization, and the LPA receptor antagonist Ki16425 blocked LPA action. LPA-mediated $BK_{Ca}$ channel activation was also attenuated by the PLC inhibitor U-73122, $IP_3$ inhibitor 2-APB, $Ca^{2+}$ chelator BAPTA, or PKC inhibitor calphostin. In addition, mutations in RCK1 and RCK2 also attenuated LPA-mediated $BK_{Ca}$ channel activation. The present study indicates that LPA-mediated activation of the $BK_{Ca}$ channel is achieved through the PLC, $IP_3$, $Ca^{2+}$, and PKC pathway and that LPA-mediated activation of the $BK_{Ca}$ channel could be one of the biological effects of LPA in the nervous and vascular systems.

Background: The biological activities of ginseng saponins (ginsenosides) are associated with type, number, and position of sugar moieties linked to aglycone skeletons. Deglycosylated minor ginsenosides are known to be more biologically active than major ginsenosides. Accordingly, the deglycosylation of major ginsenosides can provide the multibioactive effects of ginsenosides. The purpose of this study was to transform ginsenoside Rb2, one of the protopanaxadiol-type major ginsenosides, into minor ginsenosides using ${\beta}$-glycosidase (BG-1) purified from Armillaria mellea mycelium. Methods: Ginsenoside Rb2 was hydrolyzed by using BG-1; the hydrolytic properties of Rb2 by BG-1 were also characterized. In addition, the influence of reaction conditions such as reaction time, pH, and temperature, and transformation pathways of Rb2, Rd, F2, compound O (C-O), and C-Y by treatment with BG-1 were investigated. Results: BG-1 first hydrolyzes 3-O-outer ${\beta}$-$\text\tiny{D}$-glucoside of Rb2, then 3-O-${\beta}$-$\text\tiny{D}$-glucoside of C-O into C-Y. C-Y was gradually converted into C-K with a prolonged reaction time, but the pathway of Rb2 ${\rightarrow}$ Rd ${\rightarrow}$ F2 ${\rightarrow}$ C-K was not observed. The optimum reaction conditions for C-Y and C-K formation from Rb2 by BG-1 were pH 4.0-4.5, temperature $45-60^{\circ}C$, and reaction time 72-96 h. Conclusion: ${\beta}$-Glycosidase purified from A. mellea mycelium can be efficiently used to transform Rb2 into C-Y and C-K. To our best knowledge, this is the first result of transformation from Rb2 into C-Y and C-K by basidiomycete mushroom enzyme.
청국장은 한국의 전통발효식품으로서 다양한 생리활성이 보고되어 있다. 본 연구에서는 B. amyloliquefaciens 균종에 속하는 신규한 균주인 B. amyloliquefaciens(SRCM 100730)에 의한 발효 청국장이 선천면역 세포인 RAW 264.7 대식세포를 활성화하는 효과를 가지는가를 검토하였다. 청국장 추출물을 RAW 264.7 세포에 처리한 결과 처리농도에 의존하여 $TNF-{\alpha}$와 NO의 생성이 증가하였으며, 이러한 면역조절 물질의 증가는 iNOS와 $TNF-{\alpha}$ mRNA 발현에서도 확인되었다. 또한, 청국장 추출물에 의한 대식세포 활성화와 관련한 세포 내 작용기전을 해석한 결과 청국장 처리에 의해 p38과 ERK와 같은 MAPK의 인산화와 $NF-{\kappa}B$ 활성화가 유도되는 것으로 밝혀졌다. 하지만 MAPK에 속하는 JNK에 대해서는 아무런 영향을 주지 않는 것으로 나타났다. 한편 청국장 추출물을 LPS와 함께 처리한 실험에 있어서 청국장 추출물은 LPS에 의한 대식세포 활성화를 촉진하여 $TNF-{\alpha}$와 NO의 생성을 증가시키는 시너지 효과를 나타내었다. 이들 결과로부터 B. amyloliquefaciens(SRCM 100730) 균주로 발효한 청국장 추출물은 대식세포를 활성화하여 선천면역을 증가시키는 효과가 있는 것으로 확인되었다.
중금속으로 오염된 농경지 토양을 효과적으로 보권하기 위해서는 토양 매체내에서 반응과정을 거치는 중금속의 동태와 이동성에 따라 한 가지 이상의 복원기술이 선택되어야 한다. 오염토양 복원 시 중요한 토양의 물질적 수리적 요인은 투수성, 공극성, 토성과 토양조직, 오염물질의 형태와 농도, 오염물질의 동태와 이동특성이다. 따라서 중금속으로 오염된 농경지 토양에 적용할 수 있는 복합복원기술 개발 방법을 기존의 사용하고 있는 적용 가능한 화학적 기술과 물리적 기술을 중심으로 검토하여 보았다. 심층토내 중금속을 제거하는 화학적 기술로서 토양세척이 있으나 이러한 단일 기술로는 효과적 복원이 어렵다. 따라서 토양세척기술이 가지고 있는 단점을 보완하기 위한 물리적 기술로 토양파쇄 기술이 있다. 그러나 토양세척과 토양파쇄 기술은 혼용하여 오염토양에 적용할 지라도 오염물질제거율은 높은 이류 흐름 지역에서는 확산유동에 의해 비율제한적으로 된다. 그러므로 선택된 두 가지 기술을 현장에서 효과적으로 적용하기 위해서는 오염토양 복원 시 공정별로 각각의 기술이 가지는 장단점을 파악하기 위해서는 기존 현장에서 기술 적용 시 발견된 문제점과 요인들을 검토하여야 한다. 또한 복원의 효율을 예측하기 위해서는 오염물질의 역학적 탈착, 유동과 이동을 포함하는 예상수학모형을 통해 오염토양의 이질성과 복합 반응에 의한 실질 심층토내에서의 유동 경로 정확한 특성을 파악하여야 한다.

Imagine a world where we could biomanufacture hybrid nanomaterials having atomic-scale resolution over functionality and architecture. Toward this vision, a fundamental challenge in materials science is how to design and synthesize protein-like material that can be fully self-assembled and exhibit information-specific process. In an ongoing effort to extend the fundamental understanding of protein structure to non-natural systems, we have designed a class of short peptides to fold like proteins and assemble into defined nanostructures. In this talk, I will talk about new strategies to drive the self-assembled structures designing sequence of peptide. I will also discuss about the specific interaction between proteins and inorganics that can be used for the development of new hybrid solar energy devices. Splitting water into hydrogen and oxygen is one of the promising pathways for solar to energy convertsion and storage system. The oxygen evolution reaction (OER) has been regarded as a major bottleneck in the overall water splitting process due to the slow transfer rate of four electrons and the high activation energy barrier for O-O bond formation. In nature, there is a water oxidation complex (WOC) in photosystem II (PSII) comprised of the earthabundant elements Mn and Ca. The WOC in photosystem II, in the form of a cubical CaMn4O5 cluster, efficiently catalyzes water oxidation under neutral conditions with extremely low overpotential (~160 mV) and a high TOF number. The cluster is stabilized by a surrounding redox-active peptide ligand, and undergo successive changes in oxidation state by PCET (proton-coupled electron transfer) reaction with the peptide ligand. It is fundamental challenge to achieve a level of structural complexity and functionality that rivals that seen in the cubane Mn4CaO5 cluster and surrounding peptide in nature. In this presentation, I will present a new strategy to mimic the natural photosystem. The approach is based on the atomically defined assembly based on the short redox-active peptide sequences. Additionally, I will show a newly identified manganese based compound that is very close to manganese clusters in photosystem II.
Rice Oryza sativa accelerated cell death and resistance 1 (OsACDR1) encodes a putative Raf-like mitogen-activated protein kinase kinase kinase (MAPKKK). We had previously reported upregulation of the OsACDR1 transcript by a range of environmental stimuli involved in eliciting defense-related pathways. Here we apply biochemical, gain and loss-of-function approaches to characterize OsACDR1 function in rice. The OsACDR1 protein showed autophosphorylation and possessed kinase activity. Rice plants overexpressing OsACDR1 exhibited spontaneous hypersensitive response (HR)-like lesions on leaves, upregulation of defense-related marker genes and accumulation of phenolic compounds and secondary metabolites (phytoalexins). These transgenic plants also acquired enhanced resistance to a fungal pathogen (Magnaporthe grisea) and showed inhibition of appressorial penetration on the leaf surface. In contrast, loss-of-function and RNA silenced OsACDR1 rice mutant plants showed downregulation of defense-related marker genes expressions and susceptibility to M. grisea. Furthermore, transient expression of an OsACDR1:GFP fusion protein in rice protoplast and onion epidermal cells revealed its localization to the nucleus. These results indicate that OsACDR1 plays an important role in the positive regulation of disease resistance in rice.
Glucagon-like peptide-1 (GLP-1) has a broad spectrum of biological activity by regulating metabolic processes via both the direct activation of the class B family of G protein-coupled receptors and indirect nonreceptor-mediated pathways. GLP-1 receptor (GLP-1R) agonists have significant therapeutic effects on non-alcoholic fatty liver disease (NAFLD) and steatohepatitis (NASH) in animal models. However, clinical studies indicated that GLP-1 treatment had little effect on hepatic steatosis in some NAFLD patients, suggesting that GLP-1 resistance may occur in these patients. It is well-known that the gut metabolite sodium butyrate (NaB) could promote GLP-1 secretion from intestinal L cells. However, it is unclear whether NaB improves hepatic GLP-1 responsiveness in NAFLD. In the current study, we showed that the serum GLP-1 levels of NAFLD patients were similar to those of normal controls, but hepatic GLP-1R expression was significantly downregulated in NAFLD patients. Similarly, in the NAFLD mouse model, mice fed with a high-fat diet showed reduced hepatic GLP-1R expression, which was reversed by NaB treatment and accompanied by markedly alleviated liver steatosis. In addition, NaB treatment also upregulated the hepatic p-AMPK/p-ACC and insulin receptor/insulin receptor substrate-1 expression levels. Furthermore, NaB-enhanced GLP-1R expression in HepG2 cells by inhibiting histone deacetylase-2 independent of GPR43/GPR109a. These results indicate that NaB is able to prevent the progression of NAFL to NASH via promoting hepatic GLP-1R expression. NaB is a GLP-1 sensitizer and represents a potential therapeutic adjuvant to prevent NAFL progression to NASH.
'우리는 누구이며, 어디서 왔는가?' 우리 스스로에게 질문하고 우리 자신을 이해하는 것, 이것이야말로 인류 역사상 가장 위대한 혁명일 것이다. 하지만 우리는 아직 우리의 정체성을 묻는 이 물음에 명확한 답을 얻지 못하고 있다. 물론 '사회적 동물' 혹은 '이성적 동물' 같은 전형적인 정의들이 있지만 이들 역시 매우 허약한 규정들이다. 우리가 왜 사회적 동물이 되고 이성적 동물이 되었는지에 대한 해명조차 담고 있지 않기 때문이다. 이 글의 목적은 이 물음의 해답을 찾아가는 단서를 발견하여 우리의 정체성을 좀 더 명확히 하는데 있다. 실마리는 진화론적으로 우리와 가장 가까운 혈족으로 불리는 침팬지에게서 출발한다. 우리와 침팬지는 무엇이 다른가? 침팬지에게도 '자아'가 있을까? 이런 물음들을 통해 우리가 찾아낸 답은 이렇다. 즉 인간이 침팬지와 다른 존재가 된 것은 뇌의 발달과 함께 정밀하게 움직이는 운동 시스템을 갖게 되었고, 나아가 이러한 운동 시스템이 내면화되어 마음을 갖게 되었기 때문이다. 그리고 마음의 생각하는 능력도 바로 이러한 내면화 메커니즘이 진화 발전한 것이다.
농업가뭄에 대한 연구는 주로 가뭄지표의 개선과 제안 등에 초점이 맞추어져 있으며 관개기의 강수량에 중점을 둔 사례가 대부분이다. 그러나 대부분의 논 관개용수는 저수지를 통하여 공급되기 때문에 관개가 시작되는 4월 초순의 저수량은 상당히 중요한 가뭄 대응 요소이다. 이에 따라 가뭄에 대비하기 위해서는 관개가 종료되는 10월부터 이듬해 3월까지 충분한 저수량이 확보될 필요가 있다. 그러나 기후변화에 의해 대부분의 강우가 관개기에 집중될 수 있으며 이에 따라 미래에는 비관개기 동안 충분한 저수량을 확보하는데 어려움이 발생할 수 있다. 이에 본 연구에서는 기후변화에 따른 비관개기의 강수량을 확률기반으로 분석하고, 이를 관개기 필요저수량과 비교하여 저수지의 미래 가뭄 대응 능력을 분석하고자 하였다. 기후변화에 따른 비관개기의 강수량 변화를 분석하기 위하여 RCP(Representative Concentration Pathways) 시나리오를 적용하여 미래 비관개기 동안의 연도별 비관개기 누적강수량을 분석하였다. 과거와 미래시기를 30년 단위로 구분하여 1995s, 2025s, 2055s, 2085s의 비관개기 동안의 비초과확률 10%, 50%, 90%의 강수량을 분석하였다. RCP 4.5 시나리오를 기반으로 모의한 미래강우의 비초과확률 10% 누적강수량 산정 결과에 따르면 주요 곡창지대인 전라남북도는 1995s에는 10월부터 이듬해 3월까지의 누적강수량이 약 215mm 정도로 나타났으나 2025s에는 약 150mm로 줄어드는 것으로 나타났다. 즉, 비초과확률 10%의 강우가 내릴 경우 비관개기 동안에 충분한 저수량의 확보가 어렵게 되고 관개기의 심각한 가뭄을 초래할 수 있었다. 비관개기 누적강수량과 저수량변화의 관개를 모의한 개운저수지와 계룡저수지의 경우 그 관계식은 y=1.442x-198.81, y=5.8105x-752.92와 같이 나타났다. y는 비관개기의 저수량변화를 나타낸 것이고 x는 비관개기의 누적강우량을 나타낸 것이다. 식을 통해 향후 100년 중 비초과확률 10%의 강수를 가정한다면 개운저수지는 관개종료시점의 저수율이 최소 96.93% 이상이여야 다음해 관개시작시점의 저수위가 만수위가 될 수 있었고, 계룡 저수지는 최소 86.84%의 저수위를 만족해야 다음해 관개시작시점의 저수위가 만수위가 될 수 있었다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
