Influence of Growing Green Manures on Soil Microbial Activity and Diversity under Organically Managed Grape-greenhouse (시설 포도 유기농 재배지에서 녹비 생육 중 녹비 종류가 토양 미생물상에 미치는 영향)

  • The aim of present work was to assess the response of soil microbial activity and diversity to green manures under the organically managed grape-greenhouse in early spring. Hairy vetch, milk vetch, and red clover were seeded in fall, and enzymatic activities by dehydrogenase and fluorescein diacetate (FDA) hydrolase, and microbial diversities by Biolog $EcoPlate^{TM}$ and phospholipid fatty acid (PLFA) were characterized for soils sampled in early spring. Dehydrogenase activity and FDA hydrolytic activity did not differentiate the green manures but the average well color development of Biolog EcoPlate was higher in soils covered with red clover than control soil. Soil microbial functional diversity by Biolog EcoPlate differentiated the soils covered with hairy vetch and milk vetch, and Shannon diversity index by Biolog EcoPlate was higher in soils covered with hairy vetch than control soil. Principal component analysis of PLFA differentiated the soils covered with milk vetch from control soil.

A Data Base for Identification of Pseudomonas syringae pv. actinidiae, the Pathogen of Kiwifruit Bacterial Canker, Using Biolog Program (Biolog Program을 이용한 참다래 궤양병균 동정용 Data Base)

  • Reactions of Pseudomonas syringae pv. actinidiae to 95 carbon sources in a 96-well microplate (BiOLOG GN MicroPlateTM) were investigated. The bacterium used 9 carbon sources such as D-mannitol, sucrose, etc., but did not use 62 carbon sources such as $\alpha$-cyclodextrin, dextrin, etc. Based on the reactions, a user data base for identification of P. syringae pv. actinidiae was constructed in Biolog program (BiOLOG MicroLogTM 2 system). P. syringae pv. actinidiae isolates collected from kiwifruits could be identified automatically with high similarity using the user data base, which could diagnose rapidly and easily whether the tree was infected with bacterial canker or not.

Metabolic Fingerprinting of Food Wastewater Treatment System (식품폐수 처리 단계별 미생물 대사지문)

  • To determine structure and activities of microbial communities in a food wastewater treatment system, biofilm of RABC (rotating activated Bacillus contactor) and samples of aeration tanks were analyzed. Heterotrophic bacterial concentrations were similar between biofilm and stage 1 aeration tank and decreased 2-log at stage 3 aeration tank as dissolved oxygen decreased, however portions of Bacillus groups were increased at stage 3 aeration tank. It was revealed by quantitative and qualitative analysis of metabolic fingerprinting patterns of Biolog GN2 plate that RABC represented much higher activities and a different microbial community structure compared to aeration tanks. Metabolic fingerprinting showed the carbon sources that isolated Bacillus groups could or could not use, were used similarly meaning that not only Bacillus groups but also other microbial groups would contribute to the treatment of wastewater.

Effects of Organic Amendments on Soil Microbial Community in Red Pepper Field (시용 유기물의 종류가 고추 재배지 토양 미생물상에 미치는 영향)

  • Diverse organic amendments available in local areas have been used to improve soil quality in red pepper field and so the need for investigating the soil chemical and biological properties changed by the organic amendments application is increasing. Soil microbial diversities were measured by phospholipid fatty acid (PLFA) and Biolog $EcoPlate^{TM}$. Compost was most effective for improving soil chemical properties including pH, EC, total nitrogen, P, K, and Ca, and bark increased soil organic matter significantly (P=0.05). Compost increased the fatty acids indicating actinomycetes and vascular arbuscular fungi, and ratio of cy19:0/18:1w7c and monounsaturated fatty acids/saturated fatty acids in soils in PLFA analysis. Bark increased soil fungal indicators in PLFA analysis (P=0.05). Principal component analysis of Biolog EcoPlate data and PLFA differentiated the compost- and bark-amended soils from other organic matteramended soils especially the soil incorporated with compost. More researches are needed to use bark for improving soil microbial properties because the soil chemical and microbiological properties caused by compost and bark are significantly different.

  • The effect of pipe materials on biofilm communities were investigated by CLPP (community level physiology profile) using Biolog GN plates. Heterotrophic bacterial concentrations were $10^4\;-\;10^6\;CFU/cm^2$ and there was no differences between galvanized iron and carbon steel. Average optical density of Biolog plate was similar between two pipe materials. However, CLPP was different according to the type of pipe materials and exposed times to tap water, and CLPP was independent of bacterial concentration. This represents the differences of bacterial communities with pipes and water contact times.

분만후 젖소의 자궁내 미생물상 조사 및 Lipopolysaccharide의 분리

  • 분만후 젖소의 자궁내 미생물상을 조사하고 미생물로부터 분리한 Lipopolysaccharide를 적용하여 소의 번식효율 증진에 기여하고자 분만후 젖소의 도축장 유래 자궁을 채취하여 혐기적 상태에서 균분리 동정을 실시하였다. 균분리 동정을 위하여, 시료를 1cm$\times$1cm로 채취하여 혐기상태에서 거품이 생길 때까지 vortexing한 후 균액 300$\mu l$를 뽑아 혐기배지에 도말하였고 도말한 plate는 $37{\circ}C$ 혐기chamber에서 24시간 배양하였다. 혐기배지에서 자란 균의 colony를 따서 Mac, BHI+B, BHI 배지에 배양한 후 Gram stain을 실시하였다. BHI 배지에서 자란 균의 colony를 따서 BUA+B 배지에 계대배양하였고 BUA+B 배지에서 자란 균중에 가장 마지막으로 자란 균을 따서 An-IF에 넣고 탁도를 63%T로 맞춘 후 An micro plate에 100$\mu l$씩 분주하였다. 분주한 plate를 $37{\circ}C$ 혐기 chamber에서 20~24시간 동안 배양한 후 Biolog를 실시하였다. 시료의 UV측정을 위하여 Sonic Processor로 세포를 분쇄하였고 분쇄한 세포를 $4{\circ}C$에서 10,000rpm으로 10분간 원심분리한 후 상층액을 분리하여 0.45$\mu m$ 필터로 여과한 다음 여과액을 취하여 UV로 standard(E.coli O26 B6 LPS)와 sample(10배 및 20배 희석액)을 측정하였다. Effect of Agricultural Practice and Soil Chemical Properties on Community-level Physiological Profiles (CLPP) of Soil Bacteria in Rice Fields During the Non-growing Season (논의 휴한기 이용형태와 토양화학성이 토양세균의 탄소원 이용에 미치는 영향)

  • BACKGROUND: Soil bacteria play important roles in organic matter decomposition and nutrient cycling during the non-growing season. The purpose of this study was to investigate the effects of soil management and chemical properties on the utilization of carbon sources by soil bacteria in paddy fields. METHODS AND RESULTS: The Biolog EcoPlate was used for analyzing community-level carbon substrate utilization profiles of soil bacteria. Soils were collected from the following three types of areas: plain, interface and mountain areas, which were tested to investigate the topology effect. The results of canonical correspondence analysis and Kendall rank correlation analysis showed that soil C/N ratio and NH4+ influenced utilization of carbon sources by bacteria. The utilization of carbohydrates and complex carbon sources were positively correlated with NH4+ concentration. Cultivated paddy fields were compared with adjacent abandoned fields to investigate the impact of cultivation cessation. The level of utilization of putrescine was lower in abandoned fields than in cultivated fields. Monoculture fields were compared with double cropping fields cultivated with barley to investigate the impact of winter crop cultivation. Cropping system altered bacterial use of carbon sources, as reflected by the enhanced utilization of 2-hydroxy benzoic acid under monoculture conditions. CONCLUSION: These results show that soil use intensity and topological characteristics have a minimal impact on soil bacterial functioning in relation to carbon substrate utilization. Moreover, soil chemical properties were found to be important factors determining the physiological profile of the soil bacterial community in paddy fields.

Fibrinolytic Activity and Characterization of Bacillus licheniformis HK-12 Isolated from Chungkook-Jang (청국장에서 분리한 세균인 Bacillus licheniformis HK-12의 혈전용해활성 및 특징)

  • The aim of this work was to investigate the fibrinolytic activity and characterization of Bacillus licheniformis HK-12, which produces the fibrinolytic enzyme excreted from naturally fermented Chungkook-Jang. Initially, the physiological and biochemical characteristics of strain HK-12 was examined. Both physiological analysis using BIOLOG system and phylogenetic analysis using 16S rRNA sequencing were performed to identify the strain, and the strain could be assigned to Bacillus licheniformis, designated as B. lichenformis HK-12, and registered in GenBank as [EU288193]. Phylogenetic analysis of B. licheniformis HK-12 was plotted based on 16S rRNA sequence comparisons. During the incubation period of B. licheniformis HK-12, the changes of bacterial growth, fibrinolytic activity, and pH were monitored. As the results, after 36 hours of incubation, the maximum fibinolytic activity was about 2.25 times than that of plasmin used as standard. Optimal conditions on the growth of B. licheniformis HK-12 associated with the fibrinolytic activity was initial pH 7.0 and 40$^{\circ}C$, respectively.

Distribution and Antimicrobial Susceptibility Patterns of Bacteria Isolated from Genital Tract in Thoroughbred Mares (더러브렛 씨암말의 생식기내 세균의 분포 및 항생제 감수성 양상)

  • This study carried out to investigate the genital tract bacterial flora of Thoroughbred mare in Jeju province during March and July, 2006. The specimens were collected from vaginal ucosa and clitorial fossa using a culture swab (BBL, USA) from 100 Thoroughbred mares. Colonies were selected blood and MacConkey agar plate, and identified as standard biochemical properties using Biolog system (Thermo, USA). In this study, 470 gram-negative strains were isolated more frequently than 249 gram-positive strains. We were Isolated Escherichia coli (19.8%), Proteus mirabillis (14.9%), Enterobacter nimipressuralis (7.4%), Enterobacter mobilis (4.7%), Aeromonas encheleia (4.3%), Pseudomonas aeroginosa (3.0%), Staphylococcus aureus (14.9%), Staphylococcus epidermidis (11.2%), Coagulasenegative Staphylococcus spp. (10.0%), Enterococcus faecalis (9.2%), Enterococcus faecium (8.0%), Actinomyces viscosus (7.2%), Micoroccus diversus(6.8%), Streptococcus dysgalactiae subsp. equisimilis(5.2%), Streptococcus equi subsp. zooepidemicus (3.2%), Other non-beta hemolytic Streptococcus spp. (2.0%) and many others from vaginal mucosa and clitorial fossa in Thoroughbred mares. No significant bacteria (Taylorella equigenitalis and Klebsiella pneumonia) were isolated from the mare genital tract. In antimicrobial agents susceptibility test, it shows a high sensibility in the antibiotics of the most which excepts the streptomycin and neomycinm, kanamycin, spectinomycin, compound sulfonamides. Especially, Staphylococcus epidermidis, Enterococcus spp. and Streptococcus spp. were visible a high sensibility in the all antibiotics. However, Staphylococcus aureus, Pseudomonas aeruginosa, Proteus spp. and E. coli were showed a high antibiotic resistance patterns. These results may provide the basic information to establish strategies for the treatment and prevention of reproductive disease in Thoroughbred mares in Korea.

Isolation and Identification of the Origins Causing the Slime Found in Pulp and Paper Making Processes (제재공장내 슬라임 발생원의 분리와 동정)

  • The presence of slime in paper mills is practically universal. Many researches have been performed for many years to resolve the problem caused by the slime in pulp and paper mill. Many papers have been published to show the bacteria is a major cause of paper mill slime. Now that the recycling of the water has been increased and the regulations of a toxic chemical dosage have become more strengthen, the importance of the control of slime in pulp and paper mill recently has been more recognized. Therefore, to produce quality products at the lowest economic and environmental costs, a through study of the microbial ecology and the indentification of troublesome slime-forming bacteria is a quite necessary. The purpose of this paper is to indentify slime~forming bacteria isolated from the papermaking process. The samples were taken from four parts of making fine paper : machine chest, head box, wire part, white water tank. Machine chest showed the most numbers of bacteria, numbering $2.55{\times}10^7$. The different colony types were taken from the 105 dilution plate. Nine bacteria were identified u sing the Biolog system and the vitek system: 6 gram-negative bacteria, 3 gram-positive bacteria. They are Pseudomonas paucimobilis B., Staphylococcus sp., Acinetobacter calcoaceticus., Pseudomonas cepacia, Actinobaci1lus capsulatus, Acidovorax sp., Flavobacterium sp., and Staphylococcus auricularis in addition to one unidentified sp., Among them. Pseudomonas paucimobillis was found in all places where the samples were taken. And, each parts had the different predominant bacteria in it : Pseudomonas paucimobilis B. in machine chest, Acinetobactor calcoaceticus. in Wire Part and Staphylococcus sp. in head box.
