감귤의 Bioflavonoids 분리, 정제 및 혈압강하효과 (Isolation, Purification and Hypotensive Effect of Bioflavonoids in Citrus sinensis)

  • 냉동 건조한 귤(Citrus sinensis)의 껍질로 부터 methanol과 butanol추출물인 crude bioflavonoids를 건조중량으로 최초 시료대비 약 0.26%의 수율을 얻을수 있었다. 이 crude bioflavonoids를 gel filtration, HPLC를 이용하여 분리, 정제하였고 TLC, HPLC, UV spectrum, NMR spectral analysis를 통해 narirutin과 hesperidin으로 동정할 수 있었다. 또한 HPLC 정량분석 결과, narirutin과 hesperidin은 crude bioflavonoids의 g당 42mg과 530mg을 얻을 수 있었으며 귤 껍질의 주요 bioflavonoid는 hesperidin으로 판명되었다. 한편 실험 동물인 Sprague-Dawley종의 흰쥐를 대상으로 혈압 변화를 측정한 결과 귤 껍질의 주요 bioflavonoid성분인 hesperidin은 약물 투여 후 유의성있게 (p< 0.001) 혈압 저하효과가 있음을 관찰 할 수 있었다.

Functional Characteristics of Cyclodextrin Glucanotransferase from Alkalophilic Bacillus sp. BL-31 Highly Specific for Intermolecular Transglycosylation of Bioflavonoids

  • The functional characteristics of a ${\beta}$-cyclodextrin glucanotransferase (CGTase) excreted from alkalophilic Bacillus sp. BL-31 that is highly specific for the intermolecular transglycosylation of bioflavonoids were investigated. The new ${\beta}$-CGTase showed high specificities for glycosyl acceptor bioflavonoids, including naringin, rutin, and hesperidin, and especially naringin. The transglycosylation of naringin into glycosyl naringin was then carried out under the conditions of 80 units of CGTase per gram of maltodextrin, 5 g/l of naringin, 25 g/l of maltodextrin, and 1 mM $Mn^{2+}$ ion at $40^{\circ}C$ for 6 h, resulting in a high conversion yield of 92.1%.

중공사 막을 이용한 감귤 과피 bioflavonoids 분리 및 식이 섬유 회수 공정 최적화 (Optimization of Separation Process of Bioflavonoids and Dietary Fibers from Tangerine Peels using Hollow Fiber Membrane)

  • 감귤류의 가공 부산물인 감귤 과피 내에는 식이 섬유소 외에 미생물의 성장을 억제시키는 naringin과 혈압 저해 능력이 있는 hesperidin 등의 bioflavonoids가 다량 함유되어있지만, 대부분이 가공 과정 중 폐기되고 있다. 생리 활성 물질인 bioflavonoids를 회수하기 위하여 원료 투입 속도, 막 횡단 압력, 온도, pH등을 조절하여 최적 막 분리 조건을 구하였다. 감귤 과피에 7.5 배의 물을 첨가하여 균일하게 혼합한 후, 혼합 용액을 예비 여과 시스템에서 여과하였다. 예비 여과된 추출물은 중공사 막을 이용하여 한외 여과하였다. 원료 투입 속도와 flux간에는 일정한 상관 관계가 없었지만, 막 횡단 압력 8 psi 이상에서 pressure controlled region이 관찰되었다. $9^{\circ}C$에서 $25^{\circ}C$로 온도가 상승함에 따라 $10{\;}liters/m^2/hr{\;}(LMH)$씩 flux가 증가하였으나, $25^{\circ}C$에서 $33^{\circ}C$ 구간에서는 flux 증가폭이 2 LMH에 그쳤다. 모든 적용 막 횡단 압력에서 pH 4.8일 때의 flux가 가장 높았으며, pH 3.0에서의 flux가 pH 6.0, 7.0, 9.0 일 때 보다 훨씬 낮았다. 최적 운용 조건은 원료 투입 속도 49.3 L/hr, 막 횡단 압력 10psi,온도 $25^{\circ}C$, pH 4.8 이었다. 최적 분리 조건하에서 막 분리 하였을 때, 시간이 경과함에 따라 flux가 점차적으로 감소하여 약 1시간 50분 경과 후 정상 상태에 도달하였다. 각 분리 단계별로 retentate 단위 무게당 식이 섬유 함량과 permeate의 bioflavonoids농도를 측정하였다. 분리 단계별 retentate중 단위 무게당 총 식이 섬유와 수용성 식이 섬유 함량은 각각 170 mesh retentate와 prefiltered retentate에서 가장 높았다 한외 여과 permeate내의 naringin 과 hesperidin 농도는 각각 $0.45{\sim}0.65\;mg/g,\;5.15{\sim}6.86\;mg/g$으로, 감귤 과피 고유의 함량보다 각각 $15{\sim}22$배, $79{\sim}93$배 높은 수치였다. 따라서 , 감귤 과피로부터 bioflavonodsi를 회수하는데 있어서 PM10 중공사막의 운용이 매우 효과적인 분리 시스템임을 알 수 있었다.

Bioflavonoid naringin이 치아 우식증과 치주 질환 억제에 미치는 효과 (Effect of Naringin to Dental Caries and Periodontal Disease)

  • The purpose of this study was to evaluate the bioflavonoids naringin on dental caries and periondontal disease in the albinorat. Twenty-five-day-old-male rats were fed the experimental diets for 42 days in this work at the end of the 42-day experimental period. The tooth surfaces were examined under a dissecting microscope. The sulcular caries lesions were recorded : the first molars were more affected than the second and third molars. Alveolar bone loss was measured on the buccal and lingual aspects of each molar: three site measurements (mesialpoint midpoint and distalpoint) were taken on the first molars. The results showed that the bioflavonoids, naringin had beneficial effects in the prevention of periodontal disease.

  • Bioflavonoids are semi-essential food components that are ubiquitously present in nature. It has been reported that flavonoids act as anti-oxidant as well as anti-cancer agents. Quercetin is one of the most widely distributed bioflavonoids in the plant kingdom. The goal of this study was to investigate effects of quercetin analogs extracted from Lindera erythrocarpa, quercetin 3-O-${\alpha}$-arabinofuranoside and quercetin 3-O-${\alpha}$-L -rhamnoside, on oxidatie stress-induced cell death. The increase of matrix metalloproteinases expression is a key event in several pathological conditions, e.g., dermal photocarcinogenesis, tumor initiation, invasion and metastasis. A new Potential Anti-inflammatory Agent

  • Biflavonoid is one of unique classes of naturally-occurring bioflavonoids. Certain biflavonoids including amentoflavone were previously reported to have inhibitory effect on the group 11 phospholipase $A_2$ activity. Amentoflavone was also found to inhibit cyclooxygenase from guinea-pig epidermis without affecting lipoxygenase. In this study, anti-inflammatory and analgesic activities of amentoflavone were evaluated. When amentoflavone was administered intraperitoneally, it showed a potent anti-inflammatory activity as determined by amelioration of croton-oil induced mouse ear edema. It also showed a potent anti-inflammatory activity in the rat carrageenan paw edema model ($ED_{50}$=42 mg/kg) compared to the activity of prednisolone (35 mg/kg) and indomethacin (10 mg/kg). However, amentoflavone did not show a significant inhibitory activity against rat adjuvant-induced arthritis, a chronic inflammatory model. In addition, amentoflavone was found to possess a potent analgesic activity in the acetic acid writhing test ($ED_{50}$=9.6 mg/kg) compared to the activity of indomethacin (3.8 mg/kg). These results suggest that amentoflavone may be a potential lead for a new type of anti-inflammatory agents having dual inhibitory activity of group 11 phospholipase $A_2$ and cyclooxygenase.

Evaluation of the cytotoxicity of gold nanoparticle-quercetin complex and its potential as a drug delivery vesicle

  • Recently, conjugates of medicinal herb-derived bioflavonoids, such as quercetin, and gold nanoparticles (GNPs) have gained attention as targeted drug delivery systems. In the present study, because quercetin is an important flavonoid with anti-cancer, anti-inflammatory, and anti-oxidant properties, GNP-quercetin complexes (GNPQs) were synthesized to investigate possible adverse effects such as cytotoxicity. We found that while quercetin was cytotoxic, GNPQs were not cytotoxic towards the RAW 264.7 and THP-1 cell lines. Therefore, GNPQs may serve as a potential drug delivery system for cancer treatment.

Protective effects of citrus based mixture drinks (CBMDs) on oxidative stress and restraint stress

  • In the current study investigated the protective effects of citrus based mixture drinks (CBMDs) using oxidative stress in human dermal fibroblast (HDF) cells and restraint-stressed rats. The CBMDs contained citrus bioflavonoids including narirutin and hesperidin. The cell viability of HDF cells treated with $H_2O_2$ was observed at 53.9% but treated with CBMD-1 and CBMD-2 ($500{\mu}g/mL$) on $H_2O_2$ exposed HDF cells significantly increased the relative cell viability at 65.0 and 72.2%, respectively. In the treadmill test, the time spent on the electrode plate in the restraint-stressed group was analyzed 24.1 s, but restraint-stressed rats with administered CBMDs (300 mg/kg) had significantly decreased the time at 2.4 (CBMD-1) and 4.7 (CBMD-2) s, respectively. In addition, number of touches the electrode plate in restraint-stressed group was observed at 42.4 ea, but, restraint-stressed rats with administered CBMD-1 and CBMD-2 (300 mg/kg) were significantly decreased at 7.0 and 10.2 ea, respectively.
