Investigating Biochemical Properties of Bacillus aryabhattai DA2 from Diesel-Contaminated Soil

  • Petroleum energy is the major source of the world energy market, and its massive usage, and the corresponding extreme environmental pollution, imposes a serious threat on the ecological cycles. By screening oil-contaminated soil, we isolated, identified, and characterized a novel strain that represents a considerable diesel-degrading potentiality; the Bacillus aryabhattai DA2 strain is registered in the NCBI with the accession number MG571630, and it possesses an efficient tributyrin-degrading capacity. The optimal condition for diesel degradation by DA2 strain was observed at pH between 7-8 and at the temperature of $30^{\circ}C$. The strain is resistant to salt as well as the antibiotics like ampicillin and streptomycin. These results indicate B. aryabhattai is one of the potential candidates for the remediation of the diesel-contaminated sites.

  • Reviewed biosynthetic pathways in green leaves of thiamine, riboflavin and folate as disclosed by the authors, and the regulatory systems operating on their biosynthesis and biodegradation as revealed since a potent inhibitory activity of ATP was found for the phosphatase reactions in 1965. Physiological roles of ascorbic acid in plant were evidenced by its higher content in green and flower leaves; the content in persimmon leaves for example was found ten or more times of that in citrus fruits. A close relationship existing between basic and development researches was illustrated by examples chosen from the author's researches. Examples cited were as follows; basic researches on vitamins were extended to the creation of thiamine enriched rice, to the growth promotion of rice plant by foliar application of thiamine, and to the use of ascorbic acid to prevent the appearance of beer cloudness; histological and biochemical studies on protein bodies in rice endosperm turned out amino acid enriched rice; techniques acquired in the studies on catalase were successfully employed for the utilization of single cell protein (MIPRON) for human food; conception of hybernation was led up to under-water or under-ground storage of cereal grains.

  • This paper was investigated to clarify the possibility of a biodegradation of materials adsorbed on different porous granular-activated carbons (GACs) such as coal-& coconut-based GAC. Total organic carbon, humic substance and ammonia were used to compare their removal efficiencies. The objective of this study is to determine the adsorption capacity of bioregenerated GAC. When raw water reacted with chloride, the yield of THMs increased as a function of the input amount of chloride. The formation of trihalomethanes (THMs) was investigated in water treated with chlorine when humic acid was used as THM precursor. As the input amount of chloride in raw water increased by two or five-fold to remove the $NH_3$, the chloroform of the THMs significantly increased also five or ten-fold. It was found that the chloroform was significantly removed by the treatment of biological activated carbon (BAG) in comparison with the ozone treatment, and the removal efficiency of THMs in coal-typed GAC was $10-30\%$ better than coconut-typed GAC due to the biological degradation on the surface of the activated carbons.

  • In previous study, we examined poly(butylene succinate) ionomer (PBSi) and confirmed that PBSi showed acceptable mechanical and rheological properties to apply in various field, due to the physical cross-linkage formed by ion aggregation. Besides, the incorporation of ionic groups led to the change of surface properties such as the hydrophilicity and surface morphology, which could affect hydrolytic degradation. (omitted)

Preliminary Characterization of Keratinolytic Enzyme of Aspergillus flavus K-03 and Its Potential in Biodegradation of Keratin Wastes

  • Aspergillus flavus K-03 isolated from poultry forming soil in Korea was studied for its ability to produce extracellular proteases on basal medium containing 2%(w/v) chicken feathers. The fungus was observed to be a potent producer of such enzymes. Keratinolytic enzyme secretion was the best at 15 days of incubation period at pH 9 and temperature $40^{\circ}C$. No relationship existed between the enzyme yield and increase of biomass. Enzyme production was suppressed by exogenous sugars in descending order arabinose>maltose>mannose>fructose. But glucose did not influence the enzyme activity. The keratinolytic enzyme released by the fungus demonstrated the ability to decompose keratin substrates as chicken feather when exogenous glucose was present. The keratinolytic activity was inhibited by $HgCl_2$ and serine-protease inhibitors such as phenymethylsulfonyl fluoride(100%), chymostain(88%), crystalline soybean trypsin inhibtor(80%), antipain(45%) and aprotinin(40%), and was not by cystein-protease and aspartyl-protease inhibitors. The enzyme activity is only partially inhibited by metallo-protease inhibitor. Thus, the enzyme secreted by A. flavus K-03 belongs to the alkaline serine-type protease.

A Comparative Study on the Aerobic Biodegradation of the Continuous and Intermittent Aeration in Bin Composting System

  • Composting of hog manure amended with sawdust trials lasted three weeks and used pilot-scale in bin composting system. Results showed that the rise temperature and carbon dioxide evolution in compost during the composting decomposition process were affected by the aeration method, pH, C/N, moisture content, bulk density and particle size distribution. No significant differences existed in biophysical properties of the composit produced from the continuous and intermittent aeration method. The intermittent aeration was very successful in compost odor control and required less time to reach stability than the continuous aeration.

  • This study was performed to find the recycling performance and to provide design data during recycle solid composting system to reduce bulking agent cost, Dairy manure amended with rice hulls and recycled compost was composted in a laboratory -scale vessel by continuous aeration for 10 days. The temperature and ammonia emission variations according to the ratios of bulking agents during the primary aeration stage were surveyed. Also, the influence of fresh compost quality on the recycling performance were anlayzed. While recycled composting system were operatured. The temperature in compost was maintained in the range of 40∼60$^{\circ}C$ needed for biodegradation and death of pathogenic organisms, but the ammonia emission was different by the ratios of recycled compost. The ammonia emission increased due to the low C/N ratio(17.6), high pH value(8.1) of the recycled compost.

  • Chitosan and Algin are known as a natural polymers. Biodegradable films were prepared by solution blend method in the weight ratio of natural polymer(low, medium, high-Chitosan, Algin) for the purpose of useful bioimplants. The possibility of bioimplants, which were prepared from natural polymers as a skin substitute and food wrapping materials were evaluated by measuring biodegradability. This biodegradable films were inserted in the back of rats and their biodegradability was investigated by hematological change evaluation as a function of time to biotransformation. It was found that these values of biodegradable films give some good results with short period test.

Improvement of the Functional Qualities of Sea Tangle Extract through Fermentation by Aspergillus oryzae

  • This study was conducted to evaluate the potential of a microbial fermentation procedure to improve the functional qualities of seaweeds. Aspergillus oryzae, which has been used in traditional Korean fermented foods, was inoculated and cultivated in an aqueous extract of sea tangle (Laminaria japonica). Fermentation of the sea tangle extract by A. oryzae for 4 days resulted in a 3-fold increase in $\gamma$-aminobutyric acid (GABA) content. GABA is known to be a bioactive compound. Fungal fermentation of the extract also enhanced its antioxidant activity and increased its total content of phenolic compounds. It was assumed that these changes stemmed from the biodegradation of active compounds of the sea tangle packaged within its rigid structural matrix or occurred as result of fungal fermentation. These results suggested that the application of microbial fermentation to the processing of seaweeds will help in the development of processed foods to meet consumer demands.
