The Regulation of AP-1 DNA Binding Activity by Long-term Nicotine Stimulation in Bovine Adrenal Medullary Chromaffin Cells: Role of Second Messengers

  • The signal pathways involved in the regulation of AP-1 DNA binding activity in long-term nicotine stimulated bovine adrenal medullary chromaffin (BAMC) cells have not been well characterized. To understand the involvement of second messengers in the regulation of AP-1 DNA binding activity, the present study was designed to define the time-course for inhibition of nicotine-induced responses by cholinergic antagonists, $Ca^{2+}$ and calmodulin (CaM) antagonists, and calcium/calmodulin-dependent protein kinase (CaMK) II inhibitor using electrophoretic mobility shift assay. Nicotine $(10{\mu}M)$ stimulation increased AP-1 DNA binding activity at 24 hr after treatment. Posttreatment with hexamethonium (1 mM) plus atropine $(1{\mu}M)$ (HA), nimodipine $(1{\mu}M),$ or calmidazolium $(1{\mu}M)$ at 0.5, 3, and 6 hr after the nicotine treatment significantly inhibited the AP-1 DNA binding activity increased by long-term nicotine stimulation. However, posttreatment with HA, nimodipine, or calmidazolium at 9 or 12 hr after the nicotine treatment did not affect the nicotine-induced increase of AP-1 DNA binding activity. The pretreatment of BAMC cells with various concentrations of KN-62 inhibited the increase of AP-1 DNA binding activity induced by nicotine in a concentration-dependent manner. KN-62 $(10{\mu}M)$ posttreatment beginning at 0.5, 3, or 6 hr after the nicotine treatment significantly inhibited the increase of AP-1 DNA binding activity. However, KN-62 posttreatment beginning at 9 or 12 hr after the nicotine treatment did not affect the increase of AP-1 DNA binding activity. This study suggested that stimulation (for at least 6 hr) of nicotinic receptors on BAMC cells was necessary for increase of AP-1 DNA binding activity, and activation of $Ca^{2+},$ CaM, and CaMK II up to 6 hr at least seemed to be required for the increase of nicotine-induced AP-1 DNA binding activity.

Effects of Dopamine and Haloperidol on Morphine-induced CREB and AP-1 DNA Binding Activities in Differentiated SH-SY5Y Human Neuroblastoma Cells

  • In the present study, we first examined whether the changes in the DNA binding activities of the transcription factors, cAMP response element binding protein (CREB) and activator protein-1 (AP-1) mediate the long-term effects of morphine in differentiated SH-SY5Y human neuroblastoma cells. The increases in CREB and AP-1 DNA binding activities were time-dependent up to 6 days of morphine treatment (1, 4, and 6 days). However, the significant reduction in the DNA binding activities of CREB and AP-1 was observed after 10 days of chronic morphine $(10\;{\mu}M)$ administration. Secondly, we examined whether the changes of CREB and AP-1 DNA binding activities could be modulated by dopamine and haloperidol. Dopamine cotreatment moderately increased the levels of the CREB and AP-1 DNA binding activities induced by 10 days of chronic morphine treatment, and haloperidol cotreatment also resulted in a moderate increase of the CREB and AP-1 DNA binding activities. However, dopamine or haloperidol only treatment showed a significant increase or decrease of the CREB and AP-1 DNA binding activities, respectively. In the case of acute morphine treatment, the CREB and AP-1 DNA binding activities were shown to decrease in a time-dependent manner (30, 60, 90, and 120 min). Taken these together, in differentiated SH-SY5Y cells, morphine tolerance seems to involve simultaneous changes of the CREB and AP-1 DNA binding activities. Our data also suggest the possible involvement of haloperidol in prevention or reversal of morphine tolerance at the transcriptional level.

Effects of Brazilin on Glucose Metabolism in Epididymal Adipocytes from Streptozotocin induced Diabetic Rats

  • Hypoglycemic mechanism of brazilin was investigated in the streptozotocin induced diabetic rats. Plasma glucose levels of diabetic rats were significantly ame]iorated by the treatment of brazilin, but there were no changes in plasma insulin levels. Brazilin increased insulin binding capacity to epididymal adipocytes, and Scatchard analysis revealed that this increase in insulin binding was not due to the increase of insulin binding sites but that of binding affinity. 2-Deoxyglucose uptake of epididymal adipocytes was significantly augmented by the intraperitoneal administration of brazilin and the same result was obtained in in vitro study. Glucose oxidation and lipogenesis in epididymal adipocytes were significantly enhanced by the treatment of bracilin.

  • The modulatory effects of total ginseng saponin (TGS) on the 1, 6, and opioid receptor binding in morphine tolerance and dependence were examined in this study. The specific [$^{3}H$]DAGO ([D-$Ala^2$, N-$Mephe^4$, $Glyco^4$] enkephalin) binding was significantly increased in chronic morphine (10 mg/kg, i.p.) treated mouse striatum. The specific [$^{3}H$]DPDPE ([D-$Pen^2$, D-$Pen^5$] enkephalin) binding was ignificantly increased following morphine treatment in the mouse striatum and cortex. Also, an apparent decrease in the affinity of [$^{3}H$]DPN (diprenorphine) was observed after chronic morphine treatment in mouse striatum and cortex. 7GS produced a sleight increase of specific [$^{3}H$]DAGO, [$^{3}H$]DPDPE binding and a significant increase of specific [$^{3}H$]DPN binding in the mouse brain striatum. In cortex, TGS produced an inhibition of specific [$^{3}H$]DAGO and [$^{3}H$]DPDPE binding and increase of the specific [$^{3}H$]DPN binding. The prolonged administration of TGS (25, 50, 100, and 150 mg/kg, i.p., 3 wks) produced an inhibition of increased [$^{3}H$]DAGO specific binding following morphine without significant changes in the agonist binding to and receptors in mouse striatum and cortex. These contracted alterations in $\mu$, $\gamma$ and $\kappa$ opiate receptor binding were dependent in TGS dogs and brain sites.

Participation of SRE4, an URE1 Enhancer Core Sequence, in the Sterol-Mediated Transcriptional Upregulation of the Human Apolipoprotein E Gene

  • The expression of the endogenous human apolipoprotein(apo)E gene was significantly induced when HepG2 cells were treated with exogenous 25-hydroxy-cholesterol. This sterol-mediated apoE gene upregulation appears to require the participation of a positive element for the apoE gene transcription (PET) ( -169/ -140), a core sequence of upstream regulatory element (URE)1 enhancer of the human apoE gene. This PET was renamed as sterol regulatory element (SRE)4 based on its new role as a sensor for the level of intracellular sterol. Furthermore, a gel mobility shift analysis showed that binding activity of the SRE4 binding protein (BP) obtained from HepG2 cells was induced by sterol treatment, while that from either MCF7 or BT20 cells remained unchanged. Binding activity of SRE4BP was also induced in mouse macrophage cells, J774A.1, by sterol treatment, but it was drastically reduced when cells were subjected to treatment of AY-9944, a potent inhibitor for sterol synthesis. However, binding activity of Spl, which is a co-binding protein to the SRE4 region, remained the same in either condition, suggesting that SRE4BP (formally known as PETBP) may be mainly responsible for the sterol-mediated regulation of the apoE gene expression. Deletion analysis of the core binding site of SRE4BP by gel mobility shift assays showed that the minimal sequence of the SRE4BP binding appears to reside between -157 and -140, confirming the identity of SRE4 with the previously determined core sequence of URE1.

Effects of Calcium Powder Mixtures and Binding Ingredients as Substitutes for Synthetic Phosphate on the Quality Properties of Ground Pork Products

  • This study aimed to investigate the combined effect of using natural calcium mixtures and various binding ingredients as replacers for synthetic phosphate in ground pork products. We performed seven treatments: control (0.3% phosphate blend), treatment 1 (0.5% natural calcium mixtures [NCM, which comprised 0.2% oyster shell calcium and 0.3% egg shell calcium powder] and 0.25% egg white powder), treatment 2 (0.5% NCM and 0.25% whey protein concentrate), treatment 3 (0.5% NCM and 0.25% concentrated soybean protein), treatment 4 (0.5% NCM and 0.25% isolated soybean protein), treatment 5 (0.5% NCM and 0.25% carrageenan), and treatment 6 (0.5% NCM and 0.25% collagen powder). All the treatment mixtures had higher pH and lower cooking loss than the control, which was treated with phosphate. We found that NCM and binding ingredients had no negative effects on the moisture content, lightness, and yellowness of the cooked ground pork products. Treatments 3 and 4 showed significantly lower CIE $a^*$ values than the control. Treatments 2 and 6 improved the textural properties of the products. In conclusion, the combination of NCM with whey protein concentrate or collagen powder could be suitable for producing phosphate-free meat products.

이소플라본 보충이 폐경 후 여성에서 골밀도와 성호르몬에 미치는 영향 (Effects of Isoflavones Supplementation on Bone Mineral Density and Sex Hormones in Postmenopausal Women)

  • Soy isoflavones have been hypothesized to exert hormonal effects and to enhance bone mineral density in postmenopausal women. To test this hypothesis, we studied the effects of soy isoflavones supplements on bone mineral density and sex hormones (serum estradiol, sex hormone-binding globulin and testosterone) in 47 postmenopausal women. There were 24 participants in the treatment group and 23 in the control group. The treatment group consumed the isoftavones extract capsule daily (which contained 90 mg of soy isoflavones) for 12 weeks. The study compared pre- and post-isoflavones intake in the following areas: physical examination, diet survey, bone mineral density and serum sex hormone levels. The average age of the treatment group was 64.63 years and that of the control group was 66.48 years. There were no significant differences between the two groups in terms of height, weight, and body mass index. Both groups maintained regular diet patterns in terms of their average daily nutrient intake. There was no significant difference between the treatment group (18.49 mg) and the control group (21.27 mg) in terms of daily isoflavones intake based on diet. The 12-week analysis of bone mineral density change after taking isoflavones supplements demonstrated no significant differences in the following : lumbar spine BMD (0.82 g/$\textrm{cm}^2$ in pre versus 0.81 g/$\textrm{cm}^2$ in post), femoral neck BMD (0.58 g/$\textrm{cm}^2$ in pre versus 0.57 g/$\textrm{cm}^2$ in post) in the treatment group. There was no significant difference in serum estradiol in the isoflavones treatment group. The subjects indicated no significant difference in serum testosterone in the isoflavones treatment group. But the subjects indicated a significant difference in sex hormone-binding globulin (60.04 nmol/L in pre versus 52.39 nmol/L in post) in the isoflavones treatment group at the levels of p < 0.05. The significant decrease in sex hormone-binding globulin did indicate the need for long-term study on isoflavones supplementation as well as its positive effect on bone mineral density.

Solubilization of an Angiotensin II Binding Site from Rat Liver

  • The high affinity binding sites for angiotensin II were solubilized from rat liver membranes by treatment with CHAPS. The binding protein was also partially purified by angiotensin III inhibitor-coupled Affi-gel affinity chromatography. Binding to the intact membrances as well as to the solubilized preparation was specific and saturable. According to the Scatchard plot, the membrane preparations exhibited a single class of high affinity binding sites with a Kd OF 0.71 nM. The solubilized preparation also showed the presence of a single class of bindings sites with less affinity (Kd of 14 nM). Meanwhile the competition studies using angiotensin II analogues represented two separate binding sites for angiotensin II and single binding site for antagonist. These latter findings were correlated to the results provided by Garrison's research group. More works are needed to clarify this discrepancy.

마우스 선조체에서 Opioid 수용체 결합에 대한 Neuroleptics의 영향 (Effects of Neuroleptics on the Opioid Receptor Binding in the Mouse Striatum)

  • 이 연구에서는 선조체에서 opioid 신경계와 dopamine 신경계의 상호 관계를 알아보기 위해서 morphine을 5m/kg, 20 mg/kg로 10일간 복강내 투여한 후 chlorpromazine, thioridazine, haloperidol, sulpiride, pimozide를 투여하였다. Opioid ${\mu},\;{\delta},\;{\kappa}$ 수용체의 binding의 변화를 관찰하고자 $[^3H]\;DAGO$, $[^3H]\;DPDPE$, 및 $[^3H]\;DPN$ binding assay를 하였으며, 그 결과 morphine (20 mg/kg) 장기 투여된 실험군에서 $[^3H]\;DAGO$, $[^3H]\;DPDPE$, 및 $[^3H]\;DPN$ 결합이 감소되었다. Morphine 20 mg/kg 장기 투여군에 chlorpromazine, thioridazine 주사시에는 morphine 5mg/kg 투여군에 비하여 $[^3H]\;DAGO$ 결합의 감소와, $[^3H]\;DPDPE$, 및 $[^3H]\;DPN$ 결합의 증가를 나타내었고, haloperidol 주사군은 $[^3H]\;DAGO$, $[^3H]\;DPN$ 결합의 감소, 및 $[^3H]\;DPDPE$ 결합의 증가를 나타내었다. Sulpiride, pimozide 주사군은 morphine 5 m/kg 투여군에 비하여 20m/kg 투여군에서 $[^3H]\;DAGO$, $[^3H]\;DPDPE$, 및 $[^3H]\;DPN$ 결합의 증가를 나타내었다. 이상의 결과로 보아 각 약물간의 opioid 결합에 대한 차이점은 있었으나, morphine 5mg/kg 투여군보다 20m/kg 투여군에서 $[^3H]\;DPDPE$$[^3H]\;DPN$의 결합이 증가의 경향을 보임으로써, 다량의 morphine을 투여했을 때 ${\mu}\;opioid$ 수용체에 비하여 ${\delta}${\kappa}\;opioid$ 수용체가 더 활성화되는 것을 알 수 있었다.

Modulation of Phytotropin Receptors by Fluoride and ATP

  • Treatment of microsomal vesicles isolated from etiolated Pisum sativum L cv. Alaska epicotyl tissue with agents inhibiting protein dephosphorylation, namely NaF and/or ATP, resulted in increased binding of the phytotropin NPA to the putative auxin efflux carriers localized on the plasma membrane. The phytotropin effect was especially conspicuous if the vesicles were simultaneously treated with Triton X-100. Kinetic analysis of the binding indicated the existance of two distinct sites for NPA, each having different affinities. Increased binding of the phytotropin to the membrane where protein dephosphorylation was inhibited was attributable to the increased ligand affinity of both sites. Treatment of tissue segments with flubride was found to enhance in vivo auxin transport. Implications of covalent modification of the auxin efflux carrier complex for the regulation of membrane transport of auxin molecules are discussed.
