추출조건에 따른 다시마 alginate의 성질을 조사하기 위하여 추출용매인 $Na_{2}CO_{3}$의 농도를 1, 3, 및 $5\%$로 각각 조절하고 추출시간을 각각 1, 3, 5 및 10시간으로 하였을때 각 추출조건에서 추출된 alginate의 bile acids 결합능과 물리화학적 특성을 조사한 결과는 다음과 같다. 건조다시마를 미세하게 마쇄할수록, $Na_{2}CO_{3}$ 농도와 추출시간이 증가할수록 alginate 추출량은 증가하였다. alginate의 bile acids와의 결합능은 저 농도의 $Na_{2}CO_{3}$ 용액으로 단시간에 추출한 것일수록 높게 나타났다. alginate에 의한 bile acids 결합능은 cholic acid에서 가장 높게 나타났고, taurocholic acid, glycocholic acid, deoxycholic acid 순으로 조사되었다. 다시마로 부터 alginate를 추출하고 남은 잔사(RAEA)의 bile acids와의 결합능은 alginate의 경우와 같이 추출 용액의 농도가 높을수록 추출시간이 길어질수록 감소하는 경향을 보였다. Bile acids와의 결합능은 cholic acid, taurocholic acid의 경우 alginate보다 다소 낮은 값을 나타냈고, glycocholic acid의 경우 alginate와 비슷한 값을 보였으나 deoxycholic acid와의 결합능은 추출된 alginate보다도 오히려 높은 값을 보였다. 추출된 alginate의 점도와 중합도는 추출용액의 농도가 증가하고 추출시간이 길어짐에 따라 감소하였으며 이에 비례하여 bile acid와의 결합능도 감소하였다.
Bile acids are major constituents of bile and known to help absorb dietary fat and fat-soluble vitamins in the gastrointestinal tract. In the past few decades, many studies have shown that bile acids not only play a role in fat digestion but also function as broad range of signal transduction hormones by binding to various receptors present in cell membranes or nuclei. Bile acid receptors are distributed in a wide range of organs and tissues in the human body. They perform multitudes of physiological functions with complex mechanisms. When bile acids bind to their receptors, they regulate fat and glucose metabolism in a tissue-specific way. In addition, bile acids are shown to inhibit inflammation and fibrosis in the liver. Considering the roles of bile acids as metabolic regulators, bile acids and their receptors can be very attractive targets in treating metabolic disorders. In the future, if roles of bile acids and their receptors are further clarified, they will be the novel target of drugs in the treatment of various metabolic diseases.
The effects of the prevention of micelle formation and bile acid reabsorption, by using cholestyramine (CHOLN), a bile acid binding polymer, on the plasma lipid of Single Comb White Leghorn male chicks given diets containing medium chain triacylglycerol (MCT) and long chain triacylglycerol (LCT) were investigated. Corn oil and glyceryl tricaprylate were used as LCT and MCT sources, respectively. Plasma HDL cholesterol was reduced by CHOLN in all treatments. Plasma LDL cholesterol was reduced by CHOLN in chicks given LCT diet but not in MCT diet which could be accounted to the reduced plasma total cholesterol in LCT diet with CHOLN. It is concluded that bile acid binding does not alter the cholesteremic effect of MCT in the plasma of chicks.
The aim of this experiment was to compare the characterization of fatty acid digestion of Beijing Fatty (BF) and Arbor Acres (AA) chickens. One-day-old male AA and BF chickens were raised in the same house, and fed with the same diet. We first evaluated utilization of dietary fatty acids in chickens by the total collection procedure, and chickens were then killed to compare the abundance of intestinal mRNA expression of liver-fatty acid binding protein (L-FABP) and intestinal-fatty acid binding protein (I-FABP) by Real-time PCR, and also the pH of intestinal mucosa at 3 and 6 weeks of age. Another group of chickens were sampled at 6 weeks of age to compare the total bile acid concentration in serum, and lipase activity in contents of the small intestine. Results showed that compared to AA chickens, BF chickens had higher lipase activity in the content of the small intestine (p<0.05), greater total bile acid content in portal vein blood (p<0.05) at 6 weeks of age, lower intestinal mucosal pH at both 3 weeks (p<0.05) and 6 weeks (p<0.05) of age, and higher abundance of liver-fatty acid binding protein (L-FABP) mRNA expression in intestine tissues at 6 weeks of age (p<0.05), and higher digestibility of fatty acids at both 3 and 6 weeks (p<0.05) of age. There was no difference in I-FABP mRNA expression between AA and BF chickens at either age. Thus, BF chickens had greater fatty acids utilization than AA chickens that was associated with L-FABP, lipase activity, bile acid content and intestinal mucosal pH.

The fermented milk products containing that kind of lactic acid bacteria have to be recommended for human health as excellent health functional foods. This paper have been classified by 5 regions on the functions of lactic acid bacteria related to prevention of colon cancer. 1) Enhancing of host's immune response; Production of cytokines. 2) Binding and degradation of potential carcinogens; Binding and degradation of mutagenicity. 3) The changes of intestinal microflora and production of antitumorigenic or antimutagenic compounds; Production of azoxymethane. 4) Alteration of the metabolic activity of intestinal microflora; Decrease of harmful enzymes in intestinal tract. 5) Alteration of physicochemical conditions in the colon; Decrease of pH and bile acids contents.
미역과 큰 다시마 알긴산의 염산 부분가수분해에 의한 저분자화 조건을 구명하였으며, 저분자화가 알긴산의 점성, 용해도, 유화능, 흡유능, 담즙산결합능 및 금속이온결합능 등의 물성에 미치는 영향에 관하여 검토하였다. 미역에서 추출한 알긴산을 시료로 하여 저분자화의 조건을 검토한 결과, 염산에 의한 반응시간의 연장 및 염산의 농도증가와 더불어 구성 polyuronate의 중합도는 급격히 감소하였다. 그리고 0.3N HCl 부분가수분해에 의하여 분자량을 1/100로 감소시키는데 소요되는 반응시간은 약 50분이었고, 이 때 각각의 수율은 약 $75\%\~80\%$ 이었다. 각 알긴산의 M/G의 비는 저분자화에 의하여 증대하였으며, 알긴산 용액의 점성은 저분자화하므로서 급격히 저하하였고, 알긴산의 점도와 분자량은 밀접한 상관관계를 보였다($r^2=0.9890$). 온도단계별 ($5\~40^{\circ}C$)로 측정한 알긴산의 용해도는 수용성 알긴산이 산$\cdot$알칼리가용성 알긴산에 비하여 $7\~12\%$ 높았으며, 저분자화와 더불어 상승하였다. 한편, 저분자화는 유화능의 증가를 가져왔으며, 용해도와는 비슷한 관계를 보였다. 그러나 흡유능과 금속이온결합능은 저분자화에 의하여 감소하였으며, 특히 금속이온결합능에 있어서는 미역 알긴산은 $Pb^{2+},\;Cu^{2+},\;Zn^{2+},\;Co^{2+}$의 순으로, 그리고 큰 다시마 알긴산은 $Cu^{2+},\;Pb^{2+},\;Zn^{2+},\;Co^{2+}$의 순으로 각각 감소하였다. 담즙산결합능은 저분자화와 더불어 오히려 증가하는 특징을 보였다.
본 연구에서는 어린감과 성숙감의 탄닌 함량 및 담즙산과의 결합능력을 비교 조사하였다. 또한 사람의 장내 세포인 Caco-2 cell에 어린감과 성숙감 추출물을 처리한 후 고농도의 DCA로 자극시켜 생성되는 NO의 수준과 TEER의 차이를 확인하였다. 탄닌 함량과 담즙산의 결합능력은 성숙감에 비해 어린감에서 유의적으로 높은 것으로 나타났으며 어린감 산성 에탄올 추출물에서 유의적으로 가장 높은 탄닌 함량 및 담즙산 결합능력을 보였다. NO 생성 억제능으로 확인한 염증반응에서도 어린감 산성 에탄올 추출물이 유의적으로 가장 높은 효과를 보였으며 DCA에 의해 유도된 장세포 간극의 느슨함을 유의하게 막아줄 수 있음이 확인되었다. 본 연구 결과를 토대로 어린감의 산성 에탄올 추출물은 고지방 식사를 통해 고농도로 축적되는 담즙산에 의해 형성된 유해한 장내 환경을 건강하게 유지시켜줄 수 있는 기능성 소재로 개발할 가능성이 있음이 확인되었다.
우리나라 주요 사망원인중의 하나가 되고 있는 혈관질환의 주된 위험인자로 고콜레스테롤 혈증이 지적되고 있다. 콜레스테롤 저하효과가 있는 식이 인자에는 단백질의 종류, 지방의 종류와 양, 식이섬유, 우유, 칼슘, flavonoid 등이 있다. 특히 수용성 식이 섬유의 콜레스테롤 저하효과는 현저하며 동물실험과 인체실험을 통하여 확인되었다. 혈장 콜레스테롤 저하효과가 현저한 식이섬유에는 psyllium husk, pectin, oat fiber, guar gum, sodium alginate 등이 있으며, 농도에 비례하여 그 효과가 커지는 것은 아닌듯하다. 수용성 식이섬유의 콜레스테롤 저하효과에 대한 정확한 기작은 알려져 있지 않으나 몇 가지 가설이 제시되고 있다. 첫째, 식이섬유가 장에서 점성용액을 형성하여 지질 흡수를 저해함으로써 혈장과 간 콜레스테롤 농도를 낮춘다는 것이며, 둘째, 체내 콜레스테롤이 체외로 배설되는 유일한 경로인 담즙산의 소장흡수를 방해하고 변으로의 배설을 증가시켜 체내 콜레스테롤 pool 크기를 감소시킨다는 것이다. 세째, 대장미생물에 의해 생성된 식이섬유의 발효부산물인 short chain fatty acids가 콜레스테롤의 합성을 저해한다는 것이다. 이외에도 여러 가설들이 제시되고 있으며, 이러한 가설들이 서로 상호 배제적인 것은 아니며, 단독으로 혹은 복합적으로 작용하는 것으로 보인다.
Cardiovascular diseases (CVDs) are the most common cause of death in patients with nonalcoholic fatty liver disease (NAFLD) and dyslipidemia is considered at least partially responsible for the increased CVD risk in NAFLD patients. The aim of the present study is to understand how hepatic de novo lipogenesis influences hepatic cholesterol content as well as its effects on the plasma lipid levels. Hepatic lipogenesis was induced in mice by feeding a fat-free/high-sucrose (FF/HS) diet and the metabolic pathways associated with cholesterol were then analyzed. Both liver triglyceride and cholesterol contents were significantly increased in mice fed an FF/HS diet. Activation of fatty acid synthesis driven by the activation of sterol regulatory element binding protein (SREBP)-1c resulted in the increased liver triglycerides. The augmented cholesterol content in the liver could not be explained by an increased cholesterol synthesis, which was decreased by the FF/HS diet. HMG-CoA reductase protein level was decreased in mice fed an FF/HS diet. We found that the liver retained more cholesterol through a reduced excretion of bile acids, a reduced fecal cholesterol excretion, and an increased cholesterol uptake from plasma lipoproteins. Very low-density lipoproteintriglyceride and -cholesterol secretion were increased in mice fed an FF/HS diet, which led to hypertriglyceridemia and hypercholesterolemia in Ldlr-/- mice, a model that exhibits a more human like lipoprotein profile. These findings suggest that dietary cholesterol intake and cholesterol synthesis rates cannot only explain the hypercholesterolemia associated with NAFLD, and that the control of fatty acid synthesis should be considered for the management of dyslipidemia.
유산균은 일반적으로 프로바이오틱스 균주로 알려져 있다. 가능성이 있는 프로바이오틱스 균주는 세포결합과 부착능 즉, 장상피세포 부착능 및 세포표면의 소수성이 기초적으로 구비되어야 한다. 본 연구에서는 김치로부터 몇몇의 토착 유산균을 분리하였으며, 세포표면 소수성에 근거한 가능성이 있는 프로바이오틱스로서 유산균 한종을 선발하였다. 프로바이오틱스균주로서 선별한 분리균주(BCNU 9070)의 16S 리보좀DNA 염기서열을 분석한 결과 Pediococcus pentosaceus에 속하는 균주임이 확인되었다. P. pentosaceus BCNU 9070 균주는 위액과 담즙산에 대하여 내성을 가졌으며 또한 Listeria monocytogenes 및 Shigella sonnei를 포함하는 6종의 식중독 병원균에 대하여 생육저해활성도 나타내었다. 게다가 P. pentosaceus BCNU 9070 균주는 담즙산 가수분해활성 및 콜레스테롤 동화능도 있음이 확인되었다. 이들 결과를 기초로, P. pentosaceus BCNU 9070은 기능성 식품에 적용가능한 천연 프로바이오틱스 특성을 가지고 있다고 판단된다.
