  • In this study, the effect of different reaction times for thermal-alkaline pretreatment on the solubilization and biogasification of polyhydroxybutyrate (PHB) were evaluated. Thermal-alkaline pretreatment tests were performed at 73 ℃ and pH 13 at 0-120 h reaction times. The mesophilic anaerobic batch tests were performed with untreated and pretreated PHB samples. The increase in the pretreatment reaction time results in a 52.8-98.8% increase of the abiotic solubilization efficiency of the PHB samples. The reaction time required to achieve solubilization efficiencies of 50%, 90%, and 95% were 10.5, 52.0, and 89.6 h, respectively. The biogasification of the untreated PHB samples achieved a specific methane production rate of 3.6 mL CH4/g VSS/d and require 101.3 d for complete biogasification. The thermal-alkaline pretreatment significantly improved specific methane production rate (10.2-16.0 time increase), lag time (shortened by 76-81%), and time for complete biogasification (shortened by 21-83%) for the biogasification of the PHB samples when compared to those of the untreated PHB samples. The improvement was higher as the reaction time of the thermal-alkaline pretreatment increased. The findings of this study could be used as a valuable reference for the optimization of the biogasification process in the treatment of PHB wastes.

  • The effects of free ammonia (FA) concentration and temperature on nitrite accumulation were studied. To estimate the most effective ammonium oxidation and nitrite build-up condition, nitrification tests were conducted in batch conditions at various FA concentrations, and at different ammonium concentration and temperature. The activation energies of ammonium oxidizer were 81.7 KJ/mol below $20^{\circ}C$, and 32.5 KJ/mol over $20^{\circ}C$, while that of nitrite oxidizer was 35.5 KJ/mol irrespective of temperature variations. The results of nitrification tests conducted at different FA concentrations and temperatures showed that temperature strongly affects nitrite accumulation, while effects due to FA concentrations were found negligible.

  • Compaction of backfill material of Underground power lines is difficult, especially under pipeline. so it could cause structural problem because of low compaction efficiency. So various methods have been taken to solve the problem and one of them is CLSM(Controled low-strength material accelerated flow ability). But In other countries, these are already in progress for a long time to research and development and recently on practical steps. But, in our country, study for only general structures, not for underground power line structure that is being constructed at night rapidly. In this study, we performed property tests and indoor & outdoor test (3 cases). The tests showed flow ability reached at the limit construction(160 mm) flowability by 9 to 15 minute after starting to mix, and construction buoyant is lowering after placing CLSM by 70 % of theoretical buoyant that is calculated by unit weight of material. In this paper, we performed indoor tests and outdoor tests to estimate mechanical properties and to suggest construction method(using batch plant, setting spacer at 1.8 m and placing at 2m) for CLSM that using surplus soil. And the test showed good results for construction quality, workability and structure safety.

  • In this research, the decolorization mechanisms of dye wastewater were divided into two pathways, one was physicochemical sorption to biomass flocs and the other was biological removal by microbial metabolisms. Batch tests were conducted to examine the reaction conditions, anaerobic and aerobic conditions, types and dose of cosubstrates, and to confirm the mechanisms of decolorization through the biosorption tests using the activated sludge and the autoclaved deactivated sludge. From the tests, the decolorization efficiencies of dye wastewater were 102 ${\Delta}$unit/g MLSS under the aerobic condition and 123 ${\Delta}$unit/g MLSS under the anaerobic condition, and organic removals were 82 $mg{\Delta}$COD/gMLSS and 75 $mg{\Delta}$COD/gMLSS respectively. Acetate was the more efficient cosubstrate than the domestic wastewater in the decolorization step. In addition the removal of colors and organics was increased with cosubstrates dosage. And $20.3{\sim}37.3$ ${\Delta}$unit/g MLSS was removed by the autoclaved sludge and $102.0{\sim}159.0$ ${\Delta}$unit/g MLSS by the activated sludge. The physicochemical sorption was dominant in the beginning of biosorption tests, and the biological decolorization was increased with a cosubstrate in the course of time.

  • The biodegradation rates of diesel oil by a selected diesel-degrading bacterium, Pseudomonas stutzeri strain Y2G1, and microbial consortia composed of combinations of 5 selected diesel-degrading bacterial were determined in liquid and soil systems. The diesel degradation rate by strain Y2G1 linearly increased $(R^2=0.98)$ as the diesel concentration increased up to 12%, and a degradation rate as high as 5.64 g/l/day was obtained. The diesel degradation by strain Y2G1 was significantly affected by several environmental factors, and the optimal conditions for pH, temperature, and moisture content were at pH8, $25^{\circ}C$, and 10%, respectively. In the batch soil microcosm tests, inoculation, especially in the form of a consortium, and the addition of nutrients both significantly enhanced the diesel degradation by a factor of 1.5 and 4, respectively. Aeration of the soil columns effectively accelerated the diesel degradation, and the initial degradation rate was obviously stimulated with the addition of inorganic nutrients. Based on these results, it was concluded that the major rate-limiting factors in the tested diesel-contaminated soil were the presence of inorganic nutrients, oxygen, and diesel-degrading microorganisms. To resolve these limiting parameters, bioremediation strategies were specifically designed for the tested soil, and the successful mitigation of the limiting parameters resulted in an enhancement of the bioremediation efficiency by a factor of 11.

  • The influence of bacterial stress on anaerobic hydrogen-producing microorganisms was investigated in batch tests using serum bottles. Several physical and chemical stresses (i.e., heating, adding methane producing inhibitor and chemical acidification) were adapted as a pretreament of the seed sludge. In this experiment, the cultivation temperature were set at mesophilic ($35^{\circ}C$) and thermophilic conditions ($55^{\circ}C$) with adjusting pH at 5, 6, and 7 when using the mixture of food waste and activated sludge as a substrate. In conjunction with the pretreatment, hydrogen production was significantly enhanced as compared with that from untreated sludge. However, less biogas (hydrogen and methane) was produced without the pH control, resulted from the decrease of pH to below 4, mainly due to the formation of VFAs. Hydrogen and carbon dioxide gas were analyzed as main components of the biogas while methane not detected. With an application of chemical acidification, the highest hydrogen production value of 248 ml/l/day achieved at pH 7 and $35^{\circ}C$. In addition, more hydrogen gas produced when the ratio of butyric/acetic acid ratio increased. The optimum pH and temperature for hydrogen production were found to be 7 and $35^{\circ}C$, respectively.

  • Nine newly isolated mushroom strains were tested to assess both their zinc tolerance and potential for zinc removal from an aqueous solution. Four strains of ectomycorrhizal fungi, namely Clavariadelphus truncatus (T 192), Rhizopogon roseolus (T 21), Lepista nuda (T 373), and Tricholoma equestre (T 174), along with five strains of white rot fungi, Lenzites betulina (S 2), Trametes hirsuta (T 587), Ganoderma spp. (T 99), Polyporus arcularius (T 438), and Ganoderma carnosum (M 88), were investigated using zinc-amended solid and liquid media. Their biosorption properties were also determined. The colony diameter and dry weight were used as tolerance indices for fungal growth. C. truncatus and T. equestre were not strongly inhibited at the highest concentrations of (225 mg/l) zinc in solid media. The most tolerant four strains with solid media, C. truncatus, G carnosum, T. hirsuta, and T. equestre, were then chosen for tolerance tests in liquid media. An ectomycorrhizal strain, C. truncatus, was also detected as the most tolerant strain in liquid media. However, the metal-tolerant strains demonstrated weak activity in the biosorption studies. In contrast, the highest biosorption activity was presented by a more sensitive strain, G. carnosum. In addition, seven different biosorbent types from G. carnosum (M 88) were compared for their Zn (II) biosorption in batch experiments.

  • Persulfate (PS) activated with nanosized zero-valent iron (NZVI) was tested as a reagent to remove phenol from groundwater. Batch degradation experiments indicated that NZVI/PS molar ratios between 1 : 2 and 1 : 5 were appropriate for complete removal of phenol, and that the time required for complete removal varied with different PS and NZVI dosages. Chloride ions up to 100 mM enhanced the phenol oxidation rate, and nitrate of any concentration up to 100 mM did not significantly affect the oxidation rate. NZVI showed greater performance than ferrous iron did as an activator for PS. A by-product was formed along with phenol degradation but subsequently was completely degraded, which showed the potential to attain mineralization with the NZVI/PS system. Tests with radical quenchers indicated that sulfate radicals were a predominant radical. The results of this study suggest that NZVI is a promising activator of PS for treating contaminated groundwater.

  • Among 31 water-born microbial strains isolated from various sites in Korea, strain DJ-4 was selected as a test organism for toxicity measurements in that its growth was completely inhibited by the presence of 668.4 mg/L of chloroform and 297.5 mg/L of toluene in the liquid LB medium whereas others did not. It was observed that lag periods and specific growth rates of DJ-4 batch vial cultures were prolonged and decreased, respectively, by phenol, benzene, toluene, ethylbenzene, p-xylene, perchloroethylene, trichloroethylene, and chloroform at the concentrations between 3.6 and 417.8 mg/L. There changes were found to be linear with respect to the concentrations of the toxic compounds. From the first-order regression equations, 50% effective concentrations (EC50${\mu}$ for concentrations of toxic compounds causing 50% decrease of specific growth rates and EC50lag for 50% increase of length of lag periods) were calculated for each compounds. By comparing DJ-4 EC50${\mu}$ values with Daphnia LC50's from a literature for benzene, ethylbenzene, toluene, and trichloroethlyene, it was concluded that microbial specific growth could be a new, fast, and reliable parameter for toxicity tests.

  • Batch tests were carried out to evaluate the thermal treatment of low volatile organic compounds in low-permeability soil. The chemical oxidation by sodium persulfate catalyzed by heat and Fe (II) was evaluated. Enhanced persulfate oxidation of n-decane (C-10), n-dodecane (C-12), n-tetradecane (C-14), n-hexadecane (C-16), and phenanthrene was observed with thermal catalyst, indicating increased sulfate radical production. Slight enhancement of the pollutants oxidation was observed when initial sodium persulfate concentration increased from 5 to 50 g/L. However, the removal efficiency greatly decreased as soil/water ratio increased. It indicates that mass transfer of the pollutants as well as the contact between the pollutants and sulfate radical were inhibited in the presence of solids. In addition, more pollutants can be adsorbed on soil particles and soil oxidant demand increased when soil/water ratio becomes higher. The oxidation of the pollutants was significantly improved when catalyzed by Fe(II). The sodium persulfate consumption increased at the same time because the residual Fe(II) acts as the sulfate radical scavenger.
