Ar-turmerone and $\beta-atlantone$ induce internucleosomal DNA fragmentation associated with programmed cell death in human myeloid leukemia HL-60 cells

  • In the course of a search for antitumor agents, we found that the extract of Curcuma longa was effective in inducing apoptosis or programmed cell death (PCD) in human myeloid leukemia cells (HL-60). Active compounds for PCD were isolated from the hexanic extraction of the rhizome of Curcuma longa. With the several chromatographies, and spectral data, they were identified as ar-turmerone and $\beta-atlantone$. The present results demonstrate that the exposure of human myeloid leukemia HL-60 cells to clinically achievable concentrations of arturmerone (TU) or .$\beta-atlantone$(AT) produced internucleosomal DNA fragmentation of approximately 200 base-pair multiples, and the morphological changes characteristic of cells undergoing apoptosis or PCD. This findings suggest that these agents may exert their antitumoral activity, in part, through induction of apoptosis(PCD).

  • Many highway bridges in Korea need seismic retrofit because only one decade has passed since the seismic design criteria was introduced. In this experimental study, the effectiveness of base isolation bearings was discussed for the seismic retrofit of the highway bridges. Four real scale RC pier specimens were constructed for the test. These RC piers didn't have seismic details. Except for one RC pier for the pilot test, three types of bearings such as Pot bearing, Rubber bearing (RB), Lead-rubber bearing (LRB) were applied to the other RC piers respectively. The RC pier with Pot bearing means current state of the prototype bridge that is not retrofitted seismically. And two RC piers with RB or LRB mean assumed states of the prototype bridge that are retrofitted seismically. To simulate dynamic behavior of these RC piers under earthquake loads, Pseudo-dynamic test method was used.

  • The seeds of Zizyphus Jujuba have been used as an antianxiety agent for the treatment of insomnia from the earliest times. Sanjoinine-A, isolated from the seeds of Zizyphus Jujuba, have been found to have a minor tranquilizer activity. However this drug is poorly soluble in water. In order to increase the dissolution rate of sanjoinine-A, solid dispersions with PVP-MC and inclusion complex with ${\beta}-cyclodextrin$ were prepared and evaluated. All of these systems increased the dissolution rate of sanjoinine A comparing with sanjoinine-A free base. From pH-rate profile of sanjoinine-A at $60^{\circ}C$, it was found that sanjoinine A was relatively stable in acidic solution, but unstable in basic solution.

  • The volatile aroma components were isolated from Korean flue-cured tobacco (N.C. 2326) by using a vacuum steam distillation method. Individual aroma components were identified by GCIMS and comparison of gas chromatowaphic retention time with those of the authentic samples. Sensory analysis showed that a vacuum steam-distilled product of Flue-cured tobacco had a typical haylike, floral and fruity aroma. Among 62 compounds identified, major compounds included neophytadiene, benzyl alcohol, ethyl acetate, phenyl ethyl alcohol, ethyl alcohol, ethyl formate, acetic acid, solanone, 2-acetyl pyrrole, $\beta$-ionone epoxide, 2, 4-heptadienal (2 isomers), megastigmatrienone (4 isomers), furfural and total amounts of 13 compounds were about 80%.

  • Near-field ground motions exhibit special characteristics that are different from ordinary far-field ground motions. In this study the shaking table tests were conducted to evaluate the effect of earthquake ground motions with different characteristics on the response of the structure. The ground motions used in this study were the scenario earthquake, design earthquake, and Chi-Chi earthquake measured in TCU052 station. These earthquakes have different frequency contents. The test results show that the frequency content of ground motion is very important to the response of structures. The floor responses of structure were greatly affected by the higher modal frequencies, as well as the fundamental frequency. The responses of third floor were significantly reduced due to the interaction between the structure and the base isolated mass installed at the third floor.

Blind modal identification of output-only non-proportionally-damped structures by time-frequency complex independent component analysis

  • Recently, a new output-only modal identification method based on time-frequency independent component analysis (ICA) has been developed by the authors and shown to be useful for even highly-damped structures. In many cases, it is of interest to identify the complex modes of structures with non-proportional damping. This study extends the time-frequency ICA based method to a complex ICA formulation for output-only modal identification of non-proportionally-damped structures. The connection is established between complex ICA model and the complex-valued modal expansion with sparse time-frequency representation, thereby blindly separating the measured structural responses into the complex mode matrix and complex-valued modal responses. Numerical simulation on a non-proportionally-damped system, laboratory experiment on a highly-damped three-story frame, and a real-world highly-damped base-isolated structure identification example demonstrate the capability of the time-frequency complex ICA method for identification of structures with complex modes in a straightforward and efficient manner.

Molecular characterization of Plasmodium juxtanucleare in Thai native fowls based on partial cytochrome C oxidase subunit I gene

  • Avian malaria is one of the most important general blood parasites of poultry in Southeast Asia. Plasmodium (P.) juxtanucleare causes avian malaria in wild and domestic fowl. This study aimed to identify and characterize the Plasmodium species infecting in Thai native fowl. Blood samples were collected for microscopic examination, followed by detection of the Plasmodium cox I gene by using PCR. Five of the 10 sampled fowl had the desired 588 base pair amplicons. Sequence analysis of the five amplicons indicated that the nucleotide and amino acid sequences were homologous to each other and were closely related (100% identity) to a P. juxtanucleare strain isolated in Japan (AB250415). Furthermore, the phylogenetic tree of the cox I gene showed that the P. juxtanucleare in this study were grouped together and clustered with the Japan strain. The presence of P. juxtanucleare described in this study is the first report of P. juxtanucleare in the Thai native fowl of Thailand.

Application of Hybrid Structural System Using Coupled Vibration Control Structure and Seismic Isolated Structure in High-Rise Building

  • This building is a forty-eight story, 170 meters high multiple dwelling house with Dual Frame System (DFS), a coupled vibration system connecting two independent structures with hydraulic dampers. Generation of large deformation between two structures during earthquakes contributes to make the hydraulic dampers effective. To improve the aseismic performance more, this building adopts DFS hybrid system that consists of DFS and base isolation system. About typical floors, columns and beams are constructed with LRV precast concrete method that shorten the construction period greatly by integrating column-beam joints in column members.

  • Application examples of computational fluid dynamics (CFD) in the planning stage of high-rise buildings are introduced. First, we introduce examples of applications in the environmental field. The pedestrian wind environment was one of the earliest practical examples of CFD. CFD was also employed to validate the heat island mitigation measures proposed as part of the new construction plan. Second, application examples of wind-force evaluations are introduced. Prediction examples are presented for the peak wind pressure around a complex-shaped building and the wind force evaluation for a base-isolated building. The results prove that the results of the proper execution of CFD are equivalent to those of the wind tunnel experiment. As examples of CFD applications of other issues related to high-rise building planning, we introduce snow accretion on outer walls and high-temperature exhaust from emergency generators. Finally, the future prospects for the use of CFD are discussed.

  • A new bacteriocin-producing lactic acid bacteria isolated from kimchi was identified as Lactococcus lactis JNU 534, presenting preservative properties for foods of animal origin. In this study, we present the complete genome sequence of the bacterial strain JNU 534. The final complete genome assembly consists of one circular chromosome (2,443,687 bp [base pair]) with an overall GC (guanine-cytosine) content of 35.2%, one circular plasmid sequence (46,387bp) with a GC content of 34.5%, and one circular contig sequence (7,666 bp) with a GC content of 36.2%.
