Antioxidant Production by Bacillus methylotrophicus Isolated from Chungkookjang, Korean Traditional Fermented Food (한국 전통발효식품인 청국장에서 분리한 Bacillus methylotrophicus에 의한 항산화물질의 생산)

  • Although antioxidant activities of Korean traditional fermented foods were reported by many researchers, study on antioxidant activity of microorganism originated from Korean traditional fermented foods was little. Therefore, we improved condition for antioxidant production by a bacterium isolated from home-made Chungkookjang. We selected a bacterial strain, which showed the highest antioxidative activity, from Chungkookjang and then named GJ. The selected GJ strain was identified as Bacillus methylotrophicus by alignment data of 16S rRNA gene nucleotide sequences. Improved medium compositions for DPPH radical scavenging activity were 0.25% sucrose, 1% peptone, 0.01% $MgSO_4{\cdot}7H_2O$ and initial pH 6.5, respectively. Optimal culture conditions were $30^{\circ}C$, 200 rpm and 4% inoculum volume, respectively. In improved conditions, DPPH radical scavenging activity of GJ reached to 91% in a short time. The strain GJ also possessed ACE inhibition and other antioxidative activities; ACE inhibition activity (49.4%), ABTS radical scavenging activity (99.8%), metal chelating activity (67.9%), SOD-like activity (36.5%) and reducing power ($A_{700}$ = 5.982) were observed, respectively. Therefore, our results suggest that B. methylotrophicus GJ strain may be potential candidate for functional foods, cosmetic products for anti-aging and medicine for diseases caused by oxidative stress.

Functional Characterization of Lactobacillus sakei JK-17 Isolated from Long-term Fermented Kimchi, Muk Eun Ji (장기간 발효 김치인 묵은지에서 분리한 Lactobacillus sakei JK-17의 기능성 조사)

  • The purpose of this work was to investigate the several functional characteristics of Lactobacillus sakei JK-17 isolated from long-term fermented kimchi, Muk Eun Ji. Initially, phylogenetic analysis using 16S rRNA sequencing was performed to identify the isolate JK-17, and the strain could be assigned to Lactobacillus sakei and designated as L. sakei JK-17. The strain was registered in GenBank as [JX841311]. The changes of bacterial growth and residual organic acids were monitored and HPLC was used to measure quantitatively two organic acids, lactic acid and acetic acid, produced in the culture during 84 hours of incubation. During the incubation period, several functional characteristics of L. sakei JK-17 were examined. L. sakei JK-17 culture depleted nitrite concentration 94.75%. Antioxidant activity of cultural supernatants of L. sakei JK-17 was approx. 53.8%, and ${\beta}$-galactosidase activities were 0.243 units/mL at pH 7.0 and 0.387 units/mL at pH 4.1, respectively. The antibacterial activities against food-poisoning causing bacteria were examined with 20-fold concentrated culture supernatants from L. sakei JK-17 and the antibacterial effects were clearly observed against all bacteria tested in this work.

ACC Deaminase Producing Arsenic Tolerant Bacterial Effect on Mitigation of Stress Ethylene Emission in Maize Grown in an Arsenic Polluted Soil

  • Arsenic is a known hazardous metalloid not only to the animals but also to plants. With high concentrations, it can impede normal plant growth and cause even death of plants at extremely high levels. A known plant response to stress conditions such as toxic levels of metal (loids) is the production of stress ethylene, causing inhibitory effect on root growth in plants. When the effect of various arsenic concentrations was tested to maize plant, the stress ethylene emission proportionately increased with increasing concentration of As(V). The inoculation of two arsenic tolerant bacteria; Pseudomonas grimonti JS126 and Pseudomonas taiwanensis JS238 having respective high and low 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity reduced stress ethylene emission by 59% and 30% in maize grown in arsenic polluted soils. The result suggested the possible use of Pseudomonas grimonti JS126 for phytoremediation of arsenic polluted soils.

  • A bacterium hydrolysing various organic materials including cellulose, protein, starch and lipid was isolated. The isolate was identified as Bacillus subtilis, and named Bacillus subtilis CK-2 in this paper. This bacterium showed optimal growth at $40\sim45^{\circ}C$, pH 6~9, and 0~3% of NaCl. B. subtilis CK-2 seemed to synthesis highly active autolysin. The hydrolytic enzymes produced by B. subtilis CK-2 were primary enzymes because extracellular enzyme activities varied similarly to the growth curve. The hydrolytic enzymes seemed to be stable at basic pH conditions. From these results, B. subtilis CK-2 was found to bea useful bacterial agent for composting, or for use in feed-production waste in agriculture, fishery, forest materials, livestock farming, and food.

Properties of ${\beta}$-Galactosidase from Bacillus licheniformis Isolated from Cheongkookjang (청국장 유래 Bacillus licheniformis의 ${\beta}$-Galactosidase 특성)

  • A bacterial strain was isolated from homemade Cheongkookjang as a producer of the ${\beta}$-galactosidase, capable of hydrolyzing lactose to liberate galactose and glucose residues. The isolate YB-1105 has been identified as Bacillus licheniformis on the basis of its 16S rDNA sequence, morphology and biochemical properties. ${\beta}$-Galactosidase activity was detected in both the culture supernatant and the cell extract of B. licheniformis YB-1105. The enzymes of both fractions demonstrated maximum activity for hydrolysis of para-nitrophenyl-${\beta}$-D-galactopyranoside (pNP-${\beta}Gal$) under identical reaction conditions of pH 6.5 and $50^{\circ}C$. However, ${\beta}$-galactosidase activity from the culture filtrate was affected more than that from the cell free extract at acidic pHs and high temperatures. The hydrolyzing activity of both ${\beta}$-galactosidases for pNP-${\beta}Gal$ was dramatically decreased by the addition of low concentrations of galactose, but was only marginally decreased by high concentrations of glucose or mannose.

Isolation and Characterization of an Agarase-Producing Bacterial Strain, Alteromonas sp. GNUM-1, from the West Sea, Korea

  • The agar-degrading bacterium GNUM-1 was isolated from the brown algal species Sargassum serratifolium, which was obtained from the West Sea of Korea, by using the selective artificial seawater agar plate. The cells were Gram-negative, $0.5-0.6{\mu}m$ wide and $2.0-2.5{\mu}m$ long curved rods with a single polar flagellum, forming nonpigmented, circular, smooth colonies. Cells grew at $20^{\circ}C-37^{\circ}C$, between pH 5.0 and 9.0, and at 1-10% (w/v) NaCl. The DNA G+C content of the GNUM-1 strain was 45.5 mol%. The 16S rRNA sequence of the GNUM-1 was very similar to those of Alteromonas stellipolaris LMG 21861 (99.86% sequence homology) and Alteromonas addita $R10SW13^T$(99.64% sequence homology), which led us to assign it to the genus Alteromonas. It showed positive activities for agarase, amylase, gelatinase, alkaline phosphatase, esterase (C8), lipase (C14), leucine arylamidase, valine arylamidase, ${\alpha}$-chymotrypsin, acid phosphatase, naphthol-AS-BI-phosphohydrolase, ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\beta}$-glucosidase, catalase, and urease. It can utilize citrate, malic acid, and trisodium citrate. The major fatty acids were summed feature 3 (21.5%, comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH) and C16:0 (15.04%). On the basis of the variations in many biochemical characteristics, GNUM-1 was considered as unique and thus was named Alteromonas sp. GNUM-1. It produced the highest agarase activity in modified ASW medium containing 0.4% sucrose, but lower activity in rich media despite superior growth, implying that agarase production is tightly regulated and repressed in a rich nutrient condition. The 30 kDa protein with agarase activity was identified by zymography, and this report serves as the very first account of such a protein in the genus Alteromonas.

Characterization and Antifungal Activity from Soilborne Streptomyces sp. AM50 towards Major Plant Pathogens

  • BACKGROUND: Chemical fungicides not only may pollute the ecosystem but also can be environmentally hazardous, as the chemicals accumulate in soil. Biological control is a frequently-used environment-friendly alternative to chemical pesticides in phytopathogen management. However, the use of microbial products as fungicides has limitations. This study isolated and characterized a three-antifungal-enzyme (chitinase, cellulase, and ${\beta}$-1,3-glucanase)-producing bacterium, and examined the conditions required to optimize the production of the antifungal enzymes. METHOD AND RESULTS: The antifungal enzymes chitinase, cellulase, and ${\beta}$-1,3-glucanase were produced by bacteria isolated from an sawmill in Korea. Based on the 16S ribosomal DNA sequence analysis, the bacterial strain AM50 was identical to Streptomyces sp. And their antifungal activity was optimized when Streptomyces sp. AM50 was grown aerobically in a medium composed of 0.4% chitin, 0.4% starch, 0.2% ammonium sulfate, 0.11% $Na_2HPO_4$, 0.07% $KH_2PO_4$, 0.0001% $MgSO_4$, and 0.0001% $MnSO_4$ at $30^{\circ}C$. A culture broth of Streptomyces sp. AM50 showed antifungal activity towards the hyphae of plant pathogenic fungi, including hyphae swelling and lysis in P. capsici, factors that may contribute to its suppression of plant pathogenic fungi. CONCLUSION(S): This study demonstrated the multiantifungal enzyme production by Streptomyces sp. AM50 for the biological control of major plant pathogens. Further studies will investigate the synergistic effect, to the growth regulations by biogenic amines and antifungal enzyme gene promoter.

  • Streptococcus mutans (S. mutans) is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of Cyperus rotundus (Cyperaceae) methanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of C. rotundus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 0.5 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of C. rotundus showed more than 10% inhibition over the concentration of 2 mg/ml. Thus, the application of C. rotundus can be considered a useful and a practical method for the prevention of dental caries.

  • After parturition, woman in childbed's health can grow worse. Because when woman in childbed gives birth, Genital organ can be damaged, and lost a lot of blood, and also consume a lot of physical strengths. So, after parturition, woman in childbed is easy to fall in disease if do not take care in health specially. So after parturition, woman in childbed should be careful in stress, sleep, food etc. This thesis described Postpartum Spasm[痙病], Oppressive Feeling and Dizziness[鬱冒], Constipation[便秘], Abdominal Pain[腹痛], Diarrhea[下利], Annoyance[煩亂] and Vomitting Sickness[嘔逆]. Postpartum Spasm[痙病]'s major symptom is rigided body. This disease corresponds to Tetanus in modem medicine, and bacterial infection is assumed for cause. By Oriental medicine, after parturition, when blood is lacking, if patient takes wind, this disease breaks out. Oppressive Feeling and Dizziness[鬱冒]'s major symptom is dizzy head and gloomy eye. After parturition, when blood is lacking, if patient sweats too much and take cold, this disease breaks out. After parturition, if patient consumes a lot of resins of human body, Constipation[使秘] breaks out. There are four causes to Abdominal pain[腹痛]. First, due to deficiency and cold treats by Danggwisaenggangyangyuktang(當歸牛薑羊肉湯). Second, due to stagnation of Gi(氣) treats by Jisiljakyaksan(枳實芍藥散). Third, due to blood stasis treats by Ha-eohyeoltang(下瘀血湯). Fourth, due to lochiorrhea treats by Daeseunggitang(大承氣湯). After parturition, Annoyance[煩亂] and Vomitting Sickness[嘔逆] due to fever of difficiency treat by Jukpidaehwan(竹皮大丸). After parturition, Febrile diarrhea[熱性下利] treats by Baekduonggagamchoagyotang(白頭翁加甘草阿膠湯). This disease is similar with present Bacillary Dysentery. After parturition, Exteroceptive disease due to taking wind treats by Yangdaltang. Due to wind and fever treats by Jukyeoptang(竹葉湯).

  • The radurization and radicidation effects on mixed condiment with Co-60 gamma irradiation at 3-9kGy and the physicochemical aspects of irradiated samples stored at $30^{\circ}C$ for three months were investigated. The initial microbial loads of the sample were $7.5{\times}10^5/g$ in total bacterial count and $1.2{\times}10^2/g$ in coliforms, respectively. The irradiation treatment of below 9 kGy could decontaminate the sample and $D_{10}$ value of the bacteria contaminated was shown to be 1.94 kGy. The chemical components associated with the quality of the sample were little affected by the irradiation at 3-6kGy. The sensory evaluation showed that the high sterilizing dose caused appreciably the change in overall flavors of mixed condiment.
