Gold Recovery from Cyanide Solution through Biosorption, Desorption and Incineration with Waste Biomass of Corynebacterium glutamicum as Biosorbent (생체흡착, 탈착 및 회화를 이용한 시안 용액으로부터 금의 회수)

  • In this study, we propose two methods able to recover different type of gold from gold-cyanide solutions: biosorption and desorption process for mono-valent gold recovery and biosorption and incineration process for zero-valent gold recovery. The waste bacterial biomass of Corynebacterium glutamicum generated from amino acid fermentation industry was used as a biosorbent. The pH edge experiments indicated that the optimal pH range was pH 2 - 3. From isothermal experiment and its fitting with Langmuir equation, the maximum uptake capacity of Au(I) at pH 2.5 were determined to be 35.15 mg/g. Kinetic tests evidenced that the process is very fast so that biosorption equilibrium was completed within the 60 min. To recover Au(I), the gold ions were able to be successfully eluted from the Au-loaded biosorbent by changing the pH to pH 7 and the desorption efficiency was 91%. This indicates that the combined process of biosorption and desorption would be effective for the recovery of Au(I). In order to recover zero-valent gold, the Au-loaded biosorbents were incinerated. The content of zero-valent gold in the incineration ash was as high as 85%. Therefore, we claim on the basis of the results that two suggested combined processes could be useful to recover gold from cyanide solutions and chosen according to the type of gold to be recovered.

  • The foodborne illness is the important public health concerns, and the livestock feces are known to be one of the major reservoirs of foodborne pathogens. Also, it was reported that 45.5% of foodborne illness outbreaks have been associated with the animal products contaminated with the livestock feces. In addition, it has been known that the persistence of a pathogens depends on many potential virulent factors including the various virulent genes. Therefore, the first step to understanding the public health risk of livestock feces is to identify and describe microbial communities and potential virulent genes that contribute to bacterial pathogenicity. We used the whole metagenome shotgun sequencing to evaluate the prevalence of foodborne pathogens and to characterize the virulence associated genes in pig and chicken feces. Our data showed that the relative abundance of potential foodborne pathogens, such as Bacillus cereus was higher in chickens than pigs at the species level while the relative abundance of foodborne pathogens including Campylobacter coli was only detected in pigs. Also, the microbial functional characteristics of livestock feces revealed that the gene families related to "Biofilm formation and quorum sensing" were highly enriched in pigs than chicken. Moreover, the variety of gene families associated with "Resistance to antibiotics and toxic compounds" were detected in both animals. These results will help us to prepare the scientific action plans to improve awareness and understanding of the public health risks of livestock feces.

Development and Evaluation of a Next-Generation Sequencing Panel for the Multiple Detection and Identification of Pathogens in Fermented Foods

  • These days, bacterial detection methods have some limitations in sensitivity, specificity, and multiple detection. To overcome these, novel detection and identification method is necessary to be developed. Recently, NGS panel method has been suggested to screen, detect, and even identify specific foodborne pathogens in one reaction. In this study, new NGS panel primer sets were developed to target 13 specific virulence factor genes from five types of pathogenic Escherichia coli, Listeria monocytogenes, and Salmonella enterica serovar Typhimurium, respectively. Evaluation of the primer sets using singleplex PCR, crosscheck PCR and multiplex PCR revealed high specificity and selectivity without interference of primers or genomic DNAs. Subsequent NGS panel analysis with six artificially contaminated food samples using those primer sets showed that all target genes were multi-detected in one reaction at 108-105 CFU of target strains. However, a few false-positive results were shown at 106-105 CFU. To validate this NGS panel analysis, three sets of qPCR analyses were independently performed with the same contaminated food samples, showing the similar specificity and selectivity for detection and identification. While this NGS panel still has some issues for detection and identification of specific foodborne pathogens, it has much more advantages, especially multiple detection and identification in one reaction, and it could be improved by further optimized NGS panel primer sets and even by application of a new real-time NGS sequencing technology. Therefore, this study suggests the efficiency and usability of NGS panel for rapid determination of origin strain in various foodborne outbreaks in one reaction.

  • Nine hybrid rices crossed between Korean cytoplasmic-genetic male sterile rices having the WA cytoplasm and the Korean restorer lines or varieties, HR1619A/Nampungbyeo, HR1619A/Gayabyeo, HR1619A/Line234, TongilA/Nampungbyeo, TongilA/Cheongcheongbyeo, Suwon 296A/Line 209, Suwon 296A/Line 237, Suwon 296A/Line 252 and Line 201A/Line 234, and their parents were grown at Yeungnam University in 1984. The rough rice yield of the hybrids Line 201A/Line 234, TongilA/Nampungbyeo and HR1619A/Nampungbyeo were 939, 927 and 900 Kg/10a respectively. The heterosis(F$_1$/Midparent) of the above three hybrids was 40%, 20% and 19%, the heterobeltiosis(F$_1$/Better parent) was 36%, 17% and 10%, and the standard heterosis (F$_1$/Standard variety, Cheongcheongbyeo) was 19%, 17% and 14% respectively. The hybrids HR1619A/ Gayabyeo and Suwon 296A/Line 237 showed negative heterosis in grain yield. Significant heterobeltiosis for grain number per panicle was found while less or no heterobeltiosis was observed in panicle number per hill, 1000-grain weight and grain fertility. The bacterial leaf blight disease reaction of the hybrids tested was almost the same as that of one parent at least. The amylose content of the hybrids was medium to low the same as their parents. The protein content and alkali digestion value of the hybrids were almost the same as their parents.

Antifungal and carboxylesterase-producing bacteria applied into corn silage still affected the fermented total mixed ration

  • Objective: This study investigated the effects of corn silage as a source of microbial inoculant containing antifungal and carboxylesterase-producing bacteria on fermentation, aerobic stability, and nutrient digestibility of fermented total mixed ration (FTMR) with different energy levels. Methods: Corn silage was used as a bacterial source by ensiling for 72 d with an inoculant mixture of Lactobacillus brevis 5M2 and L. buchneri 6M1 at a 1:1 ratio. The corn silage without or with inoculant (CON vs MIX) was mixed with the other ingredients to formulate for low and high energy diets (LOW vs HIGH) for Hanwoo steers. All diets were ensiled into 20 L mini silo (5 kg) for 40 d in quadruplicate. Results: The MIX diets had lower (p<0.05) acid detergent fiber with higher (p<0.05) in vitro digestibilities of dry matter and neutral detergent fiber compared to the CON diets. In terms of fermentation characteristics, the MIX diets had higher (p<0.05) acetate than the CON diets. The MIX diets had extended (p<0.05) lactic acid bacteria growth at 4 to 7 d of aerobic exposure and showed lower (p<0.05) yeast growth at 7 d of aerobic exposure than the CON diets. In terms of rumen fermentation, the MIX diets had higher (p<0.05) total fermentable fraction and total volatile fatty acid, with lower (p<0.05) pH than those of CON diets. The interaction (p = 0.036) between inoculant and diet level was only found in the immediately fermentable fraction, which inoculant was only effective on LOW diets. Conclusion: Application of corn silage with inoculant on FTMR presented an antifungal effect by inhibiting yeast at aerobic exposure and a carboxylesterase effect by improving nutrient digestibility. It also indicated that fermented feedstuffs could be used as microbial source for FTMR. Generally, the interaction between inoculant and diet level had less effect on this FTMR study.

Sustained release of alginate hydrogel containing antimicrobial peptide Chol-37(F34-R) in vitro and its effect on wound healing in murine model of Pseudomonas aeruginosa infection

  • Background: Antibiotic resistance is a significant public health concern around the globe. Antimicrobial peptides exhibit broad-spectrum and efficient antibacterial activity with an added advantage of low drug resistance. The higher water content and 3D network structure of the hydrogels are beneficial for maintaining antimicrobial peptide activity and help to prevent degradation. The antimicrobial peptide released from hydrogels also hasten the local wound healing by promoting epithelial tissue regeneration and granulation tissue formation. Objective: This study aimed at developing sodium alginate based hydrogel loaded with a novel antimicrobial peptide Chol-37(F34-R) and to investigate the characteristics in vitro and in vivo as an alternative antibacterial wound dressing to treat infectious wounds. Methods: Hydrogels were developed and optimized by varying the concentrations of crosslinkers and subjected to various characterization tests like cross-sectional morphology, swelling index, percent water contents, water retention ratio, drug release and antibacterial activity in vitro, and Pseudomonas aeruginosa infected wound mice model in vivo. Results: The results indicated that the hydrogel C proved superior in terms of cross-sectional morphology having uniformly sized interconnected pores, a good swelling index, with the capacity to retain a higher quantity of water. Furthermore, the optimized hydrogel has been found to exert a significant antimicrobial activity against bacteria and was also found to prevent bacterial infiltration into the wound site due to forming an impermeable barrier between the wound bed and external environment. The optimized hydrogel was found to significantly hasten skin regeneration in animal models when compared to other treatments in addition to strong inhibitory effect on the release of pro-inflammatory cytokines (interleukin-1β and tumor necrosis factor-α). Conclusions: Our results suggest that sodium alginate -based hydrogels loaded with Chol-37(F34-R) hold the potential to be used as an alternative to conventional antibiotics in treating infectious skin wounds.

A semi-selective agar medium to detect the presence of the causal agent of anthracnose, Colletotrichum scovillei, in chilli pepper (고추에서 탄저병균 Colletotrichum scovillei를 검출하기 위한 반선택 배지)

  • A semi-selective agar medium was developed for detecting and enumerating Colletotrichum scovillei in chili (Capsicum annum) plant material. Potato-dextrose-agar(PDA) was used as the basic medium. The composition of the semi-selective agar medium was established after several attempts to favor the development of C. scovillei and inhibit the growth of other fungi and bacteria. The semi-selective agar medium contained PDA amended with pyribencarb and pydiflumetofen at 40 ㎍ mL-1 each and streptomycin at 100㎍ mL-1 for preventing bacterial growth. The pH was adjusted to 4.8 with 85% lactic acid. The inhibition of the mycelial growth of C. scovillei was significantly less than that of most other fungi including Fusarium species when grown on the semi-selective medium. C. scovillei was detected from naturally infected chili plants by plating fruit and stem tissue suspensions on the semi-selective medium, which was found to be reliable and quantifiable. This was the first report of a semi-selective agar medium to detect the presence of C. scovillei in naturally infected chili tissue.

Convergence study on the quality evaluation of ginseng sprout produced smart farm according to organic acid treatment and packing containers during storage (스마트팜 생산 새싹인삼의 유기산 처리 및 포장 용기에 따른 품질 평가에 대한 융합연구)

  • In this study, the physical quality and microbial changes of ginseng sprout according to the pretreatment process and packaging container were evaluated to improve the storage properties of ginseng sprout produced in smart farm. Quality change during storage (10 days) according to pretreatment method (ascorbic acid, citric acid, peroxyacetic acid) and packaging container (expanded polystyrene (EP), polypropylene (PP), polyethylene (PE), polypropylene + polyethylene + cast polypropylene (PP+PE+CPP)) was evaluated in terms of texture, viable cell count, water content, and color. As a result of comparison according to the type of pretreatment, the citric acid treatment group showed the lowest texture change and the effect on inhibition of bacterial growth. On the other hand, citric acid, which was most effective among pretreatments, was treated in all samples and then stored in 4 types of containers. Specially, the ginseng sprout in PP packaging container was not observed significant softening or color changes after 10 days storage, and the lowest changes in viable cell number. Therefore, this study was shown that citric acid treatment and use of PP packaging container are effective in increasing the shelf life of ginseng sprout.

Alterations in qualities of different cultivation types of garlic during storage: Changes assessed by ultrasonic and organic acid treatment (초음파 및 유기산 처리에 따른 재배유형별 마늘의 저장 중 품질변화)

  • We investigated the effects of organic acid and ultrasonication treatment in maintaining the quality of garlic during storage. Samples were exposed for 5 min to either ultrasonication at 60℃, 1% citric acid, or 0.5% fumaric acid. Presence of microorganisms and minerals, hardness, and color were compared during storage at 4℃ for 28 days. The total aerobic bacterial count remained low. No proliferation of Escherichia coli was observed after treatment with fumaric acid or ultrasonication, and mold proliferation was inhibited by ultrasonication. The mineral content of the northern type garlic was higher than that of the southern type. Exposure to fumaric acid did not result in a substantial difference in hardness until 21 days of storage, at which time there was a decrease in the L-value in each cultivation type. Our results indicate that treatment with 0.5% fumaric acid for 5 min was effective in reducing the abundance of microorganisms during storage without affecting the hardness or color in garlic.

Analysis of quality characteristics of sugar-soaked raspberry according to storage period (저장 기간에 따른 산딸기 당 침지액의 품질특성 분석)

  • Volatile and non-volatile metabolite profiles of sugar-immersed raspberry liquid during different storage periods were analyzed and comparative analysis with their general characteristics, antioxidant activity, and sensory quality was evaluated to better understand the effect of the storage period on the quality of ugar-immersed raspberry liquid. During storage, a browning reaction occurred, resulting in a change in color and the production of volatile compounds. At the beginning of storage, sucrose was completely decomposed into fructose and glucose, and the sweetness was rapidly reduced, but the increase in succinic acid increased the sour taste. Most volatile compounds increased with an increase of the storage period, and especially, the contents of citronellol, octanoic acid, and hexanoic acid, which are known as antioxidants, showed the highest content in 10 day-sample, showing the highest antioxidant activity at this time. Although a further study on bacterial profiles and browning reaction during the storage will be needed, the results of this study showed that the quality of sugar-soaked raspberry extract was significantly affected by the storage period and can be used as basic data for commercialization of ugar-immersed raspberry liquid.
