  • Objectives : Conclusion : The purpose of this study is to examine effect that KDKEJ used to atopic dermatitis disease patient get in atopy eruption control experimentally. Methods : We analyzed the expression of IgE, IL-6, II-4, IL-5 and IL-13's level in serum, and $IFN-{\gamma}'$ production by KDKEJ extract. We also analyzed KDKEJ extract get to NC/Nga mine's skin establishment analyzes neck-back skin after biopsy, and H&E method measured about epidermis and dermis part in comparison with control group. Results : In this research KDKEJ extract as treatment result to a NC/Nga mice, IgE and IL-6 content in serum decreased remarkably than control group. And decreased than result control group which measure II-4, IL-5 and IL-13's level in serum and $IFN-{\gamma}'$ production secreted in Th1 cell displayed increase by KDKEJ extract. IL-4 and $IFN-{\gamma}'$ gene revelation amount displayed marked decrease than control group in result that observe effect that get in skin of a NC/Nga mice's skin establishment analyzes neck-back skin after biposy, and dye by H&E method decreased about epidermis and inflammation of dermis part remarkable than control group. Conclusions : These results suggest that Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and $IFN-{\gamma}$ by KDKEJ extract could know that KDKEJ extract can use usefully in allergy autoimmune disease.

  • Atopic dermatitis (AD) is a chronically relapsing pruritic inflammatory skin disease. To find new anti-inflammatory products for skin inflammatory disease such as AD and contact dermatitis, we produced the effective microorganism fermentation substance (EM-S) by fermentation of medicinal plants with effective microorganisms including photosynthetic bacteria, lactic acid bacteria and yeast, screened the effects of EM-S on NC/Nga model mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinae (Df) extract. Topically applied EM-S significantly reduced clinical severity score, ear thickness and histological grade in AD-like NC/Nga mouse model by Df antigen sensitization. In addition, the serum IgE and Th2 chemokine levels (TARC/CCL17, MDC/CCL22 and CTACK/CCL27) were significantly reduced by EM-S. Futhermore, skin tissue expressions of Th2 chemokines were significantly reduced by EM-S. These results demonstrate that topical application of EM-S may be improve the AD-like skin lesion by suppressing IgE and Th2 chemokines.

Effects of SaengRyo-SaMul-tang on Atopic Dermatitis Induced by DNCB in Mice (생료사물탕(生料四物湯)이 DNCB로 유발된 생쥐의 아토피 피부염에 미치는 영향)

  • Objectives : Atopic Dermatitis (AD) is chronic skin disease characterized by allergic hypersensitivity reactions. Saeng-ryo-Samul-tang (SRSM) can treat skin disease by cooling down blood heat, cealering away congenital fever and detoxicating. This study was conducted to investigate the effects of SRSM on AD induced by 2,4-dinitrochlorobenzene (DNCB) in mice Methods : In this study, the effects of SRSM on changes in body weights, thicknesses of dorsum skin, thicknesses and weights of ear, changes of symptoms on the dorsum skin, histopathological degree of ear and dorsum skin, IL-10 and $TNF-{\alpha}$ in serum were observed. And the effects on the proliferation rates of splenocytes were also investigated in vivo and in vitro study. Results : In SRSM topical application (Topical) group, SRSM oral application (Oral) group and SRSM Combination (Combi) group thickness of dorsum skin decreased significantly. But in TPC, ORL and CBN group, weight of ear didn't show any changes, but thickness of ear decreased significantly. And TPC, ORL and CBN group showed meaningful effectiveness symptoms like desquamation and erythema on AD's clinical espect. In histopathological observation, spongiosis, edema and inflammatory cell infiltration of epidermal were remarkably diminished in TPC, ORL and CBN group. And SRSM diminished the proliferation rates of splenocytes in vivo and vitro study. Conclusions : The present study suggests that SRSM can significantly reduced symptoms of AD, therefore SRSM is effective to treatment of AD.

Effects of Anemarrhena asphodeloides Extract on Atopic-Dermatitis like Skin Lesions in DNCB-induced Balb/c Mice (DNCB로 유도한 아토피 유사 피부염에 지모 추출물이 미치는 영향)

  • Under constant environmental pollution, the incidence of Atopic Dermatitis (AD) caused by air pollutants and allergens has increased. AD is an allergy inflammatory skin disease characterized by pruritus, eczema, and skin dryness. In herbal medicine, Anemarrhena asphodeloides (Anemarrhenae Rhizoma; AR) has been utilized to treat Alzheimer's disease, osteoporosis, hypertension, and inflammation. The purpose of study evaluated the effect of AR in a mouse model of 2,4-dinitrochlorobenzene (DNCB)-induced AD-like skin lesions. After acclimatization for 5 days, the mice (6-week-old, male Balb/c) were divided into five groups (n=6/group): NC (normal control), DNCB (control), Dex (5 mg·kg-1, p.o.), AR100 (100 mg·kg-1, p.o.), and AR300 (300 mg·kg-1, p.o.). On days 1 and 3, 1% DNCB was applied to the skin and ears. After 4 days, 0.5% DNCB was applied once every 2 days for 2 weeks. Then, skin and ears eczema area and severity index (EASI); skin nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and prostaglandin E2 (PGE2) levels; and plasma immunoglobulin E (IgE) levels were examined. The AR groups showed lower EASI, skin and ear thickness, mast cell count, and IgE levels than the control groups. Moreover, AR reduced iNOS, COX-2, and PGE2 levels. Therefore, AR possesses anti-inflammatory properties and can improve skin damage, indicating its therapeutic potential against AD.

Effect of Several Herbal Medicines on Atopic Dermatitis Patients' SCORAD and Cytokine Levels (수종 한약 처방이 아토피 피부염 환자의 SCORAD 및 Cytokine 변화에 미치는 영향)

  • The purpose of this study is to determine whether the clinical activity of patients with atopic dermatitis(AD) correlate with their serum cytokine levels. 86 patients with AD were treated with KHS, YMH and GHT. On each visit to the clinic, their skin status was evaluated using the SCORAD index and serum IL-4, $IFN-{\gamma}$, IL-12, IL-13, IL-2 and IL-10 were measured by ELISA. After oral administration of herbal medicine, patients' skin status and subjective parameters such as pruritus and sleeplessness were significantly improved according to regression analysis. In particular, patients aged 0-9 showed stronger responses to treatment, which implies that the younger patients aged, the easier the treatment by herbal formula became. In analysis of the type of chosen herbal medicines, YMH treated group showed a stronger response in reduction of affected lesion, intensity of a lesion and subjective symptoms, while KHS treated group showed the least reduction. The levels of serum IL-4, $IFN-{\gamma}$ and IL-2 correlated with disease activity. Moreover, in patients under 0-9, the IL-4 level had a corrleation with disease activity; patients under 10-19 showed a correlation between the levels of IL-4 and IL-2 and disease activity; patients under 20-29 showed a correlation between the levels of IL-2, IL-12 and $IFN-{\gamma}$ and disease activity. In comparison with chosen herbal medicines, the groups treated with YMH or GHT treated group showed a correlation between the levels of IL-4, IL-12 and IL-2 and disease activity while KHS treated group did not. Our data suggest that serum levels of IL-4 and IL-13 are good indicators of clinical improvement in treating AD.

The Outbreaks, Treatment and Lifestyle in Children with Atopic Dermatitis (아토피 피부염 아동의 발병, 치료 및 생활습관)

  • Purpose: This study was performed to investigate outbreak, treatment, and lifestyle in children with atopic dermatitis (AD) for developing a health promotion education program. Method: Data were collected from 2,920 children with AD in preschool and elementary schools in K city. Result: The majority of children (43.75%) were onset in less than 1 yr after the birth. The locations of skin lesion were face, extension of extremities, and flextion of extremities, neck and trunk. The types of treatment were complement therapies, and medical treatment such as pediatrics and dermatology. Their parents chose a type of treatment for their children. Of complement therapies, the most common type was aroma oil. Herbal medicine was the most eating type among complement therapies. In lifestyle, the preferred bathing method was shower. In applying moisturizer, the most common time was within 3 min after bath. The highest frequency of taking instant/processed diet was 1-2 times per week and the most preferred type was pork. Conclusion: A Health promotion educational program with AD in preschool and elementary school children should included a specificity of disease, the recent treatment guideline, life therapy and evidenced complement therapy by child and family unit for the promotion of their health.

A Double-Blind, Placebo Controlled-Trial of a Probiotic Strain Lactobacillus sakei Probio-65 for the Prevention of Canine Atopic Dermatitis

  • Canine atopic dermatitis (CAD) is a ubiquitous, chronic inflammatory skin disorder prevalent in dogs, which results in production of abnormal levels of IgE antibodies in reciprocation to an allergen challenge. In this study, administration of the probiotic strain Lactobacillus sakei probio-65 for 2 months significantly reduced the disease severity index in experimental dogs diagnosed with CAD. In addition, one month pre-medication of L. sakei probio-65 revealed significant difference in the PVAS score in experimental dogs for both probio-65 and placebo groups. However, post 2 months treatment resulted in a significant decrease in the CASESI score values in the probio-65-treated group (p < .0.06).

Prevalence Rate and Factors Associated with Atopic Dermatitis among Korean Middle School Students

  • Purpose: The purpose of this study was to verify prevalence rates of atopic dermatitis (AD) in middle school students in Korea and to define AD and associated factors. Methods: This study was conducted using secondary analysis of raw data from the 'The 6th Korean National Adolescent Health Behavior Survey, 2010' conducted by the Korea Centers for Disease Control. The participants were 37,570 students. General characteristics and health behavior-related characteristics were analyzed using descriptive statistics, ${\chi}^2$-test and logistic regression analysis. Results: Results are as follows. 1. There were 8,494 middle school students (22.6%), who had been diagnosed with AD during the last 12 months. 2. Factors associated with AD in middle school students were gender (p<.001), grade level (p=.001), father's education (p<.001), mother's education (p=.032), regular dietary habits (breakfasts, p=.025; lunches; p=.003), stress (p<.001), recognition of subjective awareness of one's health conditions (p<.001), hand washing before meals at school (p=.013), and cessation education for both smoking (p=.002) and alcohol (p=.031). Conclusion: Results of the study indicate that in order to alleviate AD symptoms in these students, there is a need to develop nursing interventions and education programs taking into consideration these variables and verifying the effects of interventions and programs.

A Study on the Effect of Gallotannin Treatment of Ceramide-containing Fibers on Atopic Skin Diseases (세라마이드 함유 섬유의 복합갈로탄닌 처리에 의한 아토피성 피부질환 완화작용에 대한 연구)

  • In order to investigate the effect of gallotannin treatment to ceramide-containing fabrics on atopic skin diseases, the agglomeration of standard protein BSA and the deactivation of model enzyme were examined. The gallotannin treated on ceramide-containing fabrics precipitated the standard protein, BSA, and therefore deactivated the model enzyme by 70% at 6% treatment concentration. Wash durability should be improved after around 5 cycles of washing. Clinical test of the gallotannin-treated fabrics was carried out on mice for two test items, transepidermal water loss assay and severity score of diseased skin of mice. The results showed significant level of improvement of atopic skin diseases compared with the negative controled.
