SOLVING BIOANALYTICAL PROBLEMS BY THE METHOD OF MATRIX-ASSISTED LASER DESORPTION IONIZATION MASS SPECTROMETRY (MALDI-MS)

  • The method of matrix-assisted laser desorption ionization mass spectrometry has been used to solve some bioanalytical problems, which is difficult to analyse by general methods. For the selection of proper laser wavelength and matrices, eight matriees was used with laser wavelength of 226 and 355nm. The result shows that with wavelength of 355nm better results could be obtained with most of the matrices. The molecular weight of eytochrome C, which was seperated by gel electrophoresis and electro-blotted onto NC membrane is determined by MALDI. The accuracy is better than 0.1%, which is much higher than that of SDS-PAGE. Protein mixture extracted from crude peanut oil is directly determined by MALDI. The molecuiar weight of its three components are determined, and the result also demonstrated that these proteins are in free manner. As proteins arc in 2S bond, with the traditional method, SDS-PAGE, it is not able to decide whether protein exists in combination mode or in free manner. In the technique of two phase aquesous solution, which is used for separating biomaterials, water soluble polymers stained with dyes are used in this technique. By the use of MALDI the number or the dye molecules react with the polymer PEG molecule are determined, and that is difficult to determined by other methods.

  • In this paper, we introduce a robot-assisted medical diagnostic system that enables remote ultrasound (US) imaging to be applied to the conventional telemedicine, which has been possible only with interviewing or a visual exam. In particular, a master-slave robot system is developed that ultrasonic diagnosis specialist can control the position and orientation of US probe in the remote place. The slave robot is designed to be compact, lightweight, and hand-held so that it can easily transfer to the remote healthcare center. Moreover, 6-degree-of-freedom (DOF) probe motion is possible by the robot design based on Stewart platform. The master device is also based on a similar structure of the slave robot. To connect master and slave system in the wide area network (WAN) environment, a hardware CODEC was developed. In this paper, we introduce the detail of each component and the results of the recent experiments conducted in the remote sites by the developed robotic ultrasound imaging system.

Optimization of Microwave-Assisted Method for Accelerated Glycated Hemoglobin Quantification from Amino Acids to Proteins

  • Glycated hemoglobin ($HbA_{1c}$) has been commonly used to screen and diagnose for patients with diabetes mellitus. Here the accelerated procedure of microwave-assisted sample treatment from acid hydrolysis to enzyme digestion followed by isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) was optimized and applied to measure $HbA_{1c}$ in an effort to speed up analysis time. First, two signature peptides of $HbA_{1c}$ and hemoglobin $A_0$ were certified with amino acid analysis by setting optimized acid hydrolysis conditions to $150^{\circ}C$, 1.5 h and $10{\mu}M$ sample concentration in 8 M hydrochloric acid. Consequently, the accurate certified peptides above were used as calibration standards to implement the proteolytic procedure with endoproteinase Glu-C at $37^{\circ}C$, 700 W for 6 h. Compared to the traditional method, the microwave heating not only shortened dramatically sample preparation time, but also afforded comparable recovery yields. The optimized protocol and analytical conditions in this study are suitable for a primary reference method of $HbA_{1c}$ quantification with full SI-traceability and other similar proteins in complex biological samples.

자가동영상을 활용한 디브리핑 학습의 효과: 흡인간호 실습교육을 중심으로 (The Effects of Suction Care Self Video-based Debriefing-assisted Learning in the Fundamentals of Nursing Practice)

  • Purpose: This study was conducted to identify the effects of a suction care self video-based debriefing-assisted learning in the fundamentals of nursing practice for nursing students on clinical performance, self-efficacy and problem-solving process. Methods: A nonequivalent control group non-synchronized design study was used and included 97 junior nursing students at P University as participants, 48 of whom were assigned to the experimental group, and 49 of whom were assigned to the control group. The outcome measurements were clinical performance for suction care, self-efficacy, and the problem-solving process. Results: The scores of clinical performance were 23.75 in the experimental group who used the materials and 21.55 in the control group, and the difference was statistically significant (p<.000). Self-efficacy was 4.30 in the experimental group and 4.18 in the control group, which was a statistically significant difference (p=.000), and the pre-to-post differences of self-efficacy was statistically significant as well (p<.001). However, there were no significant differences in the pre-to-post differences of the problem-solving process (p<.802). Conclusions: Self video-based debriefing-assisted learning may be more effective in enhancing clinical performance and self-efficacy than that of conventional practice. Whether self-efficacy will contribute to enhanced learning motivation for nursing students needs further examination.

  • As a second part of the comparison study, microscopic features of an air-assisted fuel injector(AAFI) and a high-pressure swirl injector (HPSI) were characterized. They consist of the internal spray structure in terms of fuel mass and drop diameter, the overall atomization performance with respect to operating parameters and the drop size distribution. Large droplets are concentrated in around the head part of a spray field of the HPSI, while in the case of the AAFI, they were distributed in the tail part. Although the AAFI showed the better atomization performance, the feasible ranges of operating parameters such as injection and ambient pressure were found to be wider in the HPSI. Drop size distribution of the AAFI sprays was more dispersed than that of the HPSI. Drop size distribution of the AAFI sprays was more dispersed than that of the HPSI. However, at the well-atomized condition, it appeared to be very uniform.

  • For more than two decades, the intracytoplasmic sperm injection (ICSI) technique has been used as a valuable tool to provide opportunities for studying fertilization, treating human infertility, and producing transgenic animals. Not only in facilitating fertilization but also in propagating mammalian species, ICSI has enhanced the potential of assisted reproductive technologies in human. Polyspermic fertilization has been one of major problems in pig reproduction, but the ICSI helped to solve the problem, and used widely to generate transgenic piglets. Although the ICSI technique is considered to be a very useful tool in assisted reproductive technologies, including generation of transgenic animals, there are some disadvantages using the technique. In this review, we describe the ICSI technique and its application in animal production and human infertility, and discuss advantage and disadvantage of the technique in mammals.

  • For deposition technology using plasma, it plays an important role in improving film deposited with high ionization rate through high density plasma. Various deposition methods such as high-power impulse magnetron sputtering and ion-beam sputtering have been developed for physical vapor deposition technology and are still being studied. In this study, it is intended to control plasma using inductive coupled plasma (ICP) antennas and use properties to improve the properties of Hafnium nitride (HfN) films using ICP assisted magnetron sputtering (ICPMS). HfN film deposited using ICPMS showed a finer grain sizes, denser microstructure and better mechanical properties as ICP power increases. The best mechanical properties such as nanoindentation hardness of 47 GPa and Young's modulus of 401 GPa was obtained from HfN film deposited using ICPMS at ICP power of 200 W.

반의존 노인용 시설주거개발을 위한 기초 연구 - 미국 버지니아 주 농촌지역의 우수 노인생활보조주택 관리사례를 중심으로 - (A Study on Development of Assisted Living Facilities for Semidependent Older Koreans - Focused on Management Experiences of Best Practices in Rural Virginia -)

  • The propose of this study was to investigate the management characteristics of assisted living facility(ALF) as an housing alternative for semi-dependent elderly. For this purpose, 5 ALFs which have been well known as best practices in rural Virginia were selected for a multi- site case study. Face-to-face interview with administrators or assistant administrators of i facilities were done. In addition to this, written materials about the each facility and personal record from researcher were used to analyze the data. Six management categories including 82 open-ended questions were analyzed and successful examples and management difficulties were identified from best practices. Based on the results, some recommendations for development of ALFs in Korean elderly were suggested. In short, this study found that best practices which were selected and analyzed in this study showed that successful management was resulted from continuous efforts of the stalls to provide better services in order for improving the quality of life of their residents.

마이크로파를 이용한 바이오디젤 전환 기술 동향 분석 (A Review of Microwave-assisted Technology for Biodiesel Production)

  • Biodiesel is renewable, eco-friendly, clean burning diesel replacement that is consisted of short chain alkyl ester. Biodiesel is derived from the transesterification of vegetables oils or animal fats with alcohol. The process has some technical problems that must be resolved to reduce the high operation cost. Eco-friendly physical technologies by using microwave have successfully improved the transesterification for biodiesel production. This paper attempts to extensively review microwave-assisted technology for biodiesel production. Additionally, different types of catalyst for biodiesel production have been summarized. It is concluded that the microwave-assisted technique improves the reaction rate significantly in comparison with conventional methods. Therefore it can be a suitable method of reducing the reaction time and can also decreases the cost of biodiesel production.

Survival rate and clinical evaluation of the implants in implant assisted removable partial dentures: surveyed crown and overdenture

  • PURPOSE. The purpose of this study was to investigate survival rates of the implants used in implant assisted removable partial dentures (IARPDs). MATERIALS AND METHODS. The study was conducted on 21 patients who were treated with IARPDs. The mean follow-up period for IARPD patients ranged from 12 to 185 months (mean 47.9 months). A total of 58 implants were used for IARPDs in two different modalities: 41 for surveyed crowns and 17 for overdentures. The survival of implants was determined by clinical and radiographic evaluations considering relevant factors: location, RPD classification, opposing dentition, splinting, and implant diameter. RESULTS. The survival rate of total 58 implants was 93.1%: 95.1% for implants supporting surveyed crowns and 88.2% for implants used in overdentures. Considering only the implants supporting surveyed crown, regular diameter implants showed a higher survival rate than narrow or wide diameter implants. CONCLUSION. The survival rate of the implants used in IARPDs was 93.1% (surveyed crown: 95.1%, overdenture: 88.2%).
