Immune Stimulation and Anti-Metastasis of Crude Polysaccharide from Submerged Culture of Hericium erinaceum in the Medium Supplemented with Korean Ginseng Extracts (수삼추출물 첨가 배지에서 배양된 노루궁뎅이버섯 균사체 심부발효물 조다당획분의 면역 및 암전이 억제활성)

  • To find the new use of Korean ginseng and mushroom, crude polysaccharides were prepared from submerged cultures of Hericium erinaceum in the medium supplemented with Korean ginseng extracts. When we fractionated crude polysaccharides (HE-GE-CP-1, 3, and 5) from hot-water extracts of submerged cultures of H. erinaceum with ginseng extracts (1%, 3%, and 5% addition of total medium), the yields of HE-GE-CP-1, 3, and 5 were identified at 5.7, 5.1, and 4.8%, respectively. Among crude polysaccharide fractions, HE-GE-CP-5 was significantly higher (1.89-fold of the saline control) than those of HE-GE-CP-1 (1.64-fold) or HE-GE-CP-3 (1.76-fold) on mitogenic activity of splenocytes. HE-GE-CP-5 also had the more potent bone marrow cell proliferation (1.83-fold) rather than HE-CP or HE-GE-CP-1 or HE-GE-CP-3 (1.59- or 1.44- or 1.69-fold, respectively), and anti-metastatic activity as anti-cancer effect showed the highest prophylactic value (72.4% inhibition of tumor control) in 5% supplementation of ginseng extract. However, the lysosomal phosphatase of macrophage was significantly stimulated after HE-GE-CP-3 treatment (2.03-fold). In addition, the immunostimulating and anti-metastatic crude polysaccharide, HE-GE-CP-5, contained mainly neutral sugars (63.2%) with considerable amounts of uronic acid (19.3%) and a small amount of proteins (8.8%). HE-GE-CP-5 can stimulate immune system to inhibit tumor metastasis, and its anti-tumor metastasis may be associated with macrophages, splenocytes and Peyer's patch cells activation.

Life Style and Eating Behavior of Stroke Patients in Daegu and Gyeongbuk Province, Korea (대구.경북지역 뇌졸중 환자의 생활습관 및 식행동 특성)

  • The present study was performed to analyze the life style and eating behavior of stroke patients and to find the risk factors related to stroke incidence in Daegu and Gyeongbuk Province, Korea. The case subjects (n=100) were selected from newly diagnosed stroke patients at Kyungpook National University Hospital. The control subjects (n=150) were selected from community inhabitants who did not have stroke history and were sex and age-matched with the case subjects. The survey was conducted by individual interviews using questionnaires on the general characteristics, life-style, eating behavior, food intake frequency and food preference. The high body mass index, preexisting diseases such as diabetes and hypertension, stroke family history, smoking, overeating habit and high preference for sweet, greasy and meat foods appeared to be the risk factors for stroke incidence. On the other hand, the results suggest that life style of regular exercise and nonsmoking, food habits of green tea drinking and enough chewing, preference for Korean meal type, high intake frequency for legumes, vegetables, mushrooms, seaweeds, fishes and shell fishes, soy milk and green tea might be the protective factors for the stroke. Therefore, maintenance of healthy weight, the prevention and management of the deteriorative chronic diseases, change of life style and improvement of eating behaviors are considered to be important for stroke prevention.

Characterization of Scaled-up Low-Trans Shortening from Rice Bran Oil and High Oleic Sunflower Seed Oil with Batch Type Reactor (회분식반응기를 이용한 미강유, 팜스테아린과 고올레인산 해바라기씨유 유래 대량 제조된 저트랜스 쇼트닝의 특성 연구)

  • Scaled-up low-trans shortening (LTS) was produced by lipase-catalyzed interesterification. Blend of rice bran oil (RBO), palm stearin (PS) and high oleic sunflower seed oil (HO) with 1:2:0.9 (w/w/w) ratio was interesterified using immobilized lipase from Thermomyces lanuginosus (TLIM) in the batch type reactor at $65^{\circ}C$ for 24 hr, and physicochemical melting properties of LTS were compared with commercial shortening. Solid fat content (SFC) of commercial shortening (used as control) and LTS was similar at 9.56 and 8.77%, respectively, at $35^{\circ}C$. Major fatty acids in LTS were C16:1 (33.7 wt%), C18:1 (45.7 wt%) and C18:2 (13.4 wt%). Trans fatty acid content in the commercial shortening (4.8 wt%) was higher than that of LTS (0.5 wt%). After reverse-phase HPLC analysis, major triacylglycerol (TAG) species in LTS were POO, POP and PLO. Total tocopherol, ${\gamma}$-oryzanol and phytosterol contents in the LTS were 12.37, 0.43 and 251.38 mg/100 g, respectively. Hardness of LTS was similar to that of commercial shortening. Also, x-ray diffraction analysis showed coexistence of ${\beta}'$ and ${\beta}$ form in the LTS.

Development of a Predictive Growth Model of Staphylococcus aureus and Shelf-life Estimation of Cooked Mung Bean Sprouts Served in School Foodservice Operations (학교급식에서 제공되는 숙주나물의 Staphylococcus aureus 성장예측모델 개발 및 섭취유효기간 설정)

  • This study was conducted to estimate the shelf-life of cooked mung bean sprouts contaminated with Staphylococcus aureus according to storage temperatures after cooking in school foodservice operations. A predictive growth model of S. aureus in cooked mung bean sprouts prepared using a standard recipe was developed at 4 storage temperatures (5, 15, 25, and 35${^{\circ}C}$). To determine the effect of vinegar on the shelf-life of cooked mung bean sprouts, the growth of S. aureus in sprouts prepared using vinegar and the standard recipe were compared. The $R^2$ values of the specific growth rate (SGR) and lag time (LT) determined using the Gompertz model were greater than 0.90 at all temperatures except 5${^{\circ}C}$, which confirmed that it would be appropriate to use these parameters for a secondary model. The secondary model, which indicates changes in LT and SGR values according to storage temperatures, was calculated using response surface models. The compatibility of the developed model was confirmed by calculating $R^2$, Bf, Af and MSE values as statistic parameters. The $R^2$ values of LT and SGR were 0.94 or higher, and the MSE, Bf and Af values were 0.02 and 0.002, 0.97 and 1.03, and 1.31 and 1.10, respectively, with high statistical compatibility. The growth rate of S. aureus was higher when the standard recipe was used than when vinegar was used at all temperatures. Indeed, no growth of S. aureus was observed in mung bean sprouts prepared using vinegar. Based on the model developed, cooked mung bean sprouts prepared using the standard recipe for school foodservice should be stored at 10${^{\circ}C}$ or less. Additionally, sprouts stored at 25 or 35${^{\circ}C}$ should be consumed within 6 or 12 hours after cooking. Finally, the addition of vinegar will prevent the growth of S. aureus in cooked mung bean sprouts.

Annual Variations of Litterfall Production in a Broadleaved Deciduous Forest at the Mt. Keumsan LTER Site (금산 장기생태연구 조사지 낙엽활엽수림 낙엽낙지량의 연변동)

  • Litterfall production represents a major contribution of carbon and nutrient cycling in forest ecosystems. This study was carried out to determine the litterfall production in a broadleaved deciduous forest at the Mt. Keumsan Long Term Ecological Research (LTER) site, Southern Korea. Littefall was collected monthly or bimonthly from the site for 7 years from 2004 to2010. Leaf and reproductive (catkins) litters showed a seasonal variation, but litters of needle, branch, and barks were not changed across the seasons. Annual leaf litter of Quercus serrata and Carpinus laxiflora were significantly different (p<0.05) but that of C. cordata, Chamaecyparis obtusa, and Pinus thunbergii was not significantly changed for 7 years (p>0.05). Annual average litterfall production was 5,223 kg/ha, but annual variations were very large with minimum of 4,110 kg/ha/yr in 2004 and maximum of 6,002 kg/ha/yr in 2007. Total litterfall comprised of 2,323 kg/ha/yr in Q. serrata, 442 kg/ha/yr in C. laxiflora, 157 kg/ha/yr in C. cordata, 131 kg/ha/yr in Acer pseudosieboldianum, 390 kg/ha/yr in other deciduous tree species, 74 kg/ha/yr in P. thunbergii, 37 kg/ha/yr in C. obtusa, 672 kg/ha/yr in branches, 515 kg/ha/yr in miscellaneous, 448 kg/ha/yr in reproductive parts, and 54 kg/ha/yr in barks. respectively. The results indicate that litterfall production of the Mt. Keumsan LTER site was yearly fructurated with the positive linear relationship between leaf or total litterfall and annual mean temperature if no disturbance such as a typoon, and was lower than that of other Korean LTER sites.

Changes in the Quality of Peeled Chestnut Achieved by Browning Inhibition Treatments During Storage (갈변저해제 처리에 따른 저장 중 박피밤의 품질 변화)

  • The effects of dippin treatments using Glycyrrhiza uralensis extract (0.5%, 1.0%, 2.0%) and calcium chloride (0.5%, 1.0%, 2.0%) on the quality of peeled chestnuts were investigated. Following dipping treatment, peeled chestnuts were vacuum-packed using a 75-㎛ polyethylene PE and nylon film, and stored in a 0 ℃ incubator for six weeks. The dipping treatments of the peeled chestnuts successfully achieved browning inhibition. The browning degree following 2.0% calcium chloride treatment was the lowest at 0.68 OD. The color change (ΔE) of the peeled chestnuts was the highest (6.0) in the control, and the lowest (3.5) for the 1.0% and 2.0% calcium chloride-treated samples. G. uralensis extract and calcium chloride treatments did not impact weight, moisture loss rate, firmness, or the soluble solid content of the peeled chestnuts following storage. The decay rate was 12.0% in the control group, and 11.0%, 11.5%, and 11.0% for G. uralensis treatment at 0.5%, 1.0%, and 2.0% calcium chloride treatment, respectively, and 13.0%, 9.5%, and 9.0% at 0.5%, 1.0%, and 2.0% calcium chloride treatment, respectively. Sensory evaluation (palatability, off-odor) showed that a 2.0% G. uralensis extract treatment presented excellent results during the storage period. Texture and color indicated no differences as a result of the browning inhibition treatments. Therefore, when considered comprehensively, a 2.0% G. uralensis extract treatment was shown to be effective for maintaining the quality and providing browning inhibition of peeled chestnuts. This result isexpected to solve the problem of quality deterioration in the form of sour taste, which is a problem in chemical processing.

Yields and Fruit Quality of Different Aged Walnut Trees (Juglans sinensis Dode) Derived from Seedlings in Gimcheon (김천지역 실생묘 유래 호두나무의 수령별 수확량 및 과실 품질)

  • This study was conducted on cultivation farms in the Gimcheon area, which is the main production area of walnuts, in order to compare and analyze the yield and fruit quality of walnut trees (Juglans sinensis Dode) derived from seedlings. The official trees were classified into nine classes from 10 to 60 years old, and a total of 135 trees were selected, with 15 trees selected for each grade. The growth and fruit characteristics for each age group were then examined. The average tree height was 10.3 m, and the average crown area was 63.7 ㎡. The average number of fruit settings per tree was 573.1, with the largest number of walnuts per tree at 35 years (974.0 walnuts), and the lowest walnuts per tree at 10 years (124.7 walnuts). In addition, the number of fruit settings per bearing mother branch in 35 year old trees was the highest at 3.1. The average weight for each fruit type was found to be 58.7 ± 4.0 g of walnuts in green peel, 10.5 ± 0.5 g of dried shell walnuts, and 4.9 ± 0.2 g of walnuts, respectively. The average yield by fruit type was 32.5 ± 13.8 kg of walnuts in green peel, 5.9 ± 2.5 kg of dried shell walnut, and 2.8 ± 1.2 kg of walnuts. The yield was positively correlated with the crown area, the number of fruits setting per bearing mother branch, and the number of fruits setting per 1 ㎥. The formula used to estimate the yield per tree by age was calculated as y = 8993.9 ln (tree age-9) 8230.3, R2 = 0.8531 (walnut in green peel) and y = 1647.1 ln (tree age-9) 1442.1, R2 = 0.8527 (dried shell walnut).

Risk Assessment of Pine Tree Dieback in Sogwang-Ri, Uljin (울진 소광리 금강소나무 고사발생 특성 분석 및 위험지역 평가)

  • Extreme weather events, such as heat and drought, have occurred frequently over the past two decades. This has led to continuous reports of cases of forest damage due to physiological stress, not pest damage. In 2014, pine trees were collectively damaged in the forest genetic resources reserve of Sogwang-ri, Uljin, South Korea. An investigation was launched to determine the causes of the dieback, so that a forest management plan could be prepared to deal with the current dieback, and to prevent future damage. This study aimedto 1) understand the topographic and structural characteristics of the area which experienced pine tree dieback, 2) identify the main causes of the dieback, and 3) predict future risk areas through the use of machine-learning techniques. A model for identifying risk areas was developed using 14 explanatory variables, including location, elevation, slope, and age class. When three machine-learning techniques-Decision Tree, Random Forest (RF), and Support Vector Machine (SVM) were applied to the model, RF and SVM showed higher predictability scores, with accuracies over 93%. Our analysis of the variable set showed that the topographical areas most vulnerable to pine dieback were those with high altitudes, high daily solar radiation, and limited water availability. We also found that, when it came to forest stand characteristics, pine trees with high vertical stand densities (5-15 m high) and higher age classes experienced a higher risk of dieback. The RF and SVM models predicted that 9.5% or 115 ha of the Geumgang Pine Forest are at high risk for pine dieback. Our study suggests the need for further investigation into the vulnerable areas of the Geumgang Pine Forest, and also for climate change adaptive forest management steps to protect those areas which remain undamaged.

Soil CO2 Efflux Dynamics in Response to Fertilization in Pinus densiflora and Quercus variabilis Stands (소나무와 굴참나무 임분의 시비에 따른 토양 CO2 방출 동태)

  • This study compared soil CO2 efflux rates after fertilization, in Pinus densiflora and Quercus variabilis stands. Compound fertilizers were applied to the forest floor in March 2016, following a one-year calibration period (from March 2015 to February 2016). In situ soil CO2 efflux rates were measured every month during the two-year study periods, using an infrared gas analyzer with a closed chamber system. Mean annual soil CO2 efflux rates were higher following fertilizer application in the P. densiflora and Q. variabilis stands (P. densiflora: 2.180 μmol m-2 s-1; Q. variabilis: 1.977 μmol m-2 s-1) as compared with the rates measured during the calibration period (P. densiflora: 1.620 μmol m-2 s-1; Q. variabilis: 1.557 μmol m-2 s-1). The mean annual soil CO2 efflux rates in the unfertilized treatments of both stands were not significantly different between the two-year study periods. The Q10 values of fertilized treatments in Q. variabilis stands were higher in the fertilization period (3.41) than in the calibration period (3.14), whereas the Q10 values in P. densiflora stands did not change between the fertilization and calibration periods. The Q10 values of unfertilized treatments in the Q. variabilis stands were lower during the 2016-2017 period (3.69), than in the 2015-2016 period (3.85), whereas the Q10 values in P. densiflora stands were higher during the 2016-2017 period (3.65), than in the 2015-2016 period (3.15). These results indicate that the increase in soil CO2 efflux rates in P. densiflora stands could be more sensitive to fertilization compared with the rates in Q. variabilis stands.

Evaluation of Perception for Foodservice Hygiene by Middle School Students in Busan (부산지역 일부 중학생의 학교급식 위생에 대한 인식 평가)

  • The purpose of this study was to evaluate the importance and performance of middle school foodservice hygiene in Busan. In this study, questionnaires were administered to 1,000 students consisting of 100 students in 10 middle schools in Busan in December, 2013. The students assessed importance and performance of personal hygiene as 3.81 and 3.60 on a scale of 5.00, respectively. Males gave higher scores to importance and performance of personal hygienic than females. First grade students gave significantly higher scores than 2nd and 3rd grade students to importance and performance of personal hygienic. The mean scores of importance of foodservice hygiene were 4.47 for food hygiene, 4.25 for equipment hygiene, 4.23 for employee hygiene, and 4.19 for environment hygiene. For importance of foodservice hygiene, females gave significantly (P<0.001) higher scores than males. However, male gave significantly higher scores than females for performance of foodservice hygiene. The importance and performance grid showed important factors for personal and foodservice hygiene. 'Hand washing before eating the meal' for personal hygiene and 'cleanliness of tray' for foodservice hygiene showed the highest importance and lowest performance scores. Foodservice hygiene can be improved through customized education of students regarding meal service and food hygiene, equipment hygiene, and environment hygiene.
