Effects of Artemisia Capillaris Fructus on Fas-FasL-induced Apoptosis in Hepatocye (인진호(茵蔯蒿)가 Fas-FasL 매개형 간세포 Apoptosis에 미치는 영향)

  • Objectives: Recently, it was known that the major cause of hepatitis is apoptosis reaction mediated by Fas-FasL. Since Artemisia Capillaris Fructus has long been applied to cure the jaundice in oriental medicine. Therefore, this study was carried out to examine the effect of fractions of Artemisia Capillaris Fructus on Fas-FasL-mediated apoptosis in hepatocytes. Methods: This study employed propidium iodide negative cell count assay and some the other biochemical assays. Results : This study confirms that hepatitis has been occured by apoptosis mediated by Fas-FasL in cultured hepatocyte and fractions of Artemisia Capillaris Fructus restrain apoptosis induced Fas-FasL. Conclusions : Water-extracted fraction, methanol extracts, ether-soluble fraction, and buthanol-soluble fractions of Artemisia Capillaris Fructus restrain Fas-FasL-mediated apoptosis in hepatocyte. Silica gel chromatograph of Buthanol-soluble fraction of Artemisia Capillaris Fructus restrain Fas-FasL-mediated apoptosis in hepatocyte. Artemisia Capillaris Fructus could be applied to cure hepatitis.

Studies on the Amino Acid, Sugar Analysis and Antioxidative Effect of Extracts from Artemisia sp. (쑥 추출물의 아미노산, 당 분석 및 항산화 효과에 관한 연구)

  • This study has attempted to examine the antioxidative effect of ethanol extracts, and the analysis of amino acid and sugar of hot water extracts from Artemisia princeps, Artemisia argyi and Artemisia iwayomogi. Total phenol contents were 10.2 mg/$m\ell$, 4.7 mg/$m\ell$, and 7.0 mg/$m\ell$ in A. princeps, A. argyi and A. iwayomogi, respectively. Also, flavonoid contents were 6.1 mg%, 3.6 mg%, and 1.8 mg% in A. princeps, A. argyi and A. iwayomogi, respectively. Hot water extract from A. argyi showed highest electron donating ability toward DPPH. The contents of total amino acid by HPLC analysis were in the increasing order of A. princeps, A. argyi and A. iwayomogi. Of amino acid compositions, proline was largest in examined Artemisia sp.. The contents of total sugar by HPLC analysis were largest in A. iwayomogi. Of the sugar compositions, the glucose content was largest in examined all Artemisia sp., and the fructose content was in the increasing order of A. iwayomogi, A. argyi and A. princeps.

Comparison of Anti-inflammatory effects between Artemisia capillaris and Artemisia iwayomogi by extraction solvents (인진호(茵蔯蒿)와 한인진(韓茵蔯)의 추출용매별 항염증 효능 비교)

  • Objectives : Artemisia capillaris Thunberg (AC) and Artemisia iwayomogi Kitamura (AI) have been used without distinguishment since ancient times due to similar appearance. In this study, we compared the inhibitory effects of AC and AI on the expression of inflammatory cytokines induced by lipopolysaccharide (LPS) in murine macrophages. Methods : AC and AI were extracted by reflux with distilled water (DW) and 70% ethanol (EtOH). We investigated the inhibitory effects of AC and AI on the expression of nitric oxide (NO), inducible NO synthase (iNOS) and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) induced by LPS in macrophages. Results : Firstly, yield of the samples was higher in order of Artemisia iwayomogi DW Extract (AID), Artemisia iwayomogi 70% EtOH Extract (AIE), Artemisia capillaris DW Extract (ACD) and Artemisia capillaris 70% EtOH Extract (ACE). All of the samples were not toxic in macrophages. The inhibitory effect of the samples on LPS-induced NO expression was stronger in the order of AIE, ACE, AID and ACD. The inhibitory effect of the samples on LPS-induced inducible iNOS expression was stronger in the order of AIE, ACE and AID. Effect of ACD was same with that of AID. In addition, inhibitory effect of the samples on LPS induced $TNF-{\alpha}$expression wes stronger in the order of AIE, ACE, AID and ACD. Conclusion: These results showed that AI would be more effective than AC and 70% EtOH would be more effective than DW as an extraction solvent in inflammatory diseases.

Effect of Artemisia Capillaris Thunberg EtOH Ext. on Lowering Lipid, Anti-oxidation and Concentration of Plasma Inflammatory Mediators Using Rats Fed on High-oxidized Fat (인진호(茵蔯蒿) 추출물이 과산화지질 투여한 쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향)

  • Objectives : The present study investigated effects of Artemisia Capillaris Thunberg ethanol extract(EtOH ext). on lowering lipid, anti-oxidation and concentration of plasma inflammatory mediators using rat fed on high oxidized fat. Methods : We divided fat sprague-dawley rats fed on high oxidized into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 4 weeks. We measured concentration of plasma free fatty acid(FFA), plasma triglyceride, plasma total cholesterol, and plasma low density lipoprotein-cholesterol(LDL-cholesterol), plasma high density lipoprotein-cholesterol(HDL-cholesterol), concentration of liver total cholesterol and liver triglyceride (TG), concentration of plasma thiobarbituric acid reactive substance(TBARS) and liver thiobarbituric acid reactive substance(TBARS), glutathione peroxidase (GSH-Px) activity, superoxide dismutase(SOD) activity and catalase(CAT) activity, plasma nitric oxide(NO), ceruloplasmin and ${\alpha}-glycoprotein$. Results : 1. The Artemisia Capillaris Thunberg EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDL-cholesterol was increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 2. Concentration of liver total cholesterol and liver TG showed a significantly decrement in all Artemisia Capillaris Thunberg EtOH ext. groups than that of control group. 3. The Artemisia Capillaris Thunberg EtOH ext. groups showed lower values in concentration of plasma TBARS and liver TBARS than that of control group. The values of GSH-Px activity, SOD activity and CAT activity were increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 4. The values of plasma NO, ceruloplasmin and ${\alpha}-glycoprotein$ were decreased in Artemisia Capillaris Thunberg EtOH ext. groups. Conclusions : Based on the results in this study, the Artemisia Capillaris Thunberg EtOH ext. showed a positive effect in lowering lipid, anti-oxidation and decrement of plasma inflammatory mediators.

Studies on the Volatile Oil Constituents in Artemisia sp. -Isolation and Determination of Camphor by Gas Chromatography- (Artemisia 속 정유 성분에 관한 연구 -Gas Chromatography에 의한 Camphor의 분리 및 정량-)

  • The contents of volatile oil and camphor in the oil of Artemisia messer-schmidtiana $B_{ESSER}$ var. viridis $B_{ESSER}$ f. typica $N_{AKAI}$ and Artemisia asiatica $N_{AKAI}$ which were known to be effective as antibacterial, antifungal and antineoplastic agents by screening experimentation, were determined quantatively by gas chromatography. The volatile oil contents of Artemisia messer-schmidtiana $B_{ESSER}$ var. viridis $B_{ESSER}$ f. typica $N_{AKAI}$ and Artemisia asiatica $N_{AKAI}$ are 0.17% and 0.19%, and the camphor contents are 8.51% and 1.46%, respectively.

  • Effects of the polysaccharide fractions purified from Artemisia selengensis and Artemisia iwayomogi on the immune system was studied. The polysaccharide fractions, respectively called ASP1 and AIP1, may interact with macrophages and lymphocytes in spleen, increasing the population of those cell types in vivo and in vitro. Both ASP1 and AIP1 fractions also syuppress transplanted tumor cell growth and augment antibody production. This study suggests that ASP1 and AIP1 fractios may have immunomodulating and antitumor activities.

  • Extraction of Natural Artemisia by using super critical carbon dioxide is operated under $40-50^{\circ}C$ and 200-250 atm, thus, conversion rate is very low as 7wt% while high energy is being wasted. When Natural Artemisia Extract is applied in the control of microbe, concentration and reaction time greatly affected to the growth of microbe. Especially, when refined oil concentration from Natural Artemisia added more than 1,000ppm, staphylococcus aureus and fungi are terminated completely. Thus, it is proven that Natural Artemisia Extract has antimicrobial effect.

  • Objectives : The purpose of this study is to evaluate the effect of Artemisia capillaries on gene expression of lipid metabolism in rats. Methods : The author performed several experimental items to analyze the total cholesterol and triglyceride in liver tissue, the gene expressions of CYP7A1 and HMG-CoA reductase. Results : 1. In Artemisia capillaries group, the levels of total cholesterol in liver tissue were significantly decreased. 2. In Artemisia capillaries group, the ratios of CYP7A1, HMG-CoA reductase were as same as the normal group. Conclusions : From the above results, Artemisia capillaries can be used to treat hyperlipidemia.

  • A method was described for the analysis of Artemisia extract in cosmetic soap. Esculetin-6-methylether was used as indicator ingredient for analysis of Artemisia extract. It was isolated from the plant and purified with silicagel column. The structure was conformed with IR, NMR and MASS spectroscopy. The Artemisia extract in sample was isolated with chloroform and interfering com- pounds were eliminated with silicagel column. The Artemisia extract was determined by reverse phase high- performance liquid chromatography with a fluorescence detector and a solvent system of H2O/THF/MeOH. The recorveries of Artemisia extract were 93.9-106.1% in the cosmetic soap.

Effect of Several Zinc Solutions on Concentration of Oral Volatile Sulfur Compounds(VSCs) (수종의 Zinc 수용액이 구강내 휘발성 황화합물의 농도에 미치는 영향)

  • The purpose of this study is to investigate effects of several zinc solutions including Artemisia asiatica-containing zinc solution on concentration of oral volatile sulfur compounds(VSCs). We determined the VSCs concentration of breath of human subjects before and after use of zinc solutions(O.25% $ZnF_2$ Artemisia asiatica-containing 0.25% $ZnCl_2$ and Artemisia asiatica-containing 0.25% $ZnCl_2$ solutions) The results were as follows : 1. 0.25% $ZnCl_2$ solution was more effective than 0.25% $ZnF_2$ solution in reducing the concentration of oral VSCs and the maintenance duration of effectiveness. 2. Artemisia asiatica-containing 0.25% $ZnCl_2$ solution was more effective than Artemisia asiatica-containing 0.25% $ZnF_2$ solution in reducing the concentration of oral VSCs and the maintenance duration of effectiveness. 3. Artemisia asiatica-containing 0.25% $ZnF_2$ solution and 0.25% $ZnF_2$ solution showed no significant difference in reducing the concentration of oral VSCs and the maintenance duration of effectiveness but, Artemisia asiatica-containing 0.25% $ZnF_2$ solution was slightly more effective. 4. Artemisia asiatica-containing 0.25% $ZnCl_2$ solution and 0.25% $ZnCl_2$ solution showed no significant difference in reducing the concentration of oral VSCs and the maintenance duration of effectiveness but, Artemisia asiatica-containing 0.25% $ZnF_2$ solution was slightly more effective.
