  • In mammalian, cytochrome P4501A1 (CYP1A1) is very important for metabolism of xenobiotics such as PAHs(Polycyclic aromatic hydrocarbon) and heterocyclic amine, and it is induced by environmental contaminants such as PAHs, TCDD(2,3,7,8-tetrchlorodibenzo-p-dioxin) and 3-MC (3-methylcholanthrene). In fish, like mammalian, when it is exposed to environmental contaminants, they cause specific and sensitive induction of CYP1A. Therefore, induction of CYP1A in fish and mammalian is widely used as a biomarker for exposure of environmental contaminants. In this study, to compare the function of Cyp1a1 in fish with it in mammalian, we have used rainbow trout(Oncorhynchys mykiss) hepatoma cells (RTH-149) and mouse hepatocyte (Hepa-I). in order to examine induction of Cyp1a1 by TCDD, we have used the bioassay system. We examined effects of TCDD on the Cyp1a1-luciferase reporter gene activity, 7-ethoxyresorufin O-deethylase(EROD) activity and Cypa mRNA level.

  • Volatile flavor compounds in Korean salt fermented shrimp on the market were analyzed by vacuum simultaneous distillation solvent extraction/gas chromatography/mass spectrometry/olfac tometry(V SDE/GC/MS/O) and aroma extract dilution anlaysis(AEDA). A total of 32 volatile com pounds were detected by GC/O analysis. Of these, 18 were positively identified, and composed of S containing compounds(5), aldehydes(4), ketones(3), N containing compounds(3), ester(1), alcohol(1) and aromatic hydrocarbon(1). Predominant odorants(Log3FD$\geq$4) in salt fermented shrimp were 2,3 butanedione(sour/buttery), 1 octen 3 one(earthy/mushroom like), dimethyl trisulfide(cooked cabbage /soy sauce like) and 2 acetylthiazole(grainy/nutty). Predominant free amino acids were aspartic acid, glutamic acid(sour and umami taste), arginine, methionine(bitter) and lysine(sweet and bitter) in evaluation of taste value.

BRACKETT LINE-BASED MBH ESTIMATORS AND HOT DUST TEMPERATURES OF TYPE 1 AGNs FROM AKARI SPECTROSCOPIC DATA

  • We provide results of near-infrared (NIR) spectroscopic observations of 83 nearby (0.002< z <0.48) and bright (K <14 mag) type 1 active galactic nuclei (AGNs). For the observations, we used the Infrared Camera (IRC) on AKARI allowing us to obtain the spectrum in the rarely studied spectral range of $2.5-5.0{\mu}m$. The $2.5-5.0{\mu}m$ spectral region suffers less dust extinction than ultra violet (UV) or optical wavelength ranges, and contains several important emission lines such as $Br{\beta}$ ($2.63{\mu}m$), $Br{\alpha}$ ($4.05{\mu}m$), and polycyclic aromatic hydrocarbon (PAH; $3.3{\mu}m$). The sample is selected from the bright quasar surveys of Palomar Green and SNUQSO, and AGNs with black hole (BH) masses estimated from reverberation mapping method. We measure the Brackett line properties for 11 AGNs, which enable us to derive BH mass estimators and investigate circum-nuclear environments. Moreover, we perform spectral modeling to fit the hot and warm dust components by adding photometric data from SDSS, 2MASS, WISE, and ISO to the AKARI spectra, and estimate hot and warm dust temperatures of ~1100K and ~220 K, respectively.

Gene-Diet Interaction on Cancer Risk in Epidemiological Studies

  • Genetic factors clearly play a role in carcinogenesis, but migrant studies provide unequivocal evidence that environmental factors are critical in defining cancer risk. Therefore, one may expect that the lower availability of substrate for biochemical reactions leads to more genetic changes in enzyme function; for example, most studies have indicated the variant MTHFR genotype 677TT is related to biomarkers, such as homocysteine concentrations or global DNA methylation particularly in a low folate diet. The modification of a phenotype related to a genotype, particularly by dietary habits, could support the notion that some of inconsistencies in findings from molecular epidemiologic studies could be due to differences in the populations studied and unaccounted underlying characteristics mediating the relationship between genetic polymorphisms and the actual phenotypes. Given the evidence that diet can modify cancer risk, gene-diet interactions in cancer etiology would be anticipated. However, much of the evidence in this area comes from observational epidemiology, which limits the causal inference. Thus, the investigation of these interactions is essential to gain a full understanding of the impact of genetic variation on health outcomes. This report reviews current approaches to gene-diet interactions in epidemiological studies. Characteristics of gene and dietary factors are divided into four categories: one carbon metabolism-related gene polymorphisms and dietary factors including folate, vitamin B group and methionines; oxidative stress-related gene polymorphisms and antioxidant nutrients including vegetable and fruit intake; carcinogen-metabolizing gene polymorphisms and meat intake including heterocyclic amins and polycyclic aromatic hydrocarbon; and other gene-diet interactive effect on cancer.

Dietary Fiber Reduces Benzo[a]pyrene Hydroxylase Induced by Dietary Benzo[a]pyrene

  • This study was conducted to determine if dietary fiber would reduce exposure of the tissues to dietary benzo[a]pyrene(BP) , a well-known carcinogenic polycyclic aromatic hydrocarbon, as evaluated by benzo[a]pyrene hydroxylase (BPH) activity. The effects of three different sources of dietary fiber(pectin, polydextrose, and clellulose) on BPH activity were studied using Sprague-Dawley rats. In this study, male rats were fed a fiber-free purified diet for 7 days, whereupon they were switched to experimentla diets for 48h. After 48h, their liver, stomach , small intestinal mucosa and large intestinal mucosa were assayed for BPH activity. Thissues exposed to benzo[a]pyrene(400mg/kg diet, fiber-free) showed significant increse in the activity of BPH ; 27 times in liver, 7 times in stomach, 18 times in small intestinal mucosa and 3 times in large intestine. The inhibition in BP -induced BPH activity by dietary fiber in liver, stomach and small intestinal mucosa was observed in the decreasing order : 10 % perctin > 10% polydextrose >5 % polydextrose > 10% cellulose. Decreased BPH induction indicates that soluble dietry fibers, especially pectin and polydextrose in this study, protect the tissues of digestive system from exposure to BP.

Preparation of radiolabeled polycyclic aromatic hydrocarbon assemblies for biological assessment of diesel exhaust particulates

  • The potential health risk from inhalational exposure of diesel exhaust particulates (DEP) has gained considerable scientific interests. However, the long-term in vivo behavior of DEP have not been clearly understood due to the difficulty of accurate analysis of these substances in a living subject. We herein demonstrate a detail protocol for the preparation of radiolabeled DEP using a radioactive-iodine-tagged pyrene analog. The purified $^{125}I$-labeled pyrene ($[^{125}I]1$) was obtained with a good radiochemical yield ($32{\pm}4%$, n=3) and high radiochemical purity (>99%) from the stannylated precursor 2. Next, the purified $[^{125}I]1$ was successfully assembled into the DEP suspension in an efficient manner. The radiolabeled DEP was highly stable in a mouse serum for 7 days without significant deiodination or dissociation of $[^{125}I]1$. These results clearly indicate that the present radiolabeling method will be useful for biodistribution study of carbonaceous particulates in vivo.

다채널 액상 미세 추출 시스템 설계 및 제작 (Multichannel Liquid Phase Microextraction System)

  • In this study, a multichannel gas-liquid microextraction system is designed by integrating the automatic elution of extraction line and multichannel gas-purging liquid phase microextraction. The system uses an injection pump and inert gas to push the extraction solvent to a sample bottle of a gas-phase color autosampler and then implements multichannel gas-liquid microextraction and gas chromatography-mass spectrometry. The system also employs a three-way integrated micro-high-temperature heater, syringe pump, and microflow controller to realize the simultaneous processing of multiple groups of samples, thus improving the sample pretreatment speed and reproducibility and reducing human error. Autoinjection experiments were implemented with polycyclic aromatic hydrocarbon standard samples. The experiments show that the average recovery rate of the system exceeds 70%, and the relative standard among the channels is less than 15%.

The Effects of Liquid Butadiene Rubber and Resins as Processing Aids on the Physical Properties of SSBR/Silica Compounds

  • Highly aromatic (HA) oils are common processing aids used in tire tread compounds. However, they often bleed and evaporate from the vulcanizates during tire use. Thus, the mechanical and dynamical properties of the tire decrease. To overcome this problem, we investigated nonfunctionalized liquid butadiene rubber (LBR-305, Kuraray) and center-functionalized liquid butadiene rubber (C-LqBR), polymerized by anionic polymerization. In addition to the liquid butadiene rubbers, p-tert-octylphenol (P-Resin) and C5 hydrocarbon (H-Resin) tackifier resins, which can induce entanglement of rubber compounds, were researched as a processing aid to solve the bleeding problem. Liquid butadiene rubbers have significantly reduced extraction loss by crosslinking with the main rubber chain. They have also increased the abrasion resistance and showed similar or better mechanical and dynamical properties against HA oils. However, resin compounds did not show differences in extraction loss compared to HA oil compounds; instead, they showed increased wet traction.

The Measurement of Flash Point for Binary Mixtures of 2,2,4-Trimethylpentane, Methylcyclohexane, Ethylbenzene and p-xylene at 101.3 kPa

  • Laboratories and industrial processes typically involve the use of flammable substances. An important property used to estimate fire and explosion risk for a flammable liquid is the flash point. In this study, flash point data at 101.3 kPa were determined using a SETA closed cup flash point tester on the following solvent mixtures: {2,2,4-trimethylpentane + methylcyclohexane}, {2,2,4-trimethylpentane + ethylbenzene}, and {2,2,4-trimethylpentane + p-xylene}. The purpose of this work is to obtain flash point data for binary mixtures of 2,2,4-trimethylpentane with three hydrocarbons (methylcyclohexane, ethylbenzene, and p-xylene), which are representative compounds of the main aromatic hydrocarbon fractions of petroleum. The measured flash points are compared with the predicted values calculated using the GE models' activity coefficient patterns: the Wilson, the Non-Random Two-Liquid (NRTL), and the UNIversal QUAsiChemical (UNIQUAC) models. The non-ideality of the mixture is also considered. The average absolute deviation between the predicted and measured lower flash point s is less than 1.99 K, except when Raoult's law is calculated. In addition, the minimum flash point behavior is not observed in any of the three binary systems. This work's predicted results can be applied to design safe petrochemical processes, such as identifying safe storage conditions for non-ideal solutions containing volatile components.

Effect of Methyl Gallate on 1-Nitropyrene-Induced Keratinocyte Toxicity in a Human and Canine Skin Model

  • The skin, which is the largest organ of the human body, is in direct contact with pollutants in the surrounding atmosphere. Meanwhile, 1-nitropyrene (1-NP), the most abundant nitro-polycyclic aromatic hydrocarbon found in particulate matter, is known to have carcinogenic effects; however, studies on its toxicity in human and canine skin are still needed. In this study, we investigated 1-NP-induced apoptosis and inflammatory pathways in HaCaT cells. In addition, we also measured the cytoprotective effect of methyl gallate (MG), which is widely distributed in medicinal and edible plants and is well known for its anti-inflammatory and antioxidant properties. MG inhibited 1-NP-induced cell death and apoptosis pathways, including the cleavage of PARP and activation of caspase-3, -7, and -9. MG also suppressed 1-NP-induced COX-2 expression and phosphorylation of mitogen-activated protein kinases (MAPKs) and MAPK kinases (MAPKKs). Our findings suggest that 1-NP induces skin toxicity in human and canine through apoptosis and inflammatory responses, and moreover, that this can be prevented by treatment with MG.
