The Effect of Radix Angelicae gigantis aqua-acupuncture on the hypothyroidism induced by thiourea in rats (당귀약침(當歸藥鍼)이 흰쥐의 갑상선기능(甲狀腺機能)에 미치는 영향(影響))

  • In order to study the effect of aqua-acupuncture with Radix Angelicae gigantis extract solution by acuncture point. Radix Angelicae gigantis aqua-acupuncture was carried on every day for 3 days and 5 days 7 days respectively on the corresponding loci of Chondol(CV22) and non-acupuncture points (loci at the root of tail), after inducing hypothyroidism by thiourea administration and thereafter the contents of serum triiodothyronine$(T_3)$, thyroxine$(T_4)$, triiodothyronine uptake, free $T_3$, free $T_4$, total protein, albumin, creatinine, creatine, BUN, cholesterol, triglyceride, and total lipid were measured in hypothyroidism rats. The following results have been obtained : 1. $T_3$ was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days Chondol(CV22) and non-acupuncture points as compared with the control group. 2. $T_3$ uptake was increased with statistical significance by Radix Angelicae gigantis aquq-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22) as compared with the control group. 3. Free $T_4$ was increased with statistical significance by Radix angelicae gigantis aqua-acupuncture for 3 days, 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 4. Albumin was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 5. Creatine was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22), and non-acupuncture points respectively as compared with the control group. 6. Cholesterol was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 7. Triglyceride was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group. 8. Total lipid was decreased with statistical significance by Radix Angelicae gigantis aqua-acupuncture for 3 days, and 5 days and 7 days to Chondol(CV22) and non-acupuncture points respectively as compared with the control group.

Study on the Anti-cancer, Anti-metastasis and Effects of Immune-response of Aqua-acupuncture with Cuscutae Semen Infusion Solution (토사자약침(兎絲子藥鍼)의 항암작용(抗癌作用) 및 면역효과(免疫效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Objective : The objective of tihs study is to study the effects of anti-cancer, anti-metastasis and effects of immune-response of aqua-acupuncture with Cuscutae Semen infusion solution. Methods : We observed, in-vitro, the cytotoxicity and the effect on the expression of MMP-9 gene, and in-vivo, change of body weight, surviving number, MST, ILS, changes in amount of WBC, RBC, PLT, GOT, GPT, creatinine, glucose and LDH, number of Pulmonary colony. Results : 1. The effect on expression of MMP-9 gene was decreased in all the sample groups in B16-F10 cell line, and was decreased in Lane 1, 2, 3 in HT1080 cell, compared with control group. 2. BALB/c mice which was transpianted S-180 cancer cell line were inhibited significantly in weight increase, in all the sample groups, compared with control group. 3. The sample groups injected in vein with B16-F10 cancer cell line in C57BL/6 mice did'nt show significant change in the number of WBC, RBC, PLT. 4. In immune experiment, all the sample groups showed having more relevancy to the effect on splenic cell proliferation than normal groups. 5. $IFN-{\gamma}$ and $TNF-{\alpha}$ in cytokine-gene were increased in all the sample groups than control group. 6. In flow cytometry of spienic cell, the numbers of CD4+ cell, CD8+ cell and CD19+ cell in sample groups were increased than in control group. Conclusion : Above the results showed that aqua-acupuncture of Cuscutae Semen infusion solution has effects of anti-cancer, anti-metastasis and immune response improvement.

  • Induction of phase II enzymes such as quinone reductase (QR) or glutathione S-transferase (GST) is considered a major mechanism of protection against initiation of carcingenesis. This study was desinged to investigate the potential of Astragali Radix Aqua-acupuncture Solution (ARAS) to induce phase II enzymes and glutathione (GSH) in murine hepatoma cells grown in microtiter plate wells. ARAS was potent inducers of QR activity. ARAS was induced about 2.6-fold at concentration of $5{\times}$. In addition, GST activity was increased with ARAS. GSH levels were increased about 1.2-fold with ARAS at concentration of $0.1{\times}$. These results suggested that ARAS may act as blocking agents against carcinogenesis by induction of phase II marker enzymes.

Chemopreventive Effect of Gamdutang Aqua-Acupuncture Solution (감두약침액의 암예방 효과)

  • Gamdutang aqua-acupuncture solution (GAS), Gamdutang water-extracted solution (GWS) and Dae-Gamdutang aqua-acupuncture solution (DGAS) were prepared and tested for chemopreventive potentials. GAS was potent inducer of quinone reductase (QR) activity in Heapa1c1c7 murine hepatoma cells in culture, whereas GWS is less potent. GAS, GWS and DGAS were significantly induced QR activity in cultured rat normal liver cell, Ac2F. Glutathione (GSH) levels were increased about 1.8, 1.0 and 1.1 fold with GAS, GWS and DGAS in Hepa1c1c7 cells, respectively. In addition glutathione S-transferase (GST) activity was increased with GAS, GWS and DGAS. The effects of GAS, GWS and DGAS on the growth of Acanthamoeba castellanii were tested. Proliferation of A. castellanii was inhibited by GAS, GWS and DGAS at concentrations of 1 $\times$ and 5$\times$. These results suggest that GAS has chemopreventive potential by inducing QR and GST activities, increasing GSH levels and inhibition of polyamine metabolism.

Antioxidant Property of Aqua-Acupuncture Solution from Circium japonicum (대계 약침액의 항산화 효능)

  • Objectives : Circium japonicum is a pharmacologically active used in traditional Korean medicine. An aqua-acupuncture solution of the Circium japonicum was assessed to determine the mechanism of its antioxidant activity. Materials : Circium japonicum was obtained from a Dongguk Korean Medicine Hospital (Kyung-ju, Kyungbuk). The freeze-dry powder was collected (yield 5.1%) for the aqua-acupuncture solution. Scavenging activity on DPPH free radicals by the Circium japonicum aqua-acupuncture solution (CJAS) was assessed according to the method followed by Gyamfi et al.. and then scavenging activity orl superoxide radicals $(O_2^-{\cdot})$ was assessed by the method described by Gotoh et al. with slight modification. Deoxyribose assay to determine the rate constant for the reactions between either antioxidants and hydroxyl radicals or antioxidants and iron ions. We tested by; (1) Non-site-specific scavenging assay (hydroxyl radicals, OH), (2) Site-specific scavenging assay (chelate iron ions), and (3) Pro-oxidant effect of the CJAS on iron dependent hydroxyl radical generation. Finally, we determined hydroxyl radical-mediated DNA nicking formation. Conclusion : Our study demonstrated that CJAS has antioxidant activities and we investigated the potential effectiveness of CJAS in preventing oxidative stress-mediated disease further.

  • This experimental study was carried out to investigate on the safety of Nog-yong aqua-acupuncture solution in primary culture of adult rat hepatocytes. Primary culture of adult rat hepatocytes has been considered as a ideal model for toxicological studies because cultured hepatocytes maintained many liver-specific functions. In this research, we investigated the effects of Nog-yong aqua-aqupunture(1-10$\mu\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release assays. Hepatic glutathione level. glutathione-S-transferase activity, and albumin synthesis were not affected by treatment with Nog-yong aqua-aqupuntur alone. Nog-yong aqua-aqupuncoure solution(0.5-$10{\mu}\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release not significantly affected normal functional charaterists.

  • Taraxacunf mongofieum Hand-Mass aqua-acupuncture solution (TMAS) was prepared and investigated og, the effect on initiation of carcinogenesis. The following effe.Is as a blocking agent were measured. .(a) Indu.ction of quinone reductase, (b) Induction of glutathione S-transferase activity (c) Increase of reduced glutathione. TMAS was potent inducer of quinone reductase in Hepa Iclc7 murine hepatoma cells. Clutathione S-transferase activity was increased with TMAS. In addition glutathione levels were increased about 1.6-fold with TMAS in cultured murine hepatoma Hepa Iclc7 cells.

Effects of Cervus elaphus extract solution for Aqua-acupuncture on Antioxidation in Rat's Brain induced by ischemia and reflow (허혈(虛血)/재관류(再貫流) 뇌(腦) 조직(組織)에서 녹용(鹿茸) 약침액(藥鍼液)의 항산화작용(抗酸化作用)에 대한 연구(硏究))

  • Cervus elaphus, being known to reinforce Kidney, have tested to study the effects concerning damages of brain tissue induced by lipid peroxidation. In vitro, the level of lipid peroxide in brain tissue was decreased proportinally according to dose by Cervus elaphus extract solution for aqua-acupuncture (CESAA). It was much more decreased, when lipid peroxidation was induced with Fe(II). And, it was seen proportinally decrease according to the dose of CESAA on xanthine oxidase activities and type conversion ratio. However, I can not find special changes about aldehyde oxidase activities. And, I had observed the effects of CESAA on damages of rat's brain following ischemia and reflow. Before ischemia was caused, CESAA was applied 0.2 ml per 250 g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group. Ischemia was caused by cervical artery's clamp for 30 min and reflowed by clamp remove after 15 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CESAA was pre-appllied. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion. CESAA recovers the damage of brain due to ischemia and reflow by decreasing the lipid peroxidation through decreasing of xanthine oxidase activies and type conversion ratio.

The antimutagenic effect and genetic safety of Buthus martensi Karsch aqua-acupuncture solution (BMKAS) (전갈(全蝎) 약침액(藥鍼液)의 항돌연변이(抗突然變異) 및 항암(抗癌) 효과(效果))

  • Objective : The aim of this study is to determine the antimutagenic effect and genetic safety of Buthus martensi Karsch aqua-acupuncture solution(BMKAS) against various chemical carcinogens. Method : Ames(Salmonella typhimurium) test and Rec assay(Bacillus subtilis) were used as indicators for DNA damage and antimutagenesis. Furthermore, the levels of umu operon expression by measuring the ${\beta}$-galactosidase activity wete monitored with the SOS umu test using S. typhimurium 1535 containing plasmid pSK1002. And the host-mediated assay was used to investigate the mutagenicity and antimutagenicity of BMKAS inducing various chemical carcinogens after the activation with in vivo metabolic systems. Results : From the results, BMKAS did not atfect DNA of S. typhimurium and B. subtilis strains and showed no mutagenicity at the all concentrations of tested solution. Furthermore BMKAS dose-dependently protected the mutagenecity by AF-2, 2-AA and B[a]P. These phenomena was also similar to that after metabolic activation of BMKAS in in vivo system. Conclusion : These results suggested that BMKAS did not show the mutagenicity and protected the mutagenesis against various chemical carcinogens by four different methods used in this study.

Effect of Bupleuri Radix Aqua-Acupuncture Solution on Immune Response in Mice (시호 약침제제가 생쥐의 면역활성에 미치는 영향)

  • In order to investigate the effects of Bupleuri Radix aqua-acupuncture solution (BRAS) on immuno suppression induced by glucocorticoid, ICR mice were administrated with glucocorticoid (80 mg/kg) for 7 days, and immunized with hapten, methamphetamine-horseradish peroxidase $(10\;{\mu}g/mouse)$. And then, BRAS (0.2ml/mouse) injected into $CV_4\;and\;BL_{23}$, which are the classical acupuncture points in traditional medicine, for 7 days. And then B and T cells proliferation and cytolytic activity of natural killer (NK) cells were measured. Intraperitoneal injection of glucocorticoid decreased lysozyme activity in macrophage and cytolytic activity of NK cell. B and T cell proliferation were significantly increased in aqua-acupuncture group compared to normal group. On the other hand, BRAS significantly increased the lysozyme activity in macrophage, and the cytolytic activity of purified NK cell on K562. These results suggest that BRAS at $CV_4\;and\;BL_{23}$ may proliferate B and T cells that are suppressed by glucocorticoid and activate NK cell activity.
