In this study, phytochemicals present in Propolis Extract (PE) were employed as reducing and stabilizing reagents to synthesize silver nanoparticles. Three propolis-reduced silver nanoparticles (P-AgNPs1-3) were synthesized using increasing amounts of PE. P-AgNPs were treated with different cancer cells-lung (A549), cervix (HeLa) and colon (WiDr) - for 24, 48 and 72 h to evaluate their anti-proliferative activities. A non-cancerous cell type (L929) was also used to test whether suppressive effects of P-AgNPs on cancer cell proliferation were due to a general cytotoxic effect. The characterization results showed that the bioactive contents in propolis successfully induced particle formation. As the amount of PE increased, the particle size decreased; however, the size distribution range expanded. The antioxidant capacity of the particles increased with increased propolis amounts. P-AgNP1 exhibited almost equal inhibitory effects across all cancer cell types; however, P-AgNP2 was more effective on HeLa cells. P-AgNPs3 showed greater inhibitory effects in almost all cancer cells compared to other NPs and pure propolis. Consequently, the biological effects of P-AgNPs were highly dependent on PE amount, NP concentration, and cell type. These results suggest that AgNPs synthesized utilizing propolis phytochemicals might serve as anti-cancer agents, providing greater efficacy against cancer cells.

Background: Pereskia sacharosa is a genus of cacti widely used in folk medicine for cancer-related treatment. Anti-proliferative effects have been studied in recent years against colon, breast, cervical and lung cancer cell lines, with promising results. We here extended study of anti-proliferative effects to a blood malignancy, leukemia. Materials and Methods: Two leukemic cell lines, MV4-11 (acute myeloid leukemia) and K562 (chronic myeloid leukemia), were studied. $IC_{50}$ concentrations were determined and apoptosis and cell cycle regulation were studied by flow cytometric analysis. The expression of apoptosis and cell-cycle related regulatory proteins was assessed by Western blotting. Results: P sacharosa inhibited growth of MV4-11 and K562 cells in a dose-dependent manner. The mode of cell death was via induction of intrinsic apoptotic pathways and cell cycle arrest. There was profound up-regulation of cytochrome c, caspases, p21 and p53 expression and repression of Akt and Bcl-2 expression in treated cells. Conclusions: These results suggest that P sacharosa induces leukemic cell death via apoptosis induction and changes in cell cycle checkpoint, thus deserves further study for anti-leukemic potential.
Dietary flavonoids are currently receiving considerable attention in developing novel cancer-preventive approaches because of their potential capacities to actively induce apoptosis of cancer cells. In our previous report, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein and named RCMF (RVS chloroform-methanol fraction), was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) that is traditionally used as food additive and herbal medicine. In this study, we evaluated the effects of the RCMF on cell proliferation and apoptosis using SV40-transformed tumorigenic hepatocytes, BNL SV A.8. Tritium uptake assay showing the proliferative capacity of the cells was strongly suppressed in the presence of RCMF. This anti-proliferative effect was further confirmed through trypan blue exclusion. RCMF-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. Collectively, this study demonstrated that RCMF can be approached as a potential agent that is capable of significantly inhibiting cell growth of hepatic cancer cells.

Genistein, a phytoestrogen derived from soy isoflavone, has been shown to exert anti-proliferative activities, and have cell arrest and apoptotic effects in cultured tumor cells. However, these properties may not show at the low concentrations of genistein. Present study examined the effect of different concentrations of genistein on cell proliferative proteins, cell cycle regulators or apoptosis related protein in comparison with different concentrations of estrogen or co-treatment with estrogen.(omitted)
발효기간에 따른 미나리 발효추출액의 품질 특성을 검토하고 간암세포주에 대한 증식 억제 효과를 알아보았다. 발효 숙성 기간 중에 총 플라보노이드 함량, 색도 및 생균수는 감소하였으며, 포도당과 과당과 같은 환원당 함량은 증가하였다. 정상 간세포주와 간암 세포주를 이용하여 농도별로 시료 처리를 한 후 각 세포의 증식에 대한 효과를 알아 본 결과, 미나리 발효액이 정상 간세포의 증식에는 영향을 미치지 않았으며, 인간유래 간암 세포주인 HepG2의 증식을 농도 의존적으로 억제하였다. 특히 미나리발효액 1년에서 현저한 억제 효과를 확인하였다. 본 연구 결과는 미나리 발효액의 발효 기간에 따른 품질의 변화 및 간암세포 성장 억제 효능을 확인함으로서, 천연의 미나리 발효 추출액을 다양한 식품 산업에 활용하기 위한 기초적 데이터를 처음으로 제시했다.

Siberian ginseng (Eleutherococcus senticosus) is used primarily as an adaptogen herb and also for its immune stimulant properties in Western herbal medicine. Another closely related species used in East Asian medicine systems i.e. Kampo, TCM (Manchuria, Korea, Japan and Ainu of Hokkaido) and also called Siberian ginseng (Acanthopanax senticosus) also displays immune-stimulant and anti-cancer properties. These may affect tumour growth and also provide an anti-fatigue effect for cancer patients, in particular for those suffering from lung cancer. There is some evidence that a carbohydrate in Siberian ginseng may possess not only immune stimulatory but also anti-tumour effects and also display other various anti-cancer properties. Our study aimed to determine the inhibitory and also proliferative effects of a methanol plant extract of Siberan ginseng (E. senticosus) on various cancer and normal cell lines including: A-549 (small cell lung cancer), XWLC-05 (Yunnan lung cancer cell line), CNE (human nasopharyngeal carcinoma cell line), HCT-116 (human colon cancer) and Beas-2b (human lung epithelial). These cell lines were treated with an extract from E. senticosus that was evaporated and reconstituted in DMSO. Treatment of A-549 (small cell lung cancer) cells with E. senticosus methanolic extract showed a concentration-dependent inhibitory trend from $12.5-50{\mu}g/mL$, and then a plateau, whereas at 12.5 and $25{\mu}g/mL$, there is a slight growth suppression in QBC-939 cells, but then a steady suppression from 50, 100 and $200{\mu}g/mL$. Further, in XWLC-05 (Yunnan lung cancer cell line), E. senticosus methanolic extract displayed an inhibitory effect which plateaued with increasing dosage. Next, in CNE (human nasopharyngeal carcinoma cell line) there was a dose dependent proliferative response, whereas in Beas-2 (human lung epithelial cell line), an inhibitory effect. Finally in colon cancer cell line (HCT-116) we observed an initially weak inhibitory effect and then plateau.
Jaspine B는 석회해면류에서 추출된 sphingosine유도체로 인간 암세포에서의 항암활성이 보고되었다. 그러므로 본 연구는 다양한 인간 암세포주에서 항암활성을 비교하고, 암세포주에서의 Jaspine B의 농도를 측정하여 항암 활성과의 연관성을 확인하고자 하였다. 항암활성은 MTT 방법을 이용하여 측정하였고, $EC_{50}$ 값으로 표현하였다. 암세포주내 Jaspine B의 농도는 LC-MS/MS를 이용하여 분석하였다. 항암활성은 세포주마다 다양하게 나타났는데, 유방암과 흑색종 세포주에서 항암활성이 높게 나타났으며($EC_{50}$ 각각 $2.3{\mu}M$과 $2.6{\mu}M$), 신장암세포주에서는 $EC_{50}$ 값이 $29.4{\mu}M$이었다. 암세포주에서의 $EC_{50}$ 값은 동일한 세포에서의 Jaspine B 농도와 높은 상관성을 나타내었으며(r=0.838), 암세포내 약물농도를 조절하는 것으로 잘 알려진 P-glycoprotein과 breast cancer resistance protein 등의 배출수송계와는 관련이 없음을 확인하였다. 이상의 결과는 세포내 약물농도를 높게 유지하는 것이 항암활성에 매우 중요하며, 세포내 약물농도가 암세포주에 따라 다른 약효를 보이는 원인으로 사료된다.
본 연구에서는 연(Nelumbo nucifera)의 잎(leaf, NL), 자육(seed, NS), 자방(seedpod, NSP)의 에탄올 추출물을 제조하고 이들의 암세포 항 성장 활성과 세포사멸 활성을 연구하였다. 추출물 NL, NS, NSP는 농도의존적으로 세포 생존율을 감소시켰을 뿐 만 아니라 항암 유전자인 NAG-1 유전자와 단백질의 발현을 증가시켰다. 또한, NL은 NAG-1 단백질의 발현과 PARP cleavage를 시간 의존적으로 증가시켰다. PARP cleavage는 NAG-1 siRNA의 transfection에 의해 부분적으로 감소하였으며, 이러한 결과는 NAG-1이 NL에 의해 유도되는 apoptosis에 기여하는 유전자 중의 하나라는 것을 나타낸다. 그리고, 연근 유래의 순수물질인 neferine도 농도의존적으로 HCT116 세포 생존율을 감소시켰으며, NAG-1 단백질의 발현과 PARP cleavage를 농도의존적, 시간의존적으로 증가시켰다. Neferine에 의해 유도된 PARP cleavage는 NAG-1 siRNA의 transfection에 의해 부분적으로 회복되는 것을 확인하였으며, 이러한 결과는 NAG-1 단백질이 neferine에 의해 유도되는 apoptosis에도 기여하는 유전자 중의 하나라는 것을 시사한다. 종합적으로 본 연구 결과는 연근 부위별 추출물과 연근유래 순수물질인 neferine에 의한 암세포 항 성장 활성과 세포사멸 활성의 분자 생물학적 기전을 이해하는데 도움을 줄 것으로 생각된다.
Onchungeum, a herbal formula, which has been used for treatment of anemia due to bleeding, discharging blood and skin disease. In the present study, it was examined the effects of extract of Onchungeum (OCE) on the growth of human hepatocarcinoma cell lines Hep3B (p53 null type) and HepG2 (p53 wild type) in order to investigate the anti-proliferative mechanism by OCE. Treatment of Hep3B and HepG2 cells to OCE resulted in the growth inhibition in a dose-dependent manner, however Hep3B cell line exhibited a relatively strong anti-proliferative activity to OEC. Flow cytometric analysis revealed that OCE treatment in Hep3B cells caused G1 phase arrest of the cell cycle, which was associated with various morphological changes in a dose-dependent fashion. RT-PCR and immunoblotting data revealed that treatment of OCE caused the down-regulation of cyclin D1 expression, however the levels of cyclin E expression were not changed by OCE. The G1 arrest of the cell cycle was also associated with the induction of Cdk inhibitor p27 by OCE. Because the p53 gene is null in Hep3B cells, it is most likely that the induction of p21 is mediated through a p53-independent pathway. Moreover, p27 detected in anti-Cdk4 and anti-Cdk2 immunoprecipitates from the OCE-treated cells, suggesting that OCE-induced p27 protein blocks Cdk kinase activities by directing binding to the cyclin/Cdk complexes. Furthermore, OCE treatment potently suppresses the phosphorylation of retinoblastoma proteins and the levels of the transcription factor E2F-1 expression. Taken together, these results indicated that the growth inhibitory effect of OCE in Hep3B hepatoma cells was associated with the induction of G1 arrest of the cell cycle through regulation of several major growth regulatory gene products.
BACKGROUND/OBJECTIVES: Abundant consumption of seaweeds in the diet is epidemiologically linked to the reduction in risk of developing cancer. In larger cases, however, identification of particular seaweeds that are accountable for these effects is still lacking, hindering the recognition of competent dietary-based chemo preventive approaches. The aim of this research was to establish the antiproliferative potency and angiosuppressive mode of action of Stoechospermum marginatum seaweed methanolic extract using various experimental models. MATERIALS/METHODS: Among the 15 seaweeds screened for antiproliferative activity against Ehrlich ascites tumor (EAT) cell line, Stoechospermum marginatum extract (SME) was found to be the most promising. Therefore, it was further investigated for its anti-proliferative activity in-vitro against choriocarcinoma (BeWo) and non-transformed Human embryonic kidney (HEK 293) cells, and for its anti-migratory/tube formation activity against HUVEC cells in-vitro. Subsequently, the angiosuppressive activity of S. marginatum was established by inhibition of angiogenesis in in-vivo (peritoneal angiogenesis and chorioallantoic membrane assay) and ex-vivo (rat cornea assay) models. RESULTS: Most brown seaweed extracts inhibited the proliferation of EAT cells, while green and red seaweed extracts were much less effective. According to the results, SME selectively inhibited proliferation of BeWo cells in-vitro in a dose-dependent manner, but had a lesser effect on HEK 293 cells. SME also suppressed the migration and tube formation of HUVEC cells in-vitro. In addition, SME was able to suppress VEGF-induced angiogenesis in the chorio allantoic membrane, rat cornea, and tumor induced angiogenesis in the peritoneum of EAT bearing mice. A decrease in the microvessel density count and CD31 antigen staining of treated mice peritoneum provided further evidence of its angiosuppressive activity. CONCLUSIONS: Altogether, the data underline that VEGF mediated angiogenesis is the target for the angiosuppressive action of SME and could potentially be useful in cancer prevention or treatment involving stimulated angiogenesis.
