  • This paper concerns continuous density HMM topology optimization. There have been several researches for HMM topology optimization. BIC (Bayesian Information Criterion) is one of the well known optimization criteria, which assumes statistically well behaved homogeneous model parameters. HMMs, however, are composed of several different kind of parameters to accommodate complex topology, thus BIC's assumption does not hold true for HMMs. Even though BIC reduced the total number of parameters of HMMs, it could not improve the recognition rates. In this paper, we proposed two new model selection criteria, HBIC (HMM-oriented BIC) and BIC_Anti. The former is proposed to improve BIC by estimating model priors separately. The latter is to combine BIC and anti-likelihood to accelerate discrimination power of HMMs. We performed some comparative research on couple of model selection criteria for online handwriting data recognition. We got better recognition results with fewer number of parameters.

  • A new method for optimizing the magnetic field gradient in the exciting coil of electronic anti-fouling (EAF) system is presented based on changing exciting coil size. In the proposed method, two optimization expressions are deduced based on biot-savart law. The optimization expressions, which can describe the distribution of the magnetic field gradient in the coil, are the function of coil radius and coil length. These optimization expressions can be used to obtain an accurate coil size if the magnetic field gradient on a certain point on the coil's axis of symmetry is needed to be the maximum value. Comparing with the experimental results and the computation results using Finite Element Method simulation to the magnetic field gradient on the coil's axis of symmetry, the computation results obtained by the optimization expression in this article can fit the experimental results and the Finite Element Method results very well. This new method can optimize the EAF system's anti-fouling performance based on improving the magnetic field gradient distribution in the exciting coil.

  • In order to overcome some problems of existing anti-windup methods, this paper defines LQ (Linear Quadratic) observer and proposes a new anti-windup method using the LQ observer. LQ observer is derived by linear quadratic optimization in order to calculate controller states, which make the controller outputs equal to the plant inputs. And we propose an algorithm so that it can be implemented by a digital controller easily. The relationship between the design parameters and the anti-windup performance is shown via some numerical examples, which cover the cases with the anti-windup method using LQ observer designed and the case without it. Finally, the anti-windup performance of the proposed method is exemplified via comparison with the existing model-based conditioning scheme method[4].

  • Antibody-drug conjugates utilize the antibody as a delivery vehicle for highly potent cytotoxic molecules with specificity for tumor-associated antigens for cancer therapy. Critical parameters that govern successful antibody-drug conjugate development for clinical use include the selection of the tumor target antigen, the antibody against the target, the cytotoxic molecule, the linker bridging the cytotoxic molecule and the antibody, and the conjugation chemistry used for the attachment of the cytotoxic molecule to the antibody. Advancements in these core antibody-drug conjugate technology are reflected by recent approval of Adectris$^{(R)}$(anti-CD30-drug conjugate) and Kadcyla$^{(R)}$(anti-HER2 drug conjugate). The potential approval of an anti-CD22 conjugate and promising new clinical data for anti-CD19 and anti-CD33 conjugates are additional advancements. Enrichment of antibody-drug conjugates with newly developed potent cytotoxic molecules and linkers are also in the pipeline for various tumor targets. However, the complexity of antibody-drug conjugate components, conjugation methods, and off-target toxicities still pose challenges for the strategic design of antibody-drug conjugates to achieve their fullest therapeutic potential. This review will discuss the emergence of clinical antibody-drug conjugates, current trends in optimization strategies, and recent study results for antibody-drug conjugates that have incorporated the latest optimization strategies. Future challenges and perspectives toward making antibody-drug conjugates more amendable for broader disease indications are also discussed.

  • A dynamic system identification technique based on the topology optimization method is developed. The specific problem in consideration is the damage location identification of a plate structure using the Frequency Response Function (FRF) of a damaged structure. In this work, the identification problem is formulated as a topology optimization problem. The importance of using anti-resonance information in addition to using resonance information is addressed. Though a simple problem was considered here, the possibility of using the topology optimization for damage identification is investigated lot the first time.

  • Obesity, a condition in which an abnormally large amount of fat is stored in adipose tissue, causing an increase in body weight, has become a major public health concern worldwide. The purpose of this study was to optimize the process for fermented milk for the production of a functional product with an anti-obesity effect by using Lactobacillus plantarum Q180 isolated from human feces. We used a 3-factor, 3-level central composite design (CCD) combined with the response surface methodology (RSM). Concentration of skim milk powder (%, $X_1$), incubation temperature ($^{\circ}C$, $X_2$), and incubation time (h, $X_3$) were used as the independent factors, whereas pH (pH, $Y_1$), anti-lipase activity (%, $Y_2$) and anti-adipogenetic activity (%, $Y_3$) were used as the dependent factors. The optimal conditions of fermented milk for the highest anti-lipase and anti-adipogenetic activity with pH 4.4 were the 9.5% of skim milk powder, $37^{\circ}C$ of incubation temperature, 28 h of incubation time. In the fermentation condition, the predicted values of pH, anti-lipase activity and anti-adipogenetic activity were 4.47, 55.55, and 20.48%, respectively. However, the actual values of pH, anti-lipase activity and anti-adipogenetic activity were 4.50, 52.86, and 19.25%, respectively. These results demonstrate that 9.5% of skim milk powder and incubation at $37^{\circ}C$ for 28 h were the optimum conditions for producing functional fermented milk with an anti-obesity effect.

  • Damaged structures change the value of natural properties. The purpose of this study is to detect damage using the difference of natural properties between the healthy state and the damaged state. Anti-optimization method is used to find the conditions that maximize the difference in characteristics between the two contrasting models. In this paper, a algorithm for finding the loading conditions which can maximize the difference of strain energy between the healthy state and the damaged state of truss structures is developed. Numerical examples show that the proposed method is accurate and efficient for truss structures.

  • Ultrasound-assisted extraction (UAE) conditions, including extraction time, extraction temperature, and ethanol concentration, were optimized to increase the total flavonoid content (TFC), tyrosinase inhibitory activity (TIA), and collagenase inhibitory activity (CIA) of Spirulina platensis through central composite design (CCD). For the optimization of the three dependent variables, a quadratic regression model was derived from 17 experimental sets for the simultaneous maximization of TFC, TIA, and CIA. The predicted optimal UAE conditions were 44.0 min of extraction time, 82.8℃ of extraction temperature, and 96.0% of ethanol concentration. Under these conditions, 0.93 mg QE/g DM of TFC, 81.9% of CIA, and 92.1% of TIA were predicted. The CCD-based UAE optimization enabled an increase in TFC, CIA, and TIA, thereby confirming that the S. platensis extract can be used in the development of a cosmetic material with skin whitening and anti-wrinkle effects.
