The Effects of Daecheongryoung-tang on Anti-weight Gain and Anti-hyperlipidemic in Obese Sprague Dawley Rat Induced by High Fat Diet (대청룡탕이 비만 유도 흰쥐의 체중 감량과 혈중지질농도 개선에 미치는 영향)

  • Objectives: The aim of this study is to investigate the effects of Daecheongryoung-tang (DCR) therapy on body weight, serum total cholesterol, high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level and complete blood cell count of obese rats. Methods: 34 rats are divided into 4 groups, the rats in the normal group are 7 and the rats in the other group are 9 per group; Normal group (general fat diet and no medication), Control group (high-fat diet and no medication), DCR_L group (high-fat diet and DCR 250 mg medication) and DCR_H group (high-fat diet and DCR 500 mg medication). DCR is administrated for 6 weeks. Results: There is significant statistical difference between Control group and DCR-H group for the body weight, the total cholesterol, LDL cholesterol, triglyceride, free fatty acid level. Also, there is significant statistical difference among Control group, DCR_L group and DCR_H group for body weight, triglyceride, free fatty acid and phospholipid level. Conclusions: These results suggest that medication of DCR_L and DCR_H is effective for the treatment of obesity.

Anti-obestic Effects of Bangpoongtongsungcankamibang(BTSK) in 3%3-L1 preadipocyte cells and the Lipid Metabolism of Obesity induced Rats (방풍통성산가미방(防風通聖散加味方)이 비만유도(肥滿誘導) 백풍(白嵐)와 지방세포(脂肪細胞) 분화(分化)에 미치는 영향(影響))

  • Objective : This experimental study was designed to determine the anti-obestic effects of Bangpoongtongsungcankamibang(BTSK). Method : In vitro, BTSK extracts of various concentration (50,100, 200 $200{\mu}g/ml$)were added examination. The protein and mRNA expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ receptor was measured by western blot assay and RT-PCR. In vivo, BTSK extracts of various concentration (100, $200{\mu}g/ml$) were orally administered to induced hyperlipidemic rats by poloxamer-407 for consecutive four weeks and serum triglyceride, total cholesterol were measured. This method applied to induced hyperlipidemic rats by triton WR-1339, too. Obesity induced rats by the high fat-diet for six weeks were orally administered BTSK extracts of various concentration (100, $200{\mu}g/ml$) and serum triglyceride, total cholesterol, LDL-cholesterol, triglyceride, LDL-cholesterol, triglyceride, HDL-cholesterol, hydroxy radical, superoxide dismuatse activity were measured. Results : I. In vitro 1. The 3T3-L1 cells' differentiation was significantly decreased by BTSK. 2. expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ was was significantly decreased by BTSK. II. In vitro 1. BTSK significantly reduced serum triglyceride, total cholesterol, contents in poloxamer-407 treated rat. 2. BTSK significantly reduced serum triglyceride, contents in Triton WR-1339 treated rat. But Total cholesterol did not show a significant change. 3. BTSK significantly reduced body weight gain of rat and adipose tissue mass of rats and serum triglyceride, LDL-cholesterol, contents and significantly increased HDL-cholesterol, HTR(HDL-cholesterol/Total-cholesterol) in rats with obesity induced by the high fat-diet. 4. BTSK reduced blood lipid peroxide, hydroxy radical and increased superoxied dismuatse(SOD) activity.

Effects of Jowiseungcheung-tang Extract on the Lipid Metabolism, Anti-oxidation and Inflammatory Reflex High Fat Diet Obese Rats (조위승청탕(調胃升淸湯) 추출액이 비만유도 쥐의 지질대사, 항산화계 및 염증반응계에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effects of Jowiseungcheung-tang (JWSCT) extract on the lipid metabolism, anti-oxidation and inflammatory reflex. Methods : Male Sprague-Dawley rats were fed a high fat diet for 8 weeks and were randomly divided into 4 groups (10 mice in each group): control group, 100 mg/kg JWSCT group, 200 mg/kg JWSCT group, 300 mg/kg JWSCT group. The control group was administered 100 mg/kg of water, but the other three groups were administered 100, 200, 300 mg/kg JWSCT extract for 4 weeks. After 4 weeks, we measured lipid level, thiobarbituric acid reactive substance (TBARS), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), aspartate aminotransferase (AST), alanine aminotransferase (ALT) and cytokines in plasma and liver. The gene expression level and the ratio of apo-B and apo-E were then investigated by way of reverse transcription -polymerase chain reaction (RT-PCR). Results : In the JWSCT group, compared with the control, free fatty acid, triglyceride, total cholesterol, LDL-cholesterol, TBARS, IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ decreased significantly in plasma and liver. However HDL-cholesterol, IL-10, GSH-Px, SOD and CAT increased. In the JWSCT group, compared with the control, the gene expression level and the ratio of apo-A and apo-E decreased significantly in the RT-PCR analysis. Conclusions : The extract of JWSCT has anti-obesity, anti-inflammatory and anti-oxidant effects.

  • Aralia contientalis show several beneficial effects including anti-oxidant effects. Aralia contientalis is used as a therapeutic agent to stop haemorrhages and a tonic to promoted health in Korean and Chinese medicine. The pharmacokinetic profiles of the main Aralia contientalis is still not accurately investigated. In present study, I examined the effects of water extracts of Aralia contientalis on high cholesterol diet atherosclerosis-induced rats in serum and abdominal aorta. A total of 3-week old 9 male rats of Sprague-Dawley were divided into 3 groups and fed with the basal diet (normal group), high cholesterol diet (atherosclerosis induced group) for 8 weeks, high cholesterol diet supplemented with water extracts of Aralia contientalis (Aralia contientalis group) for 2 weeks. And rats were sacrificed, serum lipid level, abodominal aortic anti-oxidant activities and lipid peroxide were measured. These results indicated that serum total cholesterl, LDL-cholesterol, triglycerides concentration significently lowered in Aralia contientalis group than high cholesterol diet group. But HDL-cholesterol concentraion significently higher in Aralia contientalis group than high cholesterol diet group.

The Effects of Lumbricus Pharmacopuncture on the Lipid Lowering, Anti-oxidative Activity and Concentration of Proinflammatory Cytokines in Rat Fed High Fat Diet (구인약침(蚯蚓藥鍼)이 비만쥐의 지질강하능, 항산화능, 전염증성 Cytokines 농도에 미치는 영향)

  • Objectives : This study was designed to investigate effects of Lumbricus pharmacopuncture (LP) on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet. Methods : Sprague-Dawley male rats were fed high fat diet for 8 weeks and experimental groups were divided into 4 groups as follows : Control, Lumbricus Jungwan ($CV_{12}$) pharmacopuncture (T I), Lumbricus Joksamni ($ST_{36}$) pharmacopuncture (T II), Lumbricus Jungwan ($CV_{12}$) and Joksamni ($ST_{36}$) pharmacopuncture (T III). Results : The levels of plasma FFA, TG, total cholesterol, LDL-C, TBARS, IL-$1{\beta}$, IL-6, TNF-$\alpha$ and liver total cholesterol, TG, TBARS, SOD, catalase, IL-6 in more than one LP group were significantly lower than those of Control groups. The level of IL-10 in one of LP groups was significantly higher than that of Control group. Conclusions : As the results were shown, LP treatment have considerable effects on lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in rat fed high fat diet.

Immuno-stimulating Effects of BS-01 Made Using Extract of Acanthopanax Sessiliflorus on the Body Weight and Serum Lipid Level in Obesity-induced Mice (오가피를 활용한 음료인 BS-01이 비만 생쥐의 체중 및 혈청 내 지질 함량에 미치는 실험적 효과)

  • Hyperlipidemia has been treated as one of the most important etiological cause factor in 21th century. The cortex and root of Acanthopanax sessiliflorus (AR), a herbal medicine, have been used for several diseases including cardiovascular diseases in Oriental countries. Recently, we reported that AR has anti-hyperlipidemic action. BS-01 was made using extract from AR. For these reasons, we investigated the effects of BS-01 as anti-hyperlipidemic drug through measurement of body weights, cholesterol levels, total lipid, phospholipid in serum. In our results, mice induced by high fat diet elevated body weight level compared to naive group. And total lipid in serum was also elevated by induction of hyperlipidemia. In BS-01 group, body weight of mice was lowered significantly compared to that of control group. Oral administration of BS-01 decreased total cholesterol and triglyceride level back to that of naive mice. HDL and LDL cholesterol levels were not affected by BS-01. In addition, total lipid level, which elevated by induction of hyperlipidemia was also lowered by oral administration of BS-01. Finally, free fatty acid level was lowered in BS-01 group. These results demonstrate that BS-01 lowered body weight and titers involved in hyperlipdemia such as total cholesterol, triglyceride, free fatty acid and total lipid. In these results, we demonstrate that BS-01 has anti-hyperlipidemic action.

  • The effects of the water extracts from autumn pumpkin (Cucurbita maxima Duchesne, Cucurbitaceae) were evaluated on inflammation induced by carrageenan and hyperlipidemia induced by lipid rich diet, respectively in rats. The water extracts, when administered orally showed significant anti-inflammatory activity in rats treated with 1%-carrageenan and also anti-hyperlipidemia in rats treated with lipid rich diet(15% cholesterol, 1% sodium cholate and 84% corn oil).

Anti-hyperlipidemia and Anti-arteriosclerosis Effects of Laminaria japonica in Sprague-Dawley Rats

  • The anti-hyperlipidemic effects of dietary supplementation with sea tangle Laminaria japonica were investigated using an animal model in which normal rats were fed either sea tangle, sea tangle ethanol extract (EE-ST) and sea tangle extracted residue (ER-ST). Total lipid and triglyceride levels in the serum were significantly (P < 0.05) reduced in rats fed ER-ST at a dose of 200 mg/kg body weight when compared to hyperlipidemic control rats. Significant decreases in serum total cholesterol and low density lipoprotein-cholesterol levels also occurred in rats fed ER-ST at 200 mg/kg body weight. In addition, the atherosclerosis index and superoxide dismutase in blood lipids were significantly (P < 0.05) lowered in rats fed ER-ST at 200 mg/kg body weight as compared to control rats. In conclusion, sea tangle and ER-ST exhibited beneficial anti-hyperlipidemic and anti-arteriosclerosis effects.

Inhibition of Adenovirus 36 Replication and Lipid Accumulation by Distylium racemosum

  • Obesity is a worldwide disease and one of the major risk factors. Virus among many factors can lead to obesity. Adenovirus 36 (Ad-36) is the adipogenic virus linked with human obesity. Nevertheless, there is no drug to treat both Ad-36 infection and obesity associated with virus. For the precedent study on anti-cholesterol test, Distylium racemosum (D. racemosum), Quercus salicina (Q. salicina) and Raphiolepis indica (R. indica) were selected. This study was carried out to evaluate the anti-cholesterol effects, anti-lipid effects and inhibition of Ad-36 replication from three extracts. D. racemosum ($50{\mu}g/mL$) inhibited lipid accumulation on 3T3-L1 adipocyte. D. racemosum inhibited adipocyte differentiation through suppression of regulator peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR{\gamma}$) genes and adipocyte-specific genes such as adipocyte protein 2 (aP2). D. racemosum inhibited replication of Ad-36 at $50{\mu}g/mL$ of concentration. Therefore, the extract of D. racemosum could be a candidate for development of anti-Ad-36 and anti-obesity drugs.

1,8-cineole protected human lipoproteins from modification by oxidation and glycation and exhibited serum lipid-lowering and anti-inflammatory activity in zebrafish

  • We recently reported that a water extract of laurel or turmeric, 1,8-cineole enriched fractions, showed hypolipidemic activity in the zebrafish model. Therefore, the present study investigated the cineole's anti-oxidant and anti-inflammatory activities in lipoprotein metabolism in vitro and in vivo. Cineole had inhibitory effects on cupric ion-mediated oxidation of lipoproteins in general, while simultaneously enhancing ferric ion removal ability in high-density lipoprotein (HDL). Hypercholesterolemia was induced in zebrafish using cholesterol-feeding treatment, 4% cholesterol, for 3 weeks. After feeding with or without the addition of cineole, the results revealed that cineole possessed lipid-lowering and anti-inflammatory activities in hypercholesterolemic zebrafish. In addition, serum amyloid A and interleukin-6 levels were lowered and lipid accumulation was decreased in the liver. Conclusively, 1,8-cineole was found to have anti-oxidant activities in lipoprotein metabolism both in vitro and in vivo with simultaneous reduction of lipid accumulation in the liver of zebrafish.
