The protective effect of the Synadenium carinatum latex lectin (ScLL), and the possibility of using it as an adjuvant in murine model of vaccination against American cutaneous leishmaniasis, were evaluated. BALB/c mice were immunized with the lectin ScLL (10, 50, 100$[\mu}g$/animal) separately or in association with the soluble Leishmania amazonensis antigen (SLA). After a challenge infection with $10^6$ promastigotes, the injury progression was monitored weekly by measuring the footpad swelling for 10 weeks. ScLL appeared to be capable of conferring partial protection to the animals, being most evident when ScLL was used in concentrations of 50 and 100${\mu}g$/animal. Also the parasite load in the interior of macrophages showed significant reduction (61.7%) when compared to the control group. With regard to the cellular response, ScLL 50 and 100 ${\mu}g$/animal stimulated the delayed-type hypersensitivity (DTH) reaction significantly (P < 0.05) higher than SLA or SLA plus ScLL 10 weeks after the challenge infection. The detection of high levels of IgG2a and the expression of mRNA cytokines, such as IFN-$\gamma$, IL-12, and TNF-$\alpha$ (Th1 profiles), corroborated the protective role of this lectin against cutaneous leishmaniasis. This is the first report of the ScLL effect on leishmaniasis and shows a promising role for ScLL to be explored in other experimental models for treatment of leishmaniasis.
BACKGROUND: The residual analysis of polar pesticides has remained a challenge. It is even more difficult to simultaneously analyze multiple polar pesticides. Diquat, paraquat, and chlormequat are typical examples of highly polar pesticides. The existing methods for the analysis of diquat, paraquat and chlormequat are complex and time consuming. Therefore, a simple, quick and effective method was developed in the represent study for simultaneous analysis of diquat, paraquat and chlormequat in animal products, meat and fat using UPLC-MS/MS. METHODS AND RESULTS: Sample extraction was carried out using acidified acetonitrile and water and re- extracted with acidified acetonitrile and combine the extracts followed by centrifugation. The extract was then cleaned up with a HLB cartridge after reconstitution with acidic acetonitrile and water. The method was validated in quintuplicate at three different concentrations. The limits of detection (LOD) and quantification (LOQ) were 0.0015 and 0.005 mg/L, respectively. Matrix suppression effect was observed for all of the analytes. A seven point matrix matched calibration curve was constructed for each of the compound resulted excellent linearity with determination coefficients (R2) ≥ 0.991. Accuracy and precision of the method was calculated from the recovery and repeatability and ranged from 62.4 to 119.7% with relative standard deviation less than 18.8%. CONCLUSION: The recovery and repeatability of the developed method were in the acceptable range according to the Codex Alimentarius guideline. The developed method can be applied for the routine monitoring of diquat, paraquat, and chlormequat in animal products, meat and fat.
Taurine is a type of sulfur-containing amino acid having a sulfate functional group, that is biosynthesized from cysteine. It is mainly distributed in high concentrations in animal tissues and is known to have various effects such as osmotic pressure control, calcium control, anti-inflammatory, antioxidant, and hepatocellular protection. Also, taurine deficiency causes a variety of symptoms, including visual impairment. In particular, in the case of cats, taurine is not biosynthesized and must be supplied through food, so it is classified as an essential amino acid. In this study, an analysis method using mass spectrometry was developed instead of the commonly used derivatization method to quickly, environmentally, and precisely analyze taurine in various animal feeds. The developed analytical method showed good linearity (R2 > 0.99), accuracy (81.97-105.78%), and precision (0.07-12.37%). In addition, the developed method was further verified through quantitative comparison with the derivatization method. This developed method was used in the determination of taurine in 20 animal feed samples obtained from South Korea. The levels of taurine found ranged from 81.53 to 6,743.53 mg/kg. The developed analysis method will be used for the detection and quantification of taurine in domestic feed.
Purpose: Rabies is a fatal but preventable disease with proper pre-exposure anti-rabies vaccination (ARV). Dogs, as household pets and strays, are the reservoir and vector of the disease, and dog bites have been associated with human rabies cases in Sri Lanka over the past few years. However, other susceptible species having frequent contact with humans may be a source of infection. One such species is sheep and immunity following ARV has never been tested in sheep reared in Sri Lanka. Materials and Methods: We have tested serum samples from sheep reared in the Animal Centre, Medical Research Institute of Sri Lanka for the presence of anti-rabies antibodies following ARV. Sheep serum samples were tested with Bio-Pro Rabies enzyme-linked immunosorbent assay (ELISA) antibody kits used for the first time in Sri Lanka and our results were verified by a seroneutralization method on cells (fluorescent antibody virus neutralization, FAVN test) currently recommended by World Organization for Animal Health and World Health Organization. Results: Sheep received annual ARV and maintained high neutralizing antibody titers in their serum. No maternal antibodies were detected in lamb around 6 months of age. Agreement between the ELISA and FAVN test, i.e., coefficient concordance was 83.87%. Conclusion: Annual vaccination in sheep has an effect on maintaining adequate protection against rabies by measurements of anti-rabies antibody response. Lambs need to be vaccinated earlier than 6 months of age to achieve protective levels of neutralizing antibodies in their serum. Introducing this ELISA in Sri Lanka will be a good opportunity to determine the level of anti-rabies antibodies in animal serum samples.

There is a high association of heat shock on the alteration of energy and lipid metabolism. The alterations associated with thermal stress are composed of gene expression changes and adaptation through biochemical responses. Previous study showed that Angelica gigas Nakai (AGN) root extract promoted adipogenic differentiation in murine 3T3-L1 preadipocytes under the normal temperature condition. However, its effect in heat shocked 3T3-L1 cells has not been established. In this study, we investigated the effect of AGN root hot water extract in the adipogenic differentiation of murine 3T3-L1 preadipocytes following heat shock and its possible mechanism of action. Thermal stress procedure was executed within the same stage of preadipocyte confluence (G0) through incubation at $42^{\circ}C$ for one hour and then allowed to recover at normal incubation temperature of $37^{\circ}C$ for another hour before AGN treatment for both cell viability assay and Oil Red O. Cell viability assay showed that AGN was able to dose dependently (0 to $400{\mu}g/mL$) increase cell proliferation under normal incubation temperature and also was able to prevent cytotoxicity due to heat shock accompanied by cell proliferation. Confluent preadipocytes were subjected into heat shock procedure, recovery and then AGN treatment prior to stimulation with the differentiation solution. Heat shocked preadipocytes exhibited reduced differentiation as supported by decreased amount of lipid accumulation in Oil Red O staining and triglyceride measurement. However, those heat shocked preadipocytes that then were given AGN extract showed a dose dependent increase in lipid accumulation as shown by both evaluation procedures. In line with these results, real-time polymerase chain reaction (RT-PCR) and Western blot analysis showed that AGN increased adipogenic differentiation by upregulating heat shock protection related genes and proteins together with the adipogenic markers. These findings imply the potential of AGN in heat shock amelioration among 3T3-L1 preadipocytes through heat shock factor and proteins augmentation and enhanced adipogenic marker expression.

Our previous study demonstrated that methanolic extract of Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae) has the potential to induce detoxifying enzymes such as NAD(P)H:(quinone acceptor) oxidoreductase 1 (EC 1.6.99.2) (NQO1, QR) and glutathione S-transferase (GST). In this study we further fractionated methanolic extract of Chrysanthemum zawadskii and investigated the detoxifying enzyme-inducing potential of each fraction. The fraction (CZ-6) shown the highest QR-inducing activity was found to contain (+)-(3S,4S,5R,8S)-(E)-8-acetoxy-4-hydroxy-3-isovaleroyloxy-2-(hexa-2,4-diynyliden)-1,6-dioxaspiro [4,5] decane and increased QR enzyme activity in a dose-dependent manner. Furthermore, CZ-6 fraction caused a dose-dependent enhancement of luciferase activity in HepG2-C8 cells generated by stably transfecting antioxidant response element-luciferase gene construct, suggesting that it induces antioxidant/detoxifying enzymes through antioxidant response element (ARE)-mediated transcriptional activation of the relevant genes. Although CZ-6 fraction failed to induce hepatic QR in mice over the control, it restored QR activity suppressed by $CCl_4$ treatment to the control level. Hepatic injury induced by $CCl_4$ was also slightly protected by pretreatment with CZ-6. In conclusion, although CZ-6 fractionated from methanolic extract of Chrysanthemum zawadskii did not cause a significant QR induction in mice organs such as liver, kidney, and stomach, it showed protective effect from liver damage caused by $CCl_4$.

Emission factors for ammonia and particulate matters (PMs) from livestock buildings are of increasing importance in view of the environmental protection. While the existing emission factors were determined based on the emission inventory of other countries, in situ measurement of emission factors is required to construct an accurate emission inventory for Korea. This study is to report measurements of ammonia and PMs emissions from mechanically-ventilated pig houses, which are common types of pig barns in Korea. Ventilation rates and concentrations of ammonia and PMs were measured at the ventilation outlets of a weaner unit, a growing pig unit and a fattening pig unit to calculated the emission factors. The PMs emission was characterized with different aerodynamic diameters (PM2.5, PM10, and total suspended particulates (TSP)). The measured ammonia emission factors for weaners, growing pigs and fattening pigs were 0.225, 0.869 and 1.679 kg animal-1 yr-1, respectively, showing linear increase with pigs' age. The PMs emission factors for three growing stages were 0.023, 0.237 and 0.241 kg animal-1 yr-1, respectively for TSP, 0.017, 0.072 and 0.223 kg animal-1 yr-1, respectively for PM10, and 0.011, 0.016 and 0.151 kg animal-1 yr-1, respectively for PM2.5. PMs emissions were increased with pigs' age due to increasing feed supply and animal movement. The measured emission factors were smaller than those of the existing emission inventory indicating that the existing ones overestimate the emissions from pig buildings and also suggesting that long-term in situ monitoring at various livestock buildings is required to construct the accurate emission inventory.

Background and Objectives : Sphincteric function of the larynx, essential to lower airway protection, is most efficiently achieved through strong reflex adduction by both vocal cords. We hypothesize that central facilitation is an essential component of a bilateral adductor reflex and that its disturbance could result in weakened sphincteric closure. Materials and Method : Seven adult 20kg dogs underwent evoked response laryngeal electromyoraphy under 0.5 to 1.0 MAC isoflurane anesthesia. The internal branch of superior laryngeal nerve was stimulated through bipolar platinum-iridium electrodes and recording electrodes were positioned in the ipsilateral and contralateral thyroaryteonoid muscles. Results : Consistent threshold responses were obtained ipsilaterally under all anesthetic levels. However, contralateral reflex responses disappeared as anesthetic levels approached 1.0 MAC. Additionally, at 0.5 MAC, late responses (R2) were detected in one animal. Conclusion : Alteration of central facilitation by deepening anesthesia abolishes the crossed adductor reflex, predisposing to a weakened glottic closure response. Precise understanding of this effect may improve the prevention of aspiration in patients emerging from prolonged sedation or under heavy psychotropic control.
Two new species of oribatid mites, Dicastribates tenuisetosus sp. nov, and Liacarus unjangensis sp. nov. are described from Korea. Dicastribates tenuisetosus sp. nov. is easily distinguishable from a single known species, D. heterotrichus (Mahunka, 1984) from Paraguay by the presence of 14 pairs of distallv attenuate notogastral setae, the smooth rostral and lamellar setae, the relatively short and distally attenuating interlamellar setae, the relatively short sensilli with distinctly clavated head, and presence of two and six setae on epimeral region III and genital plate. Liacarus unjangensis sp. nov. can be easily distinguished from most other known species by the very wide lamellae which cover dorsally whole prodorsum, the presence of equal sized inner and outer teeth of lamellar cusps and the minute rostral, lamellar and interlamellar setae.

The objectives of this study were to categorize agriculture, to analyze perception of sustainable agriculture and to suggest the directions for improving farming practices in Korea. This study was conducted by review of literature and mail survey. The data was collected from 225 purposively sampled agricultural students by questionnaire. Major findings of the study were as follows; 1) According to the respondents, sustainable agriculture may bring positive effects on the betterment of land conservation, water management and animal protection. 2) The respondents expected that the sustainable agricultural practices may diminish the use of agricultural chemicals and increase safe agricultural products. 3) Sustainable agriculture needs more labor power, however, the number of small and middle-size farms in Korea would not be increased. 4) Even though the price of sustainable agriculture products increases, the respondents expected that rural economic condition would not be much improved because of sustainable agriculture. 5) Agricultural students responded that number of farmers in sustainable agriculture would be increased in the future.
