Anti-Helicobacter pylori Activity of Korean Propolis (국산 프로폴리스의 Helicobacter pyrori 억제효과)

  • Propolis possesses antimicrobial, anti-inflammatory, anaesthetic and immunostimulating activities. The aim of this study was to evaluated chemical composition and in vitro anti-Helicobacter pylori activity of eight propolis ethanolic extracts form different region in Korea. The chemical composition of eight Korean propolis was investicated by absorption spectrophotomety and UHPLC. Total penolic and total flavonoids content ranged between $21{\pm}1.5$ and $31{\pm}1.8$ gallic acid g/100 g and $7.2{\pm}0.9$ and $24.4{\pm}1.9$ quercetin g/100 g, respectively. The p-coumaric acid as standard components content ranged between $0.29{\pm}0.00$ and $2.34{\pm}0.02mg/g$ using UHPLC. Cinnamic acid content between $0.43{\pm}0.00$ and $1.33{\pm}0.01mg/g$. 80% ethanolic extract of propolis against H. pylori was evaluated by using the disc diffusion method. Ethanol was used as a control. Mean diameters of H. pylori growth inhibition using $10{\mu}g/mL$, $50{\mu}g/mL$ or $100{\mu}g/mL$ ethanol propolis per disc were 0, ranging from 0.9 to 2.1 mm and from 1.9 to 3.5 mm, respectively. In conclusion, variation in the chemical composition of Korean propolis were mainly due to the difference in the preferred regional plants visited by honeybees. Also Korean propolis possesses considerable antibacterial activity against H. pylori. There was not confirmed a correlation between the chemical composion contens and anti-H. pylori effect. The potential of propolis in the prevention or treatment of H. pylori infection is worth further extensive evaluation.

Anticancer Potential of Cratoxylum formosum Subsp. Pruniflorum (Kurz.) Gogel Extracts Against Cervical Cancer Cell Lines

  • Background: Most northeast Thai vegetables may play roles in human health by acting as antioxidant and anticancer agents. Recent study showed that Cratoxylum formosum subsp. pruniflorum (Kurz.) Gogel. (Teawdang) could inhibit growth of liver cancer cell lines. Cervical cancer, which has human papilloma virus as its main cause, is found at high incidence in Thailand. Due to increasing drug resistance, searches for potential anticancer compounds from natural source are required. Therefore, our purpose was to evaluate the cytotoxicity of Teawdang extracts in cervical cancer cell lines. Materials and Methods: Teawdang edible parts, purchased from Khon Kaen market during July-October 2013 was extracted with organic solvent. Phenolic profiles of crude hexane (CHE), ethyl acetate (CEE), methanol (CME) and water (CWE) extracts were performed by high performance liquid chromatographic (HPLC) techniques. Their cytotoxic effects on cervical cancer cells were investigated with HPV-non infected (C-33A) and HPV-infected (HeLa and SiHa) cell lines. Results: HPLC profiles showed that all crude extracts contained caffeine, ferulic acid and resveratrol. CME and CEE had high contents of gallic acid and quercetin. Catechin was found only in CWE. Cytotoxicity test showed that CEE had the lowest IC50 on HeLa ($143.18{\pm}13.35 {\mu}g/mL$) and SiHa cells ($106.45{\pm}15.73{\mu}g/mL$). C-33A cells were inhibited by CWE ($IC50=130.95{\pm}3.83{\mu}g/mL$). Conclusions: There were several phenolic compounds in Teawdang extracts which may have cytotoxic effects on cervical cancer cell lines. Investigation of these bioactive compounds as new sources of anticancer agents is recommended.

Helium dielectric barrier discharge-cold plasma treatment for microbiological safety and preservation of onion powder (유전체 방벽 방전 콜드 플라즈마 기술을 이용한 양파 분말 미생물 안전성 향상 및 품질 보존)

  • Efficacy of dielectric barrier discharge-cold plasma treatment (DBD-CPT) for microbial decontamination of onion powder was evaluated. Onion powder, inoculated with Escherichia coli O157:H7, Salmonella Enteritidis, or Listeria monocytogenes, was treated with helium DBD-CPT. DBD-CPT (9 kV, 20 min) inhibited E. coli O157:H7, S. Enteritidis, and L. monocytogenes by $1.4{\pm}0.5$, $2.3{\pm}0.3$, and $1.2{\pm}0.0log\;CFU/cm^2$, respectively. The inactivation levels of E. coli O157:H7, S. Enteritidis and L. monocytogenes increased by $2.2{\pm}0.1$, $2.5{\pm}0.1$ and $1.9{\pm}0.3log\;CFU/cm^2$, respectively, as water activity increased from 0.4 to 0.8, and increased by $2.3{\pm}0.4$, $2.1{\pm}0.1$ and $1.6{\pm}0.1log\;CFU/cm^2$, respectively, as the particle size increased from 0.3 to $1.0cm^2$. Neither the ascorbic acid and quercetin concentrations nor the color of onion powder was changed by DBD-CPT (p>0.05). These results demonstrate the potential for application of DBD-CPT in improving microbiological safety of onion powder while preserving the physicochemical properties.

Properties of Cheongkukjang Prepared with Admixed Medicinal Herb Powder (생약초(표고버섯, 더덕, 어성초)를 첨가한 청국장의 특성)

  • Cheongkukjang was prepared by addition of medicinal herb powder (from Lentinus edodes, Codonopsis lanceolata BENTH et HOOK, or Houttuynia cordata THUNB) to improve Cheongkukjang quality and functional properties. Redness and yellowness (measured using Hunter’s color values) increased with increasing amounts of medicinal herb powder, whereas lightness (the third Hunter's color value) and pH decreased. Crude protein and lipid levels of Cheongkukjang decreased with increasing amounts of added medicinal herb powder, whereas powder addition did not affect either the moisture or crude ash content of Cheongkukjang. The major organic acids of Cheongkukjang were lactic acid, acetic acid, succinic acid, and citric acid, and acid levels increased as the proportion of medicinal herb powder in the Cheongkukjang preparation increased. The major amino acids of Cheongkukjang were glutamic acid, aspartic acid, leucine, arginine, and lysine and the principal fatty acids were linoleic acid, oleic acid, palmitic acid, and linolenic acid. Addition of medicinal herb powder to Cheongkukjang increased both crude saponin and quercetin contents. Sensory scores of Cheongkukjang containing 2% (w/v) medicinal herb powder were optimal in terms of both quality characteristics and sensory evaluation.

Antioxidative and Antiaging Effects of Sorbus commixta Twig Extracts (마가목 가지 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.

Antioxidant activity and anti-obesity effect of Coprinus comatus in Zucker rat (fa/fa) (먹물버섯(Coprinus comatus )의 항산화 활성 및 Zucker rat에 대한 항비만 효과)

  • The aim of the study was to investigate the anti-obesity effect of Coprinus comatus (CC) in high-fat diet-fed Zucker rat (fa/fa). Obesity was induced by feeding on high-fat diet (HFD) containing 60% kcal fat for 10 weeks, in which CC extracts were administrated through the gastrointestinal tract at a concentration of 200 mg/kg BW/day for 10 weeks. The total polyphenol and flavonoid contents of CC extracts were found to be $18.5{\pm}1.1mg$ of catechin equivalent/g, and $5.24{\pm}0.54mg$ of quercetin equivalent/g extract, respectively. The DPPH, ABTS, and hydroxyl radical scavenging activities of CC extracts were 15.34 %, 17.25%, and 16.18%, respectively. In animal study, CC administration significantly reduced the body weight, while there were no significant differences in the daily food intake between the HFD-fed Zucker rats and HFD plus CC-fed rats. CC treatment decreased epididymal and perirenal fat weights in HFD-fed Zucker rats. Significant decreases in the levels of triglyceride and total cholesterol in the serum and liver were observed in the CC-treated group compared with HFD-fed Zucker rats. Serum HDL-cholesterol levels in the CC-treated group were increased compared with the HFD-fed groups. Serum AST and ALT activities in the CC group were significantly lower than those of the HFD-fed group. Taken together, these data demonstrated that CC has potential in preventing high-fat diet induced obesity and is a good candidate for an anti-obesity agent.

Antioxidant Compounds and Activities as well as Caffeine Content of Aronia melanocarpa Leaf Tea according to Pan-Roasting Conditions (아로니아잎차의 덖음조건에 따른 항산화 성분과 항산화 활성 및 카페인 함량)

  • Differences in bioactive compounds and antioxidant activities of aronia leaf (AL) extracts according to manufacturing conditions such as different number of pan-roasting and different temperatures were investigated. Both total polyphenolic compounds and total flavonoids contents were the highest in six time-pan-roasted AL tea extract ($37.96{\pm}0.48mg$ catechin equivalent/g and $19.96{\pm}0.44mg$ quercetin equivalent/g, respectively) among four tea samples. Antioxidative activities were also the highest in six time-pan-roasted AL tea extract with $IC_{50}$ 0.43 mg/mL and $IC_{50}$ 0.27 mg/mL based on DPPH and ABTS radical scavenging activities, respectively. HPLC analysis revealed that AL tea infusion did not have caffeine regardless of manufacturing conditions, whereas green tea infusion had 3.8 mg/g caffeine. Results demonstrated that AL tea can be expected as caffeine free leaf tea containing antioxidant benefits. Moreover, specific pan-roasting conditions of AL tea would be very important for its functional and sensory attributes.

Effect of Onion Consumption on Cardiovascular Disease in Human Intervention Studies: A Literature Review (국내.외 인체중재연구를 통해 살펴 본 양파의 심혈관계 질환 개선효과에 관한 고찰)

  • Onion (Allium cepa L.) production in Korea has increased gradually over the past 15 years, placing second in food consumption survey with 20.6 g daily intake in 2006. Onions, used as an ingredient in many dishes and accepted by almost all traditions and cultures, have been reported to have a range of health benefits which include anticarcinogenic, antiasthmatic, antibiotic, and antioxidative effects. These effects may be attributable to a powerful flavonoid pigment-containing compounds, such as quercetin and alk(en)yl cysteine sulphoxides (ACSOs). Although antiplatelet and antithrombotic activities of onion have been confirmed by many of in vitro or animal studies, only a few human intervention studies have been examined. The majority of human studies identified that onion improves some cardiovascular markers such as lipid profile and platelet coagulant. With regard to antioxidative effects, somewhat positive effects are confirmed through strengthening the resistance of oxidative DNA damage in lymphocyte and urine, while most studies failed to find inhibitory effects on LDL oxidation. The discrepancies among studies might be ascribed to producing area, processing methods of onion, dosage, subject characteristics, study duration, and measurement methods. In this review, we focused on the preventive effect of cardiovascular disease through onion consumption in human intervention studies.

  • Onion has been reported to contain various organosulfur compounds which have antibiotic and anticarcinogenic properties and flavonoid like quercetin which is a valuable natural source of antioxidants. Carbohydrates in onion constitute about 80% of dry matter, and the major non-structural carbohydrate of onion bulb is fructo-oligosaccharides, well known as fructan, followed by glucose, fructose, and sucrose. The sugar concentration is associated with dormancy and storage life of onion, occurring as decrease in glucose, fructose and fructan, particularly towards the end of storage. Forced air pre-drying for 15-20 days at room temperature is an essential procedure to reduce freezing injury and sprouting, then onion bulbs can be stored at $0^{\circ}C$ for 6 months to control sprouting and decay. Bacterial soft rot caused by $Erwinia$ and $Pseudomonas$ is the main postharvest disease when the bulbs are infected with the bacteria and stored at room temperature. Browning in sliced onion is due to oxidation of phenolic compounds by polyphenol oxidase and it can be inhibited by citric acid treatment, packing with nitrogen gas, and polyethylene film.

Antioxidant Capacity of Ethanol Extracts and Fractions from Rubus coreanus Miq. (복분자 에탄올 추출물 및 분획물의 항산화 효과)

  • In the present study, the total polyphenol and flavonoid contents, radical scavenging ability, and reducing power of ethanol extracts and fractions from Rubus coreanus Miq. were assessed in order to determine the antioxidant activity. The total polyphenol contents in the 70% ethanol extract and ethyl acetate fraction were 238.46 mg gallic acid equivalents (GAE)/g and 413.59 mg GAE/g, respectively; these values were higher than those in the other fractions (p < 0.05). In addition, the total flavonoid content was as high as 52.83 mg quercetin equivalents (QE)/g in the ethyl acetate fraction (p < 0.05). With regard to DPPH and ABTS radical scavenging activities, the RC50 values of the ethyl acetate fraction were 3.95 and 6.25 ㎍/mL, respectively, indicating that the scavenging activity was similar to that of ascorbic acid (control) (p < 0.05). Moreover, in the FRAP assay performed to assess the reducing power, the ethyl acetate fraction showed high activity (p < 0.05). In summary, the present findings confirmed that the ethyl acetate fraction of the R. coreanus Miq. 70% ethanol extract had high polyphenol and flavonoid contents and excellent antioxidant properties. Therefore, it was confirmed that the ethyl acetate fraction of the mugwort 70% ethanol extract had high polyphenol and flavonoid content and excellent antioxidant effect.
