Distribution of Population Densities of Heterotrophic Bacteria and Fecal-Origin Bacterial Group in Seawater and Sediments near Samcheonpo Bay (삼천포만 인근해역과 저질토에서 종속영양세균과 분변성 기원 세균의 분포)

  • Distribution of population densities of heterotrophic bacteria, E. coli, and Salmonella and Shigella in seawater and sediments at 40 stations near Samcheonpo Bay were measured for 3 times from July to December, 2003. Population densities of heterotrophic bacteria in seawater during survey periods were in the range of $1.7{\pm}0.9{\times}10^3{\sim}2.4{\pm}0.9{\times}10^5$ CFU $mL^{-1}$ and the highest density was shown at St. 34 during the sampling period of September, 2003. Population densities of heterotrophic bacteria were shown higher values on September than those of July and December at all sampling stations. Population densities of anaerobic heterotrophic bacteria in sediments during survey periods were in the range of $2.2{\pm}0.2{\times}10^3{\sim}2.0{\pm}0.2{\times}10^5$ CFU $mL^{-1}$ and their population densities at sampling stations far from Samcheonpo Bay measured lower values than those near Samcheonpo Bay. Population densities of anaerobic heterotrophic bacteria in the sediments were not affected by physico-chemical factors of upper water environment. E. coli were detected only at 8 stations in seawater and 4 stations in the sediments among 40 sampling stations on July and were not detected during September and December. Salmonella and Shigella were detected only a few stations on July and September during sampling periods.

  • In this research, characteristics of nitrification and denitrification using the microorganism attached on sponge and plates were examined. The denitrification and nitrification performance were investigated under the anaerobic and aerobic condition for about 2 months. Because the basins of denitrification and nitrification were connected in series, wastewater was flowed from denitrification basin to nitrification one. The 90% of influent flowrate was returned from nitrification basin to denitrification one. Most of organic material was removed in nitrification basin, wherease the only exact amount of organics required in denitrification process was removed in denitrification one. This experiment resulted in that heterotrophic bacteria existing in aerobic basin governed the removal efficiency of organic compounds. In case the influent BOD concentration into nitrification basin was 80mg/l, it did not affect to accumulation of nitrifying bacteria, the balance of heterotrophic bacteria was proved to be an important factor in nitrification/denitrification method such as anaerobic and aerobic cycling type.

Bacterial Diversity in the Rhizosphere of Halophyte Suaeda japonica in Western and Southern Mudflats of Korea

  • This study was carried out to investigate the population densities, R/S ratios, and identification of heterotrophic bacteria on the rhizosphere soil of halophyte Suaeda japonica found on the western and southern mudflats of Korea. The population densities of aerobic and anaerobic heterotrophic bacteria on the rhizosphere soil of Suaeda japonica were in the range of $1.3\;{\pm}\;0.3\;{\times}\;10^6\;{\sim}\;6.3\;{\pm}\;3.3\;{\times}\;10^7\;and\;2.8\;{\pm}\;1.3\;{\times}\;10^4\;{\sim}\;1.8\;{\pm}\;0.7\;{\times}\;10^7\;cfu\;g^{-1}\;d.\;wt.$, respectively. In case of physiologically specific bacteria, population densities of amylolytic bacteria on the rhizosphere soil of Suaeda japonica were in the range of $4.4\;{\pm}\;0.6\;{\times}\;10^6\;{\sim}\;2.5\;{\pm}\;1.2\;{\times}\;10^7\;cfu\;g^{-1}\;d.\;wt.$, those of cellulolytic bacteria were from $8.5\;{\pm}\;6.0\;{\times}\;10^4\;{\sim}\;2.3\;{\pm}\;1.6\;{\times}\;10^6\;cfu\;g^{-1}\;d.\;wt.$, and those of proteolytic bacteria were from $3.8\;{\pm}\;1.8\;{\times}\;10^5\;{\sim}\;4.2\;{\pm}\;2.9\;{\times}\;10^6\;cfu\;g^{-1}\;d.\;wt.$, respectively. The R/S ratios were ranged from 2.33 to 2.39. Among eleven isolates from the roots of halophyte Suaeda japonica of Goheung bay by using 16S rDNA analysis, five clones were closely related to ${\gamma}-Proteobacteria$ group and six clones were closely related to ${\alpha}-Proteobacteria$ group. Among four isolates from Suncheon bay, two strains were related to ${\gamma}-Proteobacteria$ group and another two were related to Actinobacteria and Bacilli group, respectively.

Changes of Bacterial Population during the Decomposition Process of Red Tide Dinoflagellate, Cochiodinium polykrikoides in the Marine Sediment Addition of Yellow Loess (황토첨가 해양퇴적물에서 적조생물 Cochiodinium polykrikoides 분해중 세균군집의 변동)

  • To investigate the effects of yellow loess on the microbial community after applying into C. polykrikoides as a red tide centrol method during decomposition process, we conducted this study using microcosm experiments, which consisted of sediment collected from Jinhae and Masan bay. The composition, number of bacteria and respiratory electron transport system activity (ETSA) were analyzed. The number of heterotrophic bacteria examined in the samples of both stations reached maximum value within 12 hrs with $10^7$ cells/dry g, independent with the yellow loess applied. In addition, a differenee in the variation of heterotrophic bacterial composition was not observed by adding the yellow loess, and Vibrio spp. always appeared during the culture periods, However, in day 8 culture, the sulfate reducing bacteria was $3.8\times10^7$ cells/dry g in Masan bay and $5.5\times10^6$ cells/dry g in Jinhae bay samples without yellow loess, and these were 120, 350 fold-and 160, 420 fold-increased when yellow loess was added (1 : 1, 1 : 2). The average ETSA was 6.8$\~$7.6 $\mu$g formazan $h^{-1}$ dry $g^{-1}$ independently with yellow loess in aerobic condition for both samples, but activity was decreased by addition of yellow loess in anaerobic. Thus the addition of yellow loess to marine sediment seems to have an effect to inhibit the anaerobic decomposition process and growth of sulfate reducing bacteria which lead to the bad condition of marine environments.

Isolation and Characterization of Sulfur-oxidizing Denitrifying Bacteria Utilizing Thiosulfate as an Electron Donor (황(thiosulfate)을 이용하는 탈질 미생물의 분리 및 특성 파악)

  • Sulfur-oxidizing bacteria were enumerated and isolated from a steady-state anaerobic master culture reactor (MCR) operated for over six months under a semi-continuous mode and nitrate-limiting conditions using thiosulfate as an electron donor. Most are Gram-negative bacteria, which have sizes up to 12 m. Strains AD1 and AD2 were isolated from the plate count agar (PCA), and strains BD1 and BD2 from the solid thiosulfate/nitrate medium. Based on the morphological, physiological, FAME and 16S rDNA sequence analyses, the two dominant strains, AD1 and AD2, were identified as Paracoccus denitrificans and Paracoccus versutus (formerly Thiobacillus versutus), respectively. From the physiological results, glucose was assimilated by both strains AD1 and AD2. Heterotrophic growth of strains AD1 and AD2 could be a more efficient way of obtaining a greater amount of biomass for use as an inoculum. Even though facultative autotrophic bacteria grow under heterotrophic conditions, autotrophic denitrification would not be reduced.

Effects of Chlorine Residual and Pipe Material on the Biofilm Formation in Drinking Water Distribution Pipe (수도관의 생물막 형성에 미치는 잔류염소와 파이프 재질의 영향)

  • This laboratory study examined the impact of free chlorine residual and pipe material on the formation of biofilm in drinking water distribution pipe surfaces. Result of heterotrophic plate counts(HPC) of the biofilm in the tap water-supplied reactor averaged $2.17{\times}10^5CFU/cm^2$ on PVC and $2.43{\times}10^5CFU/cm^2$ on STS 316, respectively. HPCs on the surface exposed to the tap water containing 0.2mg/L of free chlorinne residual averaged $4.24{\times}10^4CFU/cm^2$ on PVC and $6.54{\times}10^4CFU/cm^2$ on STS 316, respectively. Average of HPC/Total direct counts in the tap water-supplied reactor ranged from 1.08%(PVC) to 1.26%(STS 316) and from 0.38%(PVC) to 0.65%(STS 316) in the reactor supplemented with disinfectant, respectively. No correlation was observed between disinfectant addition and biofilm density. With regard to the biofilm formation, little difference existed between PVC and STS 316. Yellow and red pigmented bacteria were the dominant expressions in bulk fluid, whereas non-pigmented bacteria were found dominant in the biofilm. Pink/red pigmented bacteria were found to be facultative anaerobic, while yellow pigmented bacteria and non-pigmented bacteria were found to be obligate aerobic.

Nitrogen Removal in Fluidized Bed and Hybrid Reactor using Porous Media (다공성 담체를 이용한 유동상 및 하이브리드 반응기에서의 질소제거)

  • A fluidized bed reactor containing porous media has been known to be effective for nitrogen and organic matters removal in wastewater. The porous media which attached microbes plays important roles in simultaneous nitrification/denitrification (SND) due to coexistence of oxic, anaerobic and anoxic zone. For SND reaction, oxygen and organic substrates should be effectively diffused from wastewater into the intra-carrier zone. However, the overgrowth heterotrophic microbes at the surface of porous media may restrict from substrates diffusion. From these viewpoints, the existence and effect of heterotrophic bacteria at surface of porous media might be the key point for nitrogen removal. A porous media-membrane hybrid process was found to have improved nitrogen removal efficiency, due to stimulated denitrification as well as nitrification. Microelectrode studies revealed that although intra-media denitrification rate in a conventional fluidized bed was limited by organic carbon, this limitation was reduced in the hybrid process, resulting in the increased denitrification rate from 0.5 to $4.2\; mgNO_3-N/L/hr$.

Advanced Wastewater Treatment Process using Rotating Activated Bacillus Contactor (RABC) (망상형 회전식 바실러스 접촉장치를 이용한 하수의 고도처리공정에 관한 연구)

  • A new technology for advanced wastewater treatment was developed using a modified Rotating Biological Contactor (RBC) process, named as Rotating Activated Bacillus Contactor (RABC) process that utilizes Bacillus sp., the facultatively anaerobic or activated microaerophilic bacteria on multiple-stage reticular rotating carriers, as a predominant species. The RABC process for a municipal wastewater with relatively low concentrations of organics, nitrogen, and phosphorus showed stable and high removal efficiencies, less than $BOD_5$ 10 mg/L, T-N 15 mg/L, and T-P 1.5 mg/L in final effluent. The performance load of RABC process was shown to be $1.23kg{\cdot}BOD/m^2{\cdot}day$ for the first stage (average $0.31kg{\cdot}BOD/m^2{\cdot}day$ for the total stages) based on both removed BOD and converted disc area corresponding to the reticular one. The sludge produced in the RABC process is characterized by low generation rate (about $0.18kg{\cdot}MLSS/kg{\cdot}BOD$) and excellent settleability. The number ratio of Bacillus ($2.4{\times}10^6CFU/ml$) to heterotrophic bacteria ($3.6{\times}10^7CFU/ml$) inhabiting in the biofilms of the RABC process was 6.7 %, indicating that Bacillus sp. was a predominant species in the biofilms. The RABC process with reticular rotating carriers showed its excellent performance for the advanced wastewater treatment without any offensive odor problem due to organic overloading.

Influence of Electric Potential on Structure and Function of Biofilm in Wastewater Treatment Reactor : Bacterial Oxidation of Organic Carbons Coupled to Bacterial Denitrification

  • Carbon electrode was applied to a wastewater treatment system as biofilm media. The spatial distribution of heterotrophic bacteria in aerobic wastewater biofilm grown on carbon electrode was investigated by scanning electron microscopy, atomic force microscopy, and biomass measurement. Five volts of electric oxidation and reduction potential were charged to the carbon anode and cathode of the bioelectrochemical system, respectively, but were not charged to electrodes of a conventional system. To correlate the biofilm architecture of bacterial populations with their activity, the bacterial treatment efficiency of organic carbons was measured in the bioelectrochemical system and compared with that in the conventional system. In the SEM image, the biofilm on the anodic medium of the bioelectrochemical system looked intact and active; however, that on the carbon medium of the conventional system appeared to be shrinking or damaging. In the AFM image, the thickness of biofilm formed on the carbon medium was about two times of those on the anodic medium. The bacterial treatment efficiency of organic carbons in the bioelectrochemical system was about 1.5 times higher than that in the conventional system. Some denitrifying bacteria can metabolically oxidize $H_{2}$, coupled to reduction of $NO_{3}^{-}\;to\;N_{2}$. $H_{2}$ was produced from the cathode in the bioelectrochemical system by electrolysis of water but was not so in the conventional system. The denitrification efficiency was less than $22\%$ in the conventional system and more than $77\%$ in the bioelectrochemical system. From these results, we found that the electrochemical coupling reactions between aerobic and anaerobic reactors may be a useful tool for improvement of wastewater treatment and denitrification efficiency, without special manipulations such as bacterial growth condition control, C/N ratio (the ratio of carbon to nitrogen) control, MLSS returning, or biofilm refreshing.

Structure and Characteristics of Bacterial Community on Biofilm of Food Wastewater Treatment System in Winter (식품폐수 처리 공정용 생물막의 겨울철 세균군집 구조와 특성)

  • Biofilm and aeration tank of pilot and full RABC (rotating activated Bacillus contactor) plant were analyzed to characterize and determine bacterial community structure in food wastewater treatment system at winter. Concentration of heterotrophic bacteria and Bacillus group was $10^7$ and $10^5$ CFU/ml, respectively, at biofilm of pilot-plant while others represented $10^6$ and $10^4$ CFU/ml, respectively. Five and eight phyla were detected at biofilm of pilot- and full-plant, respectively, by 16S rDNA sequencing. Biofilm of pilot-plant was dominated by ${\beta}$-Proteobacteria (38.8%), ${\gamma}$-Proteobacteria (22.4%), and Bacteroidetes (12.2%), and the most dominant genus was Zoogloeae genus (22.4%). Candidate division TM7 (12.5%) was only detected at biofilm of full-plant and it was dominated by Bacteroidetes (33.3%), ${\gamma}$-Proteobacteria (29.2%), and ${\beta}$-Proteobacteria (20.8%). Clostridium genus specific primer set enabled to detect the sequences of Clostridium genus. These suggested that anaerobic and aerobic bacteria were coexisted even from the initial period of biofilm formation and ${\beta}$-Proteobacteria, ${\gamma}$-Proteobacteria and Bacteroidetes were major phyla in biofilm of food wastewater treatment system at winter.
