Synthesis of New pH-Sensitive Amphiphilic Block Copolymers and Study for the Micellization Using a Fluorescence Probe

  • This paper reports a facile synthesis of new water-soluble poly(ethylene oxide) (PEO)-based amphiphilic block copolymers showing pH sensitive phase transition behaviors. The copolymers were prepared by atom transfer radical polymerization (ATRP) of methacrylamide type of monomers carrying a sulfonamide group using a PEO-based macroinitiator and a Cu(I)Br/$Me_6TREN$ catalytic system in aqueous media. The resulting polymers were characterized by a combination of $^1H$-NMR, size exclusion chromatography, and UV/Visible spectrophotometeric analysis. The micellization of the block copolymers as a drug-loading mechanism in aqueous media using fluorescein salt was examined as a function of pH. The stable micelle formation and its loading efficacy suggest that the block copolymers can be used as precursors for drug-nanocontainers.

Supramolecular Hydrogels Instantaneously Formed by Inclusion Complexation between Amphiphilic Oligomers and $\alpha$-Cyclodextrins

  • Supramolecular hydrogels were instantaneously fabricated by mixing aqueous solutions of $\alpha$-cyclodextrins ($\alpha$-CDs) and amphiphilic methoxy (polyethylene glycol) (MPEG)-$\varepsilon$-caprolactone (CL) oligomer, which was synthesized via the ring-opening polymerization of the CL monomer using low-molecular-weight MPEG ($M_n$ of MPEG=2,000 g/mol) as an initiator. The supramolecular structure of the hydrogels was revealed by X-ray diffraction (XRD) analyses. Rheological studies of the hydrogels revealed an elastic character when the number of CL units in the oligomer was more than 2, and the obtained hydrogels showed high storage modulus but relatively low shearing viscosity due to the low-molecular-weight character of the oligomer, which was more preferable for use as an injectable delivery system. The physical properties of the hydrogels could be modulated by controlling the chain morphology and concentration of the oligomers, as well as the feed molar ratio of the oligomer to $\alpha$-CD. The components of the supramolecular hydrogels are biocompatible and can readily be eliminated from the body. These features render the supramolecular hydro gels suitable as drug delivery systems and tissue engineering scaffolds.

실란 기능화 양친성 고분자 전구체를 이용한 소수성 항균물질 담지 나노 입자 제조 및 항균 특성 (Preparation of Hydrophobic Antimicrobal Compounds Encapsulated Nanoparticles Using Alkoxysilane-functionalized Amphiphilic Polymer Precursor and Their Antimicrobial Properties)

  • 본 연구에서는 50 wt%의 매우 높은 담지량과 70%의 높은 고형분을 가지면서도 물속이나 다양한 제품에 나노 입자 형태로 분산이 가능한 새로운 형태의 소수성 항균 물질 담지 나노 입자를 제조하였다. 실란 기능화 양친성 고분자 전구체(Alkoxysilane-functionalized Amphiphilic Polymer Precursor; 이하 AAPP)와 다양한 실란 화합물을 이용하여 전형적인 Hydrolytic Sol-Gel 공정으로 제조된 수분산 유-무기 하이브리드 나노 입자들을 제조하고, 이를 이용하여서 나노 침전법을 사용하여서 소수성 항균물질을 고함량으로 담지할 수 있는 새로운 공정으로 소수성 항균물질인 Eugenol이 담지된 유-무기 하이브리드 형태의 나노입자 제조하였다. 나노 입자 제조시 제조 조건의 변화에 따른 나노 입자들의 크기, 담지량, 항균 활성 및 방출거동 등에 영향을 미치는 인자들을 조사하였다. 나노 입자의 종류에 관계없이 Minimal Inhibitory Concentration (MIC)는 50 mg/ml로 동일하였고, 모든 균주에서 99 %에 해당하는 우수한 항균력과 Pseudomonas aeruginosa (PSE)를 제외하고는 2주 이상의 항균 지속력을 나타내었다. 특히, Tetraethoxysilane (TEOS)를 첨가한 경우에는 견고한 무기물 도메인으로 인해 가장 높은 담지량 (50 wt.%)과 서방출 (Sustained release)을 나타내었고, Hexanediol (HD)을 첨가한 경우에는 HD 자체의 항균력과 용매로서의 역할도 하였기 때문에 가장 높은 항균력과 70%의 고형분을 나타내었다.

  • Amphiphilic block copolymer는 self-assembly특성을 가지고 있기 때문에 선택된 용매 속에서 nanoscale-domain 즉 micelle을 형성할 수 있다. 이러한 특성은 기타 불용성 물질을 가용화하고 colloidal particle을 안정화시키거나 micro-emulsion을 형성할 수 있어 pharmaceutics, drug delivery system등 영역, 그리고 emulsion stabilizer, thickener, dispersion agent등으로 사용될 수 있다.   • This paper describes the electrochemical and photochemical behavior of amphiphilic Os bipyridine compelx adsorbed on SnO$_2$electrodes by the Langmuir-Blodgett(LB) film as monolayer state. Theoretical equation of cyclicvoltammetry for redox species were discussed. And the cyclic voltammogram were simulated taking account theinteraction parameters. From these obtained values, we could fit almost all measured voltammograms with these parameters.

  • Saponins are glycosidic compounds present in many edible and inedible plants. Structurally, they are composed of a lipid-soluble aglycone consisting of either a sterol or, more commonly, a triterpenoid and water-soluble sugar residues differing in type and amount of sugars [1]. Because of their amphiphilic nature, they are highly surface-active.   • Membranes are a central feature of all biological systems and their ability to control many cellular processes is critically important. As a result, a better understanding of how molecules bind to biological membranes is an active area of research. In this report, the interaction between our biomimetic structures and different biological membranes is reported using both model vesicle and in vitro bacterial cell experiments. These results show that lipid composition is more important for selectivity than overall net charge. An effort is made to connect model vesicle studies with in vitro data and naturally occurring lipid compositions.
