Reactive Extraction of Succinic Acid by Amino Extractants and Comparison of Extraction Characteristics of Maleic Acid (아민계 추출제를 이용한 숙신산의 반응추출 및 말산과의 추출특성 비교)

  • Succinic acid is of interested as the raw material of biodegradable polymer. In this study, succinic acid was separated by reactive extraction using amine extractants such as TOA (trioctylamine) and Aliquat 336. The extractability of TOA for succinic acid was higher than that of Aliquat 336. The distribution of succinic acid into organic phase was decreased with increasing pH in aqueous phase. However, the effect of pH on the extractability of Aliquat 336 was little. In the case of maleic acid which has similar structure to succinic acid, the extractability for maleic acid was higher than that for succinic acid. It was mainly due to the difficulty of deprotonation of second carboxylic group by intramolecular hydrogen bonding.

Biogenic Amine Contents of Commercial Salted and Fermented Sand Lance Ammodytes personatus Sauces (시판 까나리(Ammodytes personatus) 액젓의 biogenic amines 함량)

  • This study investigated the quality of seven commercial salted and fermented sand lance Ammodytes personatus sauces by measuring their chemical compositions, bacteria concentrations, and biogenic amine contents. The sauces had a 63.97-67.32% (mean: 65.72%) moisture content, 23.16-24.03% (mean: 23.60%) salinity, 5.51-6.14 (mean: 5.81) pH, 164.76-217.98 mg/100 g (mean: 182.97 mg/100 g) volatile basic nitrogen, 0.94-1.42% (mean: 1.24%) total nitrogen, and 673.77-791.86 mg/100 g (mean: 720.76 mg/100 g) amino nitrogen content. Viable cell counts ranged from $1.2{\times}10^1$ to $2.9{\times}10^2CFU/mL$, and number of biogenic amine-forming bacteria was very low or not detected. Overall, the tested samples had average levels of histamine of 301.02 mg/kg, cadaverine of 29.78 mg/kg, tyramine of 199.21 mg/kg, putrescine of 183.00 mg/kg, and tyramine of 13.01 mg/kg. This strongly suggests that it is necessary to monitor the biogenic amine contents of commercial salted and fermented sand lance sauces carefully to ensure consumer health.

Optimization of Gene Transfection Using Fluorescence-Activated Cell Sorter(FACS) Analysis of Green Fluorescent Protein(GFP) (Green Fluorescent Protein(GFP)의 Fluorescence-Activated Cell Sorter(FACS) 분석을 통한 유전자 이입의 최적화)

  • In order to improve the transfection efficiency of CHO/dhfr- cells using cationic lipid, optimal concentrations of the cationic lipid($LipofectAmine^{TM}$) and DNA(pEGFP-C1) need to be determined. The use of green fluorescent protein(GFP) gene as a reporter gene facilitated the quantification of transfection efficiency. The green fluorescence intensity of each cell transfected at various lipid-DNA concentrations was measured using fluorescence-activated cell sorter(FACS) analysis. A combination of $2.0{\mu}L$ cationic lipid and 0.4{$\mu}g$ DNA in a well resulted in the highest trasfection efficiency. Taken together, the method using FACS analysis of GFP is simple and fast, facilitating the optimization of transfection.

  • The organic solvent mixture of 5 % trioctylamine and 5 % n-octyl alcohol in n-hexane could be loaded with tungsten to 62 g $WO_3$/1 whereas the amine alone could be loaded only to 37 g $WO_3$/1. Molecular interaction between amine and tungstate ion during the solvent extraction process and the synergistic effect of alcohol in enhancing the solubility of the amine-tungstate complex have been studied by NMR spectroscopy. Tungsten has been shown to be extracted as a polymeric tungstate ion into the organic phase to form a trioctyl ammonium polytungstate salt, which was more stabilized in the hydrocarbon medium by interacting with alcohol via hydrogen bonding between the tungstate ion and alcohol.

Prediction on the Chiral Behaviors of Drugs with Amine Moiety on the Chiral Cellobiohydrolase Stationary Phase Using a Partial Least Square Method

  • Quantitative Structure-Resolution Relationship (QSRR) using the Comparative Molecular Field Analysis (CoMFA) software was applied to predict the chromatographic behaviors of chiral drugs with an amine moiety on the chiral cellobiohydrolase (CBH) columns. As a result of the Quantitative CoMFA-Resolution Relationship study, using the partial least square method, prediction of the behavior of drugs with amine moiety upon chiral separation became possible from their three dimensional molecular structures. When a mixed mobile phase of 10 mM aqueous phosphate buffer (pH 7.0) - isopropanol (95 : 5) was employed, the best Quantitative CoMFA-Resolution Relationship, derived from the study, provided a cross-validated $q^2$ = 0.933, a normal $r^2$ = 0.995, while the best Quantitative CoMFA-Separation Factor Relationship, also derived from the study, yielded a cross-validated $q^2$ = 0.939, a normal $r^2$ = 0.991. When all of these results are considered, this QSRR-CoMFA analysis appears to be a very useful tool for the preliminary prediction on the chromatographic behaviors of drugs with an amine moiety inside chiral CBH columns.

Kinetics and Reaction Mechanism for Aminolysis of Benzyl 4-Pyridyl Carbonate in H2O: Effect of Modification of Nucleofuge from 2-Pyridyloxide to 4-Pyridyloxide on Reactivity and Reaction Mechanism

  • Pseudo-first-order rate constants $k_{amine}$ have been measured spectrophotometrically for the reactions of benzyl 4-pyridyl carbonate 6 with a series of alicyclic secondary amines in $H_2O$ at $25.0^{\circ}C$. The plots of $k_{amine}$ vs. [amine] curve upward, indicating that the reactions proceed through a stepwise mechanism with two intermediates, a zwitterionic tetrahedral intermediate $T^{\pm}$ and its deprotonated form $T^-$. This contrasts to the report that the corresponding reactions of benzyl 2-pyridyl carbonate 5 proceed through a forced concerted pathway. The $k_{amine}$ values for the reactions of 6 have been dissected into the second-order rate constant $Kk_2$ and the thirdorder rate constant $Kk_3$. The Br${\o}$nsted-type plots are linear with ${\beta}_{nuc}=0.94$ and 1.18 for $Kk_2$ and $Kk_3$, respectively. The $Kk_2$ for the reaction of 6 is smaller than the second-order rate constant $k_N$ for the corresponding reaction of 5, although 4-pyridyloxide in 6 is less basic and a better nucleofuge than 2-pyridyloxide in 5.

  • Fourteen commercial beers (eight domestic and six imported) were investigated for their biogenic amine contents. In all products, six biogenic amines, putrescine, cadaverine, tryptamine, ${\beta}-phenylethylamine$, spermine, and tyramine, were detected at different contents among the products, with the spermine content being the highest. Contents of cadaverine and ${\beta}-phenylethylamine$ in domestic beers, and ${\beta}-phenylethylamine$ and tyramine in imported ones were not significantly different. Results indicate that continuous monitoring of biogenic amine levels in commercial beers is needed to maintain chemical safety and qualities.

Photochemical Reactions of Saccharin-$\alpha$-Silylamine Systems. Desilylmethylation of $\alpha$-Silylamine via Single Electron Transfer Pathway

  • Photochemical reactions of saccharin with tertiary amines were explored. Saccharin was found to undergo an acid-base reaction with N-trimethylsilylmethyl-N,N-diethyl amine to form N-trimethylsilylmethyl-N,N-diethyl ammonium saccharin salt which is in equilibrium with free saccharin and N-trimethylsilylmethyl-N,N-diethyl amine insolution. Photoreaction of N-trimethylsilylmethyl-N,N-diethyl ammonium saccharin in $CH_3OH\;or\;CH_3CN$ results in the generation of desilylmethylated product, N,N-diethyl ammonium saccharin mainly along with benzamide. Photoreaction of N-methylsaccharin with N-trimethylsilylmethyl-N,N-diethyl amine in $CH_3OH$ leads to the production of o-(N-methylcarbamoyl)-N-ethylbenzenesulfonamid e as the major product along with N-methylbenzamide as the minor product. On the other hand, photoreaction of N,N,N-triethyl ammonium saccharin, generated from saccharin and triethylamine, produces N-methylbenzamide as the exclusive product. These photoreactions are quenched by oxygen indicating that triplets of saccharin and N-methylsaccharin are the reactive excited states. Based on the consideration of the redox potentials of saccharin and N-trimethylsilylmethyl-N,N-diethyl amine, and the nature of photoproducts, pathways involving initial triplet state single electron transfer are proposed for photoreactions of the saccharins with the ${\alpha}$-silylamine.

  • Halloysite nanotube (HNT) has a nanotube structure with the chemical formula of Al2Si2O5(OH)4 · nH2O and is a natural sediment of aluminosilicate. A lot of research has been conducted to improve the mechanical properties of epoxy composites by generating interactions between HNTs and polymers through surface treatment of HNTs, such as exchange of amine group as a terminal functional group. However, most of the surface modification methods are performed under wet conditions, which require a relatively large amount of time, manpower and solvent. In order to save time and simplify complicated procedures, a dry coating machine was designed and used for amine group exchange. Comparing the XPS results, it was found that the results of NH2-HNT prepared using a dry coating machine and the substitution through the wet method were not significantly different, and it has been confirmed that the amount of solvent used and the time savings can be made.

  • This study was conducted to evaluate the performance of 2,4-D amine salt (2,4-dichlorophenoxy acetic acid) 40EC in broadleaf weeds control under diverse conditions of rate and time of application and soil type in fall barley field. The results obtained were summarized as follows: 1) At the rate of 70cc prod./10a, 2,4-D amine salt treated during the fully tillered stage of barley was effective for the control of most broadleaf weeds without crop injury. 2) The fully tillered stage was a optimun time lot the application of 2,4-D amine salt in fall barley. Earlier(zero-to four-leaf stage) or late. (boot stage to flowering) application than this stage caused a crop injury and resulted in yield reduction. 3) It was possible to safely use 2,4-D amine salt in fall barley regardless of soil types if applied the rate of 70cc prod./10a at the fully tillered stage.
