Association of Hepatocyte Nuclear factor-4α Polymorphisms with Type 2 Diabetes in Koreans (한국인에서의 hepatocyte nuclear factor-4α의 유전자 다형성과 제2형 당뇨병과의 연관성)

  • Type 2 diabetes is a typical polygenic disease complex, for which several common risk alleles have been identified. The hepatocyte nuclear factor-$4{\alpha}$ (HNF-$4{\alpha}$), a transcription factor involved in the regulation of serum lipid and glucose levels, has recently been associated with type 2 diabetes. Therefore, we investigated the genotype for the C>T polymorphism at position 12352 of the HNF-$4{\alpha}$ gene in Koreans and compared patient genotypes with those of the control group. 100 patients (63 males, 37 females) with a history of type 2 diabetes (T2DM) and 100 controls (36 males, 64 females) participated in this study. There was no association between 12352 C>T polymorphism in the HNF-$4{\alpha}$ gene and T2DM. The present study shows that HNF-$4{\alpha}$ 12352 C>T polymorphism may not be associated with the pathogenesis of T2DM. Further studies with larger populations may be needed for the development of diagnostic methods at a genetic level such as DNA chip.

Characteristics of Seed Storage Protein Affecting the Eating Quality of Japonica and Tongil-type Rice (자포니카 및 통일형 벼 품종에서의 식미 관련 저장단백질 특성)

  • In this study, we analyzed seed storage proteins in order to investigate the main factors related to the eating quality of japonica and tongil-type rice varieties. Sensory evaluation was performed by a trained panel to assess the appearance (color and glossiness), flavor, taste, stickiness, texture, and overall score of nine japonica and three tongil-type rice cultivars. Moreover, the pattern of variation in rice storage proteins was examined by electrophoresis of protein extracts. The electrophoretic pattern of rice proteins showed 16.4 kDa albumin, 26.4 kDa globulin, 34-39 kDa and 21-22 kDa glutelin, and 14.3 kDa prolamin. In terms of storage protein, the varietal differences between japonica and tongil-type rice were found in albumin, globulin, and the ${\alpha}-1$, and ${\alpha}-2$ sub-units of acidic glutelin. Furthermore, the overall sensory evaluation score was observed to be positively correlated with albumin ($0.495^{**}$) and globulin ($0.567^{**}$), and negatively correlated with ${\alpha}-1$ glutelin ($-0.612^{**}$). Therefore, the results indicated that albumin, globulin, and ${\alpha}-1$ glutelin can affect the eating quality of japonica and tongil-type rice varieties, with the latter having lower eating quality than the former.

Short-Term High Expression of Interferon-Alpha Modulates Progression of Type 1 Diabetes in NOD Mice

  • Type I diabetes (T1D) is an organ-specific autoimmune disease caused by the T cell-mediated destruction of the insulin-producing ${\beta}$ cells in the pancreatic islets. The onset of T1D is the consequence of a progressive destruction of islet ${\beta}$ cells mediated by an imbalance between effector $CD4^+$ T helper (Th)1 and regulatory $CD4^+$ Th2 cell function. Since interferon-alpha (IFN-${\alpha}$) has been known to modulate immune function and autoimmunity, we investigated whether administration of adenoviralmediated IFN-${\alpha}$ gene would inhibit the diabetic process in NOD mice. The development of diabetes was significantly inhibited by a single injection of adenoviral-mediated IFN-${\alpha}$ gene before 8 weeks of age. Next, we examined the hypothesis that Th2-type cytokines are associated with host protection against autoimmune diabetes, whereas Th1-type cytokines are associated with pathogenesis of T1D. The expression of IFN-${\alpha}$ induced increase of serum IL-4 and IL-6 (Th2 cytokines) levels and decrease of serum IL-12 and IFN-${\gamma}$ (Th1 cytokines) levels. Therefore, overexpression of IFN-${\alpha}$ by adenoviralmediated delivery provides modulation of pathogenic progression and protection of NOD mice from T1D.

Properties of Optically Selected Supernova Remnants in M31

  • We present a study of optically emitting supernova remnants (SNRs) in M31 based on $H{\alpha}$ and [S II] images in the Local Group Survey. Using these images, we have selected objects that have [S II]:$H{\alpha}$ > 0.4 and morphology for typical SNRs. We find 76 new SNRs. We have also inspected 233 SNR candidates presented in previous studies, confirming that only 80 of them are SNRs. Combining these, we produce a master catalog of 156 SNRs in M31. We classify these SNRs according to two types of criteria: the SNR progenitor types (Type Ia and core-collapse (CC) SNRs) and the morphological types. Type Ia and CC SNRs are 23% and 77%, respectively, of the total sample. Most of CC SNRs are concentrated along the spiral arms, while Type Ia SNRs are rather spread over the entire galaxy including the inner region. CC SNRs are brighter in $H{\alpha}$ and [S II] than Type Ia SNRs. The cumulative size distribution of the SNRs with 15 < D < 50 pc is well fitted by a power law with an index, ${\alpha}=2.53{\pm}04$. It indicates that most SNRs in M31 are in Sedov-Taylor phase. Properties of these SNRs show little variation depending on the galactocentric distance. The $H{\alpha}$ and [S II] surface brightness shows a good correlation with X-ray luminosity for the SNRs that are center-bright.

Differential Responses to TGF Alpha in between Invasive Squamous Cell Carcinoma Cell Line and Noninvasive One (침투성 상피암세포주와 비침투성 상피암세포주의 TGF alpha에 대한 반응의 차이)

  • Both SCC 12 and SCC 13 cell lines were derived from squamous cell carcinoma (SCC) of the skin (Wu and Rheinwald, 1981). In the present study, we compared the inherent invasive activity in their raft cultures where most in vivo characteristics of epidermis can be reproduced by cell culture method. The raft culture of SCC 12 cell line produced many invading colonies within the collagen lattice and basal-like cells in the middle of differentiating cell layers, but no invasive activity was observed in the SCC 13 raft culture. We investigated which factors are implicated in inherent invasive activity of SCC 12 cell line by examining basal levels of type I collagenase, EGF receptor, fibronectin, and its receptor in two cell lines. Among them, only type I collagenase was significantly higher in invasive SCC 12 cells than in non-invasive SCC 13 cells. Furthermore, we tried to investigate mechanisms underlying between SCC 12 cell's inherent invasive activity and its high basal level of type I collagenase. As one of them, discrepancy in TGF alpha mediated responses between two cell lines was observed. In SCC 13 cells, TGF alpha initially stimulated type I collagenase at 12 h after TGF alpha treatment and then its down regulation was followed from 24 h even though TGF alpha was continuously present in the medium. However in SCC 12 cells, TGF alpha continuously stimulated type I collegenase up to 48 h. We propose that defect in EGF receptor's down-regulation may be involved in lack of type I collagenase's down-regulation and its possible connection to invasive activity of SCC 12 cell line.

  • In this paper, we have considered a class of constrained non-smooth multiobjective fractional programming problem involving support functions under generalized convexity. Also, second order Mond Weir type dual and Schaible type dual are discussed and various weak, strong and strict converse duality results are derived under generalized class of second order (F, ${\alpha}$, ${\rho}$, $d$)-V-type I functions. Also, we have illustrated through non-trivial examples that class of second order (F, ${\alpha}$, ${\rho}$, $d$)-V-type I functions extends the definitions of generalized convexity appeared in the literature.

Nimodipine as a Potential Pharmacological Tool for Characterizing R-Type Calcium Currents

  • Nimopidine, one of dihydropyridine derivatives, has been widely used to pharmacologically identify L-type Ca currents. In this study, it was tested if nimodipine is a selective blocker for L-type Ca currents in sensory neurons and heterologous system. In mouse dorsal root ganglion neurons (DRG), low concentrations of nimodipine $(<10\;{\mu}M),$ mainly targeting L-type Ca currents, blocked high-voltage-activated calcium channel currents by ${\sim}38%.$ Interestingly, high concentrations of nimodipine $(>10\;{\mu}M)$ further reduced the 'residual' currents in DRG neurons from ${\alpha}_{1E}$ knock-out mice, after blocking L-, N- and P/Q-type Ca currents with $10\;{\mu}M$ nimodipine, $1\;{\mu}M\;{\omega}-conotoxin$ GVIA and 200 nM ${\omega-agatoxin$ IVA, indicating inhibitory effects of nimodipine on R-type Ca currents. Nimodipine $(>10\;{\mu}M)$ also produced the inhibition of both low-voltage-activated calcium channel currents in DRG neurons and ${\alpha}_{1B}\;and\;{\alpha}_{1E}$ subunit based Ca channel currents in heterologous system. These results suggest that higher nimodipine $(>10\;{\mu}M)$ is not necessarily selective for L-type Ca currents. While care should be taken in using nimodipine for pharmacologically defining L-type Ca currents from native macroscopic Ca currents, nimodipine $(>10\;{\mu}M)$ could be a useful pharmacological tool for characterizing R-type Ca currents when combined with toxins blocking other types of Ca channels.

  • Recently, according to the low-carbon green growth policy, the Stirling Engine has been increasing the interest of some sort of external combustion engines Stirling Engine is largely divided into three types ${\alpha}$, ${\beta}$ and ${\gamma}$. Among them, a ${\gamma}$-type engine is easy to design and can be applied variously. However, owing to the connection distance between the cylinder and the crank, there was a problem that ${\gamma}$-type occupies a larger volume than the ${\alpha}$ and ${\beta}$-types. As one of the methods to solve this problem, we have to consider about the Yoke crank which have been applied mainly to the ${\alpha}$-type. Because there are two advantages in not only reduction of engine volume but also parallelizing the cylinder. In this study, leading on from the existing design techniques for the Ross Yoke crank, this paper presents a simple method that can apply the yoke crank design of the ${\gamma}$-type stirling engine.
