A taxonomic review of Korean Allium (Alliaceae) (한국산 부추속(Allium, Alliaceae)의 분류학적 재검토)

  • Allium taxa of Korea were taxonomically reviewed, and classified as three subgenera, ten sections, seventeen species, and three varieties. Among these, Korean endemics were recorded as six taxa; A. koreanum, A. taquetii, A. deltoide-fistulosum, A. linearifolium, A. thunbergii var. deltoides, A. thunbergii var. teretafolium. In addition, A. condensatum, A. splendens and A. maximowiczii proved to distribute only in North Korea. Keys to the subgenera, sections, species and infraspecies with descriptions of each taxon were provided.

  • Microscopic observations of arbuscular mycorrhizae (AM) were done with the colonizations of AM and the determinations of chitin in the plant roots of Sorghum bicolor, Cassia mimosoides, Capsicum annuum and Allium fistulosum. The intercellular and intracellular hyphae, arbuscules and vesicles were microscopically observed, according to increases of colonization of AM in the roots of four plants.   • This study was attempted to know the effect of crude extracts of Allium fistulosum on the seed germination and seedling growth in compositae. The result is as follows; The seed germination and seedling growth in the tested compositae were inhibited by the crude extracts from leaf blade, leaf sheath and root of Allium fistulosum. Generally the crude extracts from leaf blade part showed the highest inhibitory effect in the tested parts. The seed germination was inhibited by the crude extracts of Allium fistulosum extracted with 80% methyl alcohol and distilled water. But the crude extracts which had extracted with distilled water indicated more inhibitory effect than that of 80% methyl alcohol. The seedling growth was depressed by the root residues of Allium fistulosum in the soil after harvesting. The crude extracts had the character of stability in light and heat. Some out of the inhibitors contained the volatile substances.

Identification of a Tandemly Repented DNA Sequence Using Combined RAPD and FISH in Welsh Onion (Allium fistulosum)

  • A tandemly repeated DNA sequence was identified and characterized y the combined RAPD and FISH data from a total genomic DNA of Welsh onion (Allium fistulosum). A clone containing this repeating sequence was selected and sequenced. This repeating unit of 314 bp inserted into pAf 072 contained 54.1% adenine and thymine residues, and showed the primer sequence used, 5'-GAAACGGGTG-3', in both terminals of the sequence. Fluorescence in situ hybridization using this tandemly repeated sequence as a probe indicated that the detected sites were coincident with the major C-banded constitutive heterochromatin in the terminal regions of both arms of all 6 chromosomes.

Phylogenetic Analyses of Nuclear rDNA ITS Sequences of Korean Allium L. Subgenus Rhizirideum(Alliaceae)

  • Phylogenetic relationships among the Korean taxa of the genus Allium subgenus Rhizirideum and some related taxa were assessed on the basis of in sequences of nuclear ribosomal DNA. Twenty-eight accessions of the genus Allium L. consisting of subgenera Rhizirideum (19 taxa), Allium (5 taxa) and Amerallium (one taxon) were analyzed. The variation in the ITS region was informative at the levels of section except for sect. Reticulato- bulbosa which is known to be of multiple origin. The ITS 2 region was longer than the ITS 1 region, and all of the investigated Allium taxa were the same in length in the 5.8S region except for A. monanthum. Allium cyaneum var. cyaneum was the shortest (635 bp) and A. victorialis the longest (646 bp) among the investigated Korean taxa. The three morphologically similar taxa, A. thunbergii, A. sacculiferum that has been included in A. thunbergii, and A. deltoid- fistulosum, had the same ITS lengths of 641 bp, but were clearly distinguished in the phylogenetic analysis of their ITS sequences.

  • Giemsa C-banded karyotypes of A. thunbergii, A. deltoide-fistulosum, A. cyaneum and A. cyaneum var. deltoides were analyzed, and the interspecific relationships were investigated on the basis of the C-banding patterns. The banding pattern of each species was unique and made possible an easy and clear separation among them. A. thunbergii and A. deltoide-fistulosum revealed very close banding relationship but a. cyaneum and A. cyaneum var. deltoides showed significant difference in banding pattern in spite of their close plant systematic position.

Giemsa C-banded Karyotypes in Two Diploid and Two Tetraploid Allium Species (Allium속 이배체 2종과 사배체 2종의 Giemsa C-분염핵형)

  • C-banded karyotypes in A. anisopodium, A. sacculiferum, A. deltoide-fistulosum and A. splendens were investigated. The chromosome compositions were diploid of 2n=16 in A anisopodium and A. splendens, and teiraploid of 2n=32 in a. sacculiferum and A. deltoide-fistulosum. The bands were distributed in telomeric parts of the chromosomes dominantly in addition to interstitial regions sporadically, resulting in the specific C-banded karyo types according to the species. No centromeric band was observed in these species except only one chromosome in A. deltoidefistulosum. The interspecific relationship based on the C-band distribution will be discussed.

  • The structures of dioscin(1) and saponin P-d(2), isolated from the subterranean part of Allium fistulosum, were determined as diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow2)-[$\alpha$-L-rhamnopyranosyl (1longrightarrow4)]-$\beta$-D-glucopyranoside (1, ) and diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow4)-$\alpha$-L-rhamnopyranosyl(1longrightarrow4)[$\alpha$-L-rhamno-pyranosyl (1longrightarrow2)]-$\beta$-D-glucopyranoside (2) by spectroscopic and chemical degradational methods. The prosa-pogenin(4), which was provided on partial hydrolysis of 2, was not reported in previous literature.

  • Studies were conducted to control Spodoptera exigua in Allium fistulosum field using synthetic sex pheromone trap. Heavily, medium and lightly damagd fields at early transplanting time showed no differences in control effects after 8 weeks of establishment of pheromone traps in Allium fistulosum fields. Number of damaged leaves in each treated field was 0.2-3 individuals per 60 hills comparing with 33.2 individuals at non-treated field. Also, the numbers uf male adults attracted to pheromone traps were 1.8-5.0 individuals at the big inning of installation but increased to 25.8, 49.9, 74.9, and 77.4 individuals after 2, 4, 6, and 8 weeks, respectively. There was a negative correlation between amount of male adults attracted to pheromone traps and number of damaged leaves (r=-0.9572*). Number of adults attracted to various colored light trap for 30 days were 6 in red, 11 in white and none in blue comparing with 1,041 individuals in pheromone trap.

  • BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a leading cause of chronic liver disease and is closely associated with metabolic syndrome. In the present study, we observed the effect of ethanol extract of Allium fistulosum (EAF) on NAFLD and have suggested the possibility of using EAF as a natural product for application in the development of a treatment for NAFLD. MATERIALS/METHODS: The preventive effect on hepatic lipid accumulation was estimated by using an oleic acid (OA)-induced NAFLD model in vitro and a Western diet (high-fat high-sucrose; WD)-induced obese mouse model. Animals were divided into three groups (n = 7): normal diet group (ND), WD group, and WD plus 1% EAF group. RESULTS: EAF reduced OA-stimulated lipid accumulation in HepG2 cells in the absence of cellular cytotoxicity and significantly blocked transcriptional activation of sterol regulatory element-binding protein 1 and fatty acid synthase genes. Subsequently, we investigated these effects in vivo in mice fed either ND or WD in the presence or absence of EAF supplementation. In comparison to the ND controls, the WD-fed mice exhibited increases in body weight, liver weight, epididymal fat weight, and accumulation of fat in hepatocytes, and these effects were significantly attenuated by EAF supplementation. CONCLUSIONS: Allium fistulosum attenuates the development of NAFLD, and EAF elicits anti-lipogenic activity in liver. Therefore, EAF represents a promising candidate for use in the development of novel therapeutic drugs or drug combinations for the prevention and treatment of NAFLD.
