Antioxidant Potentials and Quantification of Flavonoids in Mung Bean (Vigna radiata L.) Seeds

  • Mung bean (Vigna radiata L.) is an increasingly important human food source, as well as a new functional agent, mainly due to its potent antioxidant activity. This study was conducted to determine antioxidant activity of fractions from mung bean seeds by measuring DPPH radical scavenging activity and to quantify the flavonoids by means of HPLC analysis. Vitexin and isovitexin were present in both ethanol and water extracts in highest amount. Flavonoids, vitexin and isovitexin were quantified from 195 germplasms of mung beans and their concentrations varied by 4.7 fold. Especially, the breeding line KM99004-4B-2 (Suwon28/KM94004), which has grown in Jeollanamdo Agricultural Research and Extension Services, showed the highest amount (15.88 mg/g) of total flavonoids. The vitexin portion was averaged $70.73{\pm}1.38\%$. High positive correlation $(r=0.96^{***})$ between vitexin and isovitexin contents showed. However, the flavonoid content showed very low correlation with the 24 growth and ecological characteristics. Seed coats of mung beans had the highest flavonoid amount, showing $50\~70$ times more than cotyledons. Flavonoid contents in the seed, the cotyledon, and the seed coat were decreased as the seed imbibition time increased.

Effects of Intraperitoneal Administration of Lactococcus lactis ssp. lactis Cellular Fraction on Immune Response

  • Cellular components of Lactococcus lactis ssp. lactis (heat-killed whole cells, cytoplasm, and cell walls) were tested for their in vivo immunopotentiating activities. Peritoneal macrophages from mice injected intraperitoneally with cell-wall fractions exhibited significantly greater phagocytic activity than groups injected with whole cells or cytoplasm fraction. Cytotoxicity of natural-killer cells was highest in cytoplasm fractions. Production of cytokines (IFN-${\gamma}$, IL-2, IL-6, and IL-12) in spleen cells was significantly higher when cellular components were injected intraperitoneally, and tended to be higher in whole-cell and cytoplasm groups than in cell-wall group. These results demonstrate lactic acid bacteria whole cells and their cytoplasm and cell-wall tractions have immunopotentiating activities.

Nelumbo nucifera extracts mediated synthesis of silver nanoparticles for the potential applications in medicine and environmental remediation

  • Silver nanoparticles (AgNPs) were successfully synthesized through a simple green route using the Nelumbo nucifera leaf, stem and flower extracts. These nanoparticles showed characteristic UV-Vis absorption peaks between 410-450 nm which arises due to the plasmon resonance of silver nanoparticles. The Fourier transform infrared spectroscopy (FT-IR) confirmed the presence of amides and which acted as the stabilizing agent. X-ray diffraction spectrum of the nanoparticles confirmed the Face centered cubic (FCC) structure of the formed AgNPs. Dynamic light scattering technique was used to measure hydrodynamic diameter (68.6 nm to 88.1 nm) and zeta potential (-55.4 mV, -57.9 mV and 98.9 mV) of prepared AgNPs. The scanning electron micrographs of dislodged nanoparticles in aqueous solution showed the production of reasonably monodispersed silver nanoparticles (1-100 nm). The antimicrobial activity of prepared AgNPs was evaluated against fungi, Gram-positive and Gram-negative bacteria using disc diffusion method. Anti-corrosion studies were carried out using coupon method (mild steel and iron) and dye degradation studies were carried out by assessing photo-catalytic activity of Nelumbo nucifera extracts mediated AgNPs.

유기 게르마늄 농축수로 재배한 농산물의 항염 및 함암효과 (Anti-inflammatory and Anti-cancer Effects of Agricultural Produce Grown with Organic Germanium-enriched Water)

  • The study was conducted to identify the anti-inflammatory and anti-cancer effects in sprouts of mouse-eyed bean (Rhynchosia nulubilis), ginseng (Panax ginseng), perilla (Perilla frutescens), broccoli (Brassica oleracea var. italica), and lettuce (Lactuca sativa) grown with organic germanium concentrate. Western blot analysis was performed to assess the anti-inflammatory activity of the extract. All extracts exhibited noticeable anti-oxidant activity, indicating a significant correlation between the germanium content and anti-oxidant activity (p<0.05). In particular, rat-eyed bean sprouts with the highest germanium content showed significant anti-inflammatory activity (p<0.05) by significantly inhibiting the expression of the inflammatory complexes, NLRP3, cytokines IL-1β and caspase-1. Ginseng and broccoli sprouts showed strong anti-cancer properties and had high anti-oxidant effects (p<0.001). Germanium-concentrated water allows the mass production of agricultural products containing high concentrations of organic germanium. Agricultural produce grown with germanium concentrate add organic germanium to various physiological active ingredients, increasing the anti-oxidant and anti-cancer effects. These results strongly suggest that agricultural products containing high germanium concentrations can be used as novel health supplements to improve health.

치유관광객의 여가활동 특성, 스트레스, 삶의 질의 관계 - 농촌 치유관광 관여도의 조절효과 - (The Relationships between Leisure Activity Characteristics, Stress, and Quality of Life of Healing Tourists - The Moderating Effect of Involvement on Rural Healing Tourism -)

  • Leisure activity characteristics are related to an individual's stress management as well as quality of life. This study analyzed the relationship between leisure activity characteristics, stress, and quality of life of healing tourists, and analyzed the moderating effect of involvement on rural healing tourism. For this purpose, this study analyzed 658 cases who experienced rural healing tourism. Among the characteristics of leisure activities, the conflict between work and leisure had a positive (+) effect on stress, and that stress had a negative (+) effect on the quality of life. Among the characteristics of leisure activities, social support was found to have a positive (+) effect on the quality of life. In the relationship between stress and quality of life, involvement on rural healing tourism was found to have a moderating effect. The results of this study indicate the importance of social support for work-life balance and leisure, suggesting that daily recovery through rural healing tourism has a positive effect on personal stress management as well as quality of life.

꽃공예 활동 프로그램의 치유효과 분석 - 지각된 스트레스, 우울, 자아존중감을 중심으로 - (Analysis of the Healing Effects of a Flower Craft Activity Program - Focusing on Perceived Stress, Depression, and Self-Esteem -)

  • This study aims to explore comprehensively the effects of a flower craft activity program focused on perceived stress, depression, and self-esteem of adult with mixed methods. The main results of this study are as follows. In the mixed analysis results, perceived stress was reduced by immersing oneself in completing one's own work through flower craft activities, as distracting thoughts in the head disappeared. In the case of depression, depression decreased in the relationship between group members who worked together to complete their own work through flower craft activities. In the case of self-esteem, it was analyzed that flower craft activities had the effect of increasing self-esteem through the satisfaction of the need for life and self-esteem. In future study, this suggests that the healing program applied based on this can be comprehensively reviewed, modified, and supplemented to reorganize it into a more effective flower craft activity program.

Screening and Identification of an Antifungal Pseudomonas sp. That Suppresses Balloon Flower Root Rot Caused by Rhizoctonia solani

  • A pathogenic fungus causing balloon flower root rot (Platycodon grandiflorum) was isolated from naturally infected roots. The microbial characteristics of the isolated microorganism were similar to those of Rhizoctonia solani. About 500 bacterial species from field soils were screened for a biological agent against the above-mentioned putative pathogen, and several bacteria with the antifungal activity were isolated. Among them, the isolated JS2 was identified as Pseudomonas aeruginosa. This strain showed a broad spectrum of antifungal activity potentially. When the antifungal substance was purified from a broth culture of JS2, it was identified as 2,4-diacetylphloroglucinol (Phl).

Effect of extraction conditions on chemical composition and antioxidant properties of mulberry fruit

  • Mulberry fruit (Morus alba L.) contains phytochemicals, including 1-deoxynojirimycin, quercetin-glucoside, kaempferol-glucoside, and anthocyanins, which have antioxidant effects. In this study, mulberry fruit extract was prepared at various temperatures (25-100℃) and water/ethanol solvent concentrations (0%-100% ethanol). Fourier-transform infrared spectroscopy (FT-IR) and circular dichroism (CD) data indicated that the content of bioactive compounds such as polyphenols and flavonoids was lower in 100% ethanolic extracts than in 30%-50% ethanolic extracts. Radical scavenging activity determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-di-3-ethylbenzthiazoline sulfonic acid (ABTS) assays was highly correlated with polyphenol and flavonoid content. In conclusion, 30%-50% ethanolic extracts contained the highest contents of bioactive compounds and exhibited high levels of radical scavenging activity. These findings may inform the use of mulberry fruit extract as a functional food.

녹지경관구조가 신체활동 및 건강에 미치는 영향 (Landscape Structure Influencing Physical Activity and Health)

  • The study aims to identify factors related to landscape structure and socioeconomic characteristics that influence adults' physical activity and health. The study analyzes data from 148,643 respondents from the Korea Community Health Survey conducted in 2021. The study measures various factors, including the frequency of physical activity, stress, depression, and landscape metrics of forest patches. Hierarchical multiple regression analysis was used, controlling for socio-demographic characteristics. Larger forest patches and more irregular shapes are associated with higher levels of physical activity among adults. The percentage and area of the forest patch are associated with fewer mental health complaints. Configuration-related landscape metrics such as shape or arrangement of the forest patch did not show a significant association with physical activity. Overall, the study provides insights into how specific characteristics of forest patches in urban areas may influence physical activity and mental health among adults. It emphasizes the importance of green spaces and their potential positive effects on both physical and mental well-being.

Novel Properties for Endoglucanase Acquired by Cell-Surface Display Technique

  • In order to improve the stability of endoglucanase under thermal and acidic conditions, the endoglucanase gene was fused to the N-terminus of the Saccharomyces cerevisiae pir gene, encoding the cell wall protein PIR. The fusion gene was transformed into Pichia pastoris GS115 for expression. A resulting strain with high expression and high activity was identified by examining resistance to Geneticin 418, Congo red staining, and quantitative analysis of enzyme activity. SDS-PAGE analysis revealed that the endoglucanase was successfully displayed on the yeast cell surface. The displayed endoglucanase (DEG) showed maximum activity towards sodium carboxyl methyl cellulose at approximately 275 IU/g cell dry weight. DEG exhibited greater than 60% residual activity in the pH range 2.5-8.5, higher than free endoglucanase (FEG), which had 40% residual activity at the same pH range. The highest tolerated temperature for DEG was 70℃, much higher than that of FEG, which was approximately 50℃. Moreover, DEG showed 91.1% activity at 65℃ for 120 min, while FEG only kept 77.8% residual activity over the same period. The half-life of DEG was 270 min at 65℃, compared with only 150 min for FEG. DEG could be used repeatedly at least three times. These results suggest that the DEG has broad applications as a yeast whole-cell biocatalyst, due to its novel properties of high catalytic efficiency, acid-thermal stabilities, and reusability.
