대두와 인체생리(2)-대두 이소플라본의 체내흡수

  • 이소플라본은 대두와 대두가공제품에 들어있으며 화학적으로 두가지 형태 즉 아글리콘(aglycone)과 배당체(glucoside)로 존재한다. 본 논문은 사람에게 대두의 이소플라본이 흡수되는데 아글리콘과 배당체가 차이가 있는지를 연구하였다. 이소플라본을 저농도(0.11mmol)로 1회 투여했을 때 아글리콘형은 섭취 2시간 후에 혈장의 이소플라본 농도가 최고치에 도달하였으며 배당체형은 섭취 4시간 후에 최고치에 도달하였다: 이때 피험자는 4명의 남성(41세)과 4명의 여성(45세) 이었다. 또한 8명의 피험자〔남성 4명(40세), 여성 4명(47세)〕를 대상으로 고농도(1.7mmol) 이소플라본을 1회 섭취시킨 후 혈장의 이소플라본 농도 변화를 비교하였으며, 아글리콘 섭취 후 혈장 이소플라본의 최고 농도는 배당체로 섭취한 경우보다 5배가 높았다. 저농도와 고농도의 두 실험에서 모두, 제니스테인과 다이드제인의 섭취수준은 유사함에도 불구하고 혈장의 제니스테인의 농도가 다이드제인보다 유의적으로 높았다. 또한 8명의 남성(45세)을 대상으로 4주간의 장기간 이소플라본(0.30mmol/ 일)을 섭취시킨 후 혈장의 이소플라본 농도를 측정하였다. 2주와 4주째에 혈장내 농도는 배당체형보다 아글리콘 섭취 후에 100% 이상 높은 것으로 나타났다. 이소플라본 아글리콘은 배당체보다 사람에게 더 빠르고 더 많은 량이 흡수되었다. 이소플라본 아글리콘이 풍부한 제품은 이소플라본 배당체가 풍부한 제품보다 관상 심장질환 등 만성질환 예방에 더 효과적일 수 있을 것이다.

Platycosides from the Roots of Platycodon grandiflorum and Their Health Benefits

  • The extracts and pure saponins from the roots of Platycodon grandiflorum (PG) are reported to have a wide range of health benefits. Platycosides (saponins) from the roots of PG are characterized by a structure containing a triterpenoid aglycone and two sugar chains. Saponins are of commercial significance, and their applications are increasing with increasing evidence of their health benefits. The biological effects of saponins include cytotoxic effects against cancer cells, neuroprotective activity, antiviral activity, and cholesterol lowering effects. Saponins with commercial value range from crude plant extracts, which can be used for their foaming properties, to high purity saponins such as platycodin D, which can be used for its health applications (e.g., as a vaccine adjuvant). This review reveals that platycosides have many health benefits and have the potential to be used as a remedy against many of the major health hazards (e.g., cancer, obesity, alzheimer's) faced by populations around the world. Methods of platycoside purification and analysis are also covered in this review.

Effects of citrus aglycone flavonoids, hesperetin and naringenin, on triacylglycerol metabolism in hamsters fed with a cholesterol diet

  • Effects of hesperetin and naringenin on the concentration of triacylglycerol in the serum and liver were studied in male golden hamster fed with the semipurified diet containing at 1% level of them for 3 weeks. The concentration of triacylglycerol in serum of the naringenin group decreased by 31%, whereas that in liver increased by 37% compared to the control group. The concentration of triacylglycerol in the serum and liver of the hesperetin group was slightly lower than the control group. The activity of microsomal phosphatidate phosphohydrolase in the liver, which is a key enzyme for biosynthesis of triacylglycerol, was significantly inhibited in the hesperetin group, whereas it was not affected in the naringenin group. The effect of hesperetin on phosphatidate phosphohydrolase was also measured in vitro. Hesperetin decreased the activity of phosphatidate phosphohydrolase with a dose-dependent manner. Both naringenin and hesperetin did not statistically affect the daily food consumption, body weight, liver weight, and total cholesterol in the serum. The observation accounts for the hypotriglyceridemic effect of hesperetin in the hyperlipidemic hamster.

Anti-Rheumatoid Arthritis Effect of the Kochia scoparia Fruits and Activity Comparison of Momordin Ic, its Prosapogenin and Sapogenin

  • MeOH extract of Kochia scoparia was fractionated into $CHCl_3-$, EtOAc- and BuOH extracts and the last fraction were hydrolyzed by 3%-NaOH ($MeOH-H_2O$) to compare the bioactivities on antinociceptive and anti-inflammatory effects. Silica gel column chromatography of BuOH fraction afforded a large amount of $3-Ο-{\beta}-D-xylopyranosyl {\;}(1{\rightarrow}3)-{\beta}-D-glucuronopyranosyl$ oleanolic acid (momordin Ic, 4) and that of acid hydrolysate of BuOH fraction gave $3-Ο-{\beta}-D-glucuronopyranosyl oleanolic$ acid (momordin Ib, 3), its 6'-Ο-methyl ester (2) and oleanolic acid (1). Silica gel column chromatography of alkaline hydrolysate afforded a large amount of 4. MeOH extract and both EtOAc- and BuOH fractions were active in the rheumatoidal rat induced Freund's complete adjuvant reagent (FCA) whereas $CHCl_3$ fraction was inactive. Compound 1 and 4 showed significant activities in the same assay but oleanolic acid 3-Ο-glucuronopyranoside (3) showed no activity. These fashions were also observed in carrageenan-induced edema of the rat and in the antinociceptive activity tests undertaken in hot plate- and writhing methods. These results suggest that momordin Ic and its aglycone, oleanolic acid, could be active principles for rheumatoid arthritis.

  • The GUE6 was isolated from ethyl acetate fraction of Glycyrrhiza uralensis and confirmed as liquiritin. Liquiritin(LQ) treated with ${\beta}$-glucosidase from plant(Prunus dulcis) and bacteria(Lactobacillus pentosus) crude enzymes. The ${\beta}$-glucosidase activities of crude enzymes were 229.8 U/g(Prunus dulcis) and 19.17 U/ml(Lactobacillus pentosus), respectively. Liquiritin(LQ) biotransformed into liquiritigenin(LQG) by ${\beta}$-glucosidase from crude enzymes. The EtOAc fraction(GUE6) and the converted product were identified as liquiritin and liquiritigenin, by TLC chromatogram, $^{1}H$-NMR and $^{13}C$-NMR.

양파즙 첨가 생면의 최적화 (Optimization of Wet Noodle with Onion Juice using Response Surface Methodology)

  • The noodle is one of the most popular meal as a main dish in Korea and onions are reported to contain high levels of flavonoids such as quercetin, aglycone, and quercetin 4-glycoside. This study was conducted to develop the wet noodle with onion juice and to examine the cooking quality, textural properties of it. Five sensory attributes such as gloss, chewiness, adhesiveness, odor, and taste were used for sensory evaluations. Optimal amount of onion(149.5 g) and salt (6.5 g) satisfied target sensory score(7.0) was suggested by response surface methodology. The wet noodle with optimal amount of onion juice and salt was tasted by 100 consumer and showed high score(6.67) in overall acceptability. Cooking quality was measured through absorption of water, volume, degree of turbidity. The volume, water absorption, and cooking water turbidity of cooked wet noodle with onion juice(experimental noodle) were significantly(p<0.01) higher than the control one. Texture profile analysis of experimental noodle showed a lower score of hardness, adhesiveness, springiness, and chewiness. However, it appeared that the experimental noodle showed higher score for cohesiveness than the control noodle.

  • Glycyrrhizin (18$\beta$-glycyrrhetic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL), a main component of liquore extract (Glycyrrhiza glabra), is ingested orally as a component in the oriental medicine. By human intestinal bacteria, glycyrrhizin (18$\beta$-glycyrrhetinic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL) was metabolized to glycyrrhetinic acid (GA): main pathway metabolizing GL to GA by glucuronidases of Bacteroides J-37 (Kim et al., 1997) and Eubacterium sp strain GLH (Akao et al., 1987) and minor pathway metabolizing GL to GA via 18$\beta$-glycyrrhetic acid D-glucuronic acid (GAMG) by $\beta$-glucuronidase of Streptococcus LJ-22 and glucuronidases of Bacteroides J-37 / E. coli. $\beta$-Glucuronidase from Streptococcus LJ-22 hydrolyzed GL to GAMG, not GA. $\beta$-Glucuronidase of Streptococcus LJ-22 hydrolyzed $\beta$-glucuronic acid conjugates of polysaccharides rather than aglycone-$\beta$-glucuronides Optimal pH of Streptococcus LJ-22 $\beta$-glucuronidase was 5-6 and its molecular weight was 250 kDaltons. Km for GL was 0.37mM.

Role of Metabolism by Intestinal Bacteria in Arbutin-Induced Suppression of Lymphoproliferative Response in vitro

  • Role of metabolism by intestinal bacteria in arbutin-induced immunotoxicity was investigated in splenocyte cultures. Following an incubation of arbutin with 5 different intestinal bacteria for 24 hr, its aglycone hydroquinone could be produced and detected in the bacterial culture media with different amounts. Toxic effects of activated arbutin by intestinal bacteria on lymphoproliferative response were tested in splenocyte cultures from normal mice. Lipopolysaccharide and concanavalin A were used as mitogens for B- and T-cells, respectively. When bacteria cultured medium with arbutin was treated into the splenocytes for 3 days, the medium cultured with bacteria producing large amounts of hydroquinone induced suppression of lymphoproliferative responses, indicating that metabolic activation by intestinal bacteria might be required in arbutin-induced toxicity. The results indicated that the present testing system might be applied for determining the possible role of metabolism by intestinal bacteria in certain chemical-induced immunotoxicity in animal cell cultures.

자두 Anthocyanin의 구조확인(構造確認)에 관(關)한 연구(硏究) (Studies on the Identification of the Anthocyanins in Plum)

  • 우리나라 재배종(栽培種) Santa rosa자두 중(中)에 함유(含有)된 두가지 antocyanin 색소(色素)를 분리확인(分離確認)하였다. 자두로부터 0.5% HCl methanol로 색소(色素)를 추출(抽出)하여 Amberlite IRC-50 이온 교환수지(交換樹脂) column 으로 정제(精製)하고, paper chromatography에 의하여 개별색소(個別色素)를 분리(分離)하였다. 두 색소(色素)는 다음과같은 실험(實驗)을 거쳐 얻은 결과(結果)들에 기초(基礎)하여 cyanidin-3-xyloglucoside 및 cyaniden-3-monoglucoside 임을 확인(確認)하였다. 여러 용매계(溶媒系)에 의한 paper chromatography의 Rf치(値), 부분산가수분해(部分酸加水分解), 결합당(結合糖)의 paper chromatography에 의한 확인(確認), 결합당(結合糖)의 단계가수분해(段階加水分解), aglycone의 alkali분해(分解), 및 가시부(可視部) 흡수(吸收) spectra. 자두중(中)에 함유(含有)된 주색소(主色素)는 cyanidin-3-monoglucoside였다.

Fababean 중의 Vicine과 Convicine의 안정성에 관한 연구 (Study on the Stability and Fate of Vicine and Convicine in Fababean)

  • Fababea에 존재하는 vicine과 convicine은 고온의 열처리에서 안정하였으며, vicine 용액도 약산성(pH5)과 알카리성 pH(pH10)에서 열에 대한 안정성이 높게 나타났다. 반면 산성 pH에서 vicine은 산 가수분해되어 aglycone인 divicine을 형성하며, 생성된 divicine은 매우 불안정하여 쉽게 분해되었다. 효소(${\beta}-glucosodase$)에 의해 vicine을 가수분해시킨 후 생성된 divicine의 산화에 의한 분해 반응 동안(pH5, $80^{\circ}C$), half-life time과 rate constant는 각각 14.35분과 -0.018이었다. 따라서 fababean을 함유한 식품을 가공하는 동안 효소 (${\beta}-glucosidase$)를 처리하여 vicine과 convicine을 가수분해 시키면 이들의 양은 크게 감소될 것이다.
