Bioconversion of Soybean Isoflavone by Lactobacillus plantarum and Bifidobacterium longum (Lactobacillus plantarum과 Bifidobacterium longum을 이용한 대두 이소플라본의 비배당체로의 전환)

  • In this study, phytoestrogen for the industrial production of soybean probiotics by lactic acid bacteria (LAB) was studied in a soybean extract. Soybean was fermented with LAB, Lactobacillus plantarum KCTC 3108 and Bifidobacterum longum ATCC 15707. The change in the content of various isoflavones (aglycone and glucoside) and the $\beta$-glucosidase activity in soybean during fermentation were investigated and shown to be dependent on the starter organism. Soybean extract powder fermented with L. plantarum showed the highest $\beta$-glucosidase activity and the greatest increase in the aglycone content. After 48h of fermentation, the contents of daidzin, genistin and glycitin in L. plantarum decreased from a mean initial levels of $83.03{\pm}2.17$, $168.13{\pm}8.17$ and $20.02{\pm}1.07$, respectively, to mean levels of $5.34{\pm}3.24$, $3.79{\pm}0.57$ and $1.87{\pm}1.09\;mg$/100 g. Whereas, after 48h fermentation, the contents of daidzein, genistein and glycitein increased from a mean initial levels of $8.09{\pm}0.78$, $11.20{\pm}0.84$ and $4.71{\pm}0.46$, respectively, to mean levels of $85.76{\pm}0.84$, $175.87{\pm}2.21$ and $22.41{\pm}0.91\;mg$/100 g. Taken together, these results suggested an increase of aglycones and decrease of glucoside in isoflavones occurred during fermentation, which coincided with an increase of $\beta$-glucosidase activity in the fermented soybean extract powder.

Profiles of Isoflavone and Fatty Acids in Soymilk Fermented with Lactobacilli, Bifidobacteria, or Streptococci

  • Distribution of isoflavones and fatty acids in soymilk fermented with 7 Lactobacilli (L-type), 7 Bifidobacteria (B-type), or 5 Streptococci (S-type) were monitored. Total isoflavones in fermented soymilk ranged from 5.24 to $8.59{\mu}mol/g$ dry basis while those in unfermented soymilk were $8.06{\mu}mol/g$ dry basis. Depending on the types of inoculated microorganisms, fermented soymilk showed different profiles in isoflavones, especially aglycones and $\beta$-glucosides. Four L-type fermented soymilk had significantly higher aglycone content (33.69-46.21%) and low $\beta$-glucosides compared to control (p<0.05). All B-type fermented soymilk showed significantly high aglycone levels (p<0.05). Out of 5 Streptococci, 4 strains produced over 82.2% aglycones. Lipid content ranged from 162 to 224 mg/g and linoleic acid was the highest, followed by oleic, linolenic, palmitic, and stearic acids. Average ratio of unsaturated to saturated fatty acids in control, L-, B-, and S-type fermented soymilks was 6.30, 6.09, 6.30, and 5.94, respectively. This study can help to develop a fermented soymilk containing high isoflavone aglycones and low fat content.

Production of Anthocyanin by Culture of Hairy Roots of Raphanus sativus cv. Chungpihongsim (청피홍심무우(Raphanus sativus cv. Chungpihongsim)의 모상근 배양에 의한 안토시아닌 생성)

  • The hairy root culture of Raphanus sativus cv. Chungpihongsim was established by transformation with Agrobacterium rhizogenes $A_4$. The transformed roots grew well in adjusted Murashige and Skoog medium to 1/2 basal salts, pH 5.2, 3% sucrose. Agropine and mannopine, opine synthesized in the transformed tissue were detected in the extract of hairy roots. When 2, 4-D and kinetin were added in culture medium of hairy roots, the synthesis of anthocyanin was induced with disorganization of hairy root. Especially, addition of $0.45\;\mu\textrm{M}$ 2, 4-D and $2.3\;\mu\textrm{M}$ kinetin showed the maximum synthesis of anthocyanin. Pattern of anthocyanin synthesized in transformed roots was somewhat different from that of ordinary roots. However, aglycone part of all anthocyanin was identified as pelargonidin. The content of total anthocyanin in this sample was tentatively calculated 0.49 mg/g fresh weight.weight.

Changes of Isoflavone Contents in Soybean Cultivars Pickled in Persimmon Vinegar (콩 품종별 감식초 절임 중 이소플라본 함량 변화)

  • Effects of pickling soybeans in acidic solution on soybean isoflavone, and pH and color of immersing solution were investigated. Soybean cultivars, Seoritae and Seomoktae (black beans) and Myeongju-namul (yellow bean), were soaked in persimmon vinegar for 10 days at $20^{\circ}C$. Isoflavone content increased rapidly during pickling, reaching 51.8 (Seomoktae)-106.8% (Myeongju-namul). Increase of aglycone types, from 6.2-9.3% to 20.9-50.8%, was particularly noted, while glycosidic ones were less affected. Acidity of persimmon vinegar increased from 3.4 to 4.4 up to 4 days of soaking and decreased slowly thereafter. Color 'L' and 'b' values of persimmon vinegar decreased significantly, whereas 'a' value increased in black beans. All color values of yellow beans were less affected.

Antioxidant Activity of Solvent Extract from Onion Skin (양파껍질에서 분리된 용매 추출물의 항산화효과)

  • The antioxidant activity of the solvent fractions extracted from onion skin was examined. The antioxidant activity of methanol extract at the cencentration of 0.02% and 0.03% was stronger than that of mixed tocopherol. The antioxidant activity of the fractions of methanol extract increased in the order of butanol> ethyl acetate> ethyl ether> water fraction. The antioxidant activity of each fractions was strongly related with total phenol content and HDA. Further separation of butanol fraction by TLC yielded 6 fluorescent bands with Rf values of 0.20, 0.33, 0.49, 0.60 and 0.94. The total phenol content and HDA of fluorescent band, Rf 0.96, were remarkable higher than those of the other band and exhibited a strong UV absorption at 255 nm and 317 nm, which would be specifically produced by flavonol. Spectral analyses indicated that the major antioxidant component was quercetin aglycone (3,3',4',5,7-pentahydroxyflavone).

Changes in Isoflavone and Oligosaccharides of Soybeans during Germination (콩의 발아 중 이소플라본과 올리고당의 변화)

  • Three Korean soybean varieties, Dawon, Taekwang, and Myeongiju-namul were investigated for changes in isoflavone and oligosaccharides contents, and dry weight during germination. Soybeans were soaked for 10 hr in water, followed by 8 days germination at $20^{\circ}C$ under dark condition, Highest isoflavone content measured was Myeongju-namul (1.228 mg/g), followed by Taekwang (0.671 mg/g) and Dawon (0.661 mg/g). Total isoflavone content generally increased during initial germination and decreased thereafter. Maximal increase in isoflavone was 20-50%, particularly in aglycone type such as daidzein and genistein. Raffinose and stachyose contents decreased rapidly during germination, while that of sucrose showed relatively slow decrease. Dry weights of soybeans steadly decreased.

Biotransformation of flavonoid-7-O-glucuronides by $\beta$-glucuronidases

  • $\beta$-Glucuronidases (E.C. 3.2.1.31) from Escherichia coli, Helix pomatia, and bovine liver activity have been investigated on 7-O-glucuronides (baicalin, wogonoside, and luteolin-7-O-glucuronide) and 3-O-glucuronides (quercetin-3-O-glucuronide and kaempferol-3-O-glucuronide). Bovine liver enzyme was not active on any of these substrates. E. coli and H. pomatia enzymes were active on 7-O-glucuronides, however, 3-O-glucuronides were resistant to $\beta$-glucuronidase hydrolysis. These results suggest that glucuronic acid at 7-position is more susceptible to E. coli and H. pomatia $\beta$-glucuronidases than that at 3-position. In addition, the subtle difference of aglycone structure on 7-O-glucuronides affected the preference of enzyme. E. coli enzyme was favorable for the hydrolysis of baicalin, however, H. pomatia enzyme was found to be efficient for the hydrolysis of wogonoside. Both enzymes showed the similar hydrolytic activity towards luteolin-7-O-glucuronide. When the Scutellaria baicalensis crude extract was subjected to enzymatic hydrolysis, baicalin and wogonoside were successfully converted to their aglycone counterparts with H. pomatia at 50 mM sodium bicarbonate buffer pH 4.0. Accordingly, the enzymatic transformation of glycosides may be quite useful in preparing aglycones under mild conditions.

Isoflavone Content in Korean Fermented and Unfermented Soybean Foods (대두 가공 식품 중의 이소플라본 함량)

  • Soybean isoflavones known as an anticarcinogenic factor were evaluated in some Korean soybean foods by high performance liquid chromatography. Unfermented soybean foods contained predominantly isoflavone ${\beta}-glycosides$ (genistin and daidzin), but in the fermented soybean foods isoflavones were present mainly as aglycones (genistein and daidzein). Average contents of isoflavones of tofu, soymilk and soy sprout were 1, 151, 676 and 424 mg/kg, respectively. Among the fermented soybean products, soybean paste (Chongkukjang) was highest in the isoflavone content (920 mg/kg) and the degree of hydrolysis of ${\beta}-glycosidic$ bonds was ranged from 30 to 100%. Doenjang, Chunjang and Kanjang contained lesser amounts of isoflavones and their contents were 627, 291 and 10 mg/kg, respectively. Korean traditional fermented foods, Chongkukjang and Doenjang could be regarded as the excellent sources of soy isoflavone aglycones.

Component Analysis of Persicaria hydropiper L. Extracts (여뀌 추출물의 성분 분석)

  • In the previous study, we reported the antioxidative activity, antiaging activity, antibacterial activity and moisturizing effect of cream containing Persicaria hydropiper L. extract. In this study, the components of Persicaria hydropiper L. extract were analyzed by TLC and HPLC. Aglycone fractions obtained from the deglycosylation reaction of ethyl acetate fraction among the Persicaria hydropiper L. extract, showed 2 bands and 2 peaks in TLC and HPLC experiments, respectively. Two components were identified as quercetin and kaempferol. TLC chromatogram of ethyl acetate fraction of Persicaria hydropiper L. extract revealed 6 bands and HPLC chromatogram showed 7 peaks, which were identified as quercetin, hyperin, isoquercitrin, quercitrin, kaempferol. In conclusion, with the antioxidative activity, antiaging activity, antibacterial activity and moisturizing effect reported previously, component analysis of Persicaria hydropiper L. extracts could be applicable to new cosmeceuticals.

Physiological activity and efficacy of cosmetic products in bio-converted soybean embryo extract (생물전환 콩 배아추출물의 생리활성 및 화장품의 효능 연구)

  • This study was to investigate the physiological activities of bioconversion soybean embryo extract and the effects of sleeping pack containing extract on skin. The aglycone isoflavones of the soybean embryo extract bioconversed with pectinase were identified by HPLC. in vitro tests showed that they were non-toxic, anti-inflammatory, anti-melanin and anti-aging activity. After 4 weeks of human body application with the sleeping pack using extract, the skin surface roughness and eye wrinkles were so improved by low stimulation that the sleeping pack containing bioconversion soybean embryo extract has a positive effect on the skin. We confirmed that the bioconversion soybean embryo extract was effective in enhancing the physiological activity and was useful as a cosmetic material of various formulations.
