RBC catalase activities of all experimental group were not significantly different. Total lipid and triglyceride concentration in plasma of all experimental groups were not significantly different. Total cholesterol concentrations in plasma of OJY group were significantly lower than those of control group(F=4.387, p=0.032, ANOVA test). Conclusions : According to the above results, it is considered that Ojayeonjonghwan is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.
The present study was performed to determine the effect of cold stress on myocardium of aging rat. Control groups, which aged 6, 12 and 24 months, were compared with age-matched experimental groups that were exposed to moderate cold stress for a hours daily in a week at laboratory cold room $(4{\pm}1^{\circ}C)$. The histological, histochemical and ultrastructural changes of myocardium were observed. The results were summarized as follow: 1. Age-dependent histological change of control groups was observed the formation of contraction band in 24months aged group. The experimental groups submitted to cold stress showed a similar change pattern as seen in control groups. However, the degree of change in the experimental groups was significantly larger than that of control groups. In the 34 months aged group the formation of hypercontraction band was observed. 2. Regarding age-dependent histochemical changes of control groups, we observed the increase activities of PAS and Masson's trichrome. In experimental groups the activities of PAS and Masson's trichrome were also increased with age. Compare with control group, the activities of PAS was increased but the activities of Masson's trichrome was decreased. 3. Age-dependent ultrastructural changes on vacuolization, lysosome were observed. In control groups the structural changes occur at 12 months. The accumulation of lipofuscin, contraction band, hypercontraction band and a component of connective tissue were observed in 24 months. However, the degree of change in the experimental groups was significantly larger than that of control groups. In contract, the myelin body in intercalated discs was observed in 24 months of experimental groups.
Tissue factor (TF), a principal initiator of the veertebrate coagulation cascade, is expressed in organ tissues, cells and blood. TF is konwn to be induced in endothelial cells, monocytes and macrophages by inflammatory stimuli and in many pathologic conditions. By using the modified method for in vido TF activity assay, we found that turpentine oil injection as an inflamatory stimulus also induced the TF activity in lung and brain tissues of rats. And the age-related increase in Tf activity was observed in healthy rat brain tissue.

Peroxynitrite and 4-hydroxynonenal (4-HNE) are highly reactive molecules which are generated under oxidative stress condition and during aging. Many proteins in living organism are modified by them and consequently associated with various diseases including cardiovascular and neurodegenerative diseases. We hypothesize that peroxynitrite and 4-HNE modified serum proteins are also associated with aging process. To establish information on peroxynitrite and 4-HNE adducted proteins for aging study, we used proteins methods, 2D-PAGE and MALD-TOF MS, to identify modified proteins from young (70month) and old (25-month) rat serum. Computerized image processing and analysis system was used for quantitative assessment of skin tissue components in color histological sections. The purpose of this study was to determine that the changes in the collagen fiber density and elastic fiber density in dermis in the rat skin as aging. And also to determine that the correlation between fibroblast density and collagen fiber density and elastic fiber density in the aged rat skin. Ten weeks old ($130{\sim}150g$) eight and fifty-four weeks old ($300{\sim}350g$) eight female Sprague-Dawley rats were used. The full-thickness skin biopsy specimens were prepared serial sections and stained with hematoxylin and eosin, Masson's trichrome and Verhoeff-van Gieson. The collagen fiber and the elastic fiber were identified using the image analysis processing system and then calculated the collagen fiber density rate and the elastic fiber density rate in the dermis. It also identified fibroblast and calculated fibroblast density in the dermis. By using a Student's t-test, a decrease in the collagen fiber density rate (t=-4.650, P<0.001) and the elastic fiber density rate (t=-6.494, P<0.001) of dermis can be observed in aged rats as compared with the young rats. A Student's t-test showed a significantly less fibroblast density in the aged rats than the young rats (t=-4.497, P<0.001). There were significantly positive correlation between the fibroblast density and the collagen fiber density rate (r=.69, P<0.001) and the elastic fiber density rate (r=.91, P<0.001). These results indicate that the aging may decrease the collagen fiber density and elastic fiber density due to reduced the proliferative and synthetic activity of fibroblast in the dermis.
The aging process induces a plethora of changes in the body including alterations in hormonal regulation and metabolism in various organs including the heart. Aging is associated with marked increase in the vulnerability of the heart to ischemia-reperfusion injury. Furthermore, it significantly hampers the development of adaptive response to various forms of conditioning stimuli (pre/post/remote conditioning). Aging significantly impairs the activation of signaling pathways that mediate preconditioning-induced cardioprotection. It possibly impairs the uptake and release of adenosine, decreases the number of adenosine transporter sites and down-regulates the transcription of adenosine receptors in the myocardium to attenuate adenosine-mediated cardioprotection. Furthermore, aging decreases the expression of peroxisome proliferator-activated receptor gamma co-activator 1-alpha ($PGC-1{\alpha}$) and subsequent transcription of catalase enzyme which subsequently increases the oxidative stress and decreases the responsiveness to preconditioning stimuli in the senescent diabetic hearts. In addition, in the aged rat hearts, the conditioning stimulus fails to phosphorylate Akt kinase that is required for mediating cardioprotective signaling in the heart. Moreover, aging increases the concentration of $Na^+$ and $K^+$, connexin expression and caveolin abundance in the myocardium and increases the susceptibility to ischemia-reperfusion injury. In addition, aging also reduces the responsiveness to conditioning stimuli possibly due to reduced kinase signaling and reduced STAT-3 phosphorylation. However, aging is associated with an increase in MKP-1 phosphorylation, which dephosphorylates (deactivates) mitogen activated protein kinase that is involved in cardioprotective signaling. The present review describes aging as one of the major confounding factors in attenuating remote ischemic preconditioning-induced cardioprotection along with the possible mechanisms.
Morphological difference of the renal glomerulus at different age groups have been studied in young (three month-old), adult (twelve month-old) and old (thirty month-old) Fisher strain 344 rats. Pieces of the tissues were taken from renal corticies prefixed with 2.5% glutaraldehyde-1.5% paraformaldehyde (0.1 M Millonig's phosphate buffer, pH 7.3), following by post-fixation with 1% osmium tetroxide (0.1 M Millonig's phosphate buffer, pH 7.3) and embedded within Araldite. The ultrathin sections contrasted with uranyl acetate and lead citrate were observed under a JEM 100CX electron microscope. The mean thickness of glomerular basal lamina and Bowman's capsule were determined by measuring the thinnest portion of basal lamina, and by taking the average of 50 readings from electron micrographs at different ages. The numerical changes of the slit pores were compared based upon the numbers over the length of 10um of glomerular basal lamina. The results were as follow: 1. The thickness of glomerular basal lamina is increased during aging; 140.4 nm in young rats, 270.0 nm in adult ones, and 437.8 nm in old ones. 2. The thickness of basal lamina of parietal cells of Bowman's capsule is 187.5 nm in young rats, 914.0 nm in adult ones, and 2850.0 nm in old ones. 3. The numbers of the slit pores of basal lamina are reduced during aging, 30.3 slit pores/$10{\mu}m$ in adult ones, and 24.2 slit pores/$10{\mu}m$ in old ones. 4. Accumulation of dense intracytoplasmic filamentous material in the parietal cells of Bowman's capsule is increased in the vicinity of the basal lamina during aging. The proximal tubule-like epithelial cell in Bowman's capsule is observed at one glomerulus in a young rat. 5. The endothelial cells are edematous and form balloon-like structure protruding into capillary lumen in young and old rats. 6. Cytoplasm of the podocyte shows a variety of alteration during aging, such as swelling of mitochondria and of endoplasmic reticulum, and increase of microtubules, microfilaments, lysosomes and lamellated myelin structures, etc. Accumulation of dense intracytoplasmic material in the foot processes is increased in the vicinity of the basal lamina during aging. The podocytic membrane-like structures are seen in young and o]d rats. 7. The mesangial matrices and mesangial cells are increased during aging, and slight swelling of endoplasmic reticulum and Golgi cisternae in young and old rats.
Aging causes thymus involution, and genes in thymus play an important role in the development of the immune system. In this study, we compared genes expressed in thymus of neonatal and peripubertal rats using annealing control primers (ACPs)-based GeneFishing polymerase chain reaction (PCR) and semiquantitative reverse transcription (RT)-PCR. We identified 10 differentially expressed genes (DEGs) with 20 ACPs. Of 10 DEGs, bystin-like, collagen type V alpha 1 (COL5A1), and T-cell receptor beta-chain segment 2 (TCRB2) that are related to immune-function were detected in rat thymus. Bystin-like and TCRB2 were up-regulated, while COL5A1 was down-regulated in peripubertal thymus. Semiquantitative RT-PCR confirmed postnatal changes in expression of bystin-like, COL5A1, and TCRB2. These results suggest that bystin-like, COL5A1, and TCRB2 could regulate immune function controlled in thymus as age increases.
