Ovarian cell aggregate culture in teleost, marine medaka (Oryzias dancena): basic culture conditions and characterization

  • Background: Although an understanding of the proliferation and differentiation of fish female germline stem cells (GSCs) is very important, an appropriate threedimensional (3D) research model to study them is not well established. As a part of the development of stable 3D culture system for fish female GSCs, we conducted this study to establish a 3D aggregate culture system of ovarian cells in marine medaka, Oryzias dancena. Methods: Ovarian cells were separated by Percoll density gradient centrifugation and two different cell populations were cultured in suspension to form ovarian cell aggregates to find suitable cell populations for its formation. Ovarian cell aggregates formed from different cell populations were evaluated by histology and gene expression analyses. To evaluate the media supplements, ovarian cell aggregate culture was performed under different media conditions, and the morphology, viability, size, gene expression, histology, and E2 secretion of ovarian cell aggregates were analyzed. Results: Ovarian cell aggregates were able to be formed well under specific culture conditions that used ultra-low attachment 96 well plate, complete mESM2, and the cell populations from top to 50% layers after separation of ovarian cells. Moreover, they were able to maintain minimal ovarian function such as germ cell maintenance and E2 synthesis for a short period. Conclusions: We established basic conditions for the culture of O. dancena ovarian cell aggregates. Additional efforts will be required to further optimize the culture conditions so that the ovarian cell aggregates can retain the improved ovarian functions for a longer period of time.

  • FC(Food Culture) 사업은 음식종류의 다양성과 재료의 신선도 유지를 위하여 개별적인 유통 업무를 수행하고 있다. 재료의 신선도 유지를 위한 신속성, 음식의 매출액과 재료비, 인건비, 경비 등의 집계에 대한 정확성이 부족하여 결산에 문제점이 있다. 본 논문에서는 FC사업의 유통에 대한 ERP를 구축하고 매출과 원가의 요소를 집계하여 사업장 단위의 매출액에 대비한 재료비, 인건비, 경비 등 요소를 회계 집계하고, 또한 유통 물류 구매를 위한 발주와 낙찰된 물류품의 계약과 결과 값으로 예측 Simulation을 연구한다. 사업장 단위에서 마감되면 시도단위와 전국단위의 전체 자산 및 손익계산을 가능하게 하는 ERP를 연구한다. 본 논문 연구를 통하여 FC사업 ERP 발전과 물류 유통 산업에 기여하게 될 것이다.

Particle Size, Morphology and Color Characteristics of C.I. Pigment Red 57:1 : 1. Effect of Synthesis Conditions

  • The effects of synthesis conditions on characteristics of the calcium-azo complex pigment, C.I. Pigment Red 57:1, were studied. It was mainly considered that the industrially required synthesis conditions for lowering electrical conductivity of the pigment solution keeping pigment quality such as particle size and color characteristics. Three parameters were chosen as control factors during the synthesis. The first was the amount of hydrochloric acid added to transform sodium nitrite into nitrous acid. The second was the amount of calcium chloride added to insolubilize the synthesized azo dye. The final factor was pH control during the coupling reaction. The electrical conductivity and pigment aggregate particle size were dependent on the amount of hydrochloric acid and calcium chloride. Higher HCl concentration gave brighter yellowish-red color because of smaller particle aggregate size and narrower size distribution. Amount of charged ions in the synthesis process might affect the "lake" formation resulting different particle aggregate size and color shade.

Daucus carota 현탁 배양에서 배지 조성 및 세포 응집 크기가 Carotenoid 생합성에 미치는 영향 (Effects of Nutrients and Cell Aggregate Size on the Biosynthesis of Carotenoid in Daucus carota Suspension Culture)

  • The effects of nutrients and cell aggregate size on the cell growth of D. carota and the biosynthesi of carotenoid were investigated. Highest carotenoid content was obtained with sucrose as a carbon source and the equal ratio of ammonium to nitrate. High phosphate concentration stimulated the carotenoid biosynthesis in D. corota. 2,4-D inhibited the cell growth but stimulated the specific carotenoid content at high concentration. By modifying the medium composition based on these findings, three times higher specific carotenoid content and 2.5 times higher total carotenoid content were obtained as compared with the results obtained with basic MS media. Biosynthesis of carotenoid was found to be affected by cell aggregate size; high carotenoid production was obtained from the large aggregated cells resulted from high sucrose concentration.

Factors influencing efficiency of somatic embryogenesis of Gentiana kurroo (Royle) cell suspension

  • In this paper, we would like to show unexpected morphogenic potential of cell suspensions derived from seedling explants of Gentiana kurroo (Royle). Suspension cultures were established with the use of embryogenic callus derived from seedling explants (root, hypocotyl and cotyledons). Proembryogenic mass proliferated in liquid MS medium supplemented with $0.5mg\;l^{-1}$ 2,4-D and $1.0mg\;l^{-1}$ Kin. The highest growth coefficient was achieved for root derived cell suspensions. The microscopic analysis showed differences in aggregate structure depending on their size. To assess the embryogenic capability of the particular culture, 100 mg of cell aggregates was implanted on MS agar medium supplemented with Kin ($0.0-2.0mg\;l^{-1}$), $GA_3$ ($0.0-2.0mg\;l^{-1}$) and AS ($80.0mg\;l^{-1}$). The highest number of somatic embryos was obtained for cotyledon-derived cell suspension on $GA_3$-free medium, but the best morphological quality of embryos was observed in the presence of $0.5-1.0mg\;l^{-1}$ Kin, $0.5mg\;l^{-1}$ $GA_3$ and $80.0mg\;l^{-1}$ AS. The morphogenic competence of cultures also depended on the size of the aggregate fraction and was lower when size of aggregates decreased. Flow cytometry analysis reveled luck of uniformity of regenerants derived from hypocotyl suspension and 100% of uniformity for cotyledon suspension.

배지종류가 추식 수경재배 질경이의 생장과 무기성분 함량에 미치는 영향 (Effects of the Different Substrates on the Plant Growth and Mineral Contents of Hydroponically Grown Plantago asiatica in Aggregate Culture)

  • 질경이를 공시하여 펄라이트, 펄라이트와 피트모스 1:1 혼합, 입상암면 및 피트모스 등 4종의 고형배지경에서 배지에 따른 생장반응과 무기성분 함량을 비교 분석하였다. 배지종류별 생육반응은 초장, 경경, 엽수, 최대근장, 지상부와 지하부의 생체중 및 건물중 등 전반적인 초기생장이 펄라이트와 피트모스 혼합배지>피트모스 배지>입상암면 배지>펄라이트 배지 처리구 등의 순인 것으로 나타났다. 식물체내 Ca와 Mg함량은 피트모스 배지에서, Mg와 Na 함량은 입상암면 배지에서 가장 높은 것으로 나타났다. 식물체내 $P_2O_5$ 함량은 식물 생장과 비례적으로 증가하여 펄라이트와 피트모스 1:1 혼합 배지 처리구와 피트모스 배지 처리구에서 높게 나타났다.

  • 치아 치수 세포는 치아 손상에 따르는 병리적인 상황에서 골과 상아질 기질을 형성하는 능력을 가진 것으로 생각된다. 본 연구에서는 MTA가 사람 치수세포의 성장에 미치는 영향과 상아질 형성에 관여하는 유전자의 발현을 유도하는지를 알아보고자 하였다. 또한 상아질 형성의 잠재적 지표인 alkaline phosphatase(ALP) activity에 미치는 영향을 평가하였다. 유전자 발현 검사를 위해 glyceraldehyde-3-phosphate dehydrogenase, type I collagen, alkaline phosphatase, osteonectin(SPARC), and dentin sialoprotein primer set을 이용하여 MTA 처리 2일과 4일 후 reverse transcriptase polymerase chain reaction(RT-PCR)을 시행하였다. cell viability assay(세포 생존력 측정) 에서 5일간 MTA에 노출된 치수 세포의 비율이 대조군보다 높았다. 대조군에 비해 MTA를 처리한 군에서 ALP와 SPARC가 증가되었다. 이상의 결과를 종합하여 보면, 이 연구에 사용한 dental pulp culture system은 MTA를 포함한 치과재료의 처리 후 치수세포의 성장과 분화 그리고 상아질 형성 유도 기전을 연구하는 데 유용한 모델로 사용할 수 있다. MTA 처리는 사람 치수세포에 세포독성을 유도하지 않으며, ALP 활성도와 유전자 발현 그리고 osteonectin (SPARC) 유전자 발현을 증가시켜 수복상아질을 형성할 것으로 사료된다.

Looking at Organizational Socialization from the Developmental Network Perspective

  • Paying close attention to those new to an organization, whether fresh or experienced, whose primary interest is in (re)socialization, the current study intends to (1) further the concept of mentoring from a bilateral relationship to a community and culture fostered by developmental networks, (2) propose an integrated conceptual framework for organizational socialization, and (3) suggest implications for practice and future research. This study reviews, analyzes, and integrates research assets and subsequently re-conceptualizes the aggregate information as valid propositions and a conceptual framework. The findings include (1) 11 propositions regarding the relationships among network characteristics (embeddedness, diversity), developmental functions (career support, psychosocial support, and role modeling), and socialization outcomes (learning and attitudinal outcomes); and (2) an integrated conceptual framework that depicts a comprehensive mechanism through which developmental networks conduce to organizational socialization of newcomers. Implications are that developmental networking must be an individual's fundamental competency and an essential part of organizational onboarding processes, and imperative for both members' career development and innovative organizational culture. By integrating research assets on the developmental phenomenon into conceptualizations, this study furthers the concept of mentoring to organizational culture and stimulates a substantive discourse for theory-building towards organizational socialization from the developmental network perspective.

Suspension culture of anchorage-dependent cells in serum-free medium with biodegradable polymer nanospheres

  • 본 연구에서는 생분해성 고분자 나노입자를 이용하여 부착성 세포의 세포군집 (cell aggregates) 형성을 촉진시켜 무혈청 배지에서 3차원적 부유 배양하는 방법을 개발했다. 생분해성 고분자 나노입자의 사용은 무혈청 배지 부유배양에서 부착성 동물세포인 HEK 293 세포의 세포군집 형성과 세포증식(나노입자를 사용하지 않은 대조군과 비교하여 2배 이상)을 촉진하였다. 일반적으로 무혈청배지 부유배양에 세포를 적응(adaptation)시키는 데에는 시간이 오래 걸리고 많은 비용이 드는데, 이 연구에서 개발된 방법은 이러한 세포적응 공정이 필요없다. 이 배양법은 여러 부착성 동물세포의 산업적 대량배양에 유용하게 사용될 수 있을 것이다.
