Bioconversion Strategy in Conversion of Lignocellulosic Biomass upon Various Pretreatment Methods using Sulfuric Acid and Aqueous Ammonia (황산과 암모니아를 이용한 목질계 바이오매스의 전처리 공정에 따른 당화 및 발효공정 전략)

  • This is to study the effects of various pretreatment methods of agricultural residue, corn stover, and to compare the feature and pros and cons of each method including dilute sulfuric acid (DSA), soaking in aqueous ammonia (SAA), and ammonia recycle percolation (ARP). In order to convert corn stover to ethanol, various pretreatments followed by simultaneous saccharification and co-fermentation (SSCF) were tested and evaluated in terms of ethanol yield. With 3%, w/w of glucan loading using ARP-, DSA-, and SAA-treated solids, SSCFs using recombinant E. coli strain (ATCC$^{(R)}$ 55124) with commercial enzymes (15 FPU of Spezyme CP/g-glucan and 30 CBU/g-glucan enzyme loading) were tested. In the SSCF tests, 87, 90, and 78% of theoretical maximum ethanol yield were observed using ARP-, DSA-, and SAA-treated solids, respectively, which were 69, 58, and 74% on the basis of total carbohydrates (glucan + xylan) in the untreated corn stover. Ethanol yield of SAA-treated solid was higher than those of ARP- and DSA-treated solids. In addition, SSCF test using treated solids plus pretreated hydrolysate indicated that the DSA-treated hydrolysate showed the strongest inhibition effect on the KO11 strain, whereas the ARP-treated hydrolysate was found to have the second strongest inhibition effect. Bioconversion scheme using SAA pretreatment and SSCF can make the downstream process simple, which is suggested to produce ethanol economically because utilization of hemicellulose in the hydrolysate is not necessary.

  • We present a structure and design method of the D8PSK modem compatible with the VDL mode-2 standard and performance test results of the developed modem. In VDL mode-2, the raised cosine filter is used only in the transmitter and a general low pass filter is used in the receiver. Consequently, we can not achieve ISI reduction but can have better spectrum characteristics. Although there is 1~2 dB performance degradation with an un-matched filter compared to that with a matched filter, it is more important to minimize adjacent channel interference in narrow band communications. The transmit signal is generated digitally to avoid the problems(I/Q imbalance and DC offset etc.) of analog modulators. In addition the digital down converter using digital IF sampling technique is adopted for the receiver. This paper contains the overall configuration, design method and simulation results based in part on the previously proposed structures and algorithms. It is confirmed that the modem transmits and receives messages successfully at a speed of max. 870 km/h over ranges of up to 310 km through the ground and in-flight communication tests.

Effect of Frutus gardeniae herbal acupuncture on the rat model of ankle sprain pain (치자(梔子) 약침(藥鍼)이 백서(白鼠) 모델 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響))

  • Objective : Frutus gardeniae, seed of Gardenia jasminoides Ellis is one of the crude drugs used for the treatment of inflammatory condition in oriental medicine. Methodes : The present study aimed to examine the analgesic effect and anti-inflammatory effect of Frutus gardeniae extract (FGE) on a rat model of ankle sprain pain, and the relations between FGE-induced effect and endogenous nitric oxide (NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. As a chronic pain model, ankle sprain pain model was used to test the effect of FCE injection applied to acupuncture point. After the induction of ankle sprain, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee. FGE dissolved in normal saline was injected several acupoints. Results : After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 8 hours. FGE produced significant improvement of stepping force of the hindlimb affected by the ankle sprain lasting at least 4 hours. FGE produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. In addition, FGE injection showed inhibitory effect on the paw edema induced by ankle sprain. Both NO production and iNOS, COX-2 protein expression increased by ankle sprain were suppressed by FGE. FGE on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the ankle sprain than either FGE or EA did. The present study suggest that FGE produces a potent analgesic effect on the ankle sprain pain model of the rat and that FGE-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

In vitro response of rat microglia and human polymorphonuclear cells (PMN) to immunoactive compounds

  • Although the field of study in immune enhancing compounds is relatively new, natural products from plants represent a rich and promising source of novel molecules with immunomodulating properties, Microglial cells, the main immune effector cells of the brain, usually display a ramified morphology and low expression levels of immunologically relevant antigens such as MHC class I and class II. Since any compound which participates in activation of phagocytic cells contributes to the production of potentially toxic factors, the search for convenient in vitro test-systems and study of mechanisms of action of these agents are of great interest. Human blood polymorphonuclear (PMN) cells and primary microglial cells isolated from Sprague-Dawley rats were used as cellular screening tests for study of phagocytosis-stimulating action of immunomodulating agents. Numbers of phagocytic activity were evaluated by the phagocyte ingestion of yeast cells and NO-synthase activity, nitrite production, and nitroblue tetrazolium test were determined after phagocyte stimulation. It was possible to demonstrate that indexes of phagocytic activity can be used as quantitative indicators for measurement immunomodulating activity. As a positive control, Zymosan A-induced phagocytosis in both PMN cells and primary microglial cells was used. $IFN-{\gamma}$ (0.1 -1 U/ml) stimulated phagocytosis in PMN cells 1.2 times after 2 - 3 h incubation, although at higher concentrations (10 - 100 U/ml) it strongly inhibited phagocytosis. In a similar way, at higher concentrations, $IFN-{\gamma}$ (100 - 500 U/ml) suppressed phagocytosis in zymosan-A stimulated microglial cells. When Polypodium leucotomus, cambricum and vulgare extracts were tested alone, increased levels of phagocytosis were observed in PMN. In addition, microglial cells showed both increased phagocytosis and MHC class-II antigen expressions. Surprisingly, when PMN and microglia were treated with a combination of Polypodium and $IFN-{\gamma}$, phagocytosis was not inhibited. We did not find changes in NO-synthase activity and nitrite production in both microglia and PMN cells activated by different immunomodulating agents. These results indicate that primary microglial cell cultures as well as human PMN cells can provide reproducible quantitative results in screening phagocytic activity of different immunoactive compounds. Furthermore, both inhibitory or activation mechanisms might be studied using these in vitro experimental approaches.

Baseline Sensitivity and Monitoring for the Resistance to Benomyl of Fusarium Species Causing Bakanae Disease in Korea (국내 벼 키다리병균의 Benomyl 약제에 대한 감수성 기준 및 저항성 변화)

  • To examine the changes in resistance to benomyl of Fusarium species causing bakanae disease, Fusarium isolates were collected in Korea, and pathogenicity tests were performed using rice seeds in vitro. Minimum inhibitory concentration (MIC) and effective concentration of 50% ($EC_{50}$) values of isolates were examined using the agar dilution method. High frequency distribution of MIC values to benomyl against isolates collected in 2006~2007 and 2013~2014 years were $1.5625{\sim}3.125{\mu}g/mL$ and more than $25{\mu}g/mL$, respectively. The mean $EC_{50}$ value of isolates to benomyl increased from $1.6397{\mu}g/mL$ in 2006~2007 to $2.4892{\mu}g/mL$ in 2013~2014. Based on MIC and $EC_{50}$ values of isolates, the moderate resistance of benomyl were determined as more than $25{\mu}g/mL$ of MIC and less $2.4{\mu}g/mL$ of $EC_{50}$ value, and resistant isolates to benomyl were determined as more than $2.4{\mu}g/mL$ of $EC_{50}$ value. Compared with the ratio of resistant isolates in 2006~2007, the ratio of resistance isolates in 2013~2014 increased from 12.5% to 36.4%. In addition, multiple resistant isolates to prochloraz as well as benomyl increased to 20.3% in 2013~2014.

DA-7911, $^{188}Rhenium-tin$ Colloid, as a New Therapeutic Agent of Rheumatoid Arthritis

  • Radiation synovectomy is one of the most useful methods for treating patients with refractory synovitis because of its convenience, long-term effects, repeatability and the avoidance of surgery. In this study, we investigated the toxicity, stability and biodistribution of a rhenium-188 ($^{188}$Re)-tin colloid to evaluate its suitability as a synovectomy agent. Twenty four hours after injecting the $^{188}$Re-tin colloids (74 KBq/0.1 mL) into the tail vein of ICR mice, most of the $^{188}$Retin colloidal particles was found in the lungs. In addition, there were no particle size changes at either room temperature or at $37^{\circ}C$ after injecting the $^{188}$Re-tin colloids in human plasma and synovial fluid. In vitro stability tests showed that the $^{188}$Re-tin colloid remained in a colloidal form without a critical size variation over a 2-day period. We investigated the leakage of $^{188}$Retin colloids from the intraarticular injection site with gamma counting in New Zealand white rabbits. The $^{188}$Re-tin colloids (55.5 MBq/0.15 mL) were injected at the cavum articular and the mean retention percentage of the $^{188}$Re-tin colloid was 98.7% for 1 day at the injection site, which suggests that there was neither change in the particle size nor leakage at the injection sites. In the biodistribution study with the SD rats, the liver showed the highest radioactivity (0.0427% ID/organ) except for the injected knees (99.49%). In the SD rats, mild toxicities including the skin or a synovium inflammation were observed as a result of a radioactivity of 15 mCi/kg at the intraarticular injection site. However, there was no systemic toxicity. In the Ovalbumin (OVA)-induced arthritic rabbits, the $^{188}$Re-tin colloid improved the macroscopic, the histological score and reduced the knee joint diameter when compared to the arthritic control. In conclusion, a $^{188}$Re-tin-colloid is considered as a strong candidate for radiation synovectomy with a superior efficacy and safety.

  • The algicidal effects of marine bacteria were investigated and a strain, which had the strongest algicidal activity against the harmful dinoflagellate, Cochiodinim polykrikoides was selected. The bacterium was isolated in seawater during the period of blooming of C. polykrikoides in Masan Bay. This algicidal bacterium was identified as Micrococcus sp. LG-5 by means of morphological and biochemical tests. The optimal culture conditions of Micrococcus sp, LG-5 were $25^{\circ}C,\;pH 7.0\;and\;3.0{\%}$ NaCl concentration. The algicidal activity of Micrococcus sp. LG-5 was significantly increased to maximum value in the late of logarithmic phase of cell cuture. In addition, the culture filtrate ($pore size,\;0.1{\mu}m$) of Microcoocus sp. LG-5 showed strong algicidal effects. The cell numbers of C. polykikoides were decreased from $1.2{\times}10^4 cells/ml\;to\;less\;than\;2{\times}10^3\;cells/ml$ within 3, 6, 30 hours at the concentrations of culture filtrate $10{\%},\;5{\%}\;and\;1{\%}$, respectively. These results indicated that the algicidal effect was mediated by certain substances released from Microooccus sp. LG-5.

The Structural Relationship among Sense of Coherence, Clinical Practice Stress, and Departmental Satisfaction of Dental Hygiene Students (치위생(학)과 학생의 통합성, 임상실습스트레스, 학과만족도의 구조적 관계)

  • The purpose of this study was to understand the comprehensive relationship among sense of coherence, clinical practice stress, and major satisfaction of students in the department of dental hygiene. From four local universities, departments of dental hygiene were selected: two departments represented a three-year curriculum system and two represented a four-year curriculum system. Data from the final questionnaires of 319 students were analyzed. Differences between variables were analyzed using t-tests and ANOVAs, and correlations were checked using Pearson's correlation coefficients. Path analysis was conducted to analyze the relationship among clinical practice stress, sense of coherence, and major satisfaction. The results indicated that clinical practice stress had a negative influential relationship with both sense of coherence, and major satisfaction. In addition, sense of coherence had a positive influential relationship with major satisfaction. The direct and indirect effects among factors displayed significant differences, indicating that clinical practice stress has a partial mediation effect on sense of coherence and major satisfaction. Therefore, major satisfaction can be increased by developing and implementing programs to improve sense of coherence.

Thin Hardboard Manufacture from Waste Lignocellulosic Papers as Overlay Substitutes in Low Grade Plywood and Particle Board Panels(I) (고지로부터 저급합판 및 파아티클보오드 표면단판으로 사용될 수 있는 박판 하아드보오드의 제조(I))

  • The purpose of this study was to determine the technical feasibility of making 3-dimensional thin hardboard panels for overlay substitutes of low grade particleboard and plywood panels. Experimental studies were directed at assembling bench-top apparatus, learning the characteristics of different types of lignocellulosic waste papers, for making thin hardboard with several combinations of them with and without resin addition. The raw materials used are waste corrugated cartons, cereal boxes, and old magazines which contain substantial amount of lignin in it. The experimental results showed that satisfactory thin(0.21~0.16cm) hardboard could be made from the residential mixed waste papers that have selected properties comparable to commercial 0.32cm hardboard. The significant mixing ratio effect of the waste papers was present on the thickness swelling, water absorption, linear expansion, and modulus of elasticity including Taber abrasion tests of the thin hardboard made. The mixing ratio of waste papers and resin in the thin hardboard prominently affected the specific gravity of it, which led to affect modulus of elasticity and those physical properties sensitively. And it was shown that the hardboard containing those physical properties can be used for overlay substitutes of low grade plywood and particleboard panels.

Evaluation of Combustion gas during Fire Tests of Veneers Coated with Ammonium Salts (암모늄염으로 도포시킨 베니어판의 연소 시에 발생하는 연소가스 평가)

  • This study tested the combustion characteristics of veneer specimens coated with four kinds of ammonium salts. Each veneer specimen was coated three times with 20 wt.% ammonium salt solutions at room temperature. After drying, the combustion characteristics of the specimens coated with chemicals, were investigated using the cone calorimeter (ISO 5660-1, 2). The specimens coated with monoammonium phosphate (MAPP) and, diammonium phosphate (DMPP) showed a 6.7% and, 10.0%, lower mean heat release rate ($HRR_{mean}$), respectively, than that of the uncoated specimen. On the other hand, the specimens coated with MAPP showed a 15.7% higher $CO_{peak}$ production rate and the specimens coated with DAPP showed by 8.2% lower rate than that of the uncoated specimen. The veneer coated with ammonium sulfate (AMSF) and DAPP showed a 9.6% and 33.3% lower the peak smoke production rate ($SPR_{peak}$) than that of the uncoated specimen. In addition, the time to the peak smoke extinction area ($SEA_{peak}$) was delayed by 38.4% in the specimens coated with DAPP than the uncoated specimen. Therefore, DAPP inhibited the combustion properties of the veneer and showed a tendency to reduce smoke production.
