Antimicrobial Resistance and Implicated Genes of E. coli Isolated from Commercial and Cooked Foods in Seoul (서울시내 유통식품에서 분리한 대장균의 항생제 내성 및 내성유전자)

  • Distribution of foodborne E. coli strains, antimicrobial resistant genes and antimicrobial susceptibility have been carried out on E. coli isolated from commercial and cooked foods distributed food in Seoul. Of total 1,313 samples, fifty samples(3.8%) were found E. coli that included one of the ETEC and EPEC, respectively. The serotype of ETEC in seasoning raw meat was E. coli O26 and produced Verotoxin 2. Fifty percentage of total isolates were susceptible to all antimicrobial agents. Specially, there were ampicillin(36%), amoxicillin/clavulanic acid(32%) and tetracycline(22%) etc. Resistant gene (tetB) were found in four tetracycline resistant E. coli strains, and TEM gene was found in one ampicillin resistant E. coli isolate.

  • The effect of humic acid (HA), and the roles of microfiltration (MF), PES (polyethersulfone) beads adsorption, and photo-oxidation were investigated in hybrid process of ceramic MF and PES beads loaded with titanium dioxide ($TiO_2$) photocatalyst for advanced drinking water treatment. Those were compared and studied in viewpoints of membrane fouling resistance ($R_f$), permeate flux (J), and total permeate volume ($V_T$). Because membrane fouling increased dramatically as decreasing HA, $R_f$ increased and J decreased, and finally $V_T$ was the highest at 2 mg/L HA. In the experiment to investigate the roles of photo-oxidation and adsorption at humic acid 4 mg/L and 6 mg/L. In both conditions, $R_f$ was the lowest and $V_T$ was the highest in MF + $TiO_2$ + UV process. The average treatment efficiencies of turbidity and dissolved organic matters were the highest in MF + $TiO_2$ + UV process, too.

  • ErmSF is one of the ERM proteins which transfer the methyl group to A2058 in 23S rRNA to confer the resis­tance to MLS (macrolide-lincosamide-streptogramin B) antibiotics on microorganism. Unlike other ERM pro­teins, ErmSF contains long N-terminal end region (NTER), of which $25\%$ is composed of arginine that is known to interact with RNA well. Gradual deletion of NTER leaded us to the point where mutant protein lost much of activity in vivo. Overexpressed and purified mutant protein showed much reduced activity in vitro: $2\%$ activity relative to that of wild type protein. This fact suggests that this amino acid interact with RNA close to meth­ylatable adenine to locate it at an active site properly.

  • Quantitative measurement of chloride ion concentration has an important role in various fields of electrochemistry, medical science, biology, metallurgy, architecture, etc. Among them, its importance of architecture is ever-growing due to unexpected degradations of building structure. These situations are caused by corrosion of reinforced concrete (RC) structure of buildings. And chloride ions are the most powerful factors of RC structure corrosion. Therefore, precise inspection of chloride ion concentration must be required to increase the accuracy of durability monitoring. Multi-walled Carbon nanotubes (MWCNTs) have high chemical resistivity, large surface area and superior electrical property. Thus, it is suitable for the channels of electrical signals made by the sensor. Silver nanoparticles were added to giving the sensing property. CuBTC, one of the metal organic frameworks (MOFs), was employed as a material to improve the sensing property because of its hydrophilicity and high surface area to volume ratio. In this study, sensing element was synthesized by various chemical reaction procedures. At first, MWCNTs were functionalized with a mixture of sulfuric acid and nitric acid because of enhancement of solubility in solution and surface activation. And functionalized MWCNTs, silver nanoparticles, and CuBTC were synthesized on PTFE membrane, one by one. Electroless deposition process was performed to deposit the silver nanoparticles. CuBTC was produced by room temperature synthesis. Surface morphology and composition analysis were characterized by scanning electron microscope (SEM), energy dispersive X-ray spectroscopy (EDS), respectively. X-ray photoelectron spectroscopy (XPS) was also performed to confirm the existence of sensing materials. The electrical properties of sensor were measured by semiconductor analyzer. The chloride ion sensing characteristics were confirmed with the variation of the resistance at 1 V.

Controlled Release Behavior of Bioactive Molecules from Photo-Reactive Hyaluronic Acid-Alginate Scaffolds

  • There are three important components in tissue engineering: the cells, signaling factors (cytokines and growth factors), and scaffolds. To obtain finely engineered tissue, all three components should perform their individual functions and be fully integrated with each other. For the past few years, we have studied the characteristics of photodimerizable HA (CHA)/alginate (CA) composite materials. CHA/CA complex hydrogels, which were irradiated under UV light and, then treated with calcium ions, were found to have good biocompatibility, mechanical properties and water resistance for implantable tissue scaffolds. In this study, we introduced a cell growth factor (basic fibroblast growth factor; bFGF) into the CHA/CA scaffolds and studied its release behavior. We also introduced tetracycline hydrochloride and flurbiprofen into the same scaffolds as model activation factors and evaluated their release behaviors from the scaffolds. The drug release rate from the materials was influenced by various parameters, such as the degree of crosslinking, the cross linker type, the physico-chemical properties of the drug, and the amount of the drug in the polymer. The results indicated that the negatively charged CHA/CA composite materials showed sustained release behavior and that HA has a particularly strong negative charge, making it attractive toward tetracycline hydrochloride and bFGF, but repulsive toward flurbiprofen.

Effect of Antistripping Agent on the Enhancement of Resistance to Moisture Damage of Asphalt Mixture (아스팔트 혼합물의 내수분손상 향상에 대한 박리방지제의 효과)

  • In this work, effect of antistrip additives to reduce moisture damage of asphalt mixture were studied. Asphalt antistripping agents were prepared by condensation of formaldehyde with tetraethylene pentamine (TEPA), triethylenetetramine (TETA) and bis(hexamethylene)-triamine (BHMT), respectively. And also the metal type antistripping agent was prepatred by neutralization of stearic acid or palmitic acid with metal hydroxide. Mechanical characteristics of the asphalt mixture added antistripping agent were evaluated with Marshall stability, submerging residuals and coating rate. It was found that antistripping agent prepared in this study reduced moisture damages of asphalt mixtures. In particular, asphalt mixtures added BHMT and C/S (Calcium stearate hydroxide) antistripping agent showed highest submerging and coating rate. Because BHMT and C/S type antistripping agent was to improve bonding between asphalt and aggregate owing to increase of amine concentration and role of metal surfactant.

Jet-Fuel-Resistant PVC Sealant Containing a Polyester Plasticizer (폴리에스터 가소제를 사용한 내제트유성 PVC계 실란트)

  • This work is about the development of jet-fuel resistant PVC sealant using a polyester plasticizer. PVC copolymer was compounded with adipic acid glycol(Songcizer P-3000) or DOP plasticizers. Fuel-immersed and non-immersed penetration, solubility, flow, and elongation by tensile adhesion of PVC compounds were measured. Penetration increase by fuel immersion and solubility of PVC compounds with adipic acid glycol polyester plasticizer were smaller than those of PVC compounds with DOP plasticizer. Elongation by tensile adhesion test of PVC compound containing 500 phr of Songcizer P-3000 decreased proportionally to the content of DCDP (dicyclopentadiene) base petroleum resin adhesion promoter. Calcium carbonate($CaCO_3$) filler inhibited the diffusion of fuel in all the PVC compounds and decreased the solubility of PVC compounds containing Songcizer P-3000.

  • Panax ginseng, a medicinal plant, has been used as a blood-nourishing tonic for thousands of years in Asia, including Korea and China. P. ginseng exhibits adaptogen activity that maintains homeostasis by restoring general biological functions and non-specifically enhancing the body's resistance to external stress. Several P. ginseng effects have been reported. Korean Red Ginseng, in particular, has been reported in both basic and clinical studies to possess diverse effects such as enhanced immunity, fatigue relief, memory, blood circulation, and anti-oxidation. Moreover, it also protects against menopausal symptoms, cancer, cardiac diseases, and neurological disorders. The active components found in most Korean Red Ginseng varieties are known to include ginsenosides, polysaccharides, peptides, alkaloids, polyacetylene, and phenolic compounds. In this review, the identity and bioactivity of the non-saponin components of Korean Red Ginseng discovered to date are evaluated and the components are classified into polysaccharide and nitrogen compounds (protein, peptide, amino acid, nucleic acid, and alkaloid), as well as fat-soluble components such as polyacetylene, phenols, essential oils, and phytosterols. The distinct bioactivity of Korean Red Ginseng was found to originate from both saponin and non-saponin components rather than from only one or two specific components. Therefore, it is important to consider saponin and non-saponin elements together.

Testing Investigation of Protective Coatings for Downhole Oil Tube

  • Aiming at the corrosion circumstances and corrosion prevention needs of downhole oil tubes, series protective coatings for downhole oil tubes have been developed in the authors' laboratory, including a baked type coating YG-01 and an air curing type coating YG-03, etc. The performance investigation of the coatings has been done for testing their corrosion resistance, mainly including salt fog test, immersion test in oil-field waste water and various acid solutions, high temperature and high pressure test in alkali solution or $H_2S/CO_2$ environment, as well as some other performances. The investigation results show that oil tube anti-corrosion coatings developed here can endure over 4000 hrs salt fog test, over 1000 hrs immersion in various acid solutions at room temperature and in boiling oil-field waste water. In addition, the coatings can keep intact after experiencing test in alkali solution under 70 MPa pressure at $150^{\circ}C$ for 24 hrs, and in simulative sour gas environment under the total pressure of 32 MPa ($P_{H_{2}S}=3.2MPa$, $P_{CO_{2}}=3.2MPa$) at $90^{\circ}C$ for 168 hrs, which show that the coatings can be used for corrosion prevention in downhole environments with specific high temperature and high pressure, such as sour gas wells. The other testing results show the oil tube protective coatings have excellent comprehensive performance.

Control of Erysiphe pisi Causing Powdery Mildew of Pea (Pisum sativum) by Cashewnut (Anacardium occidentale) Shell Extract

  • The effect of methanolic extract of cashewnut (Anacardium occidentale) shell extract was seen on conidial germination of Erysiphe pisi and powdery mildew development in pea (Pisum sativum). Maximum conidial germination inhibition of E. pisi on glass slides was observed at 300 ppm. Similar effect on floated pea leaves was observed after 48 h at the same concentration. Conidial germination on intact untreated pea leaves was also assessed on II and IV nodal leaves while IV and II nodal leaves were treated with the extract and vice versa. There was tremendous reduction in conidial germination on all the nodal leaves. The disease intensity of pea powdery mildew was significantly reduced by methanolic extract of cashewnut shells. Maximum reduction was observed with 200 ppm where 39% disease intensity was recorded in comparison to 96.53% in the control. The phenolic acid content of pea leaves following treatments with this extract varied and no defmite pattern was observed. Out of several phenolic compounds, namely, gallic, ferulic, chlorogenic, and cinnamic acids, only gallic acid was found to be present consistently in all the treatments with varied amounts.
