동물성 소염진통제 (II) - 반묘 및 먹가래 다당체분획의 소염.진통 및 면역조절작용 - (Anti-inflammatory Agents from Animals(II) - Anti-inflammatory, Analgesic and Immunoregulatory Activities of Mylabris sidae and Epicauta gorhami Polysaccharide Fractions -)

  • Effect of Mylabris sidae(MS) and Epicauta gorhami(EG) polysaccharide fractions on the inflammation and immune responses were studied in vivo. MS and EG contained cantharidin about 0.61 and 0.65%, respectively. It was shown that MS and EG polysaccharide fractions at a oral dose of 100 mg/kg have the significant anti-inflammatory and analgesic activity; They inhibited significantly the carrageenin-induced inflammation and acetic acid-induced writhing syndrome. They accelerated significantly the carbon clearance and the phagocytosis of colloidal carbons by Kupffer cells in liver, but they at a oral dose of 100 mg/kg suppressed significantly the Arthus reaction in the sheep red blood cell(S-RBC)-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer and plaque-forming cells. On the other hand, they at a oral dose of 200 mg/kg accelerated slightly the oxazolone-induced dermatitis in rats and delayed hypersensitivity in the S-RBC-challenzed mice in consistent with the increase of rosette forming cells. As the above results, it exhibited that MS and EG polysaccharide fractions inhibited the humoral immune responses, but they accelerated the function of macrophages and cellular immune responses. EG polysaccharide fraction had more active than MS polysaccharide fraction.

홍삼 산성 다당체의 마크로파지 및 자연살해세포의 활성화에 의한 항암작용 (Anticancer Activities of Red Ginseng Acidic Polysaccharide by Activation of Macrophages and Natural Killer Cells)

  • The composition of monosaccharides of acidic polysaccharide isolated from ethanol-insoluble and water-soluble fractions of red ginseng roots was analysed and its immunological activities were investigated. Red ginseng acidic polysaccharide (RGAP) was composed of glucose (26.1 mole %), arabinose (1.6 mole %), glucuroninc acid (51.8 mol %) and galacturonic acid (5.1 mole %) as determined by gas liquid chromatography. Addition of RGAP increased production of nitric oxide (NO) and tumor necrosis factor (TNF)-$\alpha$ in the rodent macrophage cultures. Peritoneal macrophages from RGAP-treated mice exhibited potent tumoricidal activities toward P815 and WEHI 164 tumor cells. It was also observed that concentrations of NO and TNF-$\alpha$ were high in the culture medium of macrophages from the mice administered with RGAP. Moreover, treatment of RGAP in vivo stimulated tumoricidal activities of natural killer (NK) cells. Treatment with RGAP increased life span of sarcoma 180-bearing mice and decreased tumor weights of B16-tumor-bearing mice. These results suggest that activation of macrophages and NK cells serve to enhance in vivo anticancer activities of RGAP.

Pseudomonas sp. GP32에 의해 생산된 세포 외 다당류의 생산 및 특성 (Production and Characterization of Extracellular Polysaccharide Produced by Pseudomonas sp. GP32)

  • 미생물유래 다당류 생산균주를 분리하기 위해 전국 각지의 토양시료로부터 가장 높은 점성과 다당류 생산성을 나타내는 균주 GP32를 분리하였으며, 분리균주 GP32의 동정을 분리균주의 형태학적, 생리학적 특성을 조사한 결과, Pseudomonas 속 세균으로 확인되었으며 최종적으로 Pseudomonas sp. GP32로 명명하였다. 플라스크 수준에서 Pseudomonas sp. GP32의 다당류 생산을 위한 가장 적합한 탄소원과 질소원은galactose와 (NH4)2SO4를 이용하였을 때 가장 많은 다당류를 생산하는 것으로 확인되었으며, 다당류 생산을 위한 최적의 C/N ratio는 50이었다. 다당류 생산을 위한 최적 pH와 온도는 각각 7.5와 32℃였다. 최적화된 배지를 이용한 fermentor 배양에서 다당류 생산은 배양 70시간에 최고치를 나타내었으며, 이때 다당류 생산량은 15.7 g/l이었다. Pseudomonas sp. GP32로부터 생산된 다당류는 ethanol 침전, cetylpyridimium 침전과 gel permeation chromatography를 통하여 정제하였으며, 정제된 다당류는 Biopol32로 명명하였다. Biopol 32의 분자량은 3×107 datons이었으며, Biopol32가 함유하고 있는 구성당은 galactose : glucose : gulcouronic acid : galactouronic acid 등이 1.85 : 3.24 : 1.00 : 1.42의 몰비로 함유되어있다. Biopol32 용액은 의가소성 성질을 갖는 고분자 화합물로서 Zoogloea ramigera가 생산하는 생물고분자인 zooglan보다 모든 농도에서 높은 점성을 나타내었다. Biopol32의 실제 폐수처리현장에서 응집제로의 사용 가능성을 검토하기 위하여 식품폐수, 섬유폐수와 제지폐수를 대상으로 응집효율을 조사한 결과, 높은 COD 감소율 (58.4~67.3%)과 SS제거율(82.6~91.3%)를 나타내어 실제 산업폐수에서 뛰어난 응집효율을 나타내었다.

치마버섯 균사체 배양물로부터 분리한 다당류의 이화학적 특성 (Characteristics of Polysaccharide Extracted from the Cultured Mycelia of Schizophyllum commune)

  • 자연에서 채집한 치마버섯 자실체로부터 분리한 Schizophyllum commune 균을 배양하여 얻은 균사체 배양물로부터 추출한 다당류의 물리화학적 특성에 대하여 조사하였다. 액체배양 균사체로부터 추출된 다당류는 방법에 따라 $0.19{\sim}1.63%$의 수율을 나타내었다. 이들 다당류의 총 당은 $48.9{\sim}72.0%$이고, 단백질은 $15.3{\sim}32.0%$를 함유하고 있는 것으로 나타났다. 추출한 SC-EP 다당류의 구성 당은 glucose가 69.6%로 주를 이루고, fructose가 26.1%, 소량의 xylose로 구성된 것으로 나타나는 것으로 보아 hetero 다당류인 것으로 추정되었다. 또한, 산성 아미노산인 aspartic acid, glycine 및 glutamic acid 등은 각각 12.5%, 18.1% 및 15.2%, 중성아미노산인 alanine은 9.9%, 당과 단백질의 O${\beta}-glycosidic$ 결합에 관여하고 있는 threonine과 serine의 경우는 2.9% 및 2.7%로 나타났다. IR 분석결과 이들은 ${\alpha}$${\beta}$-glucan이 혼재된 다당류인 것으로 나타났다. 한외여과장치로 제조한 다당류(SC-UP)의 0.5% 용액의 점도는 20 rpm 시 70 cps로 나타났다. 이상의 치마버섯 액체배양 균사체를 열수추출하여 에탄올 침전시켜 얻은 다당류는 당과 단백질이 결합된 단백다당류의 결합 형태를 이루고, 당은 2가지 이상의 당이 혼재하고 있으며, 아미노산은 산성아미노산의 함량이 높은 것으로 나타났다. 다당류의 결합 양식은 ${\alpha}$${\beta}$-glucan이 혼재하고 있으며, 강한 점성을 나타내었다.

꼬시래기 유래 Polysaccharide의 보습유지 및 피부개선효과 (Moisturizing and Skin Improvement Effect of Polysaccharides from Gracilariopsis sp.)

  • Polysaccharides are useful materials for keep skin moisturizing and skin improvement cosmetic materials. We have isolated polysaccharides form Gracilariopsis sp. The polysaccharide derived from Gracilariopsis sp. have various size of 300kDa, 500kDa and over 1,000kDa. The volunteers applied a cream formula with polysaccharides from Graciariopsis sp. for 4 weeks. We found significant improvement of wrinkles and skin texture. We also found an effect of improving skin elasticity. When the moisture reduction rate of the skin was 2.8% lower than the 5.1% of hyaluronic acid. The polysaccharide extract from Gracilariopsis sp. showed stable pH and viscosity. The polysaccharides derived from Gracilariopsis sp. could be used as a cosmetic material.

인진쑥 물추출물의 조다당제에 의한 Melanin 생성억제 효과 (Inhibitory Effects of Crude Polysaccharide of Water Extract of Art on Melanin Biosynthesis)

  • Melanogenesis is a physiological process resulting in the synthesis of melanin pigment. We investigated the inhibitory effect of crude polysaccharide of water extract of Artemisia iwayomgi Kitamura (AI) on melanin biosystheies in B16/F10 melanoma cells. Crude polysaccharide of water extract of Artemisia iwayomgi Kitamura significantly inhibited tyrosinase activity and melanin contents with or without $\alpha$-MSH in vivo. Melanin contents and tyrosinase activity were decreased in a dose-dependent manner,These results show that crude polysaccharide of water extract of Artemisia iwayomgi Kitamura could be developed as skin whitening components of cosmetics.

  • 유산균이 생성하는 다당류에 관한 연구의 일환으로서 발효유 제품의 종균으로 널리 사용되고 있는 Str. themophilus, Lb. bulgaricus, Lb. casei 등을 10% 환원멸균 탈지유에 배양하면서 다당류 생성에 따른, 우유 배양액 점도의 경시적 변화 및 다당류의 최적 생성 조건에 대하여 조사하였다. 1. 시험균 중에서 Str. thermophilus 510이 가장 높은 점도를 나타냈으며 $41^{\circ}C$ 배양 5일째에 5,000 CP로서 다른균에 비해서 1,000 CP 이상 높았다. 2. 인공합성배지를 사용하여 흡광도를 측정한 결과 $41^{\circ}C$에서의 흡광도가 가장 높았으며 다당류 합성을 위한 탄소원으로서는 Sucrose의 이용성이 1~5% 농도에서 가장 높았다. 3. 다당류 합성효소의 금속염에 대한 영향을 살펴본 결과 $Mg^{++}$의 효과가 인정되었다.

키랄 컬럼을 사용한 아미노산 에스테르의 니트로벤조옥사디아졸 유도체의 광학분리 (Enantiomer Separation of α-Amino Acid Esters as Nitrobenzoxadiazole Derivatives Using Chiral Columns)

  • A new convenient derivatization method of ${\alpha}$-amino acid esters as nitrobenzoxadiazole (NBD) derivatives for chiral resolution was introduced and the enantiomer separation of ${\alpha}$-amino acid esters as NBD derivatives was performed by normal HPLC using chiral columns based on polysaccharide derivatives. The NBD derivatives were readily prepared by stirring NBD-Cl and ${\alpha}$-amino acid methyl ester HCl with $NaHCO_3$ in ethanol. The performance of Chiralpak IA was superior to the other chiral stationary phases for enantiomer resolution of NBD derivatives of several ${\alpha}$-amino acid methyl esters. Owing to fluorescence detection as well as strong UV absorption, it is expected that the convenient analytical method developed in this study will be very useful for enantiomer separation of ${\alpha}$-amino acid esters as NBD derivatives on polysaccharide-derived chiral columns.

  • The chromatographic enantiomer separation of 9-anthraldimine derivatives of ${\alpha}$-amino acid methyl and ethyl esters on four polysaccharide based chiral columns was performed. The 9-anthraldehyde Schiff base derivatives of ${\alpha}$- amino acid esters were readily synthesized by stirring the ${\alpha}$- amino acid ester hydrochloride salts with 9-anthraldehyde in the presence of 1,8-diazabicyclo [5.4.0]undec-7-ene as a base and anhydrous $MgSO_4$. Chiralcel OD or Chiralcel OD-H showed the greatest enantiomer resolution of 9-anthraldimine derivatives of ${\alpha}$-amino acid methyl and ethyl esters. The L-enantiomers of all the analytes were preferentially retained on Chiralcel OD or Chiralcel OD-H. This analytical method was applied in the determination of optical purities of several commercially available D- or L-${\alpha}$-amino acid methyl esters.

  • This study was conducted to further purify and characterize a polysaccharide fraction from whole parts of Duchesnea indica (Andr.) Focke, which was previously reported to show an antitumor activity by us. Mild acid-treatment of the fraction by 5% acetic acid yielded three subtractions (Fr. A,B and C) on a Sephadex G-25. Fr. A was composed of uronic acid, hexose, pentose and phenolic substance, Fr. C mainly phenolic substance, and Fr. B contained hexose. All three fractions exhibited colony stimulating factor activity.
