버섯 단백다당체의 당뇨 유발 흰쥐의 혈당수준과 에너지원 조성에 미치는 영향 (Effects of Mushroom Protein -bound Polysaccharides on Blood Glucose Levels and Energy Metabolism in Streptozotocin-Induced Diabetic Rats)

  • The hypoglycemic effects of 2 mushrooms, Pleurotus ostreatus and Lentinus edodes, on streptozotocin(STZ) induced diabetic rats were investigated in this study . Diabets mellitus was induced in male Sprague-Dawley rats by the injection of STZinto the tail vein at a dose of 45mg/kg. Sprague-Dawley male rats(200-250g) were assigned to one control and three STZ-diabetic groups. Diabetic groups were assigned to STZ-control, pleurotus ostreatus and Lenitinus edodes groups. All groups were fed a AIN 76 diet. The two experimental groups were fed with each protein-bound polysaccharide(150mg/kg BW) for 14 days and with carboxymethyl cellulose for STZ-control group. The body weight gain was monitored and the blood levels of glucose and cholesterol were measured . Levels of protein triglyceride, and free fatty acid in plasma were analysed. Serum aminotransferase activity as also measured. The body weight gain was lower in the all diabetic groups than in the of normal group. The weight of spleen was reduced by adminstration of the Lentinus edodes protein-bound polysaccharides. The result suggest that orally administered Lentinus edodes protein-bound polysaccharides exhibited hypoglycemic effect in STZ -induced diabetic rats and that these protein-bound polysaccharides may be useful for the management of diabetes mellitus.

  • The extracellular matrix is a critical component of every human tissue. ECM not only functions as a structural component but also regulates a variety of cellular processes such as cell migration, differentiation, proliferation, and cell death. In addition, current studies suggest that ECM is critical for the pathophysiology of various human diseases. ECM is composed of diverse components including several proteins and polysaccharide chains such as chondroitin sulfate, heparan sulfate, and hyaluronic acid. Each component of ECM exerts its own functions in cellular and pathophysiological processes. One of the interesting recent findings is that ECM is involved in inflammatory responses in various human tissues. In this review, we summarized the known functions of ECM in neuroinflammation after acute injury and chronic inflammatory diseases of the central nerve systems.

Proteomic profiles and ultrastructure of regenerating protoplast of Bryopsis plumosa (Chlorophyta)

  • When a multinucleate cell of Bryopsis plumosa was collapsed by a physical wounding, the extruded protoplasm aggregated into numerous protoplasmic masses in sea water. A polysaccharide envelope which initially covered the protoplasmic mass was peeled off when a cell membrane developed on the surface of protoplast in 12 h after the wounding. Transmission electron microscopy showed that the protoplasmic mass began to form a continuous cell membrane at 6 h after the wounding. The newly generated cell membrane repeated collapse and rebuilding process several times until cell wall developed on the surface. Golgi bodies with numerous vesicles accumulated at the peripheral region of the rebuilding cell at 24 h after the wounding when the cell wall began to develop. Several layers of cell wall with distinctive electron density developed within 48-72 h after the wounding. Proteome profile changed dramatically at each stage of cell rebuilding process. Most proteins, which were up-regulated during the early stage of cell rebuilding disappeared or reduced significantly by 24-48 h. About 70-80% of protein spots detected at 48 h after the wounding were newly appeared ones. The expression pattern of 29 representative proteins was analyzed and the internal amino acid sequences were obtained using mass spectrometry. Our results showed that a massive shift of gene expression occurs during the cell-rebuilding process of B. plumosa.

키토산 마이크로캅셀 및 비드의 제조와 응용에 관한 연구 (A Study on the Preparation and Application of Chitosan Microcapsule and Bead.)

  • Glutaraldehyde를 사용하여 화학적인 가교반응에 의해 empty cross-linked chitosan microcapsule을 제조하였다. 또한 chitosan bead는 sodium hydroxide 용액을 이용한 coaervation에 의해 제조하였다. 이들의 제조를 위해 수상/유화제/유기상으로 이루어진 w/o emulsion을 형성시킨 후 에멀젼의 안정성에 영향을 미치는 요소들에 대해 조사하였다. 보조계면활선제로 n-hexanol을 첨가한 결과 유화안정성이 상승적으로 증가하는 경향을 보였다. Chitosan microcapsule은 광택이 나는 형태로 내부가 투명한 반면 chitosan bead는 백탁으로 내부가 불투명하며 매우 porous한 구조를 하고 있었다. Chitosan bead의 유리 아미노기의 분석은 picric acid titration으로 수행하였으며, 아미노산 반응의 종결여부는 ninhydrin color test에 의해 판정하였다. 또한 fluoroscamine에 의한 형광분석으로 chitosan bead의 표면에 많은 반응성 아미노기가 존재하고 있음을 정성적으로 확인할 수 있었으며, 이를 생리활성 펩티드의 커플링 반응에 이용할 수 있었다. 모델 펩티드로서 콜라겐의 receptor 부분에 해당하는 생체내 growth factor로 알려진 Gly-His-Lys를 각 아미노산 유도체를 차례로 커플링하여 chitosan bead 위에서 합성할 수 있으며, 세포 성장인자와 같은 여러 가지 생리 활성 펩티드의 운반체로서 chitosan bead가 쉽게 이용될 수 있는 가능성을 보여 주었다.

Na-alginate 투여가 白鼠의 鉛과 카드뮴 독성에 미치는 영향 (Effects of Na-alginate on the Toxicity Induced by Lead and Cadmium in Albino Rats)

  • Earlier studies have shown that the administration of Na-alginate, a water-soluble, non absorbable acidic polysaccharide, effectively reduces the absorption of radioactive strontium, cadmium and other bivalent metals. In this study the effects of Na-alginate on the Toxicity induced by Lead and Cadmium were examined in rats and the following results were obtained. 1. The elevations of the activities of serum GOT, GPT by Pb and Cd administration in the experimental animals. However, the administration of Na-alginate significantly inhibited the elevation of serum GOT, GPT. 2. Generally serum TBA values were significantly elevated in Pb and Cd group in comparison with the control group. However, the administration of Na-alginate inhibited the elevation of serum TBA values. 3. The activity of Alkaline phosphatase was also significantly elevated in Cd group in comparison with the control group. However, the administration of Na-alginate significantly inhibited the elevation of serum alkaline phsphatase. 4. Furthermore, the activity of $\delta$-aminolevulinic acid dehydratase in red blood cells was also significantly inhibited by Pb and this inhibition was stimulated by the administration of Na-alginate. 5. The concentrations of Pb in liver and Cd in kidney were markedly increased by Pb and Cd. However, the administration of Na-alginate significantly inhibited the elevation of them.

Purification, Chemical Composition, and in vitro Antioxidant Activity of Two Protein-bound Polysaccharides from Rapeseed Meal

  • Crude polysaccharides from rapeseed meal (PRM) were extracted with 0.3% NaOH aqueous solution, followed by further purifications and 2 fractions, namely PRM1 and PRM2, were separated with a DEAE-cellulose DE-52 column. Their primary compositions were analysed and antioxidant activity was determined, including scavenging activity toward superoxide anion radicals, hydroxyl radicals, and nitric oxide radicals, reducing power, and inhibitory effects against the microsomal lipid peroxidation, compared to that of L-ascorbic acid. The results indicated that PRM1 and PRM2 exhibited not only good reducing power and inhibitory effects on the microsomal lipid peroxidation, but also strong scavenging activity toward superoxide anion radicals, nitric oxide radicals, and hydroxyl radicals. In addition, positive correlations were also observed between the superoxide anion radical scavenging activity and the protein contents of the polysaccharides, and the reducing power and the sulfate contents. These findings thus clearly suggest the polysaccharides possess direct and potent antioxidant activity.

더덕과 인삼의 식이섬유소 함량의 측정 (Determination of Dietary Fiber Contents in Dodok ( Codonopsis lanceolata traut (Beneth et Gook)) and Ginseng (Panax ginseng C. A. Meyer))

  • The total dietary fiber (TDF) contents in cultivated dodok, wild dodok and ginseng were determined by AOAC method and Mongeau et al's method. Also, the nonstarch polysaccharide (NSP) contents of them were analyzed by Englyst et al's gas chromatographic (GC) method. The TDF values by AOAC method and Mongeau et al's method were 34.50% and 35.92% for wild dodok, 46.40% and 47.55% for cultivated dodok, and 14.93% and 14.03% for ginseng, respectively. The ratios of soluble dietary fiber (SDF) contents to TDF contents by Mongesu et al's method were 64.17% for wild dodok, 75.77% for cultivated dodok, and 53.74% for ginseng, respectively. The major sugar constituents of NSF in wild dodok, cultivated dodok and ginseng by GC were glucose, galactose and uronic acid. The Enalyst's TDF contents, i.e., NSP plus lignin contents in wild dodok, cultivated dodok, and ginseng were 20.65%, 20.03% , and 9.72%, respectively.

아가리쿠스 버섯(Agaricus blazei Murill)의 효능(效能) 및 연구동향(硏究動向)에 대한 고찰(考察) (Literature study on the effect and reserch trend of the Agaricus blazei Murill)

  • Basidiomycete, Agaricus blazei murill has grown Brazil naturally. It was first cultivated 1997 in Korea. Proteoglycan or polysaccharide which have the effect of immunopotency and anticancer were extracted from it. From the mycelium of cultivated mushroom, were extracted lectin, linoleic acid, FIII-2-b, and simmilar to glucomannan from fruit body of Basidiomycete Agaricus blazei. Fruit body was more studied than mycelium. The experiments were most in vivo study. The effect were shown on S-180, MethA tumor, Ehrlich ascites carcinoma, Shionogi carcinoma 42, Meth A fibrosarcoma by transferation of tumor cell line, specially effectiveness on the S-180. About immunopotenciation, were shown as activation of NK cell, pancreatic T-cell, helper T cell, enhancement of population of cytotoxity T cell. It was effect on the MethA tumor cells in vitro cell cytotoxity and has induction of apoptosis. Forthermore cytotoxity of many other tumor cell line, aneiognensis, cell cyle studies will be needed.

식물성 산성당으로부터 헤파리노이드의 제조 (Preparation of Heparinoids from Acidic Plant Polysaccharides)

  • Anticoagulant activities were tested for the fifteen kinds of medicinal plants by measuring activated partial thromboplastin time (aPTT). Of them five kinds or species (Artemisia princeps, Sanguisorba officinalis, Artemisia apiacea, Eclipa alba, Schizonepeta tenuifolia) were selected and fractionated for the preparation of acidic polysaccharides. They were extracted with water by refluxing and the extracts were precipitated with ethanol. The precipitates were separated based on charge using a DEAE-Sephadex. The low salt and high salt fractions were sulfated with anhydrous pyridine and chlorosulfonic acid complex. In vitro anticoagulant activities of sulfated polysaccharides were tested by measuring aPTT, prothrombin time (PT), and factor Xa clotting time using normal human plasma. No relationship was found between the amount of uronic acids and anticoagulant activities, but the sulfated ones show the increase of activities. In vivo anticoagulant properties of the sulfated polysaccharide from Artemisia apiacea were also tested by the intraveneous administration of three different doses (3,5 and 10 mg/kg) to rats. APTT and PT were increased significantly and the action of factor Xa and thrombin mediated through antithrombin III were inhibited slightly.

Liquid chromatographic enantioseparation of several amino acids as nitrobenzoxadiazole derivatives on polysaccharide trisphenylcarbamate derived chiral stationary phases

  • Considering the greater role of α-amino acids in our daily lives, the enantiomer resolution of seven α-amino acids derivatized with fluorogenic reagent (4-fluoro-7-nitro-2,1,3-benzoxadiazole, NBD-F) by chiral HPLC on amylose or cellulose trisphenylcarbamate derived chiral stationary phases (CSPs) under simultaneous ultraviolet (UV) and fluorescence (FL) detection was performed. The degree of enantioseparation and resolution was affected by nature and selector backbones of the CSPs as well as the kind of amino acids. Baseline enantiomer separation and resolutions were observed for the enantiomers of all analytes as NBD derivatives especially on coated type amylose tris(3,5-dimethylphenylcarbamate) derived CSPs (Chiralpak AD-H and Lux Amylose-1). The other CSPs also showed good enantioselectivity except for the CSPs (Chiralpak IB, Chiralcel OD-H and Lux Cellulose-1) having cellulose tris(3,5-dimethylphenylcarbamate) as chiral selectors. The developed analytical chiral method was applied to determine the enantiomeric purity of seven commercially available L-α-amino acids and the impurities as D-forms were found to be in the range 0.08-0.87 %, respectively. The intra- and interday accuracy and precision assays showed high accuracy and precision of the developed analytical method. This chiral HPLC method for the enantiomer resolution of amino acids using fluorescent derivatization could be useful for the determination of enantiomeric purity of pharmaceuticals and biological study for amino acid type compounds among chiral drugs.
