This experiment was carried out to develop an acclimatization system of tissue cultured seedling in Zantedeschia spp. There was not different in percentage of leaf emergence by the media compost of acclimatization but tuber was enlarged in sand(50%)+peatmoss(50%) treatment after 6 months growth. The optimum time for the acclimatizantion of Zantedeschia spp. was from the 20th of March to 20th of April. During this time, rate of acclimatization reached over 90 percentage. However on the 20th of august, acclimatization was low to 64-70.4 percentage. Size of tuber was enlarged in planting on the 20th of March. The weight of tuber was better in 10×10cm density of planting than others.
This experiment was carried out to know the effect of media and agar concentrations, aeration and growth regulators on rooting and acclimatization of the shoots harvested from bioreactor culture in Rehmannia glutinosa. Half MS media with 1.2% agar improved rooting and acclimatization of shoots. Shoots were effectively acclimatized and rooted well in case of aeration by using membrane filtered vessels. Shoots acclimatized in vessel with membrane Inter were healthier and had higher ex vitro survival rate than those without membrane Inter on plug tray. Addition of paclobutrazol 0.3-0.4 mg/L, to acclimatization media enhanced shoots growth and root development.
This study was conducted to clarify the acclimatization rate and ploidy level of anther culture-derived plants in broccoli. The acclimatization rate from 71.4 to 100% was obtained from 7 varieties in 2 years. It was possible to identify the ploidy of the Plants obtained through anther culture by measuring the number of chloroplast in the guard cell. The average numbers of chloroplasts per guard cell in haploid diploid, and tetraploid were 8.5, 13.5 and 18.5, respectively. The regenerated plants could be classified based on these results into 47.1-51.3% of haploids, 47.9∼51.7%, diploid, and 0.8∼l.2% of tetraploids.
This study was conducted to dentify the optimum plantlet type of 'Solara' potatoes (Solanum tuberosum L.) for growth in an aeroponics system. Plantlets of 'Solara' were transplanted on March 16, 2015 in a greenhouse, and growth and yield characteristics were investigated at 70 and 78 days after transplanting, respectively. Stem length was shorter in plantlet of 15-day-old stem cuttings and acclimatization of culture, and the stem length of plantlets of stem cuttings tended to increase with increasing stem cutting age. The fresh weight of plants was the highest in the plantlets of 40-day-old stem cuttings and the lowest in non-rooted stem cuttings and acclimatization of culture. The highest number of first stolons was obtained in 35-day-old stem cuttings. The number of second stolons was the highest in plantlets of 35-day-old stem cuttings, acclimatization of culture, and 30- day-old stem cuttings. The total number of tubers was higher in plantlets of 35-day-old stem cuttings and acclimatization of culture, and the number of tubers above 3 g was the highest in plantlets of 35-day-old stem cuttings. The weight of tubers above 3 g was the heaviest in plantlets of 35-day-old stem cuttings(1,947 g per 10 plants), followed by plantlets of 30-day-old stem cuttings. These results indicate that plantlets of 30 to 35-day-old stem cuttings could be the best for production of 'Solara' potato tubers in an aeroponics system.
Photosynthetic rates of the large centric diatom Coscinodiscus granii were measured by means of multicolor variable chlorophyll fluorescence imaging, single cell $^{14}C$ assimilation, and optical $O_2$ sensor measurements during light acclimatization of cultures grown at five different irradiances: 50, 150, 235, 332, and $450{\mu}mol$ photons $m^{-2}\;s^{-1}$. Photo-acclimatization was evident from changes of cellular chlorophyll a content, growth rates, and light response curves. Each of the applied methods evaluates different parts and reactions in the photosynthetic apparatus, which makes a direct quantitative comparison of rates difficult, although a different degree of correlation were found between all three methods. However, when used in combination, they provide information about the internal relationship of photosynthetic pathways as well as the variation in photosynthetic capacity between individual cells within a single algal culture.
Long-term culture of cell lines is an important issue in in vitro culture and in plant science. In this study, the regeneration ability and ex vitro acclimatization of regenerants were evaluated. The ploidy level of regenerants derived from long-term cultured cell lines was measured in Imperata cylindrica 'Rubra', Poaceae. Adventitious buds (shoots) were successfully induced from five-year-cultured calli on MS medium containing 0.1 mg/L BA or 2.0 mg/L TDZ, combined with 0.01 mg/L auxins (IAA, IBA, NAA and 2,4-D), respectively. Adventitious roots were also induced on MS medium containing 0.01 mg/L auxins (IBA, NAA and 2,4-D), respectively. Interestingly, regenerants with both red and green leaf were successfully obtained when regenerants were cultured on MS medium with 9% sucrose. Regenerants derived from long-term cultured calli were transferred to pots using an optimal acclimatization process and successfully adapted to both pot and soil conditions. Moreover, the ploidy level was measured using calli and regenerants that had been kept on MS medium containing various kinds of plant growth regulators (PGRs).
Mulberry (Morus sp.) of the family Moraceae is very economically important in Asian countries including Korea, because its leaf and fruit have been commercially used in sericulture and horticultural industries. Therefore it is necessary to develop the optimal production system for rapid and cost-effective propagation of mulberry. Our studies focused on establishing an acclimatization method for the successful plantlet production of new cultivar 'Cheongsu' which was transferred ex vitro after in vitro culture. In particular, effect of abscisic acid (ABA) addition into the last subculture medium on plantlet response to subsequent ex vitro transfer and its growth was investigated. During acclimatization, stomatal conductance and transpiration rate of ABA-pretreated plantlets were significantly lower than those of non-treated plantlets. Net photosynthetic rate of ABA-pretreated plantlets decreased after ex vitro transfer but increased after 14 days, and it was mostly higher than that of non-treated plantlets. Moreover, relative water content as well as chlorophyll contents and its ratio were also higher in ABA-pretreated plantlets. On the other hand, proline was considerably higher than in control plantlets. After 1 month of ex vitro transfer, survival rate of ABA-pretreated plantlets was 85.6%, which increased by 29.1% in comparison with control (56.5%). More vigorous growth was also observed in ABA-pretreated plantlets. From these results, it was found that application of ABA to the last subculture medium could improve acclimatization and promote survival of mulberry plantlets after ex vitro transfer, inducing water stress tolerance and alleviating abiotic stresses.
Rosa rugosa is a medicinal, ornamental, and edible plant native to Eastern Asian countries, including Korea, Japan, and China. The aim of this study was to establish a system for biomass production and secondary metabolite accumulation during in vitro culture and acclimatization of Rosa rugosa. The highest rate of multiple shoot proliferation was achieved with $8.8{\mu}M$ benzyladenine (BA) (83.3%). However, the number of shoots (14.4 per explant) at $4.4{\mu}M$ BA was higher than that at $8.8{\mu}M$ BA. Compared to BA, a combination of thidiazuron (TDZ) and indole butyric acid (IBA) exhibited significantly lower shoot induction, with only 50.0~79.2% and 4.2~16.7% relative shoot formation, respectively. During acclimatization, shoots were sampled every week and their total phenolic contents were analyzed. Among various growth factors, fresh weight showed the most dramatic increase from the 3rd week (88.0 mg/plant) to 4th week (132.7 mg/plant). Total phenolics and flavonoids contents were the highest at $1^{st}$ week of acclimatization. Depending on developmental stages, total phenolics and flavonoids contents were higher in 1-yr-old shoots grown ex vitro than in those of older field-grown or in vitro-grown plants. Amongst different ages of field grown plants, 6-year-old plants, the oldest in this study, showed the lowest content in total phenolics.
In mass production of seed-potato plantlets, the processes for in vitro propagation and ex vitro acclimatization with a high cost should be improved by a culture system with environmental control using scaled-up culture vessels. The experiment was conducted to design a hydroponic culture system for enhancement of growth and development of seed-potato (Solanum tuberosum) plantlets cultured under photoautotrophic (without sugar in culture medium) conditions with controlled light intensity and ventilation rate. The culture system was consisted of scaled-up culture vessels, ventilation pipes, a multi-cell tray and an environmental control system (ECS) for optimum controlling in temperature, light intensity, ventilation rate, and culture-medium supply. Growth and development of the plantlets was significantly increased under the ECS compared with a conventional culture system (CCS) of photomixotrophic culture (with sugar in culture medium) using small scale vessels. For 21 days, leaf area of the plantlets was expanded more than 2 times, and number of internodes also approximately 4 times greate. under the ECS. In addition, the photoautotrophic growth in sweetpotato (Ipomoea batatas) and chrysanthemum (Chrysanthemum morifolium) plantlets was greater more than 2 times compared with the CCS.
Background: Promising specific growth regulators are employed in the tissue cultures of various bamboo species. Specific natural hardening mixtures support the acclimatization and adaptation of bamboo under protected cultivation. Results: The growth regulators like 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Naphthaleneacetic Acid (NAA), Thidiazuron (TDZ), 6-Benzylaminopurine (BAP), Kinetin, Gelrite, Benzyl Adenine (BA), Indole Butyric Acid (IBA), Coumarin, Putrescine, Gibberellic acid (GA3), Indole Acetic Acid (IAA) has been widely used for callus induction, root regeneration and imposing plant regeneration in various species of bamboo such as Bambusa spp. and Dendrocalamus spp. Different combinations of growth regulators and phytohormones have been used for regenerating some of the major bamboo species. Natural hardening materials such as cocopeat, vermicompost, perlite, cow dung, farmyard manure, compost, soil, garden soil, and humus soil have been recommended for the acclimatization and adaptation of bamboo species. Standard combinations of growth regulators and hardening mixtures have imposed tissue culture, acclimatization, and adaptation in major bamboo species. Conclusions: Bamboo contributes to soil fertility improvement and stabilization of the environment. Bamboo species are also involved in managing the biogeochemical cycle and have immense potential for carbon sequestration and human use. This paper aims to review the various growth regulators, natural mixtures, and defined media involved in regenerating major bamboo species through in vitro propagation. In addition, the ecological benefits of safeguarding the environment are also briefly discussed.
