  • Bacillus subtilis KMU-13 was isolated from the Lillehammer forest soils at Norway and shown a strong antifungal activity on cucumber scab, Cladosporium cucumerinum KACC 40576. B. subtilis KMU-13 produced a maximum level of antifungal substance under incubation aerobically at $30^{\circ}C$, 180 rpm for 48 hours in LB broth containing 0.5% maltose and 0.5% bactopeptone and initial pH adjusted to 6.0. Butanol extract of cultured broth was confirmed inhibitory zone by plate assay and Rf 0.64 value substance by thin layer chromatography (TLC) represented high antifungal activity against C. cucumerinum KACC 40576 and also shown fungal growth inhibitory activity against Botytis cinerea KACC 40573, C. gloeosporioides KACC 40804, D. byoniae KACC 40669, F. oxysporum KACC 40037, F. oxysporum KACC 40052, F. oxysporum f. sp. radicis-lycopersici KACC 40537, F. oxysporum KACC 40902, M. cannonballus KACC 40940, P. cambivora KACC 40160, R. soiani AG-1 KACC 40101, R. solani AG-4 KACC 40142, and S. scleotiorum KACC by agar diffusion method.

Damage Analysis of the Bridge Structure Caused by Fire Outbreak (화재로 인한 교량구조의 손상 분석)

  • Purpose: The purpose of this study is to accurately analyze the damage of bridges in order to determine whether fire bridges can be used continuously or to provide information on maintenance augmentation data. Method: XRD, SEM and EDS analyzes of concrete were carried out to estimate the fire temperature transferred to the structure, and analyzed by depth and area from PSCI beam and bottom plate concrete surface. Results: Test results G12,11 for the fire zone concrete were confirmed to be affected by heat up to depth of 60mm and the temperature of the hydrothermal heat was above 1000℃. Also, the girder G10,9,8 was relatively weakly damaged compared to G12,11, and the degree of damage was confirmed to be affected by heat up to a depth of 40 mm. Conclusion: Based on the analyzed data, it is considered that if the repair / reinforcement and periodic inspection are carried out, the bridge can secure sufficient safety even considering the damage caused by the fire.

A Practical Algorithm to Simulate Erosion of On-Shore Zone (실용적 해안선 후퇴 반영 알고리즘)

  • An algorithm to allow shoreline movement during numerical experiment on sediment transport, deposition or resuspension for general coastal morphology is proposed here. The bed slope near shoreline, i.e. mean sea level, is influenced by bed material, tidal current, waves, and wave-induced current, but has been reported to remain within a stable range. Its annual variation is not large, either. The algorithm is adjusting the bathymetry, if the largest bed slope within shoreline band exceeds a given bed slope due to continuous erosion at zones below the shoreline. This algorithm automatically describes retreat of shoreline caused by erosion, when used within a numerical system. The algorithm was tested to a situation which includes a continuous dredging at a point, and showed satisfactory development of concentric circle contours. Next, the algorithm was tested to another situation which includes sinking of eroded part of bed plate, and produced satisfactory results, too. Finally, the algorithm was tested to a movable-bed laboratory experimental conditions. The shoreline movement behind detached breakwater was reasonably reproduced with this algorithm.

Isolation and Phylogenetic Characterization of Chitinase Producing Oligotrophic Bacteria (Chitinase생산 저영양세균의 분리 및 계통분류학적 특성)

  • Many isolates from soil of Korean ginseng rhizosphere did not show remarkable growth on full strength of the conventional nutrient broth (NB medium) but grew on its 100-fold dilution (DNB medium). Six hundred-forty strains were isolated as oligotrophic bacteria. In the course of screening for new bioactive compounds from oligotrophic bacteria from soil, 8 strains which had appeared to form of clear zone on a medium containing colloidal chitin as a sole carbon source were selected for further studies. Strain CR42 hydrolyzed a fluorogenic analogue of chitin, 4-methylumbelliferyl-D-glucosaminide (MUF-NAG) . Mo st of the culture supernatant of these isolates hydrolyzed 4-methylumbelliferyl-D-N,N'-diacetylchitobioside (MUF-diNAG). The isolates were heterogeneous and categorized to gamma- and beta-proteobacteria, Bacillaceae, Actinobactepia, and Bacteroides by 16S rRNA analysis. Two strains, WR164 and CR18, had a 16S rRNA sequence of $95-96\%$ identical to uncultured bacteria. It was observed that CR2 and CR75 could inhibit the growth of Colletotrichum gloeosporioides with hyphal extention-inhibition assay on PDA plate supplemented with $1\%$ colloidal chitin.

Isolation and Characteristics of Bacteria Showing Biocontrol and Biofertilizing Activities (생물방제 및 생물비료 활성을 가지는 세균의 분리 및 특성)

  • To develop multifunctional microbial inoculant, microorganisms with antagonistic activity and biofertilizing activity were screened. Pantoea agglomerans and Bacillus megaterium from our laboratory culture collection, and strain MF12 from soil near poultry farm in Miryang were selected. On the basis of morphological, physiological studies and 16S rDNA sequence analysis, isolate MF12 was identified as the Bacillus pumilis. Three strains were studied for insoluble phosphate solubilization, indole-3-acetic acid (IAA) and siderophore production, ammonification ability, hydrolytic enzyme production and antifungal activity against phytopathogenic fungi. P. agglomerans did not produce any visible clear zone on agar plate containing 0.5% $Ca_3(PO_4){_2}$ as a sole phosphorus source. However, this strain could solubilize insoluble phosphate in liquid medium. All strains produced IAA ranged from $3{\sim}639{\mu}g/ml$ depending on culture time and had ammonification ability. Among three strains, only P. agglomerans produced siderophore. P. agglomerans produced pectinase and lipase, B. megaterium produced amylase, protease and lipase while B. pumilis produced protease and lipase. P. agglomerans showed antifungal activities against phytopathogenic fungi, Fusarium oxysporum and Colletotrichum gloeosporioides. B. pumilis showed antifungal activities against Botrytis cinerea, Sclerotinia sclerotiorum and Phythium ultimum.

Estrogen promotes the onset and development of idiopathic scoliosis via disproportionate endochondral ossification of the anterior and posterior column in a bipedal rat model

  • This study aimed to verify the effects of estrogen on the onset and development of adolescent idiopathic scoliosis and the mechanisms associated with these effects by constructing a pubescent bipedal rat model. Experiments were conducted to investigate whether scoliosis progression was prevented by a Triptorelin treatment. One hundred twenty bipedal rats were divided into female, OVX (ovariectomy), OVX + E2, Triptorelin, sham, and male groups. According to a spinal radiographic analysis, the scoliosis rates and curve severity of the female and OVX + E2 groups were higher than those in the OVX, Triptorelin, and male groups. The measurements obtained from the sagittal plane of thoracic vertebrae CT confirmed a relatively slower growth of the anterior elements and a faster growth of the posterior elements between T11 and T13 in the female and OVX + E2 groups than in the OVX and Triptorelin groups. Histomorphometry and immunohistochemistry revealed a significantly longer hypertrophic zone of the vertebral cartilage growth plates that expressed more type X collagen and less type II collagen in the OVX and Triptorelin groups than in the female and OVX + E2 groups. Ki67 immunostaining confirmed an increase in the proliferation of vertebral growth plate chondrocytes in the OVX group compared with the female and OVX + E2 groups. In conclusion, estrogen obviously increased the incidence of scoliosis and curve severity in pubescent bipedal rats. The underlying mechanism may be a loss of coupling of the endochondral ossification between the anterior and posterior columns. Triptorelin decreased the incidence of scoliosis and curve magnitudes in bipedal female rats.

Antibacterial Effects of Atmospheric Plasma against Main Foodborne Bacteria on the Surface of Dried Filefish (Stephanolepis cirrhifer) Fillets (대기압 플라즈마 처리에 의한 쥐치포 중 주요 식중독세균의 살균 효과)

  • This study investigated the antibacterial effects of BioZone atmospheric plasma (AP) against Bacillus cereus (F4810/72) and Staphylococcus aureus (ATCC 6538) as the major foodborne bacteria on the surface of dried filefish (Stephanolepis cirrhifer) fillets. The fillets were experimentally contaminated with 7-8 log CFU/mL of B. cereus or S. aureus using a spot inoculation method. Bacterial counts were measured by standard plate method on tryptic soy agar, and were significantly reduced with the increase in the treatment time (1, 3, 5 or 20 min) of AP on the fillets (p < 0.05). The reductions of the pathogens by AP treatment ranged from 0.9 to 2.93 logCFU/g for B. cereus and from 1.04 to 2.55 logCFU/g for S. aureus. A reduction of >1-logCFU/g for B. cereus and S. aureus was observed on the fillets treated with AP for >3 min. The differences in color on the Hunter scale (L=light vs. dark, a=red vs. green, b=yellow vs. blue) of the fillets were not significantly different between the nontreated (control) and AP-treated fillets (p>0.05). This study suggested that 3 min of AP could be effective in reducing >90% of the bacteria without causing any concomitant changes in the color of the fillets.

Identification and Fermentation Characteristics of Lactic Acid Bacteria that Produce Soy Curd With Low Sour Taste (저산미의 두유 커드를 생성하는 젖산균의 동정과 발효 특성)

  • The acidity of soy curd fermented by lactic acid bacteria is a major factor degrading the sensory properties of soy curd. For preparation of soy curd with low sour taste, lactic acid bacteria were separated from kimchi. The lactic acid bacteria which showed yellow-clear zone around the colonies on BCP plate and formed soy curd with low level of acidity were selected. The selected strain was analyzed by 16S rDNA sequence and named as Pediococcus inopinatus Y2. The maximum viable cell number of the soy curd fermented by P. inopinatus Y2 was obtained at 10.73 log CFU/mL at $25^{\circ}C$ for 24 h of fermentation. By the results of panel test, the overall sensory quality of the soy curd produced by P. inopinatus Y2 was higher than that of Leuconostoc mesenteroides No. 4395 and Lactobacillus sakei strain No. 383.

SPH-Based Wave Tank Simulations (SPH 기법 기반의 파동수조 시뮬레이션)

  • Recently, large-scale offshore and coastal structures have been constructed owing to the increasing interest in eco-friendly energy development. To achieve this, precise simulations of waves are necessary to ensure the safe operations of marine structures. Several experiments are required in the field to understand the offshore wave; however, in terms of scale, it is difficult to control variables, and the cost is significant. In this study, numerical waves under various wave conditions are produced using a piston-type wavemaker, and the produced wave profiles are verified by comparing with the results from a numerical wave tank (NWT) modeled using the smoothed particle hydrodynamics (SPH) method and theoretical equations. To minimize the effect by the reflected wave, a mass-weighted damping zone is set at the right end of the NWT, and therefore, stable and uniform waves are simulated. The waves are generated using the linear and Stokes wave theories, and it is observed that the numerical wave profiles calculated by the Stokes wave theory yield high accuracy. When the relative depth is smaller than two, the results show good agreement irrespective of the wave steepness. However, when the relative depth and wave steepness are larger than 2 and 0.04, respectively, the errors are negligible if the measurement position is close to the excitation plate. However, the error is 10% or larger if the measurement position is away from the excitation location. Applicable target wave ranges are confirmed through various case studies.

The effect of antagonists produced by Paenibacillus polymyxa CK-1 on the growth of Trichoderma sp. (Paenibacillus polymyxa CK-1이 생산한 길항물질이 Trichoderma sp. 생육에 미치는 영향)

  • The separation of the bacteria inhibiting Trichoderma sp. mold, the strain causing blue mold disease that occurs frequently when cultivating mushroom while carrying out the efficient fermentation of mushroom medium, from the growth was done. In about 200 strains isolated primarily from fungus garden samples, 6 strains were secondly isolated, which had fast growth rates and a clear zone on the plate medium of SM, AM, and CM. Among the 6 strains isolated, the C-1 strain showed high enzymatic activity of cellulase, amylase, and protease, and strong antibacterial activity for the T. virens and T. harzianum, selected finally. The selected C-1 strain was identified as Paenibacillus polymyxaby the result of the identification by Bergey's Manual of Systematic Bacteriology and the analysis of the nucleotide sequence of 16S rRNA, and named as P. polymyxa CK-1. In reviewing the growth conditions of the P. polymyxa CK-1 strain, the optimum cultivation temperature was $45^{\circ}C$, and the optimum pH for growth was in the range of 6.0~7.0. Appropriate incubation time of P. polymyxa CK-1 for the growth inhibition of the fungus T. virens and T. harzianum was 22 to 36 hours. And the fungal growth was not observed, even when leaving two molds inoculated on each petri dishes, which were treated with 24 hour culture solution of P. polymyxa CK-1 strain for 10 days. As a result of studying the thermal stability of the antagonists produced by the P. polymyxa CK-1 strain, no mycelial growth of the two fungi was observed in the test group treated for 20 minutes at $60^{\circ}C$ and $100^{\circ}C$, but mycelial growth was slightly observed in the test group treated for 20 minutes at $121^{\circ}C$. As aresult of reviewing the impact of the P. polymyxa CK-1 culture medium on mushroom mycelial growth, it showed no effect on a variety of mushroom mycelial growth including enoki mushroom and shiitake mushroom.
