Determination of Eupatilin in Human Plasma by Liquid Chromatography/Electrospray Ionization Tandem Mass Spectrometry

  • A rapid, sensitive and selective liquid chromatography-tandem mass spectrometric (LC/MS/MS) method for the determination of eupatilin in human plasma was developed. Eupatilin and internal standard, (S)-[N-3-(4-(2-(1-methyl-5-tetrazolyl)-pyridine-5-y1)- 3-fluorophenyl)-2-oxo-5-oxazolidinyl]methyl acetamide (DA-7867) were extracted from human plasma by liquid-liquid extraction and analyzed on a phenyl-hexyl column with the mobile phase of acetonitrile-ammonium formate (10 mM, pH 3.0) (60:40, v/v).   • Endocrine disrupting effects of four pharmaceutical products were evaluated with fish. The test pharmaceuticals, i.e., sulfamethoxazole, sulfamethazine, oxytetracycline and tetracycline have been often detected in aquatic environment, but their ecological hazard on receptors of various trophic levels has seldom been evaluated. In the present study, we conducted acute toxicity assays with a fish, Japanese medake (Oryziα lαtipes). The vitellogenin induced in female fish normally, but a endocrine disrupting chemical could give effects even male fish. We have tried 4 pharmaceutic chemicals to find out the endocrine disrupting effects. Sulfamethoxazole 1, 0.5 ppm induced vitellogenin even at male Japanese medaka. Sulfamethazine 10, 5, 1 ppm could induced vitellogenin at male fish. Oxytetracycline 10, 5, 1ppm could induced vitellogenin With the fish. Tetracycline 10, 5 ppm could induced vitellogenin at male fish. Some pharmaceuticals such as sulfamethoxazole, sulfamethazine, oxytetracycline and tetracycline could give effects to male Oryzias latipes. They could induced vitellogenin under exposure range 0.5 ${\sim}$ 10 ppm of chemicals at male Oryzias latipes.

Development and Validation of Crocidolite Quality Control Samples for Proficiency Analytical Testing (청석면 분석 정도관리용 표준시료 개발 및 평가)

  • Crocidolite quality control (QC) sampling created by the wet generation method has been validated by validation tests such as the accuracy, precision, and storage tests. For this study we designed and developed a manufacturing apparatus and standard operating procedure for making these QC samples. The most important step in the procedure of making QC samples was the stage eliminating static electricity in asbestos fibers. This static electricity hampers the fibers clog functioning. In accuracy and precision tests by phase contrast microscopy analysis, the difference between the reference values and the studied values was at maximum 17.8%. This satisfies the AIHA proficiency analytical test criteria for asbestos. We could confirm the nearly even distribution of crocidolite fibers on the membrane filter. Also, there was no loss of fibers in the storage test after the one month.

We analyzed vitellogenin induction by qualitative (Western blot) and quantitative (ELISA) assay. $17{\beta}$ -estradiol was used as a positive control. Some pharmaceuticals could give effects to male Oryzias latipes. They could induced vitellogenin under exposure of chemicals at male Oryzias latipes.

  • In the study, the feasibility of landscape ecological measures as indices system for interpretation and evaluation of regional ecosystem was investigated through the application to Yongin City. Each patch metrics well showed the class structure and supplemented the class metrics, and class metrics also showed well the landscape structure and supplemented the landscape metrics. And the change analysis through subtraction of two set of landscape ecological measurement in two point of time showed the dynamic trends very well. One of the dynamic trends in Yongin City was the rapid fragmentation. While there was no landcover data on Yongin City, using Landsat data and remote sensing techniques were proved to be efficient and effective to produce the digital landcover data.
