Effective microorganisms (EM), which are sources of fermentation byproducts in herbal medicine, were compared with oyster mushrooms(OM) to identify alternative larval feeds for white-spotted flower chafer (Protaetia brevitarsis seulensis). The nutritional composition of each fermented feed was analyzed. The content of crude protein, crude fat, and fiber was higher in the OM fermented feed, except for crude ash. No difference in nutritional content of larvae based on fermented feed was observed. A comparative weekly analysis of the live weights of larvae was based on the fermented feed used. The average weight was significantly higher in the feeds using EM and OM from third week of observation. The larval survival rate in the presence of fermented feed was 96.7% compared with 9.8% with non-fermented feed. The results suggest that fermented food source is essential for the growth of white-spotted flower chafer, and OM was a stable alternative to EM as a fermentation source for the survival of white-spotted flower chafer.

The induced antibiotic peptides were isolatde from the white-spotted folwer chafer, Protaetia brevitarsis by injection of E. coli suspension to the larvae of the insect. The antibacterial activity of the peptides were assayed by the plate growth ingibition method, and were purified by ion-exchange chromatography, reversed-phase HPLC, ion-exchange HPLC and SDS-PAGE etc. The peptides were estimated as 9 kDa in molecular weight and named Protaecin I and Protaecin II, respectively. Protaecin I and II have strong antibacterial activities against Gram-positivie and/or Gram-negative bacteria, and they are stable in the heat treatment and in the range of pH 2-12.

Exosomes are homogenous vesicles of 40-100 nm diameter produced endogenously. Exosomes are generated by inward budding into multi-vesicular bodies (MVB) and then released to extracellular space. Exosomes contain various nucleic acid and protein cargoes from their cells of origin and this endosomal cellular molecules are used for intracellular communication and for both promotion and suppression of immune responses. Recently, they are also considered as delivery vehicle for therapeutic proteins due to their characteristics of stability in body fluids and ability for target uptake. Also, they show less immune reactivity because the isolated exosome harboring therapeutic proteins can be from the same host. White-spotted flower chafer, Protaetia brevitarsis is one of the major insect commercially reared in Korea. There are bacterial and fungal pathogens causing diseases in the beetle, and these diseases incur economic loss to the larva-rearing farms. Due to their endosomal cargoes, exosomes are good candidates in use of disease diagnosis. In this study, we isolated insect exosome from the hemolymph of P. brevitarsis, and verified it by analysis of the exosome-specific surface proteins and RNA.
In this study, the potential of infrared assisted freeze-drying (IRAFD) was tested for the production of shelf-stable edible insects: Protaetia brevitarsis larva (larva of white-spotted flower chafer). The IRAFD system was customized using an infrared lamp, K-type thermocouple, controller, and data acquisition system. The infrared lamp provided the sublimation energy for rapid freeze-drying (FD). The IRAFD conditions were continuous IRAFD-5.0 kW/㎡ and IRAFD-5.0 kW/㎡ at different weight reduction (WR) (10%, 20%, and 30%). The continuous IRAFD reduced the drying time to 247 min compared to the 2,833 min duration of FD (p<0.05). The electrical energy could be reduced by more than 90% through infrared radiation during FD (p<0.05). The Page model resulted in the best prediction among the tested drying kinetic models. In terms of quality, IRAFD showed significantly lower hardness, chewiness, and higher protein levels than hot air drying and FD (p<0.05). IRAFD better preserved the glutamic acid (6.30-7.29 g/100 g) and proline (3.84-5.54 g/100 g). The external product appearance after IRAFD exhibited more air pockets and volume expansion, which might result in a good consumer appeal. In conclusion, this study reports the potential of IRAFD in producing shelf-stable and value-added edible insects.

The white-spotted chafer, Protaetia brevitarsis (Coleoptera: Scarabaeidae), is an economically important insect in Korea. Traditionally, it has been regarded as a medicine for preventing liver-related diseases and suppressing liver cancer. Recently, this beetle was enlisted as a temporal food ingredient by Korean Ministry of Food and Drug Safety. Therefore, this beetle is focused as a one of the important insects that are commercially reared and sold in Korea. As the economic importance of this beetle is growing, the suitable rearing conditions are needed for more detailed investigation. In this study, we compared three temperature conditions and two food additives, rice chaff and soybean cake, for its effects on the body weight change of third instar larvae of P. brevitarsis. Temperature is a major environmental factor that has tremendous effects on rearing insects. In additions, rice chaff and soybean cake are byproducts of other agricultural activities. Therefore, it is easy to get, and the price is comparatively low. However, they still have meaningful amount of nutrients. With four different kinds of feed and three temperature conditions on the third instar larvae of beetles, the body weight change was tracked for 14 wk. We concluded that 27.5℃ is the optimal temperature to rear the third instar larvae among three temperatures (25, 27.5, and 30℃). Among four different feeds, conventional fermented oak saw dust with rice chaff and soybean cake was the best feed for larval weight gain during 14 wk. However, feed with soybean cake at 30 ℃ was the best condition for rearing P. brevitarsis larvae when temperatures and feeds were compared at the same time.

One of the necessary conditions for the mass production of the edible insect, the white-spotted flower chafer Protaetia brevitarsis seulensis (Kolbe), is to breed a strain with excellent egg-laying ability. To identify external morphological traits related to egg-laying ability, we investigated the effects of the weight, length, and width of female adults on egg production. Correlation and multiple regression analyses revealed a positive correlation between the weight of female adults and egg production. This study suggests that selection of heavier females is a good strategy for breeding strains with superior egg-laying characteristics. The results of this study will serve as important foundational data for future breeding of superior strains.

This study was carried out to investigate the development and oviposition characteristics of Protaetia brevitarsis fed mulberry fermented sawdust added with mulberry cake. The results of rearing of P. brevitarsis larvae on oak fermented sawdust and mulberry fermented sawdust added with different ratio of mulberry cake, the development periods of larvae feed sawdust with mulberry cake were about 41.6~48.5 days and shortened by 117 or more than that without treatment. The development periods were shorter as the ratio of mulberry cake added increased and there was no difference according to the kind of sawdust. But the weight of the larvae raised in mulberry fermented sawdust added with mulberry cake was heavier than that of the larvae raised in oak fermented sawdust added with mulberry cake. When mass rearing P. brevitarsis larvae in oak fermented sawdust, the addition of 5% mulberry cake was appropriate. Also, the average number of laying eggs of female who fed diet with mulberry cake was 12.3% higher than that of untreated.

A cuticle protein gene, PbLCP12.1, from the whitespotted flower chafer, Protaetia brevitarsis, was isolated and characterized. The gene contains an ORF of 336 nucleotides capable of encoding a 113 amino acid polypeptide with a predicted molecular mass of 12,138 Da and pI of 4.15. The PbLCP12.1 protein contained a type-specific consensus sequence identifiable in other insect cuticle proteins. The deduced amino acid sequence of the PbLCP12.1 cDNA is most similar to Bombyx mori cuticle protein BmLCP18 (37$\%$ protein sequence identity). Northern blot analysis revealed that PbLCP12.1 showed the epidermis-specific expression.

The food source of white-spotted flower chafer, Protaetia brevitarsis is important to economical performance. The P. brevitarsis larva were feed by mulberry and oak fermented sawdust. The effect of additional additives, Korean black raspberry (called bokbunja) marc and blueberry marc to fermented sawdust feed on the development and oviposition characteristics were investigated. The development periods of P. brevitarsis was decreased from 165.7 d to 40.0 d (mulberry sawdust) and 169.3 d to 39.0 d (oak sawdust), but the survival rate of larva was not affected by the addition of Korean black raspberry marc to fermented sawdust. However, blueberry marc addition to the fermented sawdust did not significant effect on the development period and survival rate of P. brevitarsis. The percentage of larva weight over 2.5 g was tend to increase by addition of Korean black raspberry marc and/or blueberry marc. The number of oviposition was also tend to increase by adding additional feed to sawdust but there was no significant different.
