Assessment of microbial quality in household water tanks in Dubai, United Arab Emirates

  • Provision of safe, accessible, and good water quality in the community is an important step towards reducing various waterborne illnesses. However, improving the quality of water should include spreading awareness to the public regarding the importance of cleaning their household water tanks. The aim of this study was to investigate the microbial quality of water of household water tanks in Dubai. The water samples from household water tanks were collected from forty houses, and a questionnaire was given to the residents to determine the history of the water tanks. The membrane filtration technique was used to quantify heterotrophic and total coliform bacteria on plate count agar and the violet red bile agar respectively. The overall results of this study have shown that 18 out of total 40 household water tanks contained different types of bacteria concentration level beyond local and widely accepted international standards. The overall results of this study indicated that there is a lack of awareness among residents regarding the importance of maintaining proper sanitation and hygiene of the household water tanks.

Coliform Regrowth and Injured Coliforms in Public Water Supply (수돗물에서의 대장균군 재생장과 손상 대장균군)

  • Densities of total and injured coliforms in treated water( TW ) and three tap water salt pies( Taps Kl, K2 and K3) of Ku- eui water treatment plant and one tap water sample( Tap T) of Tuk- do water treatment plant were measured 23 times from 1991 to 1992. Coliform regrowth in the water distribution system occurred three times during the study period. When the regrowth episode occurred, injured coliforms were always but total coliforms were not always detected in treated water. Mean densities of total coliforms in TW, and Taps Kl, K2, K3 and T were 0.8, 2.3, 1.9, 1.4 and 2.1 cfu/100mℓ by membrane filtration method using m- TF agar and those of injured coliforms were 3.4, 2.8, 2.5, 2.7 and 2.9 c1u/100mℓ using mondo- LES agar. The injury rates of TW and Taps Kl, K2, K3 and T were 89.5,77.4,67.9, 82.8 and 75.9%, respectively. The high injury rate of coliform bacteria in Seoul water supply can cause regrowth problems in distribution systems due to the repair of injured cells under an appropriate condition. It is recommended the injured coliforms should be measured in drinking water since they can lead to a significant underestimation of total coliforms and result in an inaccurate evaluation of the potential health risks.

Control of Soybean Sprout Rot Caused by Pythium deliense in Recirculated Production System

  • A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.

Mycological Characteristics of Phytophthora nicotianae var. nicotianae Causing Phytophthora Rot of Strawberry and Resistance of Strawberry Cultivars to the Pathogen (딸기 역병균 Phytophthora nicotianae var. nicotianae의 균학적 특성 및 딸기 품종간 저항성)

  • Mycological characteristics of Phytophthora nicotianae var. nicotianae SPC10 (A1 type) causing Phytophthora rot of strawberry and the resistances of 11 strawberry cultivars against the pathogen were examined. Optimum temperature for the mycelial growth of the pathogen was obtained in the range of 30~35$^{\circ}C$, and the growth was completely stopped under 13$^{\circ}C$ or over 42$^{\circ}C$. Aerial mycelia were abundant on oatmeal agar (OMA), V-8 juice agar (V8A) and lima bean agar (LBA) medium, although there were slight differences, however, on cornmeal agar (CMA) medium, it was a shape of stellate without aerial mycelia. The colony shape on potato dextrose agar (PDA) medium was rough and irregular whereas the mycelial growth was slow, and some aerial mycelia were only produced in the middle of PDA medium. Optimum temperature for sporangial formation was 3$0^{\circ}C$, and zoospores were mostly released at $25^{\circ}C$ from the sporangia. Sporangia were more produced in C/Z solution with pH 5. 0~6.$0^{\circ}C$ than sterilized distilled water (DSW) and distilled water (DW), and zoospores were also released much more than other solutions. Eleven strawberry cultivars such as Reiko, Hokowase, Eyeberry, Akaneko, Sistakara, Toyonoka, Nyoho, Sulhong, Suhong, Myhong and Wonkyo #3104 revealed the disease incidence up to 88.9~100% by the leaf inoculation with mycelial disk. However, Nyoho and Suhong showed higher level of resistance against the pathogen by root inoculation.

  • Bloom of small centric diatom Stephanodiscus is quite occasional in winter season in temperate freshwater ecosystems. It often leads to degradation of water quality and affects the quality of supplied drinking water. In a previous study, we have found that naphthoquinone (NQ) 4-6 derivate is an effective tool for efficient mitigation of natural S. hantzschii blooms. In the present research, polylactide (PLA) and agar foam were used as immobilized agent for NQ 4-6 to improve the efficiency of NQ 4-6 compound releasing process for its application under various field conditions. Mesocosm experiments at 10 ton scale suggested that the abundance of S. hantzschii was continuously increased in the control and upon treatment of the mesocosm with immobilized NQ 4-6 from PLA and agar foam. Their algicidal activities were 78.8% and 77.1%, respectively, on S. hantzschii after 10 days. In the mesocosm experiments, the dynamics of biotic (bacteria, HNFs, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. They exhibited similar trends in the control and treatment groups. Therefore, the immobilized NQ 4-6 from PLA and agar foam has potential to be used as an alternative algicidal substance to effectively mitigate natural S. hantzschii blooms under various field conditions. In addition, it not only can be used to control S. hantzschii, but also is an effective technique. The immobilized NQ 4-6 showed stable controlled release in desired system.

Effect of Acid Treatment on Extractability and Properties of Agar (산처리(酸處理) 조건(條件)이 한천(寒天)의 수율(收率) 및 성질(性質)에 미치는 영향(影響))

  • Agar was prepared from domestic agarophyte (Gelidium amansii) by a process of acid extraction. Optimal conditions in the acid treatment of the seaweed and the gelling properties of the agar thus prepared were investigated. The yield and gel strength of the control (the agar extracted with distilled water) showed 13,3% and 306g/$cm^2$, respectively. The yield of 38.7% was obtained when agar was extracted with 0.007N Hydorchloric acid while the gel strength of agar,511g/$cm^2$ was observed with the agar extracted with 0.005N Hydorchloric acid solution. The agar extracted with hydrochloric acid solution showed excellent properties of agar at the acid concentration range 0.005N-0.01N The cooking time of 60 min. was found to be effective to the yield and also to the gelling property of the agar and the cooking longer than 60 min. was proved to be destructive to agar. In general, yield and gelling properties of the agar obtained showed a close relation to the acid concentration and cooking time, but no consistent influence on the contents of ash and sulfur trioxide.

  • Agar for microbiological medium was prepared with pilot-scale for industrial application by the process of microfiltration ($0.4 {\mu}m\;pore size$) in $40{\~}50{\circ}C$, washing with sot water, and treatment with $0.25\;N NaOH\;at\;70{\circ}C$. Transparency, gel strength, viscosity, sulfate content, and syneresis ratio of agar prepared from Gelidium amansii was compared with commercial agar for microbiological medium. Gel strength and transparency were increased with processing, however, it's viscosity, sulfate content, and syneresis ratio were reduced. The final agar product was superior to commercial agar for microbiological medium.

  • Conidia and cultural characteristics of isolates of chromogenic and teleomorphic strains of Colletotrichum gloeosporioides from apple and red pepper were compared. The mycelial growth of teleomorphic strains was faster than that of chromogenic strains in potato dextrose agar and V-8 agar. The chromogenic isolates from apple and red pepper developed. white gray to gray green mycelial rings interspersed with salmon to apricot colored conidial masses in colonies on potato dextrose agar and V-8 agar and none formed on ascigerous stage in cultures. The chromogenic isolates from red pepper produced conidia, most with one apex attenuated on apple and potato dextrose agar whereas fusiform and smaller conidia were produced in V-8 agar and water agar leaf medium. The chromogenic isolates from apple produced fusiform conidia in the media tested. The teleomorphic isolates from apple and red pepper produced cylindrical conidia, most with both apices rounded, developed white gray to dark olive green in a zonate pattern with small dark spots throughout colonies and formed the ascigerous stage in cultures.

Distribution and Characterization of Heterotrophic Bacteria Isolated from Deodorizing Biofilters of a Nightsoil Treatment Plant (분뇨처리장의 질소성 악취물질처리를 위한 생물탈취탑내 종속영양세균의 분포 및 특성)

  • The distributions and characteristics of bacteria on the deodorizing biofilters were studied as a part of biofiltration research for odor gas removal. The odor gas originated from a currently operating nightsoil treatment plant were mainly nitrogenous compounds of ammonia and amines. The filter media were consisted various ratio of pine bark, nightsoil cast and compost, and were sampled before and after operation of 153 days. Before operation, the level of bacteria on nitrification agar medium, nutrient agar medium and thiosulfate agar medium were around $10^7{\sim}10^8cfu/g$ media. After operation, the level decreased but still high as $10^5{\sim}10^7cfu/g$ media. The bacteria from reactors before and after operations were either rod or coccus after growth on nitrification agar media. The 45 isolated heterotrophic bacteria were further characterized and Pseudomonas, Bacillus, Flavobacterium, Alcaligenes were dominant genera. Genus Alcaligenes was particularly dominant when the isolates were incubated on nitrification agar media first and then transfered to nutrient media. The portion of Alcaligenes in the biofilter increased after operation, and was higher in the media contained nightsoil cast, which showed better treatment than the media with pine bark only.

UV Barrier and Antimicrobial Activity of Agar-based Composite Films Incorporated with ZnO Nanoparticles and Grapefruit Seeds Extract (ZnO 나노입자와 자몽씨추출물을 첨가한 아가복합필름의 자외선차단 및 항균특성)

  • Agar-based nanocomposite films were prepared by incorporation of zinc oxide nanoparticles (ZnONP) and grapefruit seed extract (GSE). The composite films were characterized using FT-IR, UV-visible spectroscopy and thermalgravimetric analysis (TGA). The composite films showed light absorption peaks at 220 and 380 nm, characteristic for GSE and ZnONP, respectively. The UV-light transmittance of the agar film was markedly reduced from 54.4 ± 1.3% to 5.8 ± 2.5% with little sacrifice of transparency when 3 wt% ZnONP and 5 wt% GSE were added. The mechanical and water vapor barrier properties increased slightly though they were not significant statistically by the addition of ZnONP and GSE. The nanocomposite films showed stronger antibacterial activity against L. monocytogenes than E. coli O157: H7 and the antibacterial activity was affected by bacterial types as well as concentrations of ZnONP and GSE. The nano-composite film incorporated with 3 wt% of ZnONP and 5 wt% of GSE exhibited strong antibacterial activity against Listeria monocytogenes and E. coli O157: H7. The results indicate that 3 wt% of ZnONP and 5 wt% of GSE are the optimal concentrations for producing functional agar/ZnONP/GSE composite films.
