Simultaneous Determination of Eight Sugar Alcohols in Foodstuffs by High Performance Liquid Chromatography (HPLC를 이용한 식품 중 당알코올 8종 동시분석)

  • A method was established for the simultaneous determination of sugar alcohols, erythritol, xylitol, sorbitol, inositol, mannitol, maltitiol, lactitol and isomalt by High Performance Liquid Chromatography (HPLC). The sugar alcohols were converted into strong ultraviolet (UV)-absorbing derivatives with p-nitrobenzoyl chloride (PNBC). HPLC was performed on Imtakt Unison US-$C_18$ column, using acetonitrile: water (77:23) as a mobile phase and UV detection (260 nm). The calibration curves for all sugar alcohols tested were linear in the 10~200 mg/L range. The average recoveries of the sugar alcohols from three confectioneries spiked at 100 ppm of eight sugar alcohol standards ranged from 81.2 to 123.1% with relative standard deviations ranging fromo 0.2 to 4.9%. The limits of detection (LODs) were $0.5{\sim}8\;{\mu}g/L$ and the limits of quantification (LOQs) were $2{\sim}17\;{\mu}g/L$. Reproducibility of 8 sugar alcohols was 0.28~1.97 %RSD. The results of the analysis of confectioneries showed that 89 samples of 130 were detected and the sugar alcohols content of samples investigated varied between 0.4 and 693.7 g/kg. A method for the simultaneous determination of eight sugar alcohols will be used as basic data for control of sugar alcohols in confectioneries, and quality control in food manufacturing.

Comparative Analysis of Absorption Doses between Exposed and Unexposed Area on Major Organs During CT Scan (전산화 단층촬영시 주선속내 외의 주요장기 흡수선량 비교분석)

  • It is possible to obtain a fast CT scan during breath holding with spiral technique. But the risk of radiation is increased due to detailed and repeated scans. However, the limitation of X-ray doses is not fully specified on CT, yet. Therefore, the purpose of the present study is to define the limitation of X-ray doses on CT The CT unit was somatom plus 4. Alderson Rando phantom, Solenoid water phantom, TLD, and reader were used. For determining adequate position and size of organs, the measurement of distance(${\pm}$2mm) from the midline of vertebral body was performed in 40 women(20~40 years). On the brain scan for 8:8(8mm slice thickness, 8mm/sec movement velocity of the table) and 10:10(10mm slice thickness, 10mm/sec movement velocity of the table) methods, the absorption doses of exposed area of the 10:10 were slightly higher than those of 8:8. The doses of unexposed uterus were negligible on the brain scan for both 8:8 and 10:10. On the chest scan for 8:8, 8:10(8mm slice thickness, 10mm/sec movement velocity of the table), 10:10, 10:12(10mm slice thickness, 12mm/sec movement velocity of the table) and 10:15(10mm slice thickness, 15mm/sec movement velocity of the table) methods, 8:8 method of the absorption doses of exposure area was the most highest and 10:15 method was the most lowest. The absorption doses of 8:10 method was relatively lower than those of the other methods. In conclusion, the 8:10 method is the most suitable to give a low radiation burden to patient without distorting image quality.

A Study on the Efficient Measurement of Airborne Asbestos Concentrations at Demolition Sites of Asbestos Containing Buildings, etc. in Seoul (서울시내 석면함유 건축물 철거 현장 등에서의 효과적인 공기 중 석면농도 측정을 위한 연구)

  • Objectives: This study is intended to seek credible and efficient measurements on airborne asbestos concentrations that allow immediate action by establishing complementary data through comparative analysis with existing PCM and KF-100 method real-time monitoring equipment in working areas in Seoul where asbestos-containing buildings are being demolished, including living environment surroundings. Materials: We measured airborne asbestos concentrations using PCM and KF-100 at research institutes, monitoring networks, subway stations and demolition sites of asbestos-containing buildings. Through this measurement data and KF-100 performance testing, we drew a conversion factor and applied it via KF-100. Finally we verified the relationship between PCM and KF-100 with statistical methods. Results: The airborne asbestos concentrations by PCM for the objects of study were less than the detection limit(7 fiber/$mm^2$) in three (20%) out of 15 samples. The highest concentration was 0.009 f/cc. The airborne asbestos concentrations by PCM in laboratories, monitoring networks, subway stations and demolition sites of asbestos-containing buildings were respectively $0.002{\pm}0.000$ f/cc, $0.004{\pm}0.001$ f/cc, $0.009{\pm}0.001$ f/cc, and $0.002{\pm}0.000$ f/cc. As a result of KF-100 performance testson rooftops, the conversion factor was 0.1958. Applying the conversion factor to KF-100 for laboratories, the airborne asbestos concentrations ratio of the two ways was nearly 1:1.5($R^2$=0.8852). Also,the airborne asbestos concentration ratio of the two ways was nearly 1:1($R^2$=0.9071) for monitoring networks, subway stations, and demolition sites of asbestos-containing buildings. As a result of independent sample t-tests, there was no distinction between airborne asbestos concentrations monitored in the two ways. Conclusions: In working areas where asbestos-containing buildings are being demolished, including living environment surroundings, quickly and accurately monitoring airborne asbestos scattered in the air around the working area is highly important. For this, we believea mutual interface of existing PCM and a real-time monitoring equipment method is possible.

  • Germination is a well-known economical technique that has been utilized to enhance the nutritional value of brown rice. Owing to its higher nutritive quality, germinated brown rice has received significant attention in the past decade. In this study, the physicochemical and cooking properties of specialty brown rice (SBR) were analyzed before and after germination. Germination enhanced cooking properties such as water absorption, expanded volume, and increased solid solubility of cooked SBR. The SBR texture measured using tensipresser, was significantly improved by germination. The hardness of cooked SBR was decreased by germination, but stickiness was increased. Pasting analysis of the SBR flours revealed a decrease in all viscosity values (peak viscosity, breakdown, setback, and final viscosity) after germination. However, the gelatinization temperature remains unchanged upon germination. Additionally, amylose content and amylopectin chain length distribution of SBR starch were slightly changed by germination. These results indicate that germination leads to a substantial improvement in the cooking properties and texture of SBR.

Pollutant Contents with Particle Size Distribution in Bridge Road Drainage Sediment (교량도로 배수받이 퇴적물질의 입경별 오염물질 함량)

  • The purpose of this study is to present the basic data for nonpoint pollutant loads from bridge road drainage sediments using the results to analyze organic matter and heavy metals from the four bridge drainage sampling sites with sediments of different particle size ranges. The sediment sample was collected from the bridge road drainage and the masses of nine sediments fractions were obtained after drying the separated sediment in an over at $85^{\circ}C:>2,000{\mu}m$, $1,000\sim2,000{\mu}m$, $850\sim1,000{\mu}m$, $425\sim850{\mu}m$, $212\sim425{\mu}m$, $125\sim212{\mu}m$, $90\sim125{\mu}m$, $75\sim90{\mu}m$, $<75{\mu}m$. The sediment extract was analyzed water quality constituents, including chemical oxygen demand(COD), total nitrogen(T-N), total phosphorus(T-P), heavy metals and particle size distribution. The results indicate that most of particle size ranges of the bridge road sediments was $125\sim425{\mu}m$, and portion of $<75{\mu}m$ was low. But most of the pollutants are associated with the finer fractions of the load sediments. As the results of analysis, the range and average values of COD, T-N, T-P, Fe, Cu, Cr, and Pb were $177\sim198.8$ mg/kg(77.6 mg/kg), $23\sim200$ mg/kg(83 mg/kg), T-P $18\sim215$ mg/kg(129 mg/kg), and $1,508\sim5,612$ mg/kg(3,835 mg/kg), $9.2\sim69.3$ mg/kg(49 mg/kg), $19.1\sim662.2$ mg/kg(214 mg/kg), and $28.4\sim251.4$ mg/kg(114 mg/kg), respectively. The relationship between sediment size and pollutants concentration have an inverse proportion. The removal of road sediments with frequently could be reduced the significant nonpoint pollutant load, because of the bridge road sediment contains considerable micro-particles and heavy metals.

Soil Loss and Pollutant Load Estimation in Sacheon River Watershed using a Geographic Information System (GIS를 이용한 동해안 하천유역의 토양유실량과 오염부하량 평가 -사천천을 중심으로-)

  • Through the integration of USLE and GIS, the methodology to estimate the soil loss was developed, and applicated to the Sacheon river in Gangrung. Using GIS, spatial analysis such as watershed boundary determination, flow routing. slope steepness calculation was done. Spatial information from the GIS application was given for each grid. With soil and land use map, information about soil classification and land use was given for each grid too. Based upon these data, thematic maps about the factors of USLE were made. We estimated the soil loss by overlaying the thematic maps. In this manner, we can assess the degree of soil loss for each grid using GIS. Annual average soil loss of Sacheon river watershed is 1.36 ton/ha/yr. Soil loss in forest, dry field, and paddy field is 0.15 ton/ha/yr, 27.04 ton/ha/yr, 0.78 ton/ha/yr respectively. The area of dry field, which is 4% of total area, is $2.4km^2$. But total soil loss of dry field is 6561 ton/yr, and it occupies 84.9 % of total soil loss eroded in Sacheon river watershed. Comparing with the 11.2 ton/ha/yr of an average soil loss tolerance for cropland, provision for the soil loss in dry field is necessary. Run-off and water quality of Sacheon river were measured two times in flood season: from July 24, 1998 to July 28 and from September 29 to October 1. As the run-off of the river increased, SS, TN, TP concentrations and pollutant loadings increased. SS, TN, TP loads of Sacheon river discharged during the 2 heavy rains were 21%, 39%, and 19% of the total pollutant loadings generated in the Sacheon river watershed for one year. We can see that much pollutants are discharged in short period of flood season.

Relationship between a Dense Bloom of Cyanobacterium Anabaena spp. and Rainfalls in the North Han River System of South Korea (북한강 수계의 남조 Anabaena 대발생과 강우의 관계)

  • To evaluate the relationship between dynamics of Cyanobacterial bloom and rainfalls, a monthly monitoring of water quality and phytoplankton from the three serial lakes (Lake Ui-am, Lake Chung-pyeong and Lake Pal-dang) in the North Han River System were examined 12 times from May 2012 to March 2013. A dense bloom of cyanobacterium Anabaena spp., was occurred over three lakes in the summer season of 2012. In Lake Ui-am, the Anabaena population appeared in June, showed a peak in July (43,850 cells $mL^{-1}$) and disappeared in November 2012. In Lake Chung-pyeong and Lake Pal-dang, Anabaena population commonly appeared in July, showed the peaks (31,648 cells $mL^{-1}$ and 7,136 cells $mL^{-1}$, respectively) in August, and entirely disappeared in September 2012. Over the three lakes, the phytoplankton community was commonly dominated by diatoms before Monsoon, cyanobacteria during Monsoon, and diatoms after Monsoon, respectively, indicating a Monsoon-dependent succession. A correlation analysis revealed that dynamics of Anabaena population was strongly related with rainfall (r=0.72, r=0.83, r=0.88, P<0.01 for three lakes), and partly with nutrients, inflow and outflow of lakes. Therefore, this study indicates that the outbreak and destruction of Anabaena bloom in North Han River System between 2012 and 2013 was impacted by rainfalls. However, a high density of cyanobacteria in Lake Ui-am remained after Monsoon, and thus, may paroduce bad-order and toxins from phytoplankton.

Sensory Quality Assessment of Reheated Cook/Chill and Sous-Vide Spinach Soup for Foodservice Operations (단체급식소에 적용하기 위한 cook/chill 및 sous-vide 시금치국의 재가열 방법에 따른 관능적 품질평가)

  • The purpose of this study was to compare the effect of several reheating treatments (immersing the package into hot water, heating the soup in the pot and convection ovens after taking out of the opened package. heating the soup package in microwave) on the sensory characteristics of cook-chill and sous-vide spinach soup. Sensory evaluations were made on 5 sensory attributes by a 12-member panel using quantitative descriptive analysis(QDA). The fresh cooked spinach soup had a significantly(p<0.01) higher score in color than the reheated sous-vide one. The latter, however, had a significantly(p<0.05) higher score in taste than the former. Overall acceptability of the sous-vide spinach soup was not different from that of fresh cooked one, which supports the potential of the product in foodservice operation for the efficient management of meal plan. Reheating treatments didn't show any differences in taste, flavor, color, texture, appearance and overall acceptability. Therefore, any reheating treatment nay be used for cook-chill and sous-vide spinach soup.

Characteristics of Ginseng Extract and Its Effects on Sensory Properties of Pork Sausage (인삼 추출물의 사포닌 조성 특성과 추출물 첨가가 포크소시지 관능 특성에 미치는 영향)

  • This study was conducted to evaluate extraction properties of crude saponin and ginsenosides, and their effects on sensory properties of emulsified pork sausage. Non-dried ginseng root was boiled in 0 (e.g., 100% distilled water), 20, 40, 60, 80 or 100% ethanol, and powdered by a freezing dry method. Weight of dried powder for the 0% ethanol extraction was 20% of initial non-dried ginseng weight, while $20{\sim}80%$ and 100% ethanol extractions resulted in approximately 15 and 10% of their initial weights, respectively. On the other hand, crude saponin content in the dried powder was linearly increased for a higher ethanol content where 100% ethanol extraction resulted in 123.52 mg/g. LC/MS analysis of crude saponin for quantifying ginsenosides showed that Rb1, Rb2 and Rc were significantly (p<0.05) higher levels for both 80 and 100% ethanol extractions. In the case of Rg1 ginsenoside, 60, 80 and 100% ethanol extractions resulted in significantly (p<0.05) higher levels. Emulsified pork sausages containing 0, 1 or 2% ginseng extracts were smoked or non-smoked and their sensory characteristics and preference were evaluated. Smoking process significantly (p<0.05) decreased juiciness and tenderness, but the treatment significantly (p<0.05) improved flavor and consumer preference. It was particularly noticed that a 2% addition of ginseng extract prevented the adverse effects of smoking process on juiciness and tenderness while the 2% addition significantly (p<0.05) improved consumer preference. The current results implied that addition of ginseng extract in emulsified pork sausage could improve sensory quality.

Analysis of Enhancement Effect and Attachment Ability of Beneficial Intestinal Microflora in Puffed Grain Foods Using Confocal Laser Scanning Microscopy (곡물 소재 팽화식품에서 장내 유익균의 증진 효과 분석 및 공초점 현미경을 이용한 부착능 평가)

  • This study examined the adhesiveness of beneficial intestinal bacteria to whole-grains using confocal scanning laser microscopy (CLSM), to demonstrate the prebiotic effects of whole-grains, and to develop prebiotic puffed snacks with these whole-grains. CLSM has been used to observe the adhesiveness of Lactobacillus acidophilus, which belongs to beneficial intestinal bacteria, to whole-grain powders using optical sectioning techniques. The enhanced effects on the growth of beneficial intestinal bacteria with the hot water grain extract were verified using an indirect count method. Finally, a puffed snack was produced with the prebiotic effect and the quality was evaluated by checking the chromaticity and degree of hardness. As a result, L. acidophilus exhibited adhesive ability to whole-grain powders and growth of selected beneficial intestinal bacteria were improved significantly. The Hunter L value of the developed puffed snack increased when seasoning was added. The hardness of the puffed snack with seasoning was higher than that of the control. The results of a sensory evaluation showed that the puffed snack with seasoning was highly rated in the overall preference compared to the control.
