인진호(茵蔯蒿)와 한인진(韓茵蔯)의 추출용매별 항염증 효능 비교 (Comparison of Anti-inflammatory effects between Artemisia capillaris and Artemisia iwayomogi by extraction solvents)

  • Objectives : Artemisia capillaris Thunberg (AC) and Artemisia iwayomogi Kitamura (AI) have been used without distinguishment since ancient times due to similar appearance. In this study, we compared the inhibitory effects of AC and AI on the expression of inflammatory cytokines induced by lipopolysaccharide (LPS) in murine macrophages. Methods : AC and AI were extracted by reflux with distilled water (DW) and 70% ethanol (EtOH). We investigated the inhibitory effects of AC and AI on the expression of nitric oxide (NO), inducible NO synthase (iNOS) and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) induced by LPS in macrophages. Results : Firstly, yield of the samples was higher in order of Artemisia iwayomogi DW Extract (AID), Artemisia iwayomogi 70% EtOH Extract (AIE), Artemisia capillaris DW Extract (ACD) and Artemisia capillaris 70% EtOH Extract (ACE). All of the samples were not toxic in macrophages. The inhibitory effect of the samples on LPS-induced NO expression was stronger in the order of AIE, ACE, AID and ACD. The inhibitory effect of the samples on LPS-induced inducible iNOS expression was stronger in the order of AIE, ACE and AID. Effect of ACD was same with that of AID. In addition, inhibitory effect of the samples on LPS induced $TNF-{\alpha}$expression wes stronger in the order of AIE, ACE, AID and ACD. Conclusion: These results showed that AI would be more effective than AC and 70% EtOH would be more effective than DW as an extraction solvent in inflammatory diseases.

Effect of hyperthermia on cell viability, amino acid transfer, and milk protein synthesis in bovine mammary epithelial cells

  • The reduction of milk yield caused by heat stress in summer is the main condition restricting the economic benefits of dairy farms. To examine the impact of hyperthermia on bovine mammary epithelial (MAC-T) cells, we incubated the MAC-T cells at thermal-neutral (37℃, CON group) and hyperthermic (42℃, HS group) temperatures for 6 h. Subsequently, the cell viability and apoptotic rate of MAC-T cells, apoptosis-related genes expression, casein and amino acid transporter genes, and the expression of the apoptosis-related proteins were examined. Compared with the CON group, hyperthermia significantly decreased the cell viability (p < 0.05) and elevated the apoptotic rate (p < 0.05) of MAC-T cells. Moreover, the expression of heat shock protein (HSP)70, HSP90B1, Bcl-2-associated X protein (BAX), Caspase-9, and Caspase-3 genes was upregulated (p < 0.05). The expression of HSP70 and BAX (pro-apoptotic) proteins was upregulated (p < 0.05) while that of B-cell lymphoma (BCL)2 (antiapoptotic) protein was downregulated (p < 0.05) by hyperthermia. Decreased mRNA expression of mechanistic target of rapamycin (mTOR) signaling pathway-related genes, amino acid transporter genes (SLC7A5, SLC38A3, SLC38A2, and SLC38A9), and casein genes (CSNS1, CSN2, and CSN3) was found in the heat stress (HS) group (p < 0.05) in contrast with the CON group. These findings illustrated that hyperthermia promoted cell apoptosis and reduced the transport of amino acids into cells, which inhibited the milk proteins synthesis in MAC-T cells.

Effects of Pogonatherum paniceum (Lamk) Hack extract on anti-mitochondrial DNA mediated inflammation by attenuating Tlr9 expression in LPS-induced macrophages

  • BACKGROUND/OBJECTIVES: Mitochondrial DNA leakage leads to inflammatory responses via endosome activation. This study aims to evaluate whether the perennial grass water extract (Pogonatherum panicum) ameliorate mitochondrial DNA (mtDNA) leakage. MATERIALS/METHODS: The major bioactive constituents of P. paniceum (PPW) were investigated by high-performance liquid chromatography, after which their antioxidant activities were assessed. In addition, RAW 264.7 macrophages were stimulated with lipopolysaccharide, resulting in mitochondrial damage. Quantitative polymerase chain reaction and enzyme-linked immunosorbent assay were used to examine the gene expression and cytokines. RESULTS: Our results showed that PPW extract-treated activated cells significantly decrease reactive oxygen species and nitric oxide levels by reducing the p2phox and iNOS expression and lowering cytokine-encoding genes, including IL-6, TNF-α, IL-1β, PG-E2 and IFN-γ relative to the lipopolysaccharide (LPS)-activated macrophages. Furthermore, we observed that LPS enhanced the mtDNA leaked into the cytoplasm, increasing the transcription of Tlr9 and signaling both MyD88/Irf7-dependent interferon and MyD88/NF-κb p65-dependent inflammatory cytokine mRNA expression but which was alleviated in the presence of PPW extract. CONCLUSIONS: Our data show that PPW extract has antioxidant and anti-inflammatory activities by facilitating mtDNA leakage and lowering the Tlr9 expression and signaling activation.

  • The theoretical treatments on jets, in which the flow is issuing into a stagnant medium, have been based on Prandtl's mixing theory. In this study, using Prandtl's mixing length hypothesis, a theoretical relationship for the velocity profile of a single round jet is derived. Furthermore, Gaussian expression is used to approximate the theoretical relationship, in which the Gaussian coefficient is assumed to be decreasing exponentially as the flow goes far from the orifice. Two data sets for a single round jet performed by tow different techniques of measurement are used to verify the suggested relationships. The theoretical and Gaussian distribution give close results in spite of the difference in approach. The observed mean velocity distributions are in good agreements with the suggested theoretical and Gaussian distributions.

  • A finite dam under $P_{\lambda,;,T}^M-policy$ is considered, where the input of water is formed by a Wiener process subject to random jumps arriving according to a Poisson process. Explicit expression is deduced for the stationary distribution of the level of water. And the long-run average cost per unit time is obtained after assigning costs to the changes of release rate, a reward to each unit of output, and a penalty which is a function of the level of water in the reservoir.

  • 접지 시설의 효율적인 시설 계획을 위해서는 시공 전에 접지전극에 전류가 유입할 때 접지 전극주변에 발생하는 전위분포 및 전류에 의한 접지저항을 추정하는 것이 접지 설계에 있어서 매우 중요하다. 본 논문에서는 사각판상의 접지전극을 대지에 직접 시공 매설하여 지표면 전위의 분석으로 접지저항을 측정하고, 또한 사각판상 접지 저항 값의 이론적 계산식에서 구한 접지저항값과 축소된 사각판상의 접지전극 모델을 수조에 넣고 모의 실험을 통해 측정된 접지저항값을 비교·분석하였다.

Neurotrophic and Neuritogenic Effects of Water Extracts of Rhizoma of Coptis chinensis Franch in PC12 Cells

  • Coptis chinensis (CC) is one of the traditional herbs used in Oriental medicine for the treatment of gastrointestinal disorders, anxiety, and insomnia. In this study, neurotrophic and neuritogenic effects of CC on rat pheochromocytoma (PC12) cells were evaluated. Pretreatment of PC12 cells with water extracts of CC $(120{\mu}g/ml)$ produced considerable outgrowth of neurites that is comparable to the effect of nerve growth factor (NGF). Therefore, neurite outgrowth was quantified and expression of NGF mRNA was examined. Furthermore, characteristics of neurites were immunocytochemically confirmed using axon and dendrite-specific antibodies. These results suggest that water extracts of CC contain components that have neurotrophic and neuritogenic properties.

  • All human being can't exist by oneself. That is, all human being depend on intimate attachment for their survival. Physical and psychological powers and control functions are prerequisites for one's survival and these serve important functions in interpersonal relationships. There are two kinds of powers and controls. One is positive power which helps one survive effectively. Another one is negative power which disturbs one's effective survival. Introspection is one of the positive power and most important process in understanding oneself and others. Thus, the purpose of this study is to elucidate the physical and psychological meaning of the characteristics of water and based on this meaning, to suggest one way of introspection. The authors reviewed the precept of water which is described in Bible and oriental classical books such as 'the Analects of Confuicus(論語)', 'Tao Te Ching(道德經)', 'Mencius(맹자)', 'Chronicle(史記)', 'The Doctrine of the Mean(中庸)', 'University(大學)'. The author also reviewed 'The History of Three Countries(삼국사기, 삼국유사)', 'The Precept of Religion', and 'the modern poems' which includes the symbolic meaning of water. The results are summarized as follows:1) The flow of water itself is the expression of introspection. Because, water does not flow without filling a puddle. 2) Water is the symbol of 'Immortality'. 3) Water symbolizes 'Modesty and Trust'. 4) Water is the symbol of 'Freedom from Greed'. 5) Water symbolizes 'Goodness and Beauty'. 6) Water symbolizes 'Naturality and Love'. Immortality, Modesty and Trust, Freedom from Greed, Goodness and Beauty, and Naturality and Love are eternal, universal values which all human should seek and achieve. By the process of introspection through the symbolic meaning of water, we can achieve these positive aspects of human mind and spirit.

Inhibition of Bacillus cereus Growth and Toxin Production by Bacillus amyloliquefaciens RD7-7 in Fermented Soybean Products

  • Bacillus cereus is a gram-positive, rod-shaped, spore-forming bacterium that has been isolated from contaminated fermented soybean food products and from the environment. B. cereus produces diarrheal and emetic toxins and has caused many outbreaks of foodborne diseases. In this study, we investigated whether B. amyloliquefaciens RD7-7, isolated from rice doenjang (Korean fermented soybean paste), a traditional Korean fermented soybean food, shows antimicrobial activity against B. cereus and regulates its toxin gene expression. B. amyloliquefaciens RD7-7 exhibited strong antibacterial activity against B. cereus and inhibited the expression of B. cereus toxin-related genes (groEL, nheA, nheC, and entFM). We also found that addition of water extracts of soybean and buckwheat soksungjang (Korean fermented soybean paste made in a short time) fermented with B. amyloliquefaciens RD7-7 significantly reduced the growth and toxin expression of B. cereus. These results indicate that B. amyloliquefaciens RD7-7 could be used to control B. cereus growth and toxin production in the fermented soybean food industry. Our findings also provide a basis for the development of candidate biological control agents against B. cereus to improve the safety of fermented soybean food products.

Identification and Transcriptional Analysis of Priming Genes in Arabidopsis thaliana Induced by Root Colonization with Pseudomonas chlororaphis O6

  • Root colonization of Arabidopsis thaliana with Pseudomonas chlororaphis O6 induces systemic tolerance against diverse pathogens, as well as drought and salt stresses. In this study, we demonstrated that 11 genes in the leaves were up-regulated, and 5 genes were down-regulated as the result of three- to five-days root colonization by P. chlororaphis O6. The identified priming genes were involved in cell signaling, transcription, protein synthesis, and degradation. In addition, expression of selected priming genes were induced in P. chlororaphis O6-colonized plants subjected to water withholding. Genes encoding defense proteins in signaling pathways regulated by jasmonic acid and ethylene, such as VSP1 and PDF1.2, were additional genes with enhanced expression in the P. chlororaphis O6-colonized plants. This study indicated that the expression of priming genes, as well as genes involved in jasmonic acid- and ethylene-regulated genes may play an important role in the systemic induction of both abiotic and biotic stress due to root colonization by P. chlororaphis O6.
