Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

  • Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.

Effects of Fermented Total Mixed Ration and Cracked Cottonseed on Milk Yield and Milk Composition in Dairy Cows

  • Four lactating Holstein Friesian crossbred cows, with an average initial weight of 450 kg, 48${\pm}$12 days in milk and initial milk yield of 18 kg/h/d, were randomly arranged according to a 2${\times}$2 factorial arrangement in a 4${\times}$4 in Latin square design with 21-d period to investigate the effects of type of total mixed ration (TMR) and type of whole cottonseed (WCS) on intake, digestibility and milk production. The dietary treatments were i) TMR and WCS supplementation at 0.5 kg/h/d, ii) TMR and cracked WCS (cWCS) supplementation at 0.5 kg/h/d, iii) fermented TMR (FTMR) and WCS supplementation at 0.5 kg/h/d, and iv) FTMR and cWCS supplementation at 0.5 kg/h/d. Voluntary feed intake was 15.9, 15.2, 15.4 and 15.6 kg DM/d in dietary treatment 1, 2, 3 and 4, respectively. Digestibility of DM, OM, CP, EE, NDF and ADF were not significantly different among dietary treatments. Ruminal pH, $NH_{3}-N$ and volatile fatty acids in the rumen were also not significantly different among type of TMR or type of WCS. Blood urea-N concentration was not significantly different among dietary treatments. Ruminal bacteria population tended to increase but ruminal protozoa population tended to decrease with supplementation of cWCS, but they were not affected by FTMR. Milk yield and 3.5% FCM were not statistically different among treatments (16.6, 16.2, 17.0, 16.3 kg/d and 18.0, 18.6, 19.9 and 19.0 kg/d, respectively). Milk composition was not significantly different among dietary treatments. However, unsaturated fatty acids in milk fat in cows fed FTMR were lower (p<0.05) than in cows fed TMR. In conclusion, fermentation is a conceivable method to improve the quality of TMR for long-time storage and the cracking method is suitable to release the fat from cottonseed for enhancing fatty acid deposition in milk. Thus, the combination of FTMR and cWCS supplementation would be an alternative strategy to improve performance of lactating cows.

야생동물 이용빈도 및 종다양도를 활용한 생태통로 효율성 평가 (Evaluation of Wildlife Crossings Eeffectiveness on Use Frequency and Diversity Indicators of Wildlife)

  • 본 연구는 백두대간 등 주요 생태축에 위치한 생태통로 49개소를 대상으로 야생동물 일평균 이용빈도 지표와 주변 서식지와 출현종의 수를 비교한 종다양도 지표를 활용하여 생태통로 효율성을 평가하였다. 분석 결과, 전체 생태통로 일평균 이용빈도는 1.4회였으며, 평균을 상회하는 생태통로는 17개로 전체의 34.7%로 확인되었다. 주변 서식종의 수와 출현종의 수가 동일한 종다양도 1.0 이상인 생태통로는 18개로 전체의 36.7%으로 나타났다. 일평균 이용빈도와 종다양도 모두 양호한 생태통로는 10개로 전체의 20.4%로 분석되었다. 효율성이 양호한 생태통로 중 4개소의 생태통로 지침 준수율 등에 대한 현장조사 결과를 검토한 결과, 위치선정·규격·식재 및 토양·유도울타리 등에서 덕산재 육교형 생태통로의 지침 준수율이 가장 양호했다. 본 연구결과는 신규 생태통로 및 기설치된 생태통로의 기능개선 등 생태통로 효율성 연구의 기초자료로 활용될 수 있을것으로 판단된다.

  • The purpose of this study is to improve robot motion control in teaching and operating the expert system/world coordinate system (TOES/WCS) constructed in the previous study. The major contribution of this study is reduction of the inaccuracy in coordinated reading and the movement time of robots in macro motion control. This study also reduces undesirable time of micro motion control by using an unit control (UC) and a micro unit control (MUC) in micro motion control.

  • As a means of human interface, this study designs Developed-ERES/WCS with voice recognition capability and fuzzy set theory. In the advanced teleoperator system, when an error occurs on the automatic mode, the error is recovered after the automatic mode is changed into the manual mode intervened by a human. The purpose of this study is to reduce human work load and to shorten error recovery time during error recovery.

국내 포유동물에 적합한 생태통로의 유형과 규격 (The Suitable Types and Measures of Wildlife Crossing Structures for Mammals of Korea)

  • In this study, suitable types and measures of Wildlife Crossing Structures (WCS) for mammals of Korea were analyzed to suggest basic information for design guidelines of WCS. In order to find the types and measures, we conducted field surveys on 79 WCS and literature reviews on 159 WCS. The results of our study were as follows; Small-medium sized carnivores usually use all measures of wildlife underpass, meanwhile large sized herbivores as water deer need lager measures than 0.7 Openness Index(OI). Furthermore, we found that wildlife overpasses were usually used at the measure of 6.8 m width by every mammal. Therefore, we concluded that the proper measures should be lager than 0.7 OI for underpass and 7 m width for overpass.

Global Sequence Homology Detection Using Word Conservation Probability

  • Protein homology detection is an important issue in comparative genomics. Because of the exponential growth of sequence databases, fast and efficient homology detection tools are urgently needed. Currently, for homology detection, sequence comparison methods using local alignment such as BLAST are generally used as they give a reasonable measure for sequence similarity. However, these methods have drawbacks in offering overall sequence similarity, especially in dealing with eukaryotic genomes that often contain many insertions and duplications on sequences. Also these methods do not provide the explicit models for speciation, thus it is difficult to interpret their similarity measure into homology detection. Here, we present a novel method based on Word Conservation Score (WCS) to address the current limitations of homology detection. Instead of counting each amino acid, we adopted the concept of 'Word' to compare sequences. WCS measures overall sequence similarity by comparing word contents, which is much faster than BLAST comparisons. Furthermore, evolutionary distance between homologous sequences could be measured by WCS. Therefore, we expect that sequence comparison with WCS is useful for the multiple-species-comparisons of large genomes. In the performance comparisons on protein structural classifications, our method showed a considerable improvement over BLAST. Our method found bigger micro-syntenic blocks which consist of orthologs with conserved gene order. By testing on various datasets, we showed that WCS gives faster and better overall similarity measure compared to BLAST.

Bacillus thuringiensis subsp. israelensis를 이용한 느타리 재배에서 버섯파리의 생물학적 방제 (Biological Control of the Mushroom Fly, Lycoriella mali, Using Bacillus thuringiensis subsp. israelensis)

  • 국내 느타리버섯 재배지의 버섯파리 사충으로부터 분리하여 버섯파리 유충에 높은 독성을 나타낸 Bacillus thuringiensis subsp. israelensis Bti-D와 Bti-U 균주를 이용하여 느타리 버섯파리에 대한 생물학적 방제효과를 검정한 결과, 느타리버섯 병재배시는 Bti-D와 Bti-U의 방제가가 각각 74.4%와 64.2%이었으며, 소형상자재배시는 75.8%와 56.8%였다. Bti-D와 Bti-U의 제제화를 위한 대량배양용 염가배지를 개발하기 위하여 배지종류별 세균증식량을 OD를 측정하여 비교한 결과, 2균주 모두 비지배지에서 가장 높았다. 두 균주 중 방제효과가 더 높은 Bti-D 균주를 비지배지에서 대량배양하여 몇 종류의 수화형 미생물 살충제를 만들고 느타리버섯 병재배와 상자재배에서 방제효과를 검정한 결과, 병재배에서 bio-gel로서 옥수수전분을 이용하여 만든 WCS 제제의 방제가가 86.4%로 가장 높았으며, 생균처리의 69.1%보다도 효과적이었다. 또한, 이 WCS 제제를 살포하기 위하여 희석할 때 pH 8로 보정된 물을 사용할 경우에는 방제가가 93.2%로 제고되었다. 상자재배에서는 pH 8로 보정된 물을 사용한 WCS 제제와 화학살충제인 주론수화제의 방제효과와 버섯수량에 미치는 영향을 동시에 비교하였는데, WCS 제제 처리시 방제가는 74.3%로서 주론수화제 처리시 92.2% 보다 낮았다. 그러나 버섯수량 비교에서는 WCS 제제를 처리할 경우 무처리나 주론수화제 처리에 비해 수량이 증가되었다.

Effect of Bifidobacterium Cell Fractions on IL-6 Production in RAW 264.7 Macrophage Cells

  • Bifidobacterium has been previously shown to potentiate immune function, which was mediated through the stimulation of cytokine production by macrophage. This study was performed to further characterize the effective component of Bifidobacterium by measuring the level of interleukin (IL)-6 cytokine using the RAW 264.7 murine cell line as a macrophage model. RAW 264.7 cells were cultured for 24 h in the presence of whole cells (WCs), cell walls (CWs), and cell-free extracts (CFEs) from various strains of Bifidobacterium and other lactic acid bacteria at various concentrations. The most effective component was different depending on the strains and the concentrations used. When tested with each cell fraction from Bifidobacterium sp. BGN4, heat treatment of the cell fractions lowered the production of IL-6. Synergistic effect was obtained, especially when CWs and CFEs were combined. Sonicated WCs stimulated IL-6 production more than intact WCs. The in vitro approaches employed here should be useful in further characterization of the effects of Bifidobacterium on gastrointestinal and systemic immunity.

Study on sintering process of woodceramics from the cashew nutshell waste

  • In this study, the sintering mechanism of woodceramics (WCs) from cashew nut shell waste (CNSW) was studied by analyzing chemical reactions and structural changes during the sintering process of of CNSW powder, liquefied wood and green bodies of WCs at $900^{\circ}C$ for 60 minutes in the $CO_2$ atmosphere. The chemical and structural properties of the products were investigated by X-ray diffraction (XRD), Raman spectroscopy, Fourier Transform Infrared (FTIR), and scanning electron microscope (SEM). The results showed that the decomposition reactions of liquefied wood and CNSW occurred simultaneously to form the hard carbon and the soft carbon at high temperature. The sintering mechanism of WCs has been presented.
