Continuous Hydrolysis of Cod Skin Gelatin in an Ultrafiltration Reactor (한외여과막 반응기를 이용한 어피젤라틴의 연속적 가수분해)

  • A continuous stirred tank membrane reactor(CSTMR ) was developed and optimized for the production of cod skin gelatin hydrolyzates using endo-protease Alcalase. A experimental design methodology was used to optimize the four performance variables: enzyme concentration, substrate concentration, permeate flux and reactor volume. All four variables studied had an effect on substrate conversion, with enzyme and substrate concentrations being predominant. Conversion increased with the increase in enzyme concentration, with the decrease in substrate concentration, at high volumes and low flux. A strong interaction was observed between enzyme and substrate concentrations and smaller interactions between enzyme and flux and substrate and flux. The optimum operating conditions for the CSTMR process for an initial substrate concentration for 10% were $50^{\circ}C$, pH 8, flux 7.3ml/min, residence time 82 min, and Alcalase to substrate ratio 0.02(w/w). A gradual decay in reactor activity during 8 hrs was 2.1% conversion/hr. Enzyme leakage through the 10, 000 MWCO membrane was 16% at $50^{\circ}C$ and 12% at $35^{\circ}C$, 6hrs. However, there was no apparent correlation between enayme leakage and substrate conversion. The Km value for the CSTMR was 20 times higher than the batch reactor. The productivity(expressed as mg product/mg enzyme) of the CSTMR was more than six fold higher than the batch at $50^{\circ}C$. The hydrolyzate was non-bitter.

Effect of Mixed Bed Soil on Growth of Aerial Parts and Roots of Daughter Pants for Nursery Field Srawberry Seedling Raising with Expanded Rice-hull (딸기 팽연왕겨 차근육묘를 위한 혼합상토 조성이 자묘의 지상부 생육과 발근에 미치는 영향)

  • This study was performed to identify the effect of mixed bed soil on growth of aerial parts and root zone of daughter plants for nursery field strawberry seedling raising with expanded chaff. The plant height and leaf area of daughter plants were highest or largest in the mixed soil of ERH +RH (100:0, v/v), followed by ERH+RH (75:25). The higher the mixing ratio of RH, the shorter the plant height or the smaller the leaf area. A similar tendency was observed in fresh weight. Within a root diameter of 0-0.4 mm and a root height range of 0.4-0.8 mm, root surface area and volume were statistically significantly better with treatment of ERH+RH (100:0, v/v) compared to those of roots treated with ERH+RH (75:25), ERH+RH (50:50) and ERH+RH (25:75). The growth rate of aerial parts and root zone of daughter plants were noticeably lower in two mixing ratios of 50:50 and 25:75. According to the mixing ratios of ERH+CD surface treatment, the number of roots was greatest in plants treated with ERH+CD (80:20, v/v) and ERH+CD (85:15) on August 1. However, the number of roots was highest in plants treated with ERH+CD (85:15, v/v) on August 15. Root length was longest in the plant with no treatment, and drastically shortened from ERH+CD (90:10, v/v) in both surface and mixed treatment. Although root weight showed a significant difference in ERH+CD (90:10, v/v) treatment, its increase was gradual. The rate of root growth was highest in ERH+CD (85:15). These study findings suggest that the content ratios of mixed soil ERH+RH (75:25, v/v) or below and ERH+CD (85:15) are thought to be desirable for the production of high quality seedlings.

Effects of Micronization on the In situ and In vitro Digestion of Cereal Grains

  • The effects of micronization on in situ and in vitro nutrient disappearances of wheat, barley and corn were investigated in a series of experiments. In Experiment 1, chemical composition and in situ dry matter disappearance (DMD) of six varieties of wheat were determined. In addition, an in vitro study was completed using ground micronized and unmicronized wheat (var. Kansas). In Experiment 2, three varieties of wheat (Kansas, Sceptre and Laura) and in Experiment 3, three cereal grains (wheat, barley and corn) were either micronized for 1 min to attain internal kernel temperatures of 90-100$^{\circ}C$ or not (controls), and DM, protein and starch disappearances were estimated. In Experiment 2, an in vitro study was also completed using ground micronized and unmicronized wheat (var. Kansas). Wheat samples varied with respect to crude protein (10.0-21.2%), starch (61.6-73.9%), NDF (8.5-11.8%), volume weight (753-842 g/L) and kernel hardness (0.0-32.0). Rate (p = 0.003) and extent (p = 0.001) of in situ DMD differed among wheat varieties. Correlations between in situ kinetics, and chemical and physical properties of wheat varieties showed that protein content was negatively correlated with the rate of disappearance ($r^2$ = -0.77). Micronization of all grains markedly reduced (p = 0.001) the rate and extent of DM, and protein disappearances as compared to control samples. Micronization increased (p<0.05) the digestion of starch in wheat. However, release of ammonia into the incubation medium was markedly reduced (p<0.05), suggesting that micronization increased the resistance of protein to microbial digestion. Disappearances of DM, protein and starch differed (p = 0.001) among cereal grains with wheat>barley>corn. Micronization reduced the rate of DM disappearance (p = 0.011) and slowly degradable protein fractions (p = 0.03), however, increased (p = 0.004) slowly degradable starch fractions of all three cereals. Examination of in situ samples by scanning electron microscopy confirmed that microbial colonization focused on starch granules in micronized grains, and that the protein matrix exhibited resistance to microbial colonization. These results suggest that micronization may be used to increase the ruminal escape value of protein in cereal grains, but may lead to increased starch digestion if grains are finely ground.

  • Lauryl alcohol was used as extracting solvent of ethanol, and its toxicity on the free cells or immobilized cells was tested. To increase ethanol productivity, extractive fermentation method combined with ethanol fermentation and ethanol recovery was applied to the immobilized batch and continuous fermenter. As the concentration of LaOH was increased, the lag phase became longer, but specific growth rate did not change greatly. And a cell entrapment technique could protect the yeast cells against both substrate inhibition and solvent toxicity. When the glucose concentration was 400 g/l and the LaOH/fermentation medium ratio was 4, total ethanol productivity increased with the enhancement of LaOH volume, and maximum productivity was 2.75 g/l.hr in the immobilized batch fermentation.

Analysis on Cost Structure and Management Performance of Poultry Farming in AgroForestry (산지양계복합경영의 비용구조와 경영성과 분석)

  • This study is to analyze differences in gross income between only forestry and agroforestry. In addition, cost structure and management performance on poultry farming was surveyed. The study sites were three forestry households located in Chungju, Kyungsan and Hwacheon. The method of its management performance were conducted by interview. And, survey items of the performance were operating expenses items and gross income items in 2015. As a result of study, gross income of Chungju business model was 25,608,000 won. The operating cost was 20,217,571 won so that the net income was 5,390,429 won. In case of Gyeongsan, gross income was 33,950,000 won and the operating cost was 18,655,714 won. Thus, the net income was 15,494,286 won. Hwacheon business model showed that gross income was 31,850,000 won, the operating cost was 13,143,000 won. Thus, the net income was 18,707,000 won. And, break-even point of sale volume, which meets cost, was 617 chicken in Chungju, 125 chicken in Gyeongsan, and 63 chicken in Hwacheon. In terms of selling cost-per-production ratio, Chungju business model was 79%, Kyungsan was 55% and Hwacheon was 41%. The results of this study indicates that margin ratio per chicken was 21% in Chungju, 45% in Kyungsan and 59% in Hwacheon. On Agroforestry, increasing rate for gross income was evaluated by 16% in Chungju, 18% in Gyeongsan and 11% Hwacheon.

Growth and Physiological Responses of Indeciduous Quercus L. in Container by Fertilizing Treatment (시비 처리에 따른 상록 참나무속 수목의 용기 내 생장 및 생리적 반응)

  • BACKGROUND: This study was carried out in order to closely examine the influence of fertilization upon growth in container of seedling in indeciduous Quercus species (Q. mysinaefolia, Q. acuta and Q. glauca). METHODS AND RESULTS: Fertilizer level was made by adjusting water soluble compound fertilizer (N:P:K=19: 19:19, v/v) to 1000, 2000, $3000mg{\cdot}L^{-1}$ level along with non-fertilizing plot. Fertilization increased height, root collar diameter growth, and dry weight in these three species of trees. The more increase in fertilizer level led to the more rise even in growth of these species. H/D ratio and T/R ratio also showed tendency of getting bigger in the more rise in fertilizer level. Photosynthetic rate was shown to get higher in the higher fertilizer level according to fertilization in all the three species. In the analysis of root morphological traits, the total root length was surveyed to be longer in the more rise in fertilizer concentration. As even a case of root project area, surface area, and root volume is the similar tendency to characteristics in the total root length, a rise depending on fertilization was observed. CONCLUSION: In light of the results in this experiment, the fertilizer level is judged to be $2000mg{\cdot}L^{-1}$ level that is proper for production of 1-year-old container seedling in indeciduous Quercus species with excellent root development and high seedling quality index.

Effects of Irrigation Frequency, Particle Size and Depth of Perlite Medium on Growth and Flowering of Dendranthema grandiflorum Grown on Recycling System (순환식 양액재배에 있어서 관주 주기, Perlite의 입자 크기 및 깊이가 국화 '수방력'의 생육과 개화에 미치는 영향)

  • This study was carried out to investigate the effect of irrigation frequency, particle size and depth of perlite medium on the growth of Dendranthema grandiflorum (Ramat.) Kitamura 'Shuhouno-chikara' grown on recycling system. In Exp 1, the irrigation frequency (IF) was designed as 3 (IF3), 9 (IF9), and 18 (IF18) times a day. Fine and coarse particle size (PS) of perlite was used as a media, and depth of media (DM) was 15 and 10 cm contained in $34{\times}120{\times}15cm$ styrofoam bed. In Exp 2, the IF was 3 (IF3), 6 (IF6), and 9 (IF9) times a day with the same amount of nutrient solution. Fine and coarse PS were used, and DM was 15 and 7.5 cm. In this study, high IF and fine PS was favorable for plant growth. However, as the IF became frequent, difference of plant growth between fine and coarse PS became smaller. Also, shallow media showed little difference with deep media in plant growth. Thus, decreasing the amount of media was recommended to reduce the production cost.

Quality Characteristics of Garaedduk with Lentinus edodes Powder (표고버섯분말을 첨가한 가래떡의 품질 특성)

  • This research focused on investigating the effect of lentinus edodes on the Garaedduk by analyzing its quality characteristics. Through this research, it could be verified whether this improved food has the potential to develop into a functional food. 5 pieces of Garaedduk with 0, 2, 4, 6, and 8% of lentinus edodes powder were analyzed by water content, color, sensory, textural characteristics and cooking characteristics right after the production and after one day. The five Garaedduk showed significant differences in water content during storage period of 0 & 1 day. As the result of Hunter's color values, with the increase of lentinus edode during storage period of 0 & 1 day, lightness was significantly decreased and redness and yellowness were significantly increased. As the result of textural analysis, hardness and chewiness showed significant increase and cohesiveness was significantly decreased in Garaedduk with the increase of lentinus edodes. When the Garaedduk were preserved for one day, the hardness and chewiness of the Garaedduk with 2% of lentinus edodes showed similar with the control. Cohesiveness showed significant decrease in all Garaedduk. The result of the sensory evaluation showed increase in the color, aroma and taste with the increase of lentinus edodes. The 2% Garaedduk showed similar hardness with the control. Garaedduk with lentinus edodes showed no difference in cohesiveness and adhesiveness with the control. The overall acceptability was significantly increased with the increase of lentinus edodes while the Garaedduk with 6% of lentinus edodes showed the highest overall acceptability. After one day of preservation, the Garaedduk with 4% lentinus edodes also showed the highest overall acceptability. As the result of cooking properties, weight gain and volume were significantly increased with the increase of lentinus edodes. The 2% Garaedduk particularly showed similar weight gain with the control. In turbidity of soup was significantly increased with the increase of lentinus edodes. In Hunter's color values of soup, lightness was significantly decreased with the increase of lentinus edodes. Redness and yellowness were significantly increased with the increase of lentinus edodes like Garaedduk.

Development of multiplex PCR-based detection method for five approved LM canola events in Korea (Multiplex PCR 방법을 이용한 국내 승인 5개 LM 유채의 검출법 개발)

  • Canola is a crop globally used for production of oil and biofuel. Cultivation area and import volume of living modified (LM) canola have been increasing every year. As canola import dependence has reached 100% in Korea, efforts have been made for safety management of LM canola and ecological risk assessment. We developed a set of multiplex PCR method for simultaneous detection of 5 LM canola events (Topas 19/2, Rf3, Ms8, RT73 and T45) approved in Korea. The multiplex PCR assay developed allows amplification of estimated products of 5 LM canolas from event specific primer sets. Primer extension time was skipped for a time-consuming process and two annealing steps (20 cycles at $55^{\circ}C$ and 20 cycles at $60^{\circ}C$) were performed for yielding the best result which was sufficient to distinguish five LM canolas. Our results suggest that multiplex PCR method provides a cost and time-effective approach for LM canola detection.

Light and Electron Microscopical Changes of Corpus Luteum during the Course of Pregnancy in Korean Native Cows (한우의 임신경과에 따른 황체조직의 광학 및 전자현미경적 변화)

  • Corpus luteum (CL) is the primary productive organ of progesterone in pregnant cows. Progesterone levels in bovine plasma depend on the volume, weight and shape of the CL. Progesterone productions during the late stages of gestation occur both in the CL and placenta, and placentas producted more progesterone than CL on progesterone prcduction. Because division of progesterone production of these two organs is impoxxible, the CL function can not be determined by plasma progesterone levels following gestation stages. This study was carried out to evaluate histological findings on the CL spurium and CL verum, and also on the CL following the pregnant stages by histological and immunohistochemical and electron microscopical methods and then we expect to assume the functions of CL by histological findings. 1. Proliferations of luteal cells occur by day 120 of gestation, vessel hyperplasia occur by day 90 of gestation, and the walls and lumens of vessels developed by day 120 of pregnancy. 2. Sizes of CL cells increased to maximum around day 200 of gestation and similarly maintained by day 240. So these findings indicated that the function of Cl is most active around day 200 of gestation. 3. On parturation day, the number and size of luteal cells were maintained but stain intensity of the luteal cells and vessels are declined or disappeared, and fibrosis of luteal cells increased, and the vessel lumens are emptied. These findings indicate that CL is inactive. 4. In immunohistochemical findings, proliferative positive cells by PCNA antibody appeared more in number during early stages of gestation but appeared less following course of pregnant stages and not nearly appeared on day 120 of gestation. Apoptotic positive cells by TUNEL methods not nearly appeared on the early pregnant stages and a few appeared at late pregnant stages. So developments of CL proceed until day 120 of gestation and regression of CL was occurred by transform of luteal cells into fibrocytes than by luteal cell apoptosis. 5. In electron microscopical findings, the size of luteal cells increased more in CL verum than in CL spurium. During gestation stages, the size of luteal cells increased, mitochondria in the luteal cell cytoplasms densely and abundantly developed and also swelled mitochondria increased. The interspace of luteal cells are also dilated, transformation of luteal cells into fibrocytes are more number. The lumens and walls of peripheral capillaries of large luteal cells more broadened and thickened, and transformation of large and small luteal cells to fibrocytes are increased. The above findings suggest that function of pregnant CL more developed by day 120 of gestation and are most active around day 200 of gestation and similarly maintained by day 240 and are promptly regressed on paturation day.
