Genetic Relationship of the Ampelopsis brevipedunculata var. heterophylla and Vitis thunbergii var. sinuata with the Other Vitis Plants (개머루와 까마귀머루의 유전적 유연관계 분석)

  • DNA sequences of the intergenic spacer 1 and intergenic spacer 2 of the nineteen plants belonging Vitis genus were collected from the Genbank. DNA sequences of the same regions of Vitis thunbergii var. sinuata and Ampelopsis brevipedunculata var. heterophylla, both common plants in Korea, were not available in Genbank. Those two plants were collected, their genomic DNA encoding 18S rRNA, intergenic spacer 1, 5.8S rRNA, intergenic spacer 2 and part of 28S rRNA amplified and DNA sequence determined. DNA sequences of twenty-one plants including two Korean plants were aligned by the Multiple sequence comparison by log-expectation(MUSCLE) algorithm and the alignment was used to calculate neighbor-joining tree and pairwise distance. The results indicate DNA sequences of the two Korean plants are highly homologous with each other, but they are quite distantly related to the other Vitis plants. Distant relationship of the two Korean plants with the other Vitis plants might be due to independent evolution of those two plants in geographically isolated environment. Those two Korean plants are classified in different genera based on the morphology, one in Vitis genus and the other in Ampelopsis genus, providing another example of discrepancy between morphological and genetic classification.

Screening Resistance to Crown Gall Disease by Pathogen Inoculation in Muscadine and Florida Hybrid Grapes (병원균 접종에 의한 Muscadine 및 플로리다 교잡종 포도의 줄기혹병 저항성 검정)

  • It is necessary to evaluate the resistance to disease among genetic resources for development of disease resistant grapes. This study was conducted to screen the resistance to crown gall in muscadine and Florida hybrid bunch grapes by pathogen inoculation. In order to compare the responses to infection with different pathogen strains, muscadine and Florida hybrid grapes were inoculated with 3 strains of Agrobacterium vitis. Although there were different levels crown gall formation among grape cultivars, there little variation in response to inoculated strains. Among 29 muscadine cultivars tested by inoculation of A. vitis 'C4612', most of them were shown to be susceptible, and 'Gold Isle' and 'Africa Queen' were highly susceptible, and two cultivars, 'Welder' and 'Jumbo' were found to be resistant to crown gall disease. Among Florida hybrid grapes, 'Daytona', 'Stover', and 'Swanee' were susceptible and 'Blanc du Bois' was moderately susceptible to crown gall. Because muscadine grapes have been actively utilized as useful genetic resources for development of new grape varieties by intersub-genus cross, this result from the screening of resistance among muscadine grapes can provide valuable information in breeding programs of grape resistant to crown gall.

Phylogenetic analysis of the genera Azorhizobium, Bradyrhizobium, Mesorhizobium, Rhizobum and Sinorhizobium on the basis of internally transcribed spacer region (ITS 영역의 염기서열을 이용한 근류형성 질소고정균의 계통분류)

  • The phylogenetic relationships for 33 strains belonging to the genera Azorhizobium, Bradyrhizobium, Mesorhizobium, Rhizobium and Sinorhizobium were conducted by the sequence analyses of the ITS regions. The sequence homologies of these strains showed the high variations(28.0 - 94.9%). According to the phylogenetic analysis of ITS regions. 37 ITS clones from 33 strains of 32 species were classified into four groups. Group I included all strains of the genus Sinorhizobium as core members and R. giardinii as a peripheral member. The genus Rhizobium strains were clustered into group II which was very heterogeneous and the tree toplogy of this group were very unstable. Among the members of group II. the taxonomic position of R. radiobacter and R. rubi was not clearly identified on the basis of ITS I regions. R. undicola and R. vitis were remotely related with other Rhizobium strains including R. leguminosarum, R. galegae, R. gallicum, R. mongolense, R. tropici, R. hainanense, R. rhizogense and R. huautlense of group II were supposed to be loosely related to R. leguminosarum. While the stains of the genera Bradyrhizobium constituted group III with Azorhizobium caulindans, the strains of the genus Mesorhizobium formed group IV on the relatively high sequence homology level.

Data Mining and Construction of Database Concerning Effects of Vitis Genus (산머루 관련 정보수집 및 데이터베이스의 구축)

  • The database for the oriental medicine had been existed in documentation in past times and it has been developed to the database type for random accesses in the information society. However, the aspects of the database are not so diversified and the database for the bio herbal material exists in widened type dictionary style. It is a situation that the database which handles the in-depth raw herbal medicines is not sufficient in its quantity and quality. Korean wild grape is a deciduous plant categorized into the Vitaceae and it was found experimentally that it has various medical effects. It is one of the medical materials with higher potentiality of academic study and commercialization recently because it has a bigger possibility to be applied into diverse industrial fields including the medical product for health, food and beauty. We constituted the cooperative system among the Muju cluster business group for Korean mountain wild grapes, Physiology Laboratory in Kyung Hee University Oriental Medicine and Medical Classics Laboratory in Kyung Hee University Oriental Medicine with a view to focusing on such potentiality and a database for Korean wild grapes was made a touchstone for establishing the in-depth database for the single bio medical materials. First of all, the literatures based on the North East Asia in ancient times had been categorized into the classical literature (Korean literature published by government organization, Korean classical literature, Chinese classical literature and classical literature fro Korean and Chinese oriental medicine) and modern literature (Modern literature for oriental medicine, modern literature for domestic and foreign herbal medicine) to cover the eastern and western research records and writings related to Korean wild grapes and the text-mining work has been performed through the cooperation system with the Medical Classics Laboratory in Kyung Hee University Oriental Medicine. First of all, the data for the experiment and theory for Korean wild grape were collected for the Medline database controlled by the Parliament Library of USA to arrange the domestic and foreign theses with topic for Korean wild grapes and the network hyperlink function and down load function were mounted for self-thesis searching function and active view based on the collected data. The thesis searching function provides various auxiliary functions and the searching is available according to the diverse searching/queries such as the name of sub species of Korean wild grape, the logical intersection index for the active ingredients, efficacy and elements. It was constituted for the researchers who design the Korean wild grape study to design of easier experiment. In addition, the data related to the patents for Korean wild grape which were collected from European Patent Office in response to the commercialization possibility and the system available for searching and view was established in the same viewpoint. Perl was used for the query programming and MS-SQL for database establishment and management in the designing of this database. The Flora of Beomseom Island, Jeju-do (제주도 범섬의 식물상)

  • This study was conducted to prepare systematic and efficient measures for nature conservation and management in Beomseom Island, and to clarify characteristics of distribution of plant resources in southernmost islands of Korean peninsula. The numbers of vascular plants were summarized as 164 taxa included 6 families, 13 genera, and 15 species in pteridophyta, 1 family, 1 genus, and 1 species in coniferophyta, and 59 families, 126 genera, 146 species, and 2 varieties in endospermae composed of 49 families, 97 genera, 114 species, and 1 variety in dicotyledonae and 10 families, 29 genera, 32 species, and 1 variety monocotyledonae. Unrecorded taxa of the flora of Beomseom Island were 19 taxa included 4 taxa of pteridophyta, i.e. Hypolepis punctata, Cyrtomium fortunei, Dryopteris pacifica, and Thelypteris granduligera, 6 taxa of monocotyledonae, i.e. Setaria viridis, Zizania latifolia, Scirpus triangulatus, Zantedeschia aethiopica, Spirodela polyrhiza, and Dioscorea nipponica, and 9 taxa of dicotyledonae, i.e. Alnus firma, Boehmeria Platanifolia, Aconogonum ajanense, Phytolacca americana, Melandryum oldhamianum var. roseum, Vitis flexuosa, Clerodendron trichotomum, Elaeagnus glabra, and Siegesbeckia glabrescens. The two rare species, Osmanthus insularis that distributed only Jeju-do and Geomoondo Island and Orostachys iwarenge that distributed only southern parts of Jeju-do in Korea were investigated. The numbers of naturalized plants were 20 families, 33 genera, 34 species, and 1 variety.

Production of Gastrodia elata Tuber using Armillaria spp. (Armillaria 속균을 이용한 천마의 생산)

  • The genus Armillaria is important because they produce Gastrodia tubers. Seventy two isolates of Armillaria were obtained from fruit bodies grown on decayed wood in Korea. Twenty four isolates from Pinus koraiensis were identified as A. ostoyae. Two isolates from G. elata growing in the field were identified as A. mellea. Seven isolates from Acer ginnala and Quercus spp. were identified as A. tabescens. Thirty nine isolates were identified as A. gallica. Armillaria gallica was isolated from Quercus spp., Ainus japonica, Vitis amurensis and Prunus sargentii. Armillaria spp. isolates were divided into four groups based on the cultural characteristics. Group II (A. gallica KNU-A110) was better than the other groups for mycelial growth and rhizomorph formation. Isolate KNU-A110 proved to be good for production of G. elata tubers. This fungus forms mycelial fan in the plant tissue and rhizomorphs in contact with G. elata tubers. Gastrodia spp. was found in thirteen sites in Kangweon province in Korea. The plants were divided into three different kinds based on stem color. Plants with stems of brownish orange and greyish yellow were identified as G. elata, and those with greyish green colored stems were identified as G. gracilis. Gastrodia was collected mainly from humus soils rich in leaf debris, and slopes facing south from mid-May to mid-July. Once the new tubers are formed from the ancestry tuber, the ancestry tuber begins to decay. The offspring tuber, apparently gaining nutrients through rhizomorphs, begins to grow in length and slowly to enlarge. It takes three years for the offspring tuber to become ancestry tuber.

Soil Salinity and Salt Spray Drift Tolerance of Native Trees on the Coastal Windbreaks in the South-Sea, Korea (한국 남해안방풍림 자생수종의 내염성 및 내조성 수종 선발)

  • This study was carried out to investigate the soil salinity and salt spray drift of the indigenous windbreak trees, and its main purpose was to provide basic data for the selection of salt-tolerant trees in the saline coastal region in the South Sea of Korea. The soil salinity($EC_{1:5}$)was $0.18dSm^{-1}$, which was an average degree of the whole areas of investigation whose salinity degree ranged from $0.05dSm^{-1}$ to $0.58dSm^{-1}$. The level of soil salinity gradually decreased as it moved farther inland, except the belt I. The level of decreasing soil salinity was found to be in the following order: belt II, belt III, belt I, belt IV. The degree of soil salinity was $EC_{1:5}$ $0.22dSm_{1:5}$, $0.22dSm_{1:5}$ $0.19dSm^{-1}$ and $0.13dSm^{-1}$ respectively. The total 110 taxa, which consisted of 45 families, 74 genus, 101 species, and 9 varieties, were found to be tolerant to both soil salinity and salt spray drift. The trees that grow in the highest degree of salinity($EC_{1:5}$ $0.50dSm^{-1}$)were Parthenocissus tricuspidata(Siebold & Zucc.), Planch and Lonicera japonica Thunb. The next group of trees that grow in the high degree of salinity ranging from $EC_{1:5}$ 0.41 to $0.50dSm^{-1}$ was Cudrania tricuspidata(Carr.) Bureau ex Lavall$\acute{e}$e, Rubus parvifolius L., Zanthoxylum schinifolium(Siebold & Zucc.), Hedera rhombea(Miq.) Bean., Robinia pseudoacacia L., Quercus serrata Thunb., Callicarpa dichotoma(Lour.) K. Koch, and so on. The woody species which grew in the entire belts were Pueraria lobata(Willd.) Ohwi and Vitis flexuosa Thunb., and Vitex rotundifolia L. f. which was known to be highly tolerant to salt spray drift was found only in belt I. The woody species with high important value(IV) were Zelkova serrata(Thunb.) Makino., Celtis sinensis Pers., Koelreuteria paniculata Laxmann, Mallotusjaponicus(Thunb.) Muell. Arg., Trachelospermum asiaticum(Siebold & Zucc.) NAKAI, and Pueraria lobata(Willd.) Ohwi. These species were classified as native windbreak trees that are comparatively more tolerant to salt spray drift than other kinds.
