The Effect of Spent Medium Recycle on Cell Proliferation, Metabolism and Baculovirus Production by the Lepidopteran Se301 Cell Line Infected at Very Low MOI

  • The aim of this paper was to study the effect of spent medium recycle on Spodoptera exigua Se301 cell line proliferation, metabolism, and baculovirus production when grown in batch suspension cultures in Ex-Cell 420 serum-free medium. The results showed that the recycle of 20% of spent medium from a culture in mid-exponential growth phase improved growth relative to a control culture grown in fresh medium. Although both glucose and glutamine were still present at the end of the growth phase, glutamate was always completely exhausted. The pattern of the specific glucose and lactate consumption and production rates, as well as the specific glutamine and glutamate consumption rates, suggests a metabolic shift at spent medium recycle values of over 60%, with a decrease in the efficiency of glucose utilization and an increase in glutamate consumption to fuel energy metabolism. Baculovirus infection provoked a change in the metabolic pattern of Se301 cells, although a beneficial effect of spent medium recycle was also observed. Both growth rate and maximum viable cell density decreased relative to uninfected cultures. The efficiency of glucose utilization was dramatically reduced in those cultures containing the lowest percentages of spent medium, whereas glutamine and glutamate consumption was modulated, thereby suggesting that infected cells were devoted to virus replication, retaining their ability to incorporate the nutrients required to support viral replication. Recycle of 20% of spent medium increased baculovirus production by around 90%, thus showing the link between cell growth and baculovirus production.

  • Purpose : Hepatitis B virus DNA transfected cell line(HepG2.2.15) was cultured to evaluate the effect of herbs on the expression of HBeAg and the replication of HBV. HepG2.2.15 produces HBV particles as well as viral proteins into cell culture media. Methods : Extracts of herbs were adminitered to the cells on the proper concentration. Culture media was collected 48 hours after the herbal administration and HBeAg level in the media was examined by ELISA method. To confirm that the anti-viral effect was not due to direct cytotocixity of the extracts, normal cell proliferation was shown by cell counting. And as of the interference in protein synthesis of HepG2.2.15 by herb-extracts, we used the result of study that we performed before by ${\alpha}FP$ assay using EIA method. Results& Conclusion : Herb medicines like 地楡(Sanguisorbae Radix) and 覆盆子(Rubi Frusctus) showed significant inhibitory effect on HBeAg expression at p<0.01 and 五味子(Acanthopanacis Cortex) at p<0.05. Whereas, though some herbs such as ?草根(Rubiae Radix), 山査(Crataegii Fructus), 白芍藥(Paeoniae Radix Alba), and 大黃(Rhei Radix et Rhizoma) showed the tendecy to suppress HBeAg. most of them were not significant statistically. From the above, we could conclude that those herb medicines can be applied to patients effectively and further studies on effective fraction of some herbs are thought to be needed.

Studies on Cellulases of Penicillium chrysogenum (Penicillium chrysogenum에서 추출한 cellulase에 관한 연구)

  • Cellulases were isolated from both healthyl(PC) and virus0infected penicillum chrysogenum(PCV) in the wheat bean culture, and some properties of the enzymes were studied. 1) At $37^{\circ}C$, pH 6, 4 and 6day's culture the maximal enzyme yield was obtained in both PC and PCV. 2) The optimum temperature for the PC cellulase was at $50^{\circ}C$, and that for the PCV enzyme was the same. 3) The optimum pH for the PC enzyme was at 5.0, whereas the PCV enzyme was at 6.0, indicating that they are isozymes. 4) When Na-CMC was used as a substrate, PC enzyme was twice as high as the activity of PCV enzyme.

  • In 2020, due to COVID-19, all universities in Korea were conducting non-face-to-face classes. The purpose of this study is to study what factors affect academic achievement under such non-face-to-face instruction, especially for engineering students where practical training is important. Validity of the statistical hypothesis defined in this study by applying a structural equation model using questionnaires about academic achievement for engineering students at University D for this study. In addition, I would like to suggest what factors should be considered in non-face-to-face classes, especially in engineering colleges. As a result of the study, it was found that students' Q&A, feedback and e-learning system had a direct influence on academic achievement. In addition, it was confirmed that they had an indirect influence on academic achievement through the parameters of theory class and practical class.

  • To develop the fruits of cucumber (Cucumis sativus L.) producing high yields of superoxide dismutase (SOD), the MnSOD cDNA from pea (Pisum sativum) under the control of the cauliflower mosaic virus 35S promoter was introduced into cucumber using Agrobacterium tumefaciens (strain LBA 4404)-mediated transformation. The kanamycin-resistant shoots were selected on the selection medium containing MS basal salt, 1.0 mg/L zeatin, 0.1 mg/L IAA, 300 mg/L claforan, and 100 mg/L kanamycin. After 6 weeks of culture on the selection medium, the shoots were transferred to MS medium containing 0.2 mg/L NAA to induce roots. PCR analysis using the primers for neomycin phosphotransferase (NPTII) gene revealed that three plantlets were transformed. The fruits of one transgenic plant had approximately 3.2-fold higher SOD activity than those of non-transgenic plants. MnSOD isoenzyme band was strongly detected on native gel in fruits of transgenic plants.

The safety of live VHSV immersion vaccine at a temperature-controlled culture condition in juvenile olive flounder, Paralichthys olivaceus

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral diseases affecting farmed olive flounder (Paralichthys olivaceus) in Asian countries. VHS, caused by viral hemorrhagic septicemia virus (VHSV), occurs in over 80 different cultured and wild fish species worldwide. Our previous study demonstrated that VHSV infection can be restricted by adjusting the water temperature to over 17℃ from the host optima. We confirmed that the effective VHSV immersion vaccine treatment was a tissue culture infection dose (TCID) of 105.5 TCID50/mL at 17℃. However, the safety of live VHSV immersion vaccines remains unclear. The objectives of this study were to 1) demonstrate the safety of the live VHSV immersion vaccine under co-habitant conditions and 2) estimate the pathogenicity of VHSV in live VHSV-vaccinated flounder at 10℃. No mortality was observed in olive flounder treated with the live VHSV immersion vaccine, and the vaccinated flounder challenged with VHSV did not transfer VHSV to naïve fish at 10℃ through cohabitation. VHSV titration was below the detection limit (< 1.3 log TCID50/mL) in live VHSV immersion vaccine-treated flounder challenged with VHSV at 10℃. This study demonstrated that flounder treated with the live VHSV immersion vaccine were resistant to VHSV infection, and the live vaccine was also safe for naïve fish even at a water temperature known to be VHS infectious.

The Relationship between Individuals' Collectivistic Orientation and Social Distancing during the COVID-19 Crisis in Korea: The Mediating Role of Subjective Norm (코로나19 대유행 시기에 집단주의 성향과 사회적 거리두기 행동 간의 관계: 사회적 거리두기에 관한 주관적 규범의 매개효과)

  • The present study examined the mediating role of the subjective norm about social distancing in the relationship between individuals' collectivistic orientation and their social distancing behavior during the early COVID-19 crisis in Korea. A total of 445 South Korean adults residing in Korea participated in a survey during which level 2 to 2.5 prevention policies were being enforced. Results indicated, as expected, that collectivism positively predicted the perceptions of subjective norm that supports social distancing, which in turn predicted positively individuals' participation in social distancing as prescribed by the central government. This result emerged regardless of the individuals' perception of personal vulnerability to COVID-19. We discuss implications of these findings and directions for future research regarding the link between individualism-collectivism and the behavioral immune system, along with the role of social distancing in effectively curbing the spread of corona virus during the pandemic.

Evaluation of Changes in Serum Thyroid Hormone Levels in Patients with Hepatitis B Infection

  • Purpose: We aimed to determine the differences in the levels of serum thyroid hormone (free T4 [FT4]) and thyroid stimulating hormone [TSH]) as biomarkers for hepatitis B virus (HBV) infection status, with respect to age and sex. Methods: We retrospectively analyzed serum samples from 200 patients who underwent HBV testing from August 2022 to September 2022. Serum samples were collected from patients suspected of having HBV infection who visited this hospital. Thyroid hormone levels were measured, and patients were grouped according to age and sex. Results: Differences in TSH and FT4 levels in the serum of patients in the HBV-positive and -negative groups were not significant. Among the HBV-positive patients in the younger age group (<60 years), TSH and FT4 levels were 1.78 ± 0.09 µIU/mL (normal: 0.4-5.0 µIU/mL) and 1.24 ± 0.02 ng/mL (normal: 0.8-1.9 ng/mL), respectively, whereas among the HBV-positive patients in the older age group (≥60 years), TSH and FT4 levels were 2.22 ± 0.17 µIU/mL and 1.24 ± 0.07 ng/mL, respectively. Conclusions: The presence of HBV did not markedly affect serum thyroid hormone levels. Our findings shed light on the conflicting evidence on the association between thyroid hormone levels and HBV infection. Effect of Medium Composition on in Vitro Shoot Regeneration from Leaves of Cassava (Manihot esculenta Crantz) Through Somatic Embryogenesis and Callus Induction (카사바 잎 절편 유래 체세포배 배양시 배지조성이 기내 식물체 재분화에 미치는 영향)

  • The Cassava(Manihot esculenta Crantz) is a tropical root crop, originally from Amazonia, that provides the staple food of an estimated 800 million people worldwide. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. So we tried to optimize protocols for mass production of somatic embryo amenable to large-scale vegetative propagation of Cassava. After in vitro eight-week culture of leaves of Cassava, the medium which contained the 2,4-D, BAP and IBA showed the highest callus induction rate, embryogenesis callus formation rate and somatic embryo formation in Cassava culture. In the medium with GA3 and myo-inositol, shoots were most vigorously regenerated from somatic embryos of Cassava. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.
